The aim of this study was to evaluate the effect of a 20% increase in dietary energy during short-term estrus induction treatment on the reproductive parameters of Santa Inês ewes. Females (n=43) were allocated into two experimental groups according to the amount of energy inclusion in the diet: maintenance diet or maintenance diet plus 20% energy. Ultrasound examinations were performed in order to detect ovulation. To assess sexual behavior, ewes were teased and further mated. Blood samples were collected for the determination of glucose and insulin concentrations. There was no difference (P>0.05) between groups in the following categories: ovulation rate (80.00% vs. 60.00%), largest follicle diameter (6.00 ± 0.20 vs. 5.90 ± 0.60), interval from device removal to ovulation (52.80 ± 14.87 vs. 59.01 ± 8.34 hours), animals in estrus (75.00% vs. 65.21%), interval from device removal to estrus (30.00 ± 15.49 vs. 30.00 ± 13.35 hours) and conception rate (50.00% vs. 21.73%). There were differences (P<0.05) in the concentrations of insulin and glucose. It can be concluded that the 20% increase in energy in the diet during short-term estrus induction treatment did not affect the reproductive parameters studied.

This study compared new and previously used (and autoclaved) progesterone devices for synchronisation of oestrus during the breeding season in Toggenburg goats. Nulliparous (n = 17) or lactating (n = 50) received new intravaginal devices containing 0.3 g progesterone (CONTROL), or similar devices previously used for either 6 (USED6) or 12 (USED12) days and subsequently autoclaved. All goats received 5 mg dinoprost at device insertion and 200 IU equine chorionic gonadotropin 5 days later and all devices were removed after 6 days. After device removal, females were mated by fertile bucks. Ovarian ultrasonography was performed every 12 h after device removal until ovulation detection. Blood samples were collected for determination of plasma progesterone concentration in different moments and intervals (from 7 days before device insertion to 3 days after its removal). There was no difference (P > 0.05) among groups CONTROL, USED6 or USED12 for: oestrus response [75% (18/24), 77% (17/22) or 71% (15/21), respectively]; duration of oestrus (30.7 ± 3.4, 31.8 ± 1.7 or 32.8 ± 3.4 h), percentage of ovulating goats [67% (6/9), 78% (7/9) or 56% (5/9)], ovulation rate (1.3 ± 0.2, 1.4 ± 0.2 or 1.8 ± 0.4 units of corpora lutea), average follicle diameter (6.2 ± 0.1, 6.7 ± 0.1 or 6.8 ± 0.3 mm) and pregnancy rate [54% (13/24), 50% (11/22) or 48% (10/21)]. Plasma progesterone concentrations were not different (P > 0.05) for does among treatments and between nulliparous and lactating females. In conclusion, autoclaved, previously used intravaginal progesterone devices are effective in synchronising oestrus and ovulation in cyclic goats during the breeding season.

The aim of this study was to verify the efficacy of reusing intravaginal progesterone (P4) devices on the reproductive parameters in Santa Inês ewes. Females received intravaginal P4 devices for their first, second or third use for five days plus 300 IU eCG IM and 5mg dinoprost laterovulvar 24h before device removal. Blood was collected at different moments. Transrectal ultrasonography was performed from device removal to ovulation. Part of the ewes were submitted to artificial insemination by laparoscopy (IAL - n=55) with fresh semen, whereas the rest were bred by fertile rams (n=41). On the initial 18 h, ewes that received devices for the first time showed higher P4 concentrations (5.1±1.8 vs 3.5±1.4 vs 2.4±1.1 - P<0.05). However, after the first 48h no difference was observed among all treatments and P4 supraluteal concentrations were detected in all ewes upon device removal. Estrous response, interval from device removal to estrus, rate of ovulating animals, number of ovulations, time from device removal to ovulation and average conception rates after IAL or natural mating were similar among all 3 groups. Intravaginal progesterone devices can be used up to three times without altering reproductive parameters in Santa Inês ewes.

Hormonal oestrus induction treatment is an important tool for the success of reproductive biotechniques in anoestrus goats. In the nonbreeding season, oestrus induction can be performed by the use of several protocols, including those varying in their duration. This study aimed to compare short-term (for either 5 or 6 days) progestogen-based oestrus synchronization protocols in dairy [Toggenburg (n=12) and Saanen (n=12)] goats during the nonbreeding season. This study was performed in Minas Gerais State, Brazil (21°35′S and 43°15′W), with 12 nulliparous (12–14 months old) and 12 pluriparous (2–4 years old in final third of lactation) goats, all with body condition scores (BCS) of 3.0 to 3.5. They were equally assigned to both treatments according to their order of parturition, BCS, and breed. On a random day (Day 0), 24 goats received an intravaginal device of 60mg of medroxyprogesterone acetate (Progespon®, Zoetis) which remained for 5 (G5; n=12) or 6 (G6; n=12) days. On Day 5, goats from both groups received 30µg of cloprostenol intramuscularly (IM) (Prolise®, ARSA S.R.L.) and 200IU of equine chorionic gonadotrophin (eCG) IM (Novormon®, Schering Plough Animal Health). Both groups received cloprostenol and eCG on the same day (regardless of device removal) to ensure they were administered at the same follicular wave. Both device insertion and removal were performed in the morning. Ultrasound evaluations were performed every 12h from device removal to ovulation detection. Ovulations were considered as the average interval between the last observation of follicles and the first exam in which it was no longer seen. The normal distribution of variables was determined by Shapiro-Wilk test and homocedasticity by Levene’s test. All variables were not normally distributed and thus analysed by Mann–Whitney U test, with P&lt;0.05 considered significantly different. Values are presented in median±interquartile range. There was no difference between G5 and G6, respectively, in the interval to oestrus (48.0±0.0 vs. 48.0±12.0h), interval from oestrus onset to ovulation (18.0±9.0 vs. 18.0±12.0h), interval from device removal to ovulation (66.0±18.0 vs. 66.0±24.0h), number of ovulated follicles (2.0±2.0 vs. 2.5±2.0), or mean diameter of ovulated follicles (6.6±1.6 vs. 6.6±1.4mm). All ovulations were detected in the morning. In conclusion, our data demonstrate that short-term hormonal treatment for 5 or 6 days is similarly efficient to induce synchronous oestrus in Saanen and Toggenburg goats. ABSTRACT The aim of this study was to evaluate hCG treatment on ovarian response and on pregnancy rate using a 9-day oestrus synchronization protocol in Santa Ines ewes. On a random oestrus cycle day, ewes received an intravaginal progesterone device (Primer-PR®, Tecnopec, Brazil). Nine days later (Day 9), 30µg of d-cloprostenol (Prolise®, Syntex, Argentina) and 250IU of eCG (Folligon®, Intervet, Brazil) were administered and the progesterone device was removed. This moment, the ewes were randomly assigned on two groups: Control Group and hCG Group. In the hCG Group, the ewes received 500IU of hCG (Vetecor®, Hertape-Calier, Spain) 24h after device removal. In the Control Group, the ewes did not receive any ovulation inductor. Control and hCG Groups ewes were inseminated 60h and 48h after device removal, respectively. There was no difference between the groups regarding the first ovulatory follicle diameter and the second ovulatory follicle. hCG Group ewes had shorter interval between device removal and ovulation (Control: 79.9±15.4h and hCG: 54.7±4.9h; P=0.001) and more synchronized ovulations. However, the treatment with hCG decreased the pregnancy rate after TAI (P=0,009). In conclusion, hCG administration improves ovulatory synchronisation, but causes a decrease in the pregnancy rate.

A new, simple, sensitive and rapid spectrophotometric method is proposed for the determination of trace amount of Nickel (II). The method is based on the formation of a 1:2 complex with 4-(4-((2-hydroxy-6-nitrophenyl) diazenyl) -3-methyl-5-oxo-2, 5-dihydro-1H-pyrazol-1-yl) benzenesulfonic acid (2-ANASP) as a new reagent is developed. The complex has a maximum absorption at 516 nm and εmax of 1. 84 X 105 L. mol-1. cm-1. A linear correlation (0. 25 – 4. 0μg. ml-1) was found between absorbance at λmax and concentration. The accuracy and reproducibility of the determination method for various known amounts of Nickel (II) were tested. The results obtained are both precise (RSD was 1. 2 %) and accurate (relative error was 0. 787 %). The effect of diverse ions on the determination of Nickel (II) to investigate the selectivity of the method were also studied. The stability constant of the product was 0. 399 X 106 L. mol-1. The proposed method was successfully applied to the analysis of diabetes blood and normal human blood.

An ever-increasing need exists within the forensic laboratories to develop analytical processes for the qualitative and quantitative determination of a broad spectrum of new psychoactive substances. Phenylethylamine derivatives are among the major classes of psychoactive substances available on the global market and include both amphetamine analogues and synthetic cathinones. In this work, an ultra-high-performance liquid chromatography-positive ion electrospray ionization tandem mass spectrometric method (UHPLC-ESI-MS/MS) has been developed and fully validated for the determination of 19 psychoactive substances, including nine amphetamine-type stimulants and 10 synthetic cathinone derivatives, in premortem and postmortem whole blood. The assay was based on the use of 1 mL premortem or postmortem whole blood, following solid phase extraction prior to the analysis. The separation was achieved on a Poroshell 120 EC-C18 analytical column with a gradient mobile phase of 0.1% formic acid in acetonitrile and 0.1% formic acid in water in 9 min. The dynamic multiple reaction monitoring used in this work allowed for limit of detection (LOD) and lower limit of quantitation (LOQ) values of 0.5 and 2 ng mL−1, respectively, for all analytes both in premortem and postmortem whole blood samples. A quadratic calibration model was used for the 12 quantitative analytes over the concentration range of 20–2000 ng mL−1, and the method was shown to be precise and accurate both in premortem and postmortem whole blood. The method was applied to the analysis of real cases and proved to be a valuable tool in forensic and clinical toxicology.

Abstract. Seven patients suffering from severe endometriosis were treated with danazol 200 mg × 3 daily for 6 months. Clinical symptoms were alleviated and menses disappeared in response to the treatment. After cessation of the treatment the menstrual bleedings returned in 1–3 months. Blood samples for determination of gonadotrophins, prolactin (Prl), oestradiol (E2), progesterone, thyroid hormones and thyrotrophin in radioimmunoassays were taken and a combined TRF and LRF test carried out in the follicular phase before treatment, at the 6th month of treatment and after reappearance of the first menses. There were no statistically significant changes in the basal levels of serum FSH, LH or TSH during the danazol treatment. Neither was there any change in episodic secretions of FSH, LH or Prl, as determined by the mean coefficients of variation of the hormone levels in seven consecutive samples taken at 20 min intervals. On the other hand, serum E2, Prl and thyroid hormone levels were significantly decreased in the 6th month of treatment. In the TRF-LRF test the responses of serum FSH and LH were significantly higher and those of serum Prl and TSH significantly lower during danazol treatment than before. Prl responses remained lowered after the treatment. It appears that low serum oestrogen levels, induced by the danazol treatment, sensitize the pituitary gonadotrophs to exogenous LRF, but make the sensitivity of thyrotrophs and lactotrophs lower to exogenous TRF. These results thus indicate that danazol does not make the pituitary gonadotrophs insensitive to LRF, but danazol may rather inhibit the secretion of hypothalamic LRF.
