Numerous studies have demonstrated that food shapes the structure and composition of the host’s oral and gut microbiota. The disorder of oral and gut microbiota may trigger various host diseases. Here, we collected oral and gut samples from wild water monitor lizards (Varanus salvator) and their captive conspecifics fed with bullfrogs, eggs, and depilated chicken, aiming to examine dietary correlates of oral and gut microbiota. We used the 16S rRNA gene sequencing technology to analyze the composition of the microbiota. Proteobacteria and Bacteroidota were the dominant phyla in the oral microbiota, and so were in the gut microbiota. The alpha diversity of microbiota was significantly higher in the gut than in the oral cavity, and the alpha diversity of oral microbiota was higher in captive lizards than in wild conspecifics. Comparing the relative abundance of oral and gut bacteria and their gene functions, differences among different animal groups presumably resulted from human contact in artificial breeding environments and complex food processing. Differences in gene function might be related to the absolute number and/or the taxonomic abundance of oral and gut microorganisms in the wild and the water environment. This study provides not only basic information about the oral and gut microbiota of captive and wild water monitor lizards, but also an inference that feeding on frogs and aquatic products and reducing human exposure help water monitor lizards maintain a microbiota similar to that in the wild environment.

Monitor lizards are acclimatized to a variety of environments. Most of the monitor species are terrestrial, although there are arboreal and semiaquatic monitors. Such accommodation requires unique cellular structure and regulatory devices in various organs, particularly their lungs. This study aimed to report the pulmonary guardians and special regulatory devices that may guard and promote the function of the lungs of the Nile monitor lizards (Varanus niloticus). Specially structured vessels were recorded in the pulmonary tissue involving atypical glomus vessels, vessels with variable wall thickness, and a venule with specialized internal elastic membrane. Moreover, numerous lung resident guardians could be identified including both alveolar and interstitial macrophages, dendritic cells, mast cells, and B- and T-lymphocytes. Pericytes were demonstrated surrounding the capillary endothelium with a characteristic direct hetero-cellular junction with telocytes. Telocytes established a microenvironment through an indirect hetero-cellular junction with the interstitial macrophage, dendritic cells, and pneumocyte type II. Collectively, these data indicate a significant role played by the specially structured vessels and the resident immune cells in guarding the pulmonary tissue of the Nile monitor lizards and promoting its function. Telocytes are suggested to play a key role in angiogenesis and cellular communication to promote the function of the immune cells.

Environmental enrichment has been shown to enhance the behavioural repertoire and reduce the occurrence of abnormal behaviours, particularly in zoo-housed mammals. However, evidence of its effectiveness in reptiles is lacking. Previously, it was believed that reptiles lacked the cognitive sophistication to benefit from enrichment provision, but studies have demonstrated instances of improved longevity, physical condition and problem-solving behaviour as a result of enhancing husbandry routines. In this study, we evaluate the effectiveness of food- and scent-based enrichment for three varanid species (Komodo dragon, emerald tree monitor lizard and crocodile monitor). Scent piles, scent trails and hanging feeders resulted in a significant increase in exploratory behaviour, with engagement diminishing ≤330 min post provision. The provision of food- versus scent-based enrichment did not result in differences in enrichment engagement across the three species, suggesting that scent is just as effective in increasing natural behaviours. Enhancing the environment in which zoo animals reside is important for their health and wellbeing and also provides visitors with the opportunity to observe naturalistic behaviours. For little known and understudied species such as varanids, evidence of successful (and even unsuccessful) husbandry and management practice is vital for advancing best practice in the zoo industry.

Abstract Background African trypanosomiasis, caused by protozoa of the genus Trypanosoma and transmitted by the tsetse fly, is a serious parasitic disease of humans and animals. Reliable data on the vector distribution, feeding preference and the trypanosome species they carry is pertinent to planning sustainable control strategies. Methodology We deployed 109 biconical traps in 10 villages in two districts of northwestern Uganda to obtain information on the apparent density, trypanosome infection status and blood meal sources of tsetse flies. A subset (272) of the collected samples was analyzed for detection of trypanosomes species and sub-species using a nested PCR protocol based on primers amplifying the Internal Transcribed Spacer (ITS) region of ribosomal DNA. 34 blood-engorged adult tsetse midguts were analyzed for blood meal sources by sequencing of the mitochondrial cytochrome c oxidase 1 (COI) and cytochrome b (cytb) genes. Results We captured a total of 622 Glossina fuscipes fuscipes tsetse flies (269 males and 353 females) in the two districts with apparent density (AD) ranging from 0.6 to 3.7 flies/trap/day (FTD). 10.7% (29/272) of the flies were infected with one or more trypanosome species. Infection rate was not significantly associated with district of origin (Generalized linear model (GLM), χ2 = 0.018, P = 0.895, df = 1, n = 272) and sex of the fly (χ2 = 1.723, P = 0.189, df = 1, n = 272). However, trypanosome infection was highly significantly associated with the fly’s age based on wing fray category (χ2 = 22.374, P < 0.001, df = 1, n = 272), being higher among the very old than the young tsetse. Nested PCR revealed several species of trypanosomes: T. vivax (6.62%), T. congolense (2.57%), T. brucei and T. simiae each at 0.73%. Blood meal analyses revealed five principal vertebrate hosts, namely, cattle (Bos taurus), humans (Homo sapiens), Nile monitor lizard (Varanus niloticus), African mud turtle (Pelusios chapini) and the African Savanna elephant (Loxodonta africana). Conclusion We found an infection rate of 10.8% in the tsetse sampled, with all infections attributed to trypanosome species that are causative agents for AAT. However, more verification of this finding using large-scale passive and active screening of human and tsetse samples should be done. Cattle and humans appear to be the most important tsetse hosts in the region and should be considered in the design of control interventions.

As indicated by archeological and literary sources, the pot drum has been a membranaphone of popular use since the Anuradhapura kingdom. It has been seen in various forms as Kumbha beraya, kala beraya, bummadiya or bimbisaka. This drum, being made out of clay in the shape of a gourd with an elongated neck or in that of a clay pot had a stretched skin of goat hide, monitor lizard hide or monkey hide. The use of these raw materials and how it brings harmony between the pottery industry and villagers in the processes of making the bummadiya is of importance. Through this research, understanding the Sri Lankan pot drum as a primary musical instrument and studying its historical and cultural aspects served as a foundation. After which, the objectives of identifying the role the pot-drum plays in sustainability were discussed. Finally, an investigation into an evolutionary stage of the pot drum in modern times, the thunpata beraya, was investigated. While this is a qualitative research, literary and archeological sources were used to collect data through discussions and field visits. The cultural texts and articles written throughout the ages on the Sri Lankan pot drum were used as secondary sources. Judgment sampling was the background behind the selection of data sources. Audio-visual methods were also used in collecting data. The human activity of using a material such as clay to express creativity in the form of music is of significance here.

Reptile sex determination is attracting much attention because the great diversity of sex-determination and dosage compensation mechanisms permits us to approach fundamental questions about sex chromosome turnover and evolution. However, reptile sex chromosome variation remains largely uncharacterized and no reptile master sex determination genes have yet been identified. Here we describe a powerful and cost-effective chromosomics approach, combining probes generated from the microdissected sex chromosomes with transcriptome sequencing to explore this diversity in non-model Australian reptiles with heteromorphic or cryptic sex chromosomes. We tested the pipeline on a turtle, a gecko, and a worm-lizard, and we also identified sequences located on sex chromosomes in a monitor lizard using linked-read sequencing. Genes identified on sex chromosomes were compared to the chicken genome to identify homologous regions among the four species. We identified candidate sex determining genes within these regions, including conserved vertebrate sex-determining genes pdgfa, pdgfra amh and wt1, and demonstrated their testis or ovary-specific expression. All four species showed gene-by-gene rather than chromosome-wide dosage compensation. Our results imply that reptile sex chromosomes originated by the independent acquisition of sex-determining genes on different autosomes, as well as translocations between different ancestral macro- and micro-chromosomes. We discuss the evolutionary drivers of the slow differentiation, but rapid turnover, of reptile sex chromosomes.

In Borneo, oil palm plantations have replaced much of natural resources, where generalist species tend to be the principal beneficiaries, due to the abundant food provided by oil palm plantations. Here, we analyse the distribution of the Asian water monitor lizard (Varanus salvator) population within an oil palm-dominated landscape in the Kinabatangan floodplain, Malaysian Borneo. By using mark-recapture methods we estimated its population size, survival, and growth in forest and plantation habitats. We compared body measurements (i.e. body weight and body length) of individuals living in forest and oil palm habitats as proxy for the population’s health status, and used general least squares estimation models to evaluate its response to highly fragmented landscapes in the absence of intensive hunting pressures. Contrary to previous studies, the abundance of lizards was higher in the forest than in oil palm plantations. Recruitment rates were also higher in the forest, suggesting that these areas may function as a source of new individuals into the landscape. While there were no morphometric differences among plantation sites, we found significant differences among forested areas, where larger lizards were found inhabiting forest adjacent to oil palm plantations. Although abundant in food resources, the limited availability of refugia in oil palm plantations may intensify intra-specific encounters and competition, altering the body size distribution in plantation populations, contrary to what happens in the forest. We conclude that large patches of forest, around and within oil palm plantations, are essential for the dynamics of the monitor lizard population in the Kinabatangan floodplain, as well as a potential source of individuals to the landscape. We recommend assessing this effect in other generalist species, as well as the impact on the prey communities, especially to reinforce the establishment of buffer zones and corridors as a conservation strategy within plantations.

Background and Aim: The high prevalence of sparganosis has been reported globally, especially in Asian countries where the majority of individuals consume raw meat from wild-caught reptiles. In Indonesia, similar cases regarding the high prevalence of sparganosis were recorded from wild reptiles such as snakes that utilized for culinary purposes, whereas, there are no data regarding other species such as water monitor lizard (Varanus salvator), which also provided as daily culinary with a high number of trades. Therefore, this study aims to investigate the prevalence of sparganosis in water monitor lizard (V. salvator), which is frequently utilized as culinary products in East Java Province, Indonesia. Materials and Methods: A total of 313 living wild-caught and captive-bred Asian water monitor lizards were collected from the reptile markets and breeders. All samples were euthanized and observed for the presence of plerocercoid. Identification of the plerocercoid as larval infective stage was made using carmine staining method. Results: The total prevalence of sparganosis was recorded at 69.64%. A total of 393 plerocercoids were collected in which divided 280 (71.24%) infecting muscles and 113 (28.75%) located in subcutaneous tissues. Conclusion: To the best of our knowledge, this study not only recorded as the first evidence but also confirms the role of monitor lizards as sparganosis transmitters in Asia and reveals additional routes of sparganosis transmission in Indonesian reptiles. Local conservation laws should be strengthened to effectively control or ban wildlife trade in traditional markets. Moreover, public awareness regarding sparganosis as a neglected zoonotic disease should be applied to prevent disease transmission in Indonesia.
