Purpose: to determine the relationship of the damage level of cell genetic apparatus with reproductive health disorder degree under conditions of harmful production.Materials and methods: the study involved 36 poultry farm female workers of childbearing age. The first group was made up by 22 workers of the main production. The control group included the other 14 employees who were administrative and managerial personnel. Clinical statistical study of reproductive health was carried out with simultaneous investigation of the cariological indices of buccal epithelium using a fluorescence microscope. At least 1000 cells in a monolayer were analyzed on each micropreparation. Statistical data analysis was made using Statistica 6.0.Results: in workers of the main production, there was a high spread of inflammatory gynecological diseases (90.9 % versus 57.1 % in the control group), uterine fibroids (9.1 %, versus 7.1 %). At the same time a statistically significant increase in the number of cells with micronuclei was revealed in this group by 2.6 times (p < 0.05), with protrusions by 1.8 times (p < 0.05), an increase in the total proliferation index by 2.3 times (p < 0.05), and the two-nuclear cells made the greatest contribution to these differences. The integral indicator of cytogenetic disorders of women of the 1st group was also significantly higher by 2.0 times (p < 0.05). Conclusion: negative influence of factors of poultry house production environment on cell genome stability disturbance was revealed. It has been established that the reproductive health disorder degree has a direct proportional dependence on the damage level of cell genetic apparatus.

ABSTRACT: Erythrina velutina Willd. (Fabaceae) is a Brazilian native tree with economic, ecological and social potential. The aim of this study was to evaluate isoenzyme activity and changes in DNA integrity during germination of E. velutina at different temperatures. The seeds were placed to germinate at 5, 15, 25, 35 and 45 °C, evaluating isoenzyme activity and degradation of the DNA during germination. Isoenzyme expression occurs differently for seed germination under different temperatures, with varied expression between seedlings and cotyledons. The esterase enzyme was more sensitive to express the response of the E. velutina germination seeds at different temperatures. DNA repair is more efficient during the germination of E. velutina seeds when submitted to a temperature of up to 25 °C, with damage to the genetic apparatus with an increase higher than this temperature.

At cryopreservation, the issue of preserving the integrity and cytogenetic stability of dermal papilla (DP) cells of the hair follicle is topical. The study examined the effect of various concentrations of the cryoprotectant dimethyl sulfoxide (DMSO) (5-15%), as well as its combination with 5% bovine serum albumin (BSA), on the mitotic index (MI) and the number of pathological mitoses in the cell culture of DP on the first and second passages. It has been established that the treatment of cells with DMSO in concentrations of 10% and above leads to a significant increase in the number of pathological mitosis. This is accompanied by the decrease in MI (when processing concentrations of DMSO below 10%) at the first passage or a complete cessation of proliferation at the second passage (when processing concentrations of DMSO above 10%). The combination of 5% DMSO with BSA is optimal for minimizing toxic effects on the genetic apparatus of DP cells and can be used as part of cryopreservation media.

The authors present a review of literature on experimental osteoarthritis (OA) within searching for its universal model. Classification, advantages, disadvantages, methodological inaccuracies of various authorial approaches from the point of view of conformity to human disease were identified based on analysis of the array of literature. Among techniques for OA modeling traumatic techniques were in most common use; among them "local" surgical manipulations, local devascularization, intraarticular injections of chemical-and-mechanical inducers and adjuvants which were followed up by gross changes in joint structures and, probably, severe pain. The influence of age and some genetic characteristics of cartilage structure were studied during experiments on laboratory animals with modified genetic apparatus. The methods of indirect "systemic" influence on joint structures are the most advanced in simulation of non-traumatic OA; however the etiological adequacy of proposed models is debatable. The concept of searching for the "gold standard" of the experimental model of OA formed earlier requires reconsideration because the modern identification of different OA subtypes leads to the development of new approaches on its reproducing in animals with ultimate imitation of a particular phenotype.
