Abstract Background The aim of this study was to provide an information about the homogeneity on the level of enterokinase productivity in P. pastoris depending on different suppliers of the media components. Results In previous studies, we performed the optimisation process for the production of enterokinase by improving the fermentation process. Enterokinase is the ideal enzyme for removing fusion partners from target recombinant proteins. In this study, we focused our optimization efforts on the sources of cultivation media components. YPD media components were chosen as variables for these experiments. Several suppliers for particular components were combined and the optimisation procedure was performed in 24-well plates. Peptone had the highest impact on enterokinase production, where the difference between the best and worst results was threefold. The least effect on the production level was recorded for yeast extract with a 1.5 fold difference. The worst combination of media components had a activity of only 0.15 U/ml and the best combination had the activity of 0.88 U/ml, i.e., a 5.87 fold difference. A substantially higher impact on the production level of enterokinase was observed during fermentation in two selected media combinations, where the difference was almost 21-fold. Conclusions Results demonstrated in the present study show that the media components from different suppliers have high impact on enterokinase productivity and also provide the hypothesis that the optimization process should be multidimensional and for achieving best results it is important to perform massive process also in terms of the particular media component supplier .

Purpose – The purpose of this paper is to develop alternative analytical measures for the degree of operating leverage (DOL) that reflect the impact of uncertain demand shocks in the product's market on optimal production levels, sales and profits of the firm. Design/methodology/approach – The elasticity measures are constructed according to a theoretical formulation of optimal production level that corresponds to demand shocks for given predetermined levels of fixed cost. Findings – The paper suggests two main findings. First, the analytical marginal DOL is at least twice the traditional DOL depending on the structure of the shock, the production function and demand's elasticity. The traditional DOL is equal to the measure only when large-scale negative demand prompts the firm to abandon production. Second, the paper also provides an analytical measure of DOL in terms of elasticity of profit to sales rather than to production level. Both theoretically and empirically elasticity of profit to sales can be better measured and better reflects risk. Research limitations/implications – This paper should be extended to encompass multiple shocks on demand and supply while investigating the empirical multi variants distribution of the shocks. Practical implications – The model can be used by managers who are well informed about the fixed and variable costs of their firm. The model determines the mean profit- risk trade off which is an important factor in all investment decision problems. Originality/value – Surprisingly and according to the best knowledge, this paper is the first attempt in the literature for alternative analytical DOLs’ formulations that is coherent with basic economic theories of optimal production level under risk.

Resonance inspection uses the natural acoustic resonances of a part to identify anomalous parts. Modern instrumentation can measure the many resonant frequencies rapidly and accurately. Sophisticated sorting algorithms trained on sets of good and anomalous parts can rapidly and reliably inspect and sort parts. This paper aims at using finite-element-based modal analysis to put resonance inspection on a more quantitative basis. A production-level automotive steering knuckle is used as the example part for our study. First, the resonance frequency spectra for the knuckle are measured with two different experimental techniques. Next, scanning laser vibrometry is used to determine the mode shape corresponding to each resonance. The material properties including anisotropy are next measured to high accuracy using resonance spectroscopy on cuboids cut from the part. Then, finite element model (FEM) of the knuckle is generated by meshing the actual part geometry obtained with computed tomography (CT). The resonance frequencies and mode shapes are next predicted with a natural frequency extraction analysis after extensive mesh size sensitivity study. The good comparison between the predicted and the experimentally measured resonance spectra indicate that finite-element-based modal analyses have the potential to be a powerful tool in shortening the training process and improving the accuracy of the resonance inspection process for a complex, production level part. The finite element based analysis can also provide a means to computationally test the sensitivity of the frequencies to various possible defects such as porosity or oxide inclusions especially in the high stress regions that the part will experience in service.

The present study sheds some light on the conversation as an optimal form of communication. Also, it tries to illustrate Iraqi EFL learners’ aptitude for producing conversation and specifying the errors types committed by them in its progression. To accomplish these purposes, it is hypothesized that the learners may face formidable problems in sharing their experiences with each other and are unable to keep a conversation going. In the face of such problems, a sample of (50) learners is selected to a diagnostic test administration. The subjects are at the Fourth year, Department of English, College of Education for Humanities, University of Babylon. Data analysis has proved that the leaners encounter more difficulty on the production level than the recognition one. This manifestation is manifested itself in the number of the learners’ correct responses as compared with their incorrect ones on both levels.

Abstrak : Harga jual merupakan salah satu faktor yang sangat berpengaruh terhadap petani, salah satunya yaitu tingkat pendapatan yang di peroleh oleh para petani, yang selanjutnya akan berpengaruh terhadap motivasi dan produktivitas kerja dari para petani. Saluran pemasaran yang terlalu panjang, dan kebijakan harga yang tidak memihak pada petani mengakibatkan keuntungan yang di peroleh oleh petani sangat sedikit. Tujuan dari penelitian ini adalah untuk mengetahui faktor- faktor yang mempengaruhi harga jual kedelai tingkat petani di Kecamatan Peudada Kabupaten Bireuen. Berdasarkan hal tersebut, penelitian ini dilakukan untuk mengetahui faktor-faktor apa saja yang berpengaruh terhadap harga jual kedelai tingkat petani. Penelitian ini dilakukan di Kecamatan Peudada Kabupaten Bireuen, Lokasi penelitian dipilih secara sengaja (purposive). Ruang lingkup penelitian ini terbatas pada faktor tingkat produksi, kualitas, penanganan pascapanen dan saluran distribusi pemasaran. Populasi dalam penelitian ini sebanyak 30 orang yang diambil dari tiga desa tersebut yaitu desa Cot Kruet, Pinto Rimba, Ara Bungong. Sedangkan besarnya sampel yang digunakan dalam penelitian ini adalah 15% dari total populasi penelitian. Model analisis yang digunakan adalah fungsi linier berganda. Hasil analisis koefisiensi determinasi ( R² ) diperoleh nilai sebesar 0,827 yang berarti bahwa tingkat produksi, kualitas, penanganan pascapanen, dan saluran ditribusi pemasaran mempengaruhi harga jual kedelai sebesar 82,7%, sedangkan sisanya 17,3% dipengaruhi oleh faktor lain. Hasil analisis secara serempak (uji-f) bahwa tingkat produksi, Kualitas, Penanganan pascapanen, dan saluran distribusi pemasaran berpengaruh nyata terhapap harga jual kedelai tingkat petani di Kecamatan Peudada Kabupaten Bireuen. Analisis secara parsial (uji-t) menyatakan bahwa faktor kualitas dan penanganan pascapanen yang berpengaruh nyata terhadap harga jual kedelai tingkat petani.Analysis Factors Affecting The Selling Price Soybean Level Farmer in Central Production in Kecamatan Peudada Kabupaten Bireuen.Abstract : The selling price is one factor that would influence the farmers, one of them is income levels in get the farmers, which later will have an influence to motivation and labor productivity from farmers. Marketing outlets is too long, and policy the price of being impartial the farmers resulting in an advantage in get by farmers very few. The purpose of this study is to find of factors affect the selling price of soybean the farm gate in Kecamatan Peudada Kabupaten bireuen. Based on it, the study is done to find out what factors had an influence on the selling price of soybean farm gate. The study is done in Kecamatan Peudada Kabupaten Bireuen, the research areas was selected deliberately (purposive). Scope this research limited to factors production level, the quality of, post-harvest handling of crops and distribution channels marketing. Population in this study as many as 30 people who is taken from the three the village the village Cot Kruet, Pinto Rimba, Ara Bungong. The amount of sample used in this research was 15 % of the total number of research. Model the analysis used is a function linear multiple.The results of the analysis koefisiensi determination ( R2 ) obtained value of 0,827 which means that production level, the quality, post-harvest handling of crops, and channels ditribusi marketing affect the selling price soybean of 82.7 %, while the rest 17.3 % influenced by other factors. The results of the analysis in unison ( uji-f ) that production level, the quality, post-harvest handling of crops, marketing and distribution channels may had have real impact against the selling price soybean the farm gate in Kecamatan Peudada Kabupaten Bireuen. Analysis in partial ( uji-t ) said that factors the quality and post-harvest handling of crops who had have real impact on the selling price soybean the farm gate.

Concerning the red clover breeding, it is essential to choose as genitors those varieties that possess good characteristics. In order to identify potential genitors, two red clover varieties were studied at Cluj-Napoca: Roxana, diploid variety and Napoca-tetra, tetraploid variety. The analyzed traits comprised the number of capitula/plant, number of seeds/capitulum and the binding process, under the aspect of their contribution to the seed crop and the interaction between those components, in order to provide important clues about di-and tetraploid red clover seed production, for plant breeding. The highest variability (s%=30) presented regarding the number of flowers/capitulum, diploid variety comparatively with a lower variability (s%=26) regarding the number of flowers number/capitulum, tetraploid varieties. Several traits were compared between di-and tetraploid red clover varieties in order to find genitors which can be used for creating new genotypes with high seed production level.

Abstract Clostridium butyricum is a spore-forming probiotic existing in the intestines of humans and animals which can promote the enhancement of beneficial bacteria and maintain intestinal microecological balance. However, it is difficult to improve the production level of C. butyricum by conventional fermentation process. In this study, a co-fermentation process of C. butyricum DL-1 and Bacillus coagulans ZC2-1 was established to improve the viable counts and spore yield of C. butyricum, and the formula of coculture medium was optimized by flask fermentation. The results showed that the optimum medium composition is bran 10 g/L, corn steep powder 15 g/L, peptone 15 g/L, K2HPO4 1 g/L and MnSO4 0.5 g/L. Cultured stationarily in the optimal medium for 36 h, the number of viable bacteria of C. butyricum DL-1 reached 1.5×108 CFU/mL and the spore forming rate was 92.6%. The results revealed an economical and effective medium composition for the coculture of C. butyricum and B. coagulans. The co-fermentation process established in this study provides a new fermentation mode for the industrial production of other absolute anerobic bacteria.
