In this paper, the hot water extraction conditions of mycelium polysaccharide ofTricholoma mongoliumwere investigated. By single-factor experiments, ethanol amount was determined at three times and the other factors (liquid–solid ratio, extraction temperature and time) were chosen to further optimize extraction conditions using response surface methodology (RSM). The Center Composite experimental results showed the optimum extraction conditions as follows: a temperature of 85.38°C, a liquid–solid ratio of 31.78 g/ mL and a extraction time of 2.18 h. Under these conditions, two extractions sufficiently reached the maximal the rate of mycelium polysaccharide 7.23%.

In this study, a simultaneous water extraction process for baicalin, wogonoside, and chlorogenic acid has been optimized. The effect of extraction temperature, extraction time, and liquid–solid ratio was scrutinized by single factor experiments and further analyzed by Box–Behnken design (BBD) approach using response surface methodology (RSM). The extraction yield of investigated compounds was determined by high performance liquid chromatography (HPLC). Single-factor experiments and response surface analysis results revealed that the optimized conditions are: Liquid to solid ratio 25:1 (mL/g), extraction temperature 93 °C, extraction time 2.4 h, and the extraction cycle two. Importantly, it has been noted that under the above conditions, concentrations of baicalin, wogonoside, and chlorogenic were 0.078, 0.031, and 0.013 mg/mL, respectively, and the overall desirability (OD) value was 0.76 which was higher than the non-optimized conditions and the deviation from the predicted OD value was only 2.44%. Conclusively, it has been suggested that the model was stable and feasible, and fit for extraction of baicalin, wogonoside, and chlorogenic acid from Scutellariae Radix and Lonicerae (L.) japonicae Flos.

The experiment of extraction of polysaccharide from foxtail millet was investigated. Response Surface Methodology (RSM),based on a threelevel, three variablesBox-Behnken design (BBD), was employed to obtain the best possible combination of liquid-solid ratio(X1: 15.0-25.0 mL?g-1),extraction time (X2: 1.5-2.5h), and extraction temperature (X3: 65.0-75.0?C)for maximum polysaccharide yields. The experimental data obtained were fitted to a second-order polynomial equation using multiple regression analysis and also analyzed by appropriate statistical methods (ANOVA). The optimum extraction conditions were as follows: liquid-solid ratio 20.7mL?g-1, extraction time 2.0h, extraction temperature 72.3?C. Under these conditions, the experimental yield was 8.08mg?g-1, which is well in close agreement with8.02mg?g-1predicted value by the model.

The research in this paper optimized the extraction technique of lentinan with ultrasonic assistant method on the basis of hot water extraction technique, and investigated the promoting function of ultrasound to polysaccharides extraction. Extraction condition was selected by means of orthogonal experimental design, four factors and three levels L9(34), after key elements were respectively chosen through single factor experiments. Comparison between optimal extraction parameters of two method, hot water extraction technique and ultrasonic assistant extraction technique, showed decreased extraction temperature and significantly shortened extraction time, which existed in the second means, improved the extraction efficiency. Lentinus edodes polysaccharide extracted with ultrasonic assistant technique, the extraction rate and polysaccharide content percentage increased 6.22% and 8.66% respectively, comparative to which extracted with hot water extraction technique.

Saffron, which has many kinds of biological activities, has been widely used in medicine, cosmetics, food, and other fields of health promotion industries. Crocins are the main component of saffron (Crocus sativus L.). At present, most of the extraction methods for crocins require long time or special instruments to complete the process and some of them are not suitable for industrial production at present. In this article, homogenate extraction technology which is a convenient and efficient method was developed for crocins extraction from saffron. Firstly, the influences of extraction voltage, extraction time, ethanol concentration, and temperature on crocins yield were studied by single factor experiments; and then response surface methodology (RSM) was used to optimize levels of four variables based on the result of single factor experiments. Results showed that the optimum extraction process conditions for crocins were as follows: extraction voltage, 110 V; ethanol concentration, 70%; extraction temperature, 57°C; and extraction time, 40 s. Based on these conditions, the extraction yield of crocins can reach 22.76% which is higher than ultrasonic extraction method. Therefore, homogenate extraction is an effective way to extract crocins from saffron with higher extraction yield and shorter extraction time.

An efficient ultrasonic-assisted extraction of inulin from Jerusalem artichoke was investigated by Response Surface Methodology (RSM). The results showed that the yield of ultrasonic-assisted extraction was 62.07 ± 0.39% over the microwave (40.85 ± 0.28%) and hot water extraction (27.42 ± 0.42%). The Box-Behnken Design (BBD) was used to optimize the effects of three parameters (temperature-X1, ultrasonic power-X2 and time-X3) on inulin yield. Analysis of variance showed that the contributions of X1, X3, X12, X13, X22 were significant. The optimal yield of inulin was 82.93 ± 1.03% at 82 °, 120 W and 18 min.

Response surface methodology was applied to optimize the extraction of phenolic compounds from fresh Pouzolzia zeylanica plant using hot water as a solvent. A central composite design (CCD) in form (23+star) was used to investigate the effects of two independent variables, namely, extraction temperature (70 to 90oC) and extraction time (20 to 40 min). The dependent variables were the content of anthocyanin, flavonoid, polyphenol, tannin and total soluble solids of extracted solution. A second-order polynomial model was used for predicting the response. The results showed that the optimal extraction process was obtained at 84.4oC for 31.7 min. The experimental values agreed with predicted within a 95% confidence interval. Consequently, the contents of anthocyanin, flavonoid, polyphenol and tannin were 38.66 mgCE/100 g, 3.01 mgQE/g, 5.17 mgGAE/g, 4.07 mgTAE/g fresh weight, and total soluble solids content was 0.73%, respectively.

Ultrasonic-assisted extraction (UAE) was optimized using response surface methodology (RSM) to maintain the cyto-protective activity of M.toringoides against oxidative stress. The optimal conditions for UAE were a 58 mL/g liquid-solid ratio, a 38 °C extraction temperature, an 85% solvent concentration, and a 19-min extraction time, which resulted in a protection rate of 54.57% against hydrogen peroxide-induced oxidative stress in human umbilical vein endothelial cells (HUVECs). These results were comparable to the predicted value of 53.75%. The extracts showed excellent antioxidant activity, and phlorizin was detected in the dried leaves of Malus.toringoides. The highest yield of phlorizin (101.239 mg/g) was also obtained using these conditions. Taken together, these results showed that the method successfully integrated RSM and partial least squares regression methods to optimize M.toringoides extraction to yield the highest cyto-protective activity and effectively increase the yield of phlorizin from M.toringoides.

Moringa oleifera is considered one of the most useful plants in the world because it's rich in bioactive substances, which employing on modern medical treatment, also can be used for many fields: pharmaceutical, food and cosmetics purposes. In this study, the response surface methodology (RSM) based on a Box–Behnken design (BBD) was employed to optimize the extraction time (X1: 20–60 min), extraction temperature (X2: 15–45 °C) and solvent-solid ratio (X3: 5–7 ml/g), to obtain a high crude of flavonoids yield from Moringa oleifera Leaves by ultrasonic-assisted extraction technique (UAE). The optimum conditions were an extraction time 23 (min), extraction temperature 44 (°C) and solvent-solid ratio 5.05 (ml/g). Under these conditions, the experimental yield was 72.65 (mg ER/g), well matched with the predicted yield 74.34 (mg ER/g) with the coefficients of determination (R2= 0.9861), thus indicating the suitability of response surface methodology in optimizing the ultrasound-assisted extraction of flavonoids from Moringa oleifera Leaves.

An efficient procedure for ultrasound-assisted enzymatic extraction of crude polysaccharides fromTrichosanthes Fructus(crude TFP) using response surface methodology (RSM) was developed. The Box–Behnken design was applied to optimize the effects of pH (X1), enzyme amount (X2), extraction temperature (X3), and liquid-to-solid ratio (X4) on the extraction. The statistical analysis indicated that the independent variables (X4,X2, andX3), the quadratic coefficients (X12,X22,X32, andX42), and the interaction coefficient (X1X3) had significant impact on the yield of crude TFP. The optimal conditions were determined as follows: pH 4.5, enzyme amount 5000 u/g, extraction temperature 45°C, and liquid-to-solid ratio 30 ml/g. The experimental yield of crude TFP was 6.58%, which was very close to the predicted yield of 6.71%. TFPI was then purified and characterized with Sephadex G-100 column, UV-Vis, GPC, and FT-IR. The average molecular weight of TFPI was calculated to be 1.49 × 105 Da. TFPI exhibited strong reducing power and possessed not only remarkable scavenging activities against ABTS•+and DPPH radicals, but also high antitumor activities in C4-2, DU145, and PC3 cells. The results suggest thatTrichosanthes Fructusand TFPI could be a novel potent natural medicine with antioxidant and antitumor activities.
