Antioxidants are helpful in prevention of several diseases related with oxidative stress including neurodegenerative disorders. In recent studies, carbazoles were given proof of promising antioxidant activities. In this article, 9-ethyl-9H-carbazole hydrazone derivatives were synthesized, characterized and their in vitro antioxidant activity and possible cytotoxic effects were investigated. Furthermore, protective effect of the synthesized derivatives against amyloid β-induced damage in PC12 neuronal cells was examined by using MTT assay. The newly synthesized carbazoles were found to have radical scavenging activity with a varying potency both in cell-free and cell-based in vitro assays. Several compounds, especially such as 3d and 3e, 3m and 3n bearing two halogen groups on the phenyl ring, were found to have cytotoxic activity. However, their cytotoxic activities were not higher than that of melatonin. Several compounds also significantly protected neuronal PC12 cells against amyloid β-induced damage, which can be defined as neuroprotective agents. (4-(2-((9-Ethyl-9H-carbazol-3-yl)methylene)hydrazinyl)benzonitrile) 3r was found as the most active compound with both radical scavenging activity and neuroprotective effects against amyloid β-induced damage. These findings might provide an alternative strategy for developing novel carbazole derivatives for management of neurodegenerative diseases, such as Alzheimer's disease.

Background: Cataract is a major cause of visual impairment and blindness around the world. This study evaluated the in vitro antioxidant and anti-cataract activities of Cnidoscolus aconitifolius leaves extract and fractions. Antioxidant activities were evaluated by 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azinobis (3-ethylbenzothiozoline-6-sulfonic acid) (ABTS), total reducing power, and hydrogen peroxide scavenging assays. Anti-cataract potential was evaluated in vitro using goat lenses divided into eight groups of different treatments and incubated in artificial aqueous humor at 37 °C for 72 hours. Glucose-induced opacity in the lenses was observed and biochemical indices quantified (catalase, Malondialdehyde (MDA) and total protein in the lens homogenate). Results: The crude extract and its fractions possess substantial antioxidant activities. The aqueous fraction exhibited the best DPPH radical scavenging activity (IC50 value 78.599 µg/ml); while the dichloromethane fraction exhibited the highest ABTS radical scavenging activity with IC50 66.68 µg/ml. The anti-cataract evaluation of crude and fractions at 250 μg/ml showed a significant increase (p<0.05) in the total protein and catalase activity compared to the cataract control group. The malonaldehyde level decreased significantly (p<0.05) in all the treated groups.

The study was designed to investigate the antioxidant activities of the methanolic extract of leaves of Begonia roxburghii by five different in vitro methods namely Folin-Ciocalteu, total flavonoid content, phosphomolybdenum, total reducing power determination, and DPPH scavenging activity. Phenol content (Gallic acid equivalent) was found as 180.40±0.03 mg/g of the dry sample by the Folin-Ciocalteu method. Flavonoid assay method revealed the content of flavonoids (Quercetin equivalent) as 60.43±0.27 mg/g of the dry sample. Total antioxidant capacity of the extract was observed 243.43±0.11 mg/g ( ascorbic acid equivalent) by phosphomolybdenum method. Free radical scavenging activity (measured by the DPPH test) of the methanolic extract was found 158.36±0.22 μg/mL whereas ascorbic acid, the standard, showed 19.53±0.04 μg/mL. The methanolic extract of Begonia roxburghii leaves exhibited positive and linear dose dependent reducing power activity and its activity was comparable to the standard ascorbic acid for a given dose. Phytochemical screening of the extract showed positive results for alkaloids, carbohydrates, flavonoids, glycosides, saponins, steroids, and tannins, which demonstrate definite pharmacological actions on human body. Dryopteris filix mas (D filix-mas) is wildly used in ethnomedicine for the management of rheumatoid arthritis, wounds and other diseases. We investigated the anti-oxidant activities of its leaf extract, and chromatographic fractions. The ethanol leaf extract was partitioned into four fractions; n-hexane, ethyl acetate, n-butanol and water. Ferric reducing anti-oxidant power (FRAP), 1, 1-diphenyl-2-picrylhydrazil (DPPH) and nitric oxide (NO) scavenging in vitro assays were carried out on the extract and fractions at 6.25, 12.5, 25, 50, 100, 200, 400 and 800 µg/mL. The most active fraction (ethyl acetate fraction) was further purified using chromatographic techniques to isolate its major compound whose structure was elucidated using ID nuclear magnetic resonance (NMR) and mass spectrometry. The ethyl acetate fraction produced the highest free radical scavenging activity among the other fractions. The fraction (VLC-E7) from which the bioactive compound, quercetin-3-O-αL-rhamnopyranoside, was isolated had the best FRAP and DPPH scavenging activities with EC50 and IC50 values of 88.81 ± 3.41 and 26.87 ± 0.24 respectively more than the ethyl acetate fraction. This study revealed that the polyphenol flavonoid, quercetin-3-O-αL-rhamnopyranoside could be responsible for antioxidant activity of ethno-medicinal property of D filix-mas leaf.

The antioxidant and hepatoprotective activities of ethyl acetate extract was carefully investigated by the methods of DPPH radical scavenging activity, Hydroxyl radical scavenging activity, superoxide radical scavenging activity, hydrogen peroxide radical scavenging activity, and its reducing power ability. All thesein vitroantioxidant activities were concentration dependent, which were compared with standard antioxidants such as BHT, α-tocopherol. The hepatoprotective potential ofPodophyllum hexandrumextract was also evaluated in male Wistar rats against carbon tetrachloride- (CCl4-) induced liver damage. Pretreated rats were given ethyl acetate extract at 20, 30, and 50 mg/kg dose prior to CCl4administration (1 mL/kg, 1:1 in olive oil). Rats pretreated withP. hexandrumextract remarkably prevented the elevation of serum AST, ALT, LDH, and liver lipid peroxides in CCl4-treated rats. Hepatic glutathione levels were significantly increased by the treatment with the extract in all the experimental groups. The extract at the tested doses also restored the levels of liver homogenate enzymes (glutathione peroxidase, glutathione reductase, superoxide dismutase, and glutathione-S-transferase) significantly. This study suggests that ethyl acetate extract ofP. hexandrumhas a liver-protective effect against CCl4-induced hepatotoxicity and possessin vitroantioxidant activities.

AbstractIn vitro antioxidant activities of three sweet dihydrochalcone glucosides from the leaves of Lithocarpus pachyphyllus (Kurz) Rehd. (Fagaceae), trilobatin 2″-acetate (1), phloridzin (2) and trilobatin (3), were investigated. The IC50 (50% inhibitory concentration) values for compounds 1-3 of lipid peroxidation in rat liver homogenate were 261, 28, 88 μm, respectively. Compounds 1-3 increased superoxide dismutase (SOD) activity with EC50 (50% effective concentration) values of 575, 167, 128 μm, and glutathione peroxidase (GSH-Px) activity with EC50 values of 717, 347, 129 μm, respectively, and showed only weak DPPH (1,1- diphenyl-2-picrylhydrazyl) radical scavenging activity

The antioxidant activities of extracts and residuum of Perillafrutescens fruits from supercritical CO2 extraction were determined in vitro. The residuum was extracted in turn with water, propyl alcohol and ethyl acetate. The antioxidant activities of the extracts were assayed with antioxidant capacity in linoleic acid model system, reducing powers, radical scavenging activity using 1,1-diphenyl-2-picrylhy-drazyl (DPPH) method. The results show that the ethyl acetate extract of Perillafrutescens possesses strongest DPPH radical scavenging activity and reducing power in a concentration-dependent manner.

The aim of this work was the analytical characterization of the ?-orcinol depside, baeomycesic acid in lichens extracts. The extract of Thamnolia vermicularis var. subuliformis was analyzed by the two different methods, namely HPLC-UV and 1H NMR analysis. The results showed that baeomycesic acid was the most abundant depside in the lichens. These results could be of use for rapid identification of this metabolite in other lichen species. Besides baeomycesic acid, three depsides and one monocyclic phenolic compound were isolated from the lichen extract on the chromatographic column. The structure of baeomycesic acid was confirmed by HPLC-UV and spectroscopic methods. In addition, antioxidant and cytotoxic activities of baeomycesic acid were determined. The result of the testing showed that baeomycesic acid exhibited a moderate radical scavenging activity (IC50 = 602.10 ? 0.54 ?g/mL) and good cytotoxic activity. This is the first report of detailed analytical characterization, isolation, as well as antioxidant and cytotoxic activities of baeomycesic acid from Thamnolia vermicularis. These results may be helpful in future industrial production of herbal medicines that include this important natural product.

Second-grade dates (Phoenix dactyliferaL.), with hard texture, from three selected Tunisian cultivars (Allig, Deglet Nour, and Bejo) were analysed from their antioxidant activities using DPPH radical scavenging activity, FRAP assay, H2O2scavenging activity, and metal chelating activity. Date extracts showed strong and concentration-dependant activity in all tested methods. The results showed that the best antioxidant activity was obtained in Allig, followed by Bejo and Deglet Nour. Total phenolics, total flavonoids, carotenoids, and tannins were determined spectrophotometrically in three date extracts. Results indicated that date contained significantly different amounts of these compounds. In fact, Allig presented the highest antioxidant compounds, followed by Bejo and Deglet Nour. Correlation analyses indicated a positive linear relationship between antioxidant activities and total phenolic and flavonoid content of date. This study demonstrates the potential antioxidant activity with Tunisian date, where we can use these natural extracts as food additives in replacement of synthetic compounds.

The current study was carried out to determine the antioxidant potential, anti-inflammatory activity, and cellular viability of Pterospermum rubiginosum (PR), a tropical tree endemic to the Western Ghats. The antioxidant activities of the PR bark methanolic (PRME) and aqueous extract (PRAQ) were tested using ABTS as well as superoxide, nitric oxide, and hydroxyl radical assays. Total antioxidant activity was evaluated by adopting the colorimetric method and correlation with their antioxidant activities was derived by Pearson co-efficient analysis. The PRME showed the highest ABTS radical scavenging activity, EC50 (46.09µg/ml) followed by PRAQ (52.08µg/ml). Furthermore, the PRME exhibited the highest scavenging activity against superoxide, nitric oxide, and hydroxyl radicals. The MTT assay results revealed good cellular viability up to a concentration of 100µg/ml with an EC50 (106.869µg/ml). The inflammatory mediators such as Cox-2, IL-1β, IL-6, and NF-kB were reduced during the treatment of PRME in LPS stimulated RAW cells. The stress marker in rat liver cells such as glutathione reductase (GR), glutathione peroxidase (GPx), and reduced glutathione (GSH) levels was found in normal levels when compared to the untreated group of rats. The antioxidant enzyme superoxide dismutase and catalase also exhibited notable bioactivity in PRME treated groups up to a concentration of 1000µg/ml. The present study showed excellent In vitro and In vivo antioxidant activity; the potent anti-inflammatory ability of PRME in reducing the LPS induced inflammation in cell culture conditions.
