Diabetes mellitus has been reported to affect functions of the hippocampus. We hypothesized thatCentella asiatica, a herb traditionally being used to improve memory, prevents diabetes-related hippocampal dysfunction. Therefore, the aim of this study was to investigate the protective role ofC. asiaticaon the hippocampus in diabetes.Methods. Streptozotocin- (STZ-) induced adult male diabetic rats received 100 and 200 mg/kg/day body weight (b.w)C. asiaticaleaf aqueous extract for four consecutive weeks. Following sacrifice, hippocampus was removed and hippocampal tissue homogenates were analyzed for Na+/K+-, Ca2+- and Mg2+-ATPases activity levels. Levels of the markers of inflammation (tumor necrosis factor, TNF-α; interleukin, IL-6; and interleukin, IL-1β) and oxidative stress (lipid peroxidation product: LPO, superoxide dismutase: SOD, catalase: CAT, and glutathione peroxidase: GPx) were determined. The hippocampal sections were visualized for histopathological changes.Results. Administration ofC. asiaticaleaf aqueous extract to diabetic rats maintained near normal ATPases activity levels and prevents the increase in the levels of inflammatory and oxidative stress markers in the hippocampus. Lesser signs of histopathological changes were observed in the hippocampus ofC. asiaticaleaf aqueous extract treated diabetic rats.Conclusions.C. asiaticaleaf protects the hippocampus against diabetes-induced dysfunction which could help to preserve memory in this condition.

Protective effects of Fenugreek seeds (Trigonella foenum graecum L), added in the diet, upon oxidative stress and dysfunctions in kidney, thyroid and liver of alloxan-diabetic rats were investigated.In our study, the alloxan-induced diabetes triggered 1) increased levels of glucose, total cholesterol and triglycerides in blood, 2) increased activities of alkaline phosphatase and transaminases in blood, 3) increased levels of creatinine, urea and protein in blood, 4) a decreased level of TSH and an increased level of free thyroxin in plasma.In addition, an oxidative stress, evidenced by an increase of lipids peroxidation level and superoxide dismutase activity associated with a decrease of glutathione peroxidase and catalase activities in hepatic and renal tissues, was observed.When Fenugreek seeds powder (100g/kg) was added in the food for 30 days, all this parameters were significantly shifted to more normal values.In conclusion, fenugreek seeds powder displays beneficial effects upon hepatotoxicity, nephropathy, thyroid dysfunction and oxidative stress in alloxan-diabetic rats. This property could be attributed to the presence of antioxidant components, such as complex polysaccharides and phenolic acids, as confirmed by analyses.

The present study investigated the effects of oral administration of annatto carotenoids (bixin (BIX) and norbixin (NBIX)) on glucose levels, lipid profiles, and oxidative stress parameters in streptozotocin (STZ)-induced diabetic rats. Animals were treated for 30 days in the following groups: nondiabetic control, diabetic vehicle, diabetic 10 mg/kg BIX, diabetic 100 mg/kg BIX, diabetic 10 mg/kg NBIX, diabetic 100 mg/kg NBIX, diabetic metformin, and diabetic insulin. Blood glucose, LDL cholesterol, and triglyceride levels were reduced in the diabetic rats treated with BIX. BIX treatment prevented protein oxidation and nitric oxide production and restored superoxide dismutase activity. NBIX treatment did not change most parameters assessed, and at the highest dose, it increased LDL cholesterol and triglycerides levels and showed prooxidant action (increased protein oxidation and nitric oxide levels). These findings suggested that BIX could have an antihyperglycemic effect, improve lipid profiles, and protect against damage induced by oxidative stress in the diabetic state. Because NBIX is a water-soluble analog of BIX, we propose that lipophilicity is crucial for the protective effect of annatto carotenoids against streptozotocin-induced diabetes.

Background: Hyperlipidemia and inflammation are critical components in the pathophysiology of endothelial disorder, which can lead to vascular complications. Our study aimed to evaluate the effects of immunomodulatory therapy (methotrexate and infliximab) in a diet-induced hyperlipidemia rat model. Methods: Sprague-Dawley (wild type (WT), male, n = 32) rats were divided into four groups: one group fed with standard diet (SD), one group fed with high lipid diet (HLD), and two groups that received HLD and drug treatment (methotrexate (Mtx) or infliximab (Ifx)). In order to evaluate if modifications to the endothelial cells may influence the risk of vascular complications following hyperlipidemia or treatment reactivity, each group was doubled by a rats group that overexpressed beta-3 receptors on the endothelial cells (transgenic (TG-beta 3), male, n = 32). Serum lipid profile, liver enzymes, oxidative stress, and inflammation markers were determined. Histopathologic analysis of the liver and aorta was performed. Results: After 9 weeks of HLD, rats exhibited significant pathologic serum lipid profiles, elevated oxidative stress, and pro-inflammatory markers. Additionally, the aortic histopathological analysis revealed aorta media-intima thickening (p < 0.05) in the transgenic group. Methotrexate and infliximab significantly decreased inflammation and oxidative stress parameters, but presented opposing effects on lipid profiles (methotrexate decreased, whereas infliximab increased the atherosclerosis index). Drug treatment decreased the aorta media-intima thickness (p < 0.05) only in transgenic rats. Conclusions: HLD was associated with hyperlipidemia, inflammation and oxidative stress. The overexpression of beta-3 receptors on endothelial cells increased aortic thickening in response to the HLD. Methotrexate and infliximab reduced oxidative stress and inflammation in all groups, but led to favorable histopathologic vascular results only in the transgenic groups.

This study investigated the effects of methanol extract of Cussonia arborea on serum lipid and oxidative stress biomarkers of alloxan-induced diabetic rats. A total of 72 male albino rats assigned into 6 groups of 12 rats per group were used. Groups 1-5 were made diabetic while group 6 were normal. Groups 1-4 were treated with 62.5, 125, 250 mg/kg of the extract and 2 mg/kg glibenclamide respectively while groups 5 and 6 received 10 ml/kg distilled water each. Total cholesterol, triglyceride, high density lipoprotein (HDL), malondiadehyde, catalase, and superoxide dismutase (SOD) were assayed on days 28, 56 and 84 post treatment. The results indicated that the extract significantly (p<0.05) reduced the levels of total cholesterol, triglyceride, malondialdehyde but significantly (p<0.05) increased the activities of SOD, catalase and the levels of HDL when compared to negative control. It was therefore concluded that methanol extract of C. arborea mitigated dyslipidaemia and oxidative stress.
