Changes in the abundance of individual proteins in the proteome can be elicited by modulation of protein synthesis (the rate of input of newly synthesized proteins into the protein pool) or degradation (the rate of removal of protein molecules from the pool). A full understanding of proteome changes therefore requires a definition of the roles of these two processes in proteostasis, collectively known as protein turnover. Because protein turnover occurs even in the absence of overt changes in pool abundance, turnover measurements necessitate monitoring the flux of stable isotope labeled precursors through the protein pool such as labeled amino acids or metabolic precursors such as ammonium chloride or heavy water. In cells in culture, the ability to manipulate precursor pools by rapid medium changes is simple, but for more complex systems such as intact animals, the approach becomes more convoluted. Individual methods bring specific complications, and the suitability of different methods has not been comprehensively explored. In this study we compare the turnover rates of proteins across four mouse tissues, obtained from the same inbred mouse strain maintained under identical husbandry conditions, measured using either [13C6]lysine or [2H2]O as the labeling precursor. We show that for long-lived proteins, the two approaches yield essentially identical measures of the first order rate constant for degradation. For short-lived proteins, there is a need to compensate for the slower equilibration of lysine through the precursor pools. We evaluate different approaches to provide that compensation. We conclude that both labels are suitable, but careful determination of precursor enrichment kinetics in amino acid labeling is critical and has a considerable influence on the numerical values of the derived protein turnover rates.

The rapid development of drug resistant micro-organisms is a challenge to the mankind. Nano formulated compounds have proved to be effective strategy to combat bacterial drug resistance. Currently nanoparticulate systems such as nanoantibiotics are getting major attention due to their low inherent toxicity, biodegradability, bioincompatibility and tuneable mechanical characteristics. Nano formulated antibiotics are generally obtained by emulsification and gelification techniques. The effective uses of polymers in encapsulation of antibiotics show enhancement of the efficacy of antibiotics. Combined with techniques like diffraction laser spectroscopy (DLS), electron microscopy (EM) and atomic force microscopy (AFM), morphological research of nanoformulated antibiotics are conducted. The detailed study of the polymers used in the preparation of antibiotics nanoparticles as well as their impact on interactions is done by bio-analytical techniques. Antibiotics attached to nanoparticles can avoid the action of enzymes produced by drug resistant bacteria.  Nano antibiotics show higher efficacy and bioavailability so a lot of new formulations using nano methods can be developed with the help of bioanalytical techniques. The development as well as the estimation of antibiotics prepared as nano-formulations as per the recent advanced techniques is illustrated in this review.

The substantiation of the methodological viability and hermeneutic potential of the category conflict is given. The role of conflict as a category of world modeling in fiction is pointed out. Consideration of the conflict in the aspect of dialogism is suggested. Conflict in fiction is a special kind of communicative act arising from the antagonism of misunderstanding of the opposing sides, but providing a new level of understanding to the perceiving consciousness of the reader, making it intellectually armed (R. Jacobson) in the face of conflicting structures. Analytical strategies for the study of conflict are presented in the semantic aspect (conflict as an object of artistic depiction) and the aspect of structure (conflict as a fundamental structural principle). It is proved that the dual nature of the conflict presupposes taking into account in research practice the possibilities of textocentric and anthropological approaches to the study of conflict, understood as antagonistic relations of oppositional units. The development of epistemology, axiology and poetics of conflict in fiction is seen in the way of studying all levels of conflict dialogue.

Abstract BackgroundEpidemiologists need tools to measure effects of gender, a complex concept originating in the humanities and social sciences which is not easily operationalized in the discipline. MethodsWe conducted a conceptual analysis and applied causal and mediation analysis methodology to standard questions in order to propose a methodologically appropriate strategy for measuring sex and gender effects in health.ResultsWe define gender as a set of norms prescribed to individuals according to their attributed-at-birth sex. Gender pressure creates a systemic gap, at population level, in behaviors, activities, experiences, etc. between men and women. A pragmatic individual measure of gender would correspond to the level at which an individual complies with a set of elements constituting femininity or masculinity in a given population, place and time. However, defining and measuring gender is not sufficient to isolate the effects of sex and gender on a health outcome. We should also think in terms of pathways to define appropriate analysis strategies. Gender could also be examined as a mechanism rather than through its realization in the individual, by considering it as an interaction between sex and environment. ConclusionsBoth analytical strategies have limitations relative to the impossibility of reducing a complex concept to a single or a few measures, and of capturing the entire effect of the phenomenon. However, these strategies could lead to more accurate and rigorous analyses of the mechanisms underlying health differences between men and women, and ultimately limit the sex and/or gender bias encountered in epidemiological and clinical research studies.

There is a tight nexus between visual literacy and textbook picture representations. This is of paramount importance when textbooks in general and ELT textbooks, in particular, are under question. To conduct a visual and verbal discourse analysis based on modes of communication, ELT textbook pictures were analyzed under the assumption that visual and verbal discourse interacts with reflected modes of communication. To this end, 50 ELT textbook pictures were used as the corpus and analyzed according to KvL's (2006) visual images analytical strategies in multimodal texts and Halliday’s (1985) transitivity system for verbal analysis of textbook pictures. The analysis of multimodal resources revealed that the analyzed visual images were used to represent non-human images; close-up images, frontal images, left-right compositions were the most frequent visual modes in the selected pictures. In the case of verbal mode, the relational main-type and verbal minor-type level with 39% and 2% were the most and least frequent verbal strategies, respectively. The findings might have significant theoretical and pedagogical implications for scholars, L2 teachers, and ELT textbook designers to consider the potential of using multimodal resources for non-pedagogical purposes while integrating textbook visual images and verbal strategies to create meanings.
