Marigold flower (Tagetes erecta L.) produces lutein compounds which present biological activities such as antioxidant, antiinflammatory, antimutagenicity, and immunomodulatory effects. The study was to investigate the antioxidant activity of the lutein of T. erecta L. and the effect of lutein on the activity and phagocytic capacity of macrophage cells. The antioxidant screening was carried out using diphenyl picrylhydrazyl (DPPH), 2,2′-and-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging assay with serial concentrations and ferric-reducing antioxidant power (FRAP) method. For the observation of activity and phagocytic capacity of peritoneal macrophages, twenty-eight mice were used and divided into seven groups each comprising four replicates, i.e., Group (I) normal controls, mice were untreated (II) a negative control, mice were induced by Staphylococcus aureus (III) positive control, mice were induced by S. aureus and treatment of meniran extract (Phyllanthus niruri). The treatment group (IV-VII) mice were induced by S. aureus and treated crude lutein, respectively: 0.15 mg, 0.30 mg, 0.60 mg, and 0.90 mg. 20 g−1 of body weight. The lutein extracted from T. erecta shows an antioxidant activity against DPPH radical with an IC50 value of 53.58 μg.ml−1, while the antioxidant activity against ABTS has an IC50 value of 72.91 μg.ml−1. The antioxidant activity test results by the FRAP method at each lutein concentrations of 10, 25, 50, and 75 ppm were obtained respectively of 33, 88, 185.5, and 288.5 μmol Fe2+/g extract. The data were analyzed using one-way ANOVA and Duncan’s multiple range test (DMRT) after. The phagocytic activity was 45.5%; 54.75%; 57.50% and 67.0%, respectively, while the phagocytic capacity values ​​were 355; 519; 611 and 767 S. aureus bacterial cells per 50 macrophage cells. The lutein from marigolds (T. erecta L.) is capable of scavenging free radicals and reducing oxidants. Lutein can increase the activity and capacity of phagocytic of peritoneum macrophage cells in mice.

ABSTRAK: Pada penelitian ini dilakukan pengembangan formula sediaan hand and body cream dengan memanfaatkan sumber antioksidan yang dimiliki oleh dua jenis ekstrak etanol yaitu ekstrak bunga pacar air merah (Impatiens balsamina L.) dan bunga gemitir (Tagetes erecta L.) dengan taraf perlakuan sebesar 1% dari masing-masing ekstrak (FPM dan FG) dan campuran 0,5% ekstrak pacar air merah dan 0,5% ekstrak gemitir (FCPMG). Pengujian terhadap  produk dilakukan pada sifat fisikokimia  meliputi pH, viskositas, aktivitas antioksidan, angka lempeng total mikroba, dan ukuran partikel. Berdasarkan hasil pengujian produk hand and body cream yang dihasilkan memiliki antioksidan yang cukup tinggi sebagai produk perawatan tubuh dengan penambahan 1% ekstrak etanol pacar air merah, gemitir dan campuran dari kedua ekstrak etanol. Kualitas produk hand and body cream yang dihasilkan baik dan memenuhi standar mutu SNI dari aspek nilai pH, viskositas, dan nilai ALT. Warna yang dihasilkan mulai dari warna kuning kemerahan sampai dengan warna kuning, dan partikel yang dimiliki oleh produk adalah nanopartikel dengan sistem polidispersi. ABSTRACT: Formulations of hand and body creams using ethanol extracts of flowers of red Impatiens balsamina L. And Tagetes erecta L. As antioxidant sources have been developed.  One percent of  extracts (FPM and FG) and the mixture of 0.5% each extracts (FCPMG) were added to the basic formula. The analysis of physicochemical properties of the products was conducted including pH, viscosity, antioxidant activity, the size of the particles, and total microbial plate numbers. Based on the results, the quality of the hand and body creams meet the national standard of Indonesia (SNI) regarding to pH, viscosity, and the total microbial numbers. The colours of the hand and body creams resulted are reddish yellow to yellow with polydispersion nanoparticles.

<p>Immunomodulatory activity assay and characterization of exopolysaccharide (EPS) from Lactic Acid Bacteria (LAB) was done in Bogor. Bacteria used in this study was LAB strains of Lactobacillus delbrueckii subsp. bulgaricus and Streptococcus thermophilus. Exopolysaccharide was extracted from L. delbrueckii subsp. bulgaricus and S. thermophilus then characterized with FT-IR spectrophotometer to determine the functional group. IR spectrum analysis using Fourier Transform-Infra Red (FT-IR) showed that EPS from both LAB isolates were carbohydrate compounds. Immunomodulatory activity in vivo from EPS was measured using phagocytic activity and phagocytic capacity macrophage cells from mice peritoneal cavity fluid. Exopolysaccharide were given orally to mice in concentrations of 100 μg/ml, 200 μg/ml and 300 μg/ml for 14 days then the mice were infected with Staphylococcus aureus. Result showed that EPS from both LAB isolate enhanced either phagocytic activity and phagocytic capacity macrophage cell from mice peritoneal fluid. EPS from L. delbrueckii subsp. bulgaricus concentration 300 μg/ml showed the highest phagocytic activity of macrophage cells and EPS from S. thermophilus concentration 300 μg/ml showed the highest phagocytic capacity. It is concluded that EPS potency tested as immunomodulatory derived from a culture of L. delbrueckii and S. thermophilus subsp.bulgaricus are able to increase the activity and phagocytosis murine peritoneal macrophages.</p>

Pyrazoline compounds are reported to have many useful biological activities such as antimicrobials, antihyperglycemia, antioxidants, anti-inflammatory, anticancer and anticonvulsants. The pyrazoline analogues of 3- (2-methoxy-phenyl) -5-naphthalene-1-yl-4,5-dihydro-1H-pyrazol were synthesized usingglacial acetic acid as catalyst and assisted by microwave irradiation. The structures of the compounds were obtainedby UV, IR and HRMS spectroscopy data. Antioxidant activity test using DPPH method showed that pyrazoline compound had IC50 value of 27.41 μg/mL.

Tagetes erecta L. is a popular ornamental plant of the Asteraceae family, which is widely cultivated not only for its decorative use, but also for the extraction of lutein. Besides carotenoid representatives, which have been extensively studied, other important classes of secondary metabolites present in the plant, such as polyphenols, could exhibit important biological activities. The phytochemical analysis of a methanolic extract obtained from T. erecta inflorescences was achieved using liquid chromatography–mass spectrometry (LC-MS) techniques. The extract was further subjected to a multistep purification process, which allowed the separation of different fractions. The total extract and its fractions contain several polyphenolic compounds, such as hydroxybenzoic and hydroxycinnamic acid derivatives, flavonols (especially quercetagetin glycosides), and several aglycons (e.g., quercetin, patuletin). One of the fractions, containing mostly quercetagitrin, was subjected to two different antioxidant assays (metal chelating activity and lipoxygenase inhibition) and to in vitro cytotoxicity assessment. Generally, the biological assays showed promising results for the investigated fraction compared to the initial extract. Given the encouraging outcome of the in vitro assays, further purification and structural analysis of compounds from T. erecta extracts, as well as further in vivo investigations are justified.

Kumis kucing (Orthosiphon stamineus), pegagan (Centella asiatica), seledri (Apium graveolens), kunyit (Curcuma domestica), temulawak (Curcuma xanthorrhiza), tempuyung (Sonchus arvensis) and meniran (Phyllanthus niruri) are herbs that commonly used in the Indonesia folk medicine. The constituents that responsible for several important biological activities are phenolic and flavonoid compounds which also possess antioxidant activity. Antioxidant activity of those seven Indonesian herbal extracts was evaluated using DPPH, ABTS and FRAP methods. The extraction was done with the reflux method by using 80% ethanol as a solvent. The total phenol and total flavonoids from each herbal extract were measured using Folin–Ciocalteu reagent and spectrophotometry. Antioxidant activity results by DPPH method on O. stamineus, C. asiatica, A. graveolens, C. domestica, C. xanthorrhiza, S. arvensis, and P. niruri showed IC50 value at 132; ND; 2221; 361; 538; 1118; and 102 ppm, respectively. Results from ABTS method, showed IC50 value at 22; 1199; 169; 100; 82; 143; and 20 ppm respectively. While results from the FRAP method showed that the ethanolic extract of P. niruri at a concentration of 20 ppm possesses the strongest antioxidant activity (17.41 ppm AEAC/ppm extract). The content of total phenolic compounds are 22.50; 0.67; 2.16; 11.40; 7.80; 7.22; and 2.62% GAE, while the total flavonoid compounds were 19.88; 6.67; 4.06; 71.02; 34.62; 3.78; and 8.34% QE, respectively. It can be concluded that ethanolic extract of P. niruri and O. stamineus obtain the highest antioxidant activity based on DPPH, ABTS and FRAP method.

Kandungan kimia flavonoid dan fenolik dalam ekstrak daun T.erecta diketahui memiliki aktivitas antibakteri yang baik. Pengunaan ekstrak daun T.erecta sebagai zat aktif sediaan hidrogel akan sangat mempermudah pemanfaatan dalam proses terapi oleh masyarakat. Tujuan dalam penelitian ini adalah melihat kemampuan aktivitas antibakteri sediaan hidrogel dengan zat aktif ekstrak daun T.erecta. Dalam penelitian ini digunakan 6 variasi konsentrasi ekstrak yang terkandung pada sediaan hidrogel yaitu 1.5; 2.0; 2.5; 3.0; 3.5; dan 4.0 %. Pengujian aktivitas antibakteri menggunakan metode difusi sumuran terhadap bakteri Staphylococcus aureus dan Escherichia coli. Aktivitas antibakteri terbesar ditunjukkan oleh sediaan hidrogel dengan konsentrasi ekstrak daun T.erecta 2.5%. Diameter zona hambat yang terbentuk terhadap pertumbuhan E.coli adalah 15.33 mm sedangkan diameter zona hambat terhadap S.aureus adalah 17.87 mm. Untuk kontrol positif digunakan bioplacenton® dan untuk kontrol negatif digunakan basis hidrogel tanpa kandungan ekstrak T.erectaFlavonoid and phenolic on ethanolic extract of T.erecta L has been antibacterial activity. The aim of this research was to determinate of antibacterial from hydrogel with ethanolic extract of T.erecta L. This research was begun making hydrogels with six variations of ethanolic extract of T.erecta L were 1.5; ; 2.0; 2.5; 3.0; 3.5; and 4.0 %.  Antibacterial activity used hole diffusion method against Staphylococcus aureus and Escherichia coli. Hydrogel containing 2.5 % ethanolic extract of T.erecta L was shown the highest antibacterial activity. The diameters of the inhibition zone against E.coli was 15.33 mm and against S.aureus was 17.87 mm. Positive control used bioplacenton® and negative control was hydrogel without ethanolic extract of T.erecta content
