<p><strong>Abstract.</strong> Despite the consistent development of approaches aimed at the virtual reconstruction of whole houses or archaeological monuments, the variety of technologies involved in virtual reconstruction procedures and the complexity of a rigorous process to provide validation models, seems to limit a univocal and shared standards adoption. For example, compared to the large number of contributions on the virtual reconstruction of whole architectures or cultural heritage sites, only a few works have proposed a rigorous workflow specific to mosaics and ancient floors and to their virtual reconstruction. The goal of this work is to present the first results on the virtual reconstruction of the "Temple of Isis" in Marsala (Italy); in particular, the work is aimed at the virtual reconstruction of the mosaic and the <i>opus spicatum</i> floor still preserved <i>in situ</i>. A close-range photogrammetric survey was carried out in order to provide a detailed floors 3D reconstruction; a structure-from-motion photogrammetry approach was employed to obtain a detailed 2D and 3D documentation with millimetric accuracy and high level of colour fidelity. Thanks to the photogrammetric products, accurate virtual restoration/reconstruction of the whole floor surfaces was conducted in a rigorous way and as close as possible to the historical reality.</p>

This paper presents a methodological contribution in the field of the archaeological restitution’s process. This latter is based on virtual anastylosis, which concerns the reconstruction of fragments of the ruins of the nameless Temple of Tipasa in Algeria. We have to mention that our work focused more particularly on the virtual restitution of the three access doors of the sacred courtyard of the temple. Here, we have found many fragments including the voussoirs, which were revealed during the excavation work, encouraging the proposition of our hypothesis on the initial state of the temple. The protocol followed is based on the photogrammetric survey of the blocks which has allowed us to generate 3d models of the elements constituting the entrance facade to the sacred courtyard. The historical documentation as well as the architectural treatises made it possible to fill the gaps with the aim of communicating the most relevant image of our temple. The main objective of the research was to provide a corpus of data in 2d and 3d of all the blocks which has served, at first the documentation and the study of the remains; but also for the proposal of virtual reconstitution hypothesis for valorization and knowledge of part of the history of the site of Tipasa.

<p><strong>Abstract.</strong> Movable heritage preserved in our museums are an invaluable evidence of our past. In order to properly respond to the need of 3D documentation of these significant assets, in the last few years both range-based and image-based solutions have been developed by researchers operating in the framework of Geomatics with a special focus on reaching a high level of detail and on texture radiometric quality, taking into consideration the intrinsic fragility of these kinds of objects which during the survey require a contactless approach. During the presented research a collection of architectural models representing ancient Nubian temples from “Museo Egizio di Torino” had been digitalized using different techniques; in particular, the wooden maquette of the temple of El-Hilla has been acquired using a new structured light handheld laser scanner, the Stonex F6 SR, and applying a close-range photogrammetric approach. In this paper a comparison between the two approaches is proposed as regards acquisition workflow, final results and suitability as regards digitisation of objects belonging to movable heritage and museum collections.</p>

This paper reviews the literature concerning technology used for creating and delivering accessible museum and cultural heritage sites experiences. It highlights the importance of the delivery suited for everyone from different areas of expertise, namely interaction design, pedagogical and participatory design, and it presents how recent and future artificial intelligence (AI) developments can be used for this aim, i.e.,improving and widening online and in situ accessibility. From the literature review analysis, we articulate a conceptual framework that incorporates key elements that constitute museum and cultural heritage online experiences and how these elements are related to each other. Concrete opportunities for future directions empirical research for accessibility of cultural heritage contents are suggested and further discussed.

Polyploidization, or whole genome duplication (WGD), has an important role in evolution and speciation. One of the biggest challenges faced by a new polyploid is meiosis, in particular, discriminating between multiple related chromosomes so that only homologs recombine to ensure regular chromosome segregation and fertility. Here, we report the production of two new hybrids formed by the genomes of species from three different genera: a hybrid between Aegilops tauschii (DD), Hordeum chilense (HchHch), and Secale cereale (RR) with the haploid genomic constitution HchDR (n = 7× = 21); and a hybrid between Triticum turgidum spp. durum (AABB), H. chilense, and S. cereale with the constitution ABHchR (n = 7× = 28). We used genomic in situ hybridization and immunolocalization of key meiotic proteins to establish the chromosome composition of the new hybrids and to study their meiotic behavior. Interestingly, there were multiple chromosome associations at metaphase I in both hybrids. A high level of crossover (CO) formation was observed in HchDR, which shows the possibility of meiotic recombination between the different genomes. We succeeded in the duplication of the ABHchR genome, and several amphiploids, AABBHchHchRR, were obtained and characterized. These results indicate that recombination between the genera of three economically important crops is possible.

A variety of bedrock weathering features—both modern and remnant—including surface grus, polygonal cracks, siliceous glaze, tors, weathering pits, and tafoni typify upland outcrops on the Cumberland Peninsula. Tor ridges are particularly prevalent and at lower elevations they show significant modification and streamlining by flowing ice. On summit areas at elevations above 750 m, however, remnant corestones are preserved in situ, suggesting selective preservation of upland surfaces. Bedrock structure and composition, topographic position, and intensity of process strongly influence tor development. Weathering pits are common on high level, open summit surfaces where weathering occurs in response to both climate and continued removal of derived debris. Pit enlargement through lateral undercutting has been favoured by accumulation of protective bottom residua, mechanical weathering, and the presence of exfoliation crusts. It is postulated that salt crystallization plays a role in outcrop microweathering under present upland arctic conditions.

AbstractMammalian and Drosophila genomes are partitioned into topologically associating domains (TADs). Although this partitioning has been reported to be functionally relevant, it is unclear whether TADs represent true physical units located at the same genomic positions in each cell nucleus or emerge as an average of numerous alternative chromatin folding patterns in a cell population. Here, we use a single-nucleus Hi-C technique to construct high-resolution Hi-C maps in individual Drosophila genomes. These maps demonstrate chromatin compartmentalization at the megabase scale and partitioning of the genome into non-hierarchical TADs at the scale of 100 kb, which closely resembles the TAD profile in the bulk in situ Hi-C data. Over 40% of TAD boundaries are conserved between individual nuclei and possess a high level of active epigenetic marks. Polymer simulations demonstrate that chromatin folding is best described by the random walk model within TADs and is most suitably approximated by a crumpled globule build of Gaussian blobs at longer distances. We observe prominent cell-to-cell variability in the long-range contacts between either active genome loci or between Polycomb-bound regions, suggesting an important contribution of stochastic processes to the formation of the Drosophila 3D genome.

SUMMARYThe electric potential of the contractile vacuole (CV) of Paramecium multimicronucleatum was measured in situ using microelectrodes,one placed in the CV and the other (reference electrode) in the cytosol of a living cell. The CV potential in a mechanically compressed cell increased in a stepwise manner to a maximal value (approximately 80 mV) early in the fluid-filling phase. This stepwise change was caused by the consecutive reattachment to the CV of the radial arms, where the electrogenic sites are located. The current generated by a single arm was approximately 1.3×10-10 A. When cells adapted to a hypotonic solution were exposed to a hypertonic solution, the rate of fluid segregation, RCVC, in the contractile vacuole complex (CVC) diminished at the same time as immunological labelling for V-ATPase disappeared from the radial arms. When the cells were re-exposed to the previous hypotonic solution, the CV potential, which had presumably dropped to near zero after the cell's exposure to the hypertonic solution, gradually returned to its maximum level. This increase in the CV potential occurred in parallel with the recovery of immunological labelling for V-ATPase in the radial arm and the resumption of RCVC or fluid segregation. Concanamycin B, a potent V-ATPase inhibitor, brought about significant decreases in both the CV potential and RCVC. We confirm that (i) the electrogenic site of the radial arm is situated in the decorated spongiome, and (ii) the V-ATPase in the decorated spongiome is electrogenic and is necessary for fluid segregation in the CVC. The CV potential remained at a constant high level(approximately 80 mV), whereas RCVC varied between cells depending on the osmolarity of the adaptation solution. Moreover, the CV potential did not change even though RCVC increased when cells adapted to one osmolarity were exposed to a lower osmolarity, implying that RCVC is not directly correlated with the number of functional V-ATPase complexes present in the CVC.

Purpose The purpose of this study is to explore the hedonic experience and its formation at heritage attractions. Design/methodology/approach A qualitative and exploratory approach was applied, using data from 21 semi-structured interviews and three in-situ focus groups. Findings Findings highlight that senses, imagery and emotions are stimulated by the physical landscape and by triggers of memorable experiences. Research limitations/implications To further explore this topic, a broader range of heritage attractions and perspectives from the diverse stakeholders involved in the management and consumption of these sites is needed. Originality/value Given the scarcity of research dedicated to the hedonic experience at heritage sites, this study provides a contribution by exploring the visitor’s perspective and points out relevant insights. As the hedonic feelings of pleasure, comfort and related affective responses impact the quality of memorable experiences, relevant implications for theory and practice are discussed.

Abstract. Organic matter preserved in Lake Ohrid sediments originates from aquatic and terrestrial sources. Its variable composition reflects climate-controlled changes in the lake basin's hydrology and related organic matter export, i.e. changes in primary productivity, terrestrial plant matter input and soil erosion. Here, we present first results from lipid biomarker investigations of Lake Ohrid sediments from two near-shore settings: site Lz1120 near the southern shore, with low-lying lands nearby and probably influenced by river discharge, and site Co1202 which is close to the steep eastern slopes. Variable proportions of terrestrial n-alkanoic acids and n-alkanols as well as compositional changes of ω-hydroxy acids document differences in soil organic matter supply between the sites and during different climate stages (glacial, Holocene, 8.2 ka cooling event). Changes in the vegetation cover are suggested by changes in the dominant chain length of terrestrial n-alkanols. Effective microbial degradation of labile organic matter and in situ contribution of organic matter derived from the microbes themselves are both evident in the sediments. We found evidence for anoxic conditions within the photic zone by detecting epicholestanol and tetrahymanol from sulphur-oxidising phototrophic bacteria and bacterivorous ciliates and for the influence of a settled human community from the occurrence of coprostanol, a biomarker for human and animal faeces (pigs, sheep, goats), in an early Holocene sample. This study illustrates the potential of lipid biomarkers for future environmental reconstructions using one of Europe's oldest continental climate archives, Lake Ohrid.
