SummaryThe uptake and binding of antimitotic alkaloid colchicine has been demonstrated in washed preparations of human platelets. A silicone oil technique was adapted so that both uptake and binding of 14C-colchicine were examined in the same platelet preparations. The time dependence and amount of colchicine taken up and bound by different platelet preparations during a 90 to 120 min incubation period were highly reproducible. Both colchicine uptake and binding by intact platelets, and colchicine binding by preparations of lysed platelets were specific and temperature dependent. Colchicine uptake was slowly reversible. Magnesium and GTP enhanced colchicine binding by lysed platelet preparations but calcium decreased binding.Exposure of platelets to either cold (4° C) or to thrombin, which disrupt platelet microtubules, produced significant increases in colchicine uptake and binding. The thrombin effect was maximal at 37° C and resulted in a greater increase in uptake and binding than that produced by either cold treatment alone or, by cold treatment followed by incubation with thrombin at 37° C. The amount of increase in uptake and binding produced by thrombin was independent of both thrombin (1–5 Units/109 platelets) and colchicine concentrations (1–50 × 10−6M).It is postulated that thrombin may initiate the formation, or make available, colchicine binding sites (microtubule subunits) within platelets.

AbstractObjective:To search the English language literature for original research addressing the effect of cryotherapy on motor performance and recovery.Data Sources:We searched MEDLINE, the Physiotherapy Evidence Database, SPORT Discus, Pubmed, and the Cochrane Reviews database, from 1976 to 2009 to identify randomized clinical trials of cryotherapy, systematic reviews, original articles and methods of cryotherapy. Key words used were cryotherapy, return to participation, cold treatment, ice, injury.Data Synthesis:Brief review including assessment of cryotherapy as a tool of performance and a recovery method.Conclusions:Most studies suggest that a short rewarming time would be beneficial (a couple minutes), which is very reasonable in sports. Also, cooling techniques differ in its result accordingly to the procedures and objectives used. Finally, the type of tissue cooled plays a large role (ie. Joint vs. Muscle).

Seed banks of two seashore meadows were studied on the west coast of Finland (latitude 61°30'-61°33'N, longitude 21°28'-21°41'E). Samples were taken in June to a depth of 10 cm in the geolittoral zone of the grazed and ungrazed transects. The grazed samples were halved lengthwise: one half was grown immediately, the other after cold treatment. One third of the all samples was treated as controls, one third was watered with brackish water, and one third was given a pesticide treatment. Altogether, 13 926 seedlings germinated and 25 species were identified (three annuals, two biennials, and the rest perennials). Most seedlings were perennial monocots, with Juncus gerardii Loisel. the most abundant species. The seed bank was significantly larger and richer in the ungrazed site than in the grazed site. Cold treatment reduced the number of germinating species and seedlings. In the grazed and non-cold-treated samples, the numbers of species and seedlings were highest in the pesticide treatment. In ungrazed samples there were no significant differences among treatments. After the cold treatment, the least number of species and seedlings was produced by the salt-water treatment. Changing brackish water to tap water led to a burst of germination, especially of J. gerardii. The seed bank of the upper geolittoral zone was richer than that of the middle geolittoral. The multivariate classification and ordination groupings are based on the abundances of J. gerardii and Glaux maritima L.; different treatments were not distinguishable. There was a low resemblance between the seed bank and the aboveground vegetation.Key words: seed bank, salinity, pesticide, seashore meadow, cold treatment, vegetation.

Trihelix transcription factors play important roles in plant growth, development and various stress responses. In this study, we identified 32 trihelix family genes (DoGT) in the important Chinese medicinal plant Dendrobium officinale. These trihelix genes could be classified into five different subgroups. The gene structure and conserved functional domain of these trihelix genes were similar in the same subfamily but diverged between different subfamilies. Various stresses responsive cis-elements presented in the promoters of DoGT genes, suggesting that the trihelix genes might respond to the environmental stresses. Expressional changes of DoGT genes in three tissues and under cold treatment suggested that trihelix genes were involved in diverse functions during D. officinale development and cold tolerance. This study provides novel insights into the phylogenetic relationships and functions of the D. officinaletrihelix genes, which will aid future functional studies investigating the divergent roles of trihelix genes belonging to other species.

Abstract Cultivated grapevine (Vitis) is a highly valued horticultural crop, and cold stress affects its growth and productivity. Wild Amur grape (Vitis amurensis) PAT1 (Phytochrome A signal transduction 1, VaPAT1) is induced by low temperature, and ectopic expression of VaPAT1 enhances cold tolerance in Arabidopsis (Arabidopsis thaliana). However, little is known about the molecular mechanism of VaPAT1 during the cold stress response in grapevine. Here, we confirmed the overexpression of VaPAT1 in transformed grape calli enhanced cold tolerance. Yeast two-hybrid and bimolecular fluorescence complementation assays highlighted an interaction between VaPAT1 with INDETERMINATE-DOMAIN 3 (VaIDD3). A role of VaIDD3 in cold tolerance was also indicated. Transcriptome analysis revealed VaPAT1 and VaIDD3 overexpression and cold treatment coordinately modulate the expression of stress-related genes including lipoxygenase 3 (LOX3), a gene encoding a key jasmonate biosynthesis enzyme. Co-expression network analysis indicated LOX3 might be a downstream target of VaPAT1. Both electrophoretic mobility shift and dual luciferase reporter assays showed the VaPAT1-IDD3 complex binds to the IDD-box (AGACAAA) in the VaLOX3 promoter to activate its expression. Overexpression of both VaPAT1 and VaIDD3 increased the transcription of VaLOX3 and JA levels in transgenic grape calli. Conversely, VaPAT1-SRDX (dominant repression) and CRISPR/Cas9-mediated mutagenesis of PAT1-ED causing the loss of the C-terminus in grape calli dramatically prohibited the accumulation of VaLOX3 and JA levels during cold treatment. Together, these findings point to a pivotal role of VaPAT1 in the cold stress response in grape by regulating JA biosynthesis.

Successful results of textile materials finishing process in order to gain desired properties to the fabric according to customers’ demands mainly depend on properly-carried pre-treatment stages. The present study covers twill weave cotton/polyester fabric’s modification in alkaline medium at different temperatures and treatment durations through exhaustion (hot and cold treatment) and pad-steam process. An influence of the reduction agent addition was ascertained. Weight loss, water absorbency, vertical wicking, dye uptake, colour measurements, surface morphology and psysicomechanical characteristics were determined. Samples and recommendations of optimal alkaline treatment technologies are developed.
