A new rainfrog of the genus Pristimantis (Anura, Brachycephaloidea) from central and eastern Panama

Substantial molecular and morphological character differences lead us to the description of a new species of the genus Pristimantis from the cloud forest of Cerro Chucantí, Maje Mountains, Darien Province, as well as from several other mountain ranges in eastern and central Panama. Pristimantis gretathunbergaesp. nov. is a sister species to the allopatric P. erythropleura-penelopus group from northern Colombia with a mtDNA sequence divergence of > 4.4% at 16S and > 14.6% at COI. Its closest congener in sympatry is P. cruentus that differs by a large sequence divergence of > 9.6% in 16S mtDNA and 19.0% at COI, and from which it differs also by ventral and groin coloration, unusually prominent black eyes, a contrasting light upper lip, commonly a single conical to spine-like tubercle on the upper eyelid, and a larger head. While the habitat continuity at most sites in eastern Panama is moderate, habitats in central Panama are severely fragmented. Cerro Chucantí and the surrounding Maje Mountains are highly threatened by rapid deforestation and replaced by plantations and cattle pastures. Thus, investigations on the ecology of the new species and its population status, especially at the type locality, are highly recommended. As a flagship species, this new frog can help to preserve the Chucantí cloud forest including several recently described species known only from this isolated area in eastern Panama.

Deforestation scenarios show the importance of secondary forest for meeting Panama’s carbon goals

Context Tropical forest loss has a major impact on climate change. Secondary forest growth has potential to mitigate these impacts, but uncertainty regarding future land use, remote sensing limitations, and carbon model accuracy have inhibited understanding the range of potential future carbon dynamics. Objectives We evaluated the effects of four scenarios on carbon stocks and sequestration in a mixed-use landscape based on Recent Trends (RT), Accelerated Deforestation (AD), Grow Only (GO), and Grow Everything (GE) scenarios. Methods Working in central Panama, we coupled a 1-ha resolution LiDAR derived carbon map with a locally derived secondary forest carbon accumulation model. We used Dinamica EGO 4.0.5 to spatially simulate forest loss across the landscape based on recent deforestation rates. We used local studies of belowground, woody debris, and liana carbon to estimate ecosystem scale carbon fluxes. Results Accounting for 58.6 percent of the forest in 2020, secondary forests (< 50 years) accrue 88.9 percent of carbon in the GO scenario by 2050. RT and AD scenarios lost 36,707 and 177,035 ha of forest respectively by 2030, a carbon gain of 7.7 million Mg C (RT) and loss of 2.9 million Mg C (AD). Growing forest on all available land (GE) could achieve 56 percent of Panama’s land-based carbon sequestration goal by 2050. Conclusions Our estimates of potential carbon storage demonstrate the important contribution of secondary forests to land-based carbon sequestration in central Panama. Protecting these forests will contribute significantly to meeting Panama’s climate change mitigation goals and enhance water security.

High 3He/4He in central Panama reveals a distal connection to the Galápagos plume

It is well established that mantle plumes are the main conduits for upwelling geochemically enriched material from Earth's deep interior. The fashion and extent to which lateral flow processes at shallow depths may disperse enriched mantle material far (>1,000 km) from vertical plume conduits, however, remain poorly constrained. Here, we report He and C isotope data from 65 hydrothermal fluids from the southern Central America Margin (CAM) which reveal strikingly high 3He/4He (up to 8.9RA) in low-temperature (≤50 °C) geothermal springs of central Panama that are not associated with active volcanism. Following radiogenic correction, these data imply a mantle source 3He/4He >10.3RA (and potentially up to 26RA, similar to Galápagos hotspot lavas) markedly greater than the upper mantle range (8 ± 1RA). Lava geochemistry (Pb isotopes, Nb/U, and Ce/Pb) and geophysical constraints show that high 3He/4He values in central Panama are likely derived from the infiltration of a Galápagos plume–like mantle through a slab window that opened ∼8 Mya. Two potential transport mechanisms can explain the connection between the Galápagos plume and the slab window: 1) sublithospheric transport of Galápagos plume material channeled by lithosphere thinning along the Panama Fracture Zone or 2) active upwelling of Galápagos plume material blown by a “mantle wind” toward the CAM. We present a model of global mantle flow that supports the second mechanism, whereby most of the eastward transport of Galápagos plume material occurs in the shallow asthenosphere. These findings underscore the potential for lateral mantle flow to transport mantle geochemical heterogeneities thousands of kilometers away from plume conduits.

Chromosome-Scale Assembly of the Complete Genome Sequence of Porcisia hertigi, Isolate C119, Strain LV43

Porcisia hertigi is a parasitic kinetoplastid first isolated from porcupines ( Coendou rothschildi ) in central Panama in 1965. We present the complete genome sequence of P. hertigi , isolate C119, strain LV43, sequenced using combined short- and long-read technologies. This complete genome sequence will contribute to our knowledge of the parasitic genus Porcisia .

Rubiaceae Americanarum Magna Hama Pars XLVII. New Species and a New Combination for Rudgea in Panama and Western South America (Palicoureeae)

Review of specimens of Rudgea Salisb. (Rubiaceae, Palicoureeae) has discovered some species new to science and clarified the identity of one previously described taxon. Here we raise R. viburnoides (Cham.) Benth. subsp. megalocarpa Zappi of the western Amazon basin to species status, as R. megalocarpa (Zappi) Bruniera & C. M. Taylor. We also describe 11 new species found variously from lowland to montane elevations in Panama and western South America: R. barbosae C. M. Taylor from scattered locations on sandstone in Colombia; R. campanana C. M. Taylor from central Panama; R. cardenasii C. M. Taylor from the Caribbean area of northwestern Colombia; R. chocoana C. M. Taylor from the Pacific drainage of western Colombia and northwestern Ecuador; R. elegans C. M. Taylor from sandstone formations in the Andes of central Peru; R. homeieri C. M. Taylor from the Andean slopes of central Ecuador; R. inflata C. M. Taylor from the northwestern Amazon basin in Colombia and Brazil; R. retiniphylloides C. M. Taylor from northern to north-central Colombia; R. sanluisensis C. M. Taylor & Cogollo from the lower Río Magdalena valley of northern Colombia; R. suberosa C. M. Taylor & Bruniera from cloud forest and pajonal in the Andes from southern Ecuador through southern Peru; and R. zappiae C. M. Taylor & Bruniera from central western Ecuador.

Isolation and genetic characterization of a relapsing fever spirochete isolated from Ornithodoros puertoricensis collected in central Panama

Tick-borne relapsing fever (TBRF) spirochetes are likely an overlooked cause of disease in Latin America. In Panama, the pathogens were first reported to cause human disease in the early 1900s. Recent collections of Ornithodoros puertoricensis from human dwellings in Panama prompted our interest to determine whether spirochetes still circulate in the country. Ornithodoros puertoricensis ticks were collected at field sites around the City of Panama. In the laboratory, the ticks were determined to be infected with TBRF spirochetes by transmission to mice, and we report the laboratory isolation and genetic characterization of a species of TBRF spirochete from Panama. Since this was the first isolation of a species of TBRF spirochete from Central America, we propose to designate the bacteria as Borrelia puertoricensis sp. nov. This is consistent with TBRF spirochete species nomenclature from North America that are designated after their tick vector. These findings warrant further investigations to assess the threat B. puertoricensis sp. nov. may impose on human health.

Molecular Epidemiology of Escherichia coli Clinical Isolates from Central Panama

Escherichia coli represents one of the most common causes of community-onset and nosocomial infections. Strains carrying extended spectrum β-lactamases (ESBL) are a serious public health problem. In Central America we have not found studies reporting the molecular epidemiology of E. coli strains implicated in local infections, so we conducted this study to fill that gap. Materials and Methods: We report on an epidemiological study in two reference hospitals from central Panama, identifying the susceptibility profile, associated risk factors, and molecular typing of E. coli strains isolated between November 2018 and November 2019 using Pasteur’s Multilocus Sequence Typing (MLST) scheme. Results: A total of 30 E. coli isolates with antimicrobial resistance were analyzed, 70% of which came from inpatients and 30% from outpatients (p < 0.001). Two-thirds of the samples came from urine cultures. Forty-three percent of the strains were ESBL producers and 77% were resistant to ciprofloxacin. We identified 10 different sequence types (STs) with 30% of the ESBL strains identified as ST43, which corresponds to ST131 of the Achtman MLST scheme—the E. coli pandemic clone. Thirty-eight percent of the E. coli strains with the ESBL phenotype carried CTX-M-15. Conclusions: To the best of our knowledge, this is the first report confirming the presence of the pandemic E. coli clone ST43/ST131 harboring CTX-M-15 in Central American inpatients and outpatients. This E. coli strain is an important antimicrobial-resistant organism of public health concern, with potential challenges to treat infections in Panama and, perhaps, the rest of Central America.

Divergent regional evolutionary histories of a devastating global amphibian pathogen

Emerging infectious diseases are a pressing threat to global biological diversity. Increased incidence and severity of novel pathogens underscores the need for methodological advances to understand pathogen emergence and spread. Here, we use genetic epidemiology to test, and challenge, key hypotheses about a devastating zoonotic disease impacting amphibians globally. Using an amplicon-based sequencing method and non-invasive samples we retrospectively explore the history of the fungal pathogen Batrachochytrium dendrobatidis ( Bd ) in two emblematic amphibian systems: the Sierra Nevada of California and Central Panama. The hypothesis in both regions is the hypervirulent Global Panzootic Lineage of Bd ( Bd GPL) was recently introduced and spread rapidly in a wave-like pattern. Our data challenge this hypothesis by demonstrating similar epizootic signatures can have radically different underlying evolutionary histories. In Central Panama, our genetic data confirm a recent and rapid pathogen spread. However, Bd GPL in the Sierra Nevada has remarkable spatial structuring, high genetic diversity and a relatively older history inferred from time-dated phylogenies. Thus, this deadly pathogen lineage may have a longer history in some regions than assumed, providing insights into its origin and spread. Overall, our results highlight the importance of integrating observed wildlife die-offs with genetic data to more accurately reconstruct pathogen outbreaks.

Dengue and hantavirus coinfection with good outcomes: a clinical case from Panama

Dengue and hantavirus are endemic in central Panama. We present a case report of a patient coinfected with dengue and hantavirus who presented overlapping clinical manifestations of both infections. The patient did not require intensive care—as it is commonplace during hantavirus pulmonary syndrome—and he had a good outcome. Patient’s care was positively impacted due to correct diagnosis of coinfection. This case highlights that in endemic areas, coinfection with dengue virus and hantavirus should be suspected. To the best of our knowledge, this case is the first documented case of coinfection with dengue virus and hantavirus in Central America.
