Abstract BackgroundWe previously identified six drought-inducible CC-type glutaredoxins in cassava cultivars, however, less is known about their potential role in the molecular mechanism by which cassava adapted to abiotic stress. ResultsHerein, we investigate one of cassava drought-responsive CC-type glutaredoxins, namely MeGRXC3, that involved in regulation of mannitol-induced inhibition on seed germination and seedling growth in transgenic Arabidopsis. MeGRXC3 overexpression up-regulates several stress-related transcription factor genes, such as PDF1.2, ERF6, WRKY40, and WRKY53 in Arabidopsis. Protein interaction assays show that MeGRXC3 interacts with Arabidopsis TGA2 and TGA5 in the nucleus. Eliminated nuclear localization of MeGRXC3 failed to result mannitol-induced inhibition of seed germination and seedling growth in transgenic Arabidopsis. Mutation analysis of MeGRXC3 indicates the importance of conserved motifs for its transactivation activity in yeast. Additionally, these motifs are also indispensable for its functionality in regulating mannitol-induced inhibition of seed germination and enhancement of the stress-related transcription factors in transgenic Arabidopsis.ConclusionsMeGRXC3 overexpression confers mannitol sensitivity in transgenic Arabidopsis possibly through interaction with TGA2/5 in the nucleus, and nuclear activity of MeGRXC3 is required for its function.

Boron (B), an essential micronutrient, helps the plants to complete their life cycle successfully. Therefore, the present experiment was conducted to study (1) the role of B in seed germination and seedling growth, (2) the toxicity effect of B in seed germination and seedling growth and (3) the role of B in tolerance of barley (Hordeum vulgare L. var. ‘Bakore’) to NaCl stress. Under NaCl stress and non-stress conditions, application of high levels of B (100 µM) decreased parameters of germination (G%, VI, GI and MGT), growth (RL, SL, RFW, SFW, RDW and SDW), except the accumulation of Pro and MDA in barley seedlings. Also, a fluorescence study reveals that production of ROS (H2O2 and O2 •—) and non-viable cells increased in roots of barley seedlings treated with NaCl and high dose of B. An alteration in anatomical structure of barley seedlings was observed with the application of NaCl and high dose of B. However, a low concentration of B (50 µM) proved best and increased all germination and growth traits of barley seedlings by increasing further accumulation of Pro. Also, 50 µM of B significantly increased the biosynthesis of photosynthetic pigments (Chl a, b and total Chl) and deceased formation of ROS and viable cells in roots. Therefore, concluded that sufficient dose of B could be beneficial for barley plant in improving the tolerance to NaCl stress.

The role of L-lysine (Lys) on some cytogenetic and physiological parameters in Allium cepa L. seeds exposed to salt was evaluated. NaCl stress on the other hand showed a significantly inhibitory effect on the seedling growth and seed germination of Allium cepa. Besides, it significantly reduced the mitotic index in the root tip meristems of seeds and increased micronuclei which are the most effective and simplest indicator of cytological damage and chromosomal abnormalities. However, the effects of salinity on chromosomal aberrations, seedling growth, seed germination and mitotic activity have decreased significantly with Lys application.

The aim of the study was to determine the role of the seed coat in the presence of aqueous extracts from Stellaria media (L.) Vill. on germination and early growth stages of bean seeds Phaseolus vulgaris L. Dry shoots of the chickweed aqueous extracts were prepared, with which the bean seeds with coat and without coat were treated. The control group was seeds watered only with distilled water. After 7 days of the experiment, seed germination parameters, seed germination rate (SE), seed vitality index (SVI), seedling growth inhibition index (IP), fresh and dry mass values and percentage water content were determined. The experiment showed the germination capacity of bean seeds was varied, in relation to seeds from the control. With increasing concentrations of extracts, a significant reduction in the seed germination rate was observed, both for those with seed coat and without seed coat. The seed vitality index increased only in seeds with coat, and decreased in each of the applied concentrations of extracts in seeds without seed coat. The seedling growth inhibition index reached negative values in both groups of seeds tested only at a concentration of 5%. In comparison to the control, IP was positive for seedlings watered with 15% extracts. For P. vulgaris seedlings grown on 5% of extracts the highest increase in the fresh mass was observed, in relation to the value of control mass. For seedlings grown from seeds with seed coat the diferences in the dry mass values primarily were noted. The percentage of water content in bean seedlings varied depending on the group of seeds studied and the concentration of allelopathic substances in the chickweed aqueous extracts. The examined indexes of seed germination and seedling growth showed that in the case of P. vulgaris seeds without seed coat. The role of seed coat in the germination and early stages of growth of bean (Phaseolus vulgaris L.) in the presence of chickweed (Stellaria media (L.) Vill.) were more sensitive to aqueous extracts from dry shoots of S. media. Compared to the control group, in low concentrations of allelopathic substances the seeds germinated similarly to the distilled water, and at higher concentrations, the seeds germination, the seedlings growth and fresh and dry mass values were inhibited.

In laboratory bioassay, conducted to study the allelopathic influence of aqueous extracts (0, 2.5, 5.0, 7.5, 10.0% concentration) of twelve hedgerow plant species (Amorpha fruticosa L., Citrus reticulate Banco., Coronilla varia L., Eriobotrya japonica Thunb., Eulaliopsis binata Retz., Gynura bicolor DC., Hemerocallis citrine Baroni., Medicago sativa L., Morus alba L., Toona sinensis Roem., Vetiveria zizanioides L., Zanthoxylum schinifolium Sieb.) which planted in the purple hilly area of Sichuan basin in China was on the germination and seedling growth of wheat (Triticum astivum L.). At 10.0% concentration, The aqueous extracts of twelve hedgerow plant species significantly inhibited the seed germination and seedling growth of wheat except that A. fruticosa, E. binata and V. zizanioides extracts had no effect or slight inhibition on germination of wheat, but the degree of inhibition was species dependent. The allelopathic effect of aqueous extracts from hedgerow plants on wheat was maximum with G. bicolor, H. citrine, C. varia and M. sativa in terms of germination, and G. bicolor , M. sativa, M. alba and H. citrine in terms of shoot length, root length and dry weight, respecitively. The degree of inhibition increased with increase in concentration. Duplication of this work under field conditions is needed, for which the results of this study will form a valuable base.
