The initial aggregation of trypsin-dissociated cells from the skeletal muscle tissue of 9-day-old chick embryos in the presence of cytochalasins A and B was studied in order to discover the effects of these agents on contact and adhesion. Cytochalasin B (3 µg/ml) had a negligible effect on the rate of aggregation of cells over an 8-h period, but cytochalasin A at concentrations between 3 and 20 µg/ml markedly inhibited aggregation. Both agents altered the shape and size of aggregates and caused cells at their periphery to appear more spherical. The oxygen uptake of the treated cells was not noticeably different from that of the controls, despite the severe inhibition of isotopic carbon dioxide evolution. The effect of cytochalasin B on cell aggregation was reversible and although the cytochalasin A effect could not be abolished on return to medium free of A, the unaltered oxygen consumption was taken as an indication that permanent cellular injury did not occur. The effect of the cytochalasins on aggregate structure was interpreted on the basis of arrested cellular motility, but the singular inhibition by cytochalasin A of the rate of aggregation must await final confirmation of its site of action.

Background: To differentiate ovarian mass as benign or malignant could change clinical approach. Finding a screening and diagnostic method for ovarian cancer is challenging due to high mortality and insidious symptoms. Risk malignancy index (RMI) has the advantage of rapid and exact triage of patients with ovarian mass.Methods: Prospective study carried for 2 years at NRI Medical College and General Hospital, Chinakakani, Mangalagiri, Andhra Pradesh, India. 79 patients with ovarian mass were investigated and risk malignancy index (RMI-3 and RMI-4) calculated. Final confirmation was done based on histopathological report. Sensitivity, specificity, positive predictive value and negative predictive value were calculated for RMI 3 and RMI 4 taking histopathology as control and comparison was done.Results: (n=79); 50 (63.29%) cases were benign and 29 (36.70%) were malignant based on histopathology. RMI 4 is more sensitive (68.96%) than RMI 3 (62.06%), but RMI 3 is more specific (94%) than RMI 4 (92%).The positive predictive value of RMI-3 and RMI-4 were 85.71%  and 83.33% respectively. The negative predictive value for RMI-4 and RMI-3 were 83.63% and 81.03% respectively.Conclusions: With increasing age, chance of malignancy increases. RMI 4 was more sensitive than RMI-3, however less specific than RMI 3 in differentiating benign and malignant tumors. The positive predictive value is slightly more for RMI 3, than RMI 4. Negative predictive value is slightly more for RMI 4, than RMI 3.

The reports of nontuberculous mycobacteria (NTM) associated with extrapulmonary diseases are increasing in tertiary care hospitals. Despite a significant increase in knowledge about NTM infections, they still represent a diagnostic and therapeutic challenge. The aim of this study is to know the prevalence of NTN among extrapulmonary tuberculosis cases in tertiary care centers in Northern India. A total of 227 culture positive isolates from 756 cases were tested for niacin production and catalase assay. BIO-LINE SD Ag MPT64 TB test and final identification and differentiation between MTBC and different species of NTM were further confirmed by GenoType Mycobacterium CM/AS assay. 71 cases (9.3%) were positive for AFB by ZN staining and 227 cases (30.1%) were positive for mycobacteria by culture. Niacin production and catalase activity were negative in 62/227 (27.4%) strains and after using a panel of different biochemicals and final confirmation by GenoType Mycobacterium CM assay. Out of 227 cultures tested, 165 (72.6%) strains were confirmed asM. tuberculosiscomplex, and 62 (27.4%) were confirmed as NTM. The most common NTM species identified wereM. fortuitum17 (27.5%) andM. intracellulare13 (20.9%). The rapid identification of NTM species may help in targeted therapy and management of the diseases.

In vivo fiber photometry is a powerful technique to analyze the dynamics of population neurons during functional study of neuroscience. Here, we introduced a detailed protocol for fiber photometry-based calcium recording in freely moving mice, covering from virus injection, fiber stub insertion, optogenetical stimulation to data procurement and analysis. Furthermore, we applied this protocol to explore neuronal activity of mice lateral-posterior (LP) thalamic nucleus in response to optogenetical stimulation of primary visual cortex (V1) neurons, and explore axon clusters activity of optogenetically evoked V1 neurons. Final confirmation of virus-based protein expression in V1 and precise fiber insertion indicated that the surgery procedure of this protocol is reliable for functional calcium recording. The scripts for data analysis and some tips in our protocol are provided in details. Together, this protocol is simple, low-cost, and effective for neuronal activity detection by fiber photometry, which will help neuroscience researchers to carry out functional and behavioral study in vivo.

Background: Hepatitis C virus is one of the significant causes of morbidity and mortality in the World. Surprisingly, despite national screening campaigns, new cases of HCV are still on the rise. Methods and Results: A total of 5914 healthy blood donors was included in this study after informed consent. Anti-HCV ELISA was used to check presence of antigen in participant’s plasma.using Monlisa HCV Antigen-Antibody Ultra kit. Final confirmation was done by using real time PCR considered as a gold standard. Results: 0.5% of anti-HCV ELISA negative samples showed presence of antigen in plasma, when checked through core Ag detection method. Conclusion: Our result suggested that HCV core antigen detection and/or combo testing are far safer screening methods for the detection of HCV and the use of these methods can avoid/reduce further spread of this deadly disease.

It is well known that the value of full feeding of farm animals is an important condition for the production of livestock products. Because of the researches of many scientists was found that the value of feed is determined by the content of proteins, fats, carbohydrates, as well as vitamins, minerals and so on. However, the true value of the feed is determined by the part that is digestible and can be used by the body in the metabolism process. In the event of a violation of feeding level associated with the lack of nutrients or minerals of animals or their deficiency, the development of osteodystrophic processes is observed, which directly contribute to changes in the tendon-ligamentous apparatus of the finger of animals, there is an occurrence of prolonged osteotendinitis of the finger flexors, which causes to fingers extension, partially or completely removed from the hoof wall support, and overload of crumbs. Tightening of the crumb support is accompanied by the appearance of the skin of the hoof inflammatory process, excessive horn growth and deformation of the horn capsule. The final confirmation of osteodystrophy is the results of a multivariate mass volumetric analysis of the skeleton state and determination of the magnitude of the angles of the joints of the fingers. On the side of the hoof horn, there is an increase in the moisture content and SH-groups and a decrease in g-keratosis, which affects the biophysical properties of the hoof horn, which are shown by a decrease in density and hardness. Such changes indicate a deterioration in the quality of the hoof horn, a confirmation of which is a decrease in the intensity of erasing of the sole horn, the development of deformation and the occurrence of aseptic inflammation of the base of the skin of the sole horn.

In theory there are only 2 types of business typologies, namely craftsman entrepreneur and opportunistic entrepreneur. Both have unique and different characteristics in carrying out the pace of their business, including the technical and strategic steps to determine a sustainable business. In this case the pattern of leadership and knowledge of business and business orientation certainly plays its role which in turn will provide final confirmation of the sustainability of a business activity. The purpose of this study is to analyze the extent of leadership roles, knowledge of management and business orientation towards business performance. This study took a sample of business people with SME status in Makassar City. Research approach with quantitative analysis with linear regression analysis method. The results of this study stated that leadership strategy, knowledge management and business orientation had no significant effect on the performance of SMEs.

Chromocult coliform agar (CCA) was compared with Petrifilm Escherichia coli count plate (PEC) for identifying coliforms and E. coli in a variety of meat products. Products examined included 45 raw beef samples, 12 sausage emulsion samples, 11 samples of meat-based ready-to-eat appetizers, and 8 pork trimming samples. Coliforms from CCA and PEC were confirmed by gassing in brilliant green lactose broth plus a positive reaction on purple broth agar plus lactose after incubation at 35°C for 48 h. Lauryl sulfate tryptose plus methylumbelliferyl-β-glucuronide and tryptophan broth were used to confirm E. coli from CCA and PEC with 48-h incubations at 35 and 42.5°C, respectively. API 20E test strips were inoculated for final confirmation. The overall respective confirmation percentages (CFU/g) for the PEC and the CCA methods were 93.1 and 93.7% for coliforms and 99.8 and 98.1% for E. coli, although the CCA method yielded significantly (P < 0.001) higher mean CFU/g values for both coliforms and E. coli. Regression analyses of these data indicated a strong positive linear relationship existed between the two methods over a wide CFU/g range for both coliforms and E. coli. The respective correlation coefficients obtained for coliforms and E. coli of 0.89 and 0.86 indicate that the CCA method provides a reliable optional method for these determinations in meat products.

Abstract LacTekTM tests are competitive enzyme-linked immunosorbent assays intended for rapid detection of antimicrobial residues in bovine milk. In this study, the LacTek test protocol was modified for use with extracts of bovine tissue to detect β-lac-tam, tetracycline, and sulfamethazine residues. Testperformance characteristics—precision, accuracy, ruggedness, practicability, and analytical specificity and sensitivity—were investigated. Results suggest that LacTek tests can be easily adapted to detect antimicrobial residues in extracts of lean ground beef. However, positive samples may not contain residues at violative concentrations (i.e., Canadian maximum residue limits), and therefore, additional analysis would be required for final confirmation and quantitation (e.g., chromatography).
