Introduction: White pomegranate (P. granatum) is a species of the Punicaceae which is thought as hepatoprotector based on its antioxidant secondary metabolite compoundst. Aims: The purpose of study was to determine the hepatoprotector activity of white pomegranate fruit and seeds ethanol extract on male white rats Wistar strain. Methods: This research is laboratory experimental which rats were grouped into 5 groups randomly. Normal, negative (given Carboxymethylcellulose sodium 1% orally), test dose 1,2,3 (given extract at a dose of 50 mg /200 g bodyweight of rat, 100 mg/200 g bodyweight of rat and 200 mg/200 g bodyweight of rat in Carboxymethylcellulose sodium 1% orally). Negative, test dose 1,2,3 induced with paracetamol 180 mg/200 g bodyweight of rat. Results: From the statistical analysis, it was found that a significant difference between the negative with test dose 1, 2, and 3. The best hepatoprotector activity was produced by the test dose 3 with the percentage reduction in AST levels 91.62% and ALT 90.20%.

ABSTRACTBuah pepino memiliki kandungan seyawa yaitu alkaloid, flavonoid, vitamin C yang dapat berkhasiat sebagai hepatoprotektor. Penelitian ini bertujuan untuk mengetahui aktivitas hepatoprotektor ekstrak etanol buah pepino (Solanum muricatum) terhadap tikus putih jantan  yang diinduksikan ccl4 dengan dosis ekstrak dibagi menjadi tiga kelompok yaitu 100 mg/kgBB, 200 mg/kgBB dan 400 mg/kgBB. Penelitian ini merupakan jenis eksperimental menggunakan rancangan  penelitian True Experimental design. Pengujian aktivitas hepatoprotektor dengan parameter acuan nya adalah kadar SGOT dan SGPT.  Analisis hasil pengukuran SGOT dan SGPT dihitung dengan menggunakan ANOVA satu arah.  Hasil pengukuran  kadar SGPT dan SGOT dengan dosis 100 mg/kgBB, 200 mg/kgBB dan 400 mg/kg BB terjadi penurunan yang berbeda-beda .Pada SGOT penurunan tertinggi kadar terdapat pada ekstrak etanol buah pepino 100 mg/kgBB diikuti ekstrak etanol buah pepino 200 mg/kgBB lalu kelompok ekstrak etanol buah pepino 400 mg/kgBB kemudian kelompok positif dan terakhir kelompok negative .Dan pada SGPT penurunan kadar SGPT tertinggi pada kelompok control positif diikuti kelomok ekstrak etanol  buah pepino dengan konsentrasi 100 mg/kgBB, lalu ekstrak etanol buah pepino dengan konsentrasi 200 mg/kgBB, kemudian konsentrasi 400 mg/kgBB dan terakhir kelompok control negative. Berdasarkan penelitian yang telah dilakukan, dapat disimpulkan bahwa buah pepino (Solanum muricatum) memiliki aktivitas hepatoprotektor pada konsentrasi 100 mg/kgBB pada kadar SGPT dan SGOT . Pepino fruit contains compounds such as alkaloids, flavonoids, vitamin C which can be efficacious as a hepatoprotector. This study aims to study the activity of the ethanol hepatoprotector extract of pepino fruit (Solanum muricatum) against male white rats induced by ccl4 with extract extract into three groups namely 100 mg / kg, 200 mg / kg and 400 mg / kg. This research is an experimental type using a true experimental design research design. Testing of hepatoprotector activity with reference parameters is SGOT and SGPT levels. Analysis of the results of the SGOT and SGPT measurements were calculated using one-way ANOVA. The results of the measurement of SGPT and SGOT levels with a dose of 100 mg / kgBW, 200 mg / kgBW and 400 mg / kgBW were different. 200 mg / kgBB then pepino fruit ethanol extract group 400 mg / kgBB then the positive group and finally the negative group. pepino with a concentration of 200 mg / kg, then a concentration of 400 mg / kg and finally the negative control group. Based on research that has been done, it can be concluded that the pepino fruit (Solanum muricatum) has hepatoprotector activity at a concentration of 100 mg / kgBW at SGPT and SGOT levels. Keywords : CCL4, Hepatoprotektor, Solanum muricatum.

Background: Drug induced liver injury (DILI) is known as the damage of liver cells due to chronic administrations of drug. The chronic administration of paracetamol could be trigger the damage of liver cells.The hepatoprotector agents are still limited worldwide.  Gambier(Uncaria gambir Roxb) is an Indonesia’traditional medicine which have many benefits as antioxidant, antiseptic, antidiarrhoea, etc  that commonly used in society.  Method: The present study was conducted to investigate the hepatoprotector effect ofgambier in wistar rats induced by paracetamol.  The wistar rats were divided into seven groups and received the treatment orally for 12 days. Group I (aquadest), II(curcuma,400 mg/kgBW),III (gambier,26 mg/200gr), IV(gambier, 53 mg/200gr), V(gambier,106 mg/200gr),VI(gambier,212 mg/200gr) and VII(gambier,424mg/200gr). Termination, blood and liver organ collection were done after all group induced by paracetamol for two days. Histopatology changes of liver were examined using Hematoxycilline (HE) staining. AST and ALT levels were analyzed. Results: There were significant differentiation of AST levels among the groups, especially between group I and group IV and between group II and group IV. The ALT levels were statistically significant between group II and group V using Mann-Whitney test (p<0,05). In histopatology examination, there were significant differentiation between group I with another group, not only group II but also group III-VII (p<0,05). In the treatment group, group III and IV had been showed the improvement of liver cells damage than group I by using One-way Annova, post hoc Bonferroni (p<0,05). Conclusion: Uncaria gambir Roxb has hepatoprotector activity start at dose 53 mg/200grBWin rats.  The hepatoprotector activity was not superior than curcuma.  Keywords: hepatoprotector, Uncaria gambir Roxb, AST,ALT,histopatology

Many plants have been reported to possess hepatoprotective activity. The mechanism has not yet been revealed, but it was predicted due to the antioxidants that could prevent the formation of free radicals produced by hepatotoxins. Of the 30 plants studied, Crocus sativus gave the largest hepatoprotector activity at a dose of 20 mg / kg, followed by Uncaria gambier Roxb. with a dose of 30 mg / kg and Melastoma malabathricum Leaf at a dose of 50 mg / kg. Keywords: Liver, CYP450, antioxidant, free radicals
