The efficacy of meso-2,3-dimercaptosuccinic acid (DMSA) was evaluated in workers occupationally exposed to lead (Pb; blood level >50 mg //dL). Ten men were given 600 mg of DMSA per orem daily, for five days. Pb concentrations of whole blood and urine were determined throughout therapy. Hematology analyses, blood chemistry, and urinalysis were obtained at the start of the study, at the end of the DMSA treatment, and at 72 hours after the administration of the final dose. DMSA therapy had no influence on hepatic, hematologic, or renal functions and was effective in decreasing the concentration of blood Pb in all the subjects without adverse drug reactions.

ABSTRACT Background The purpose of this study is to evaluate the effects of different immunosuppressant agents on gingival overgrowth. Materials and methods About 60 patients medicated with cyclosporin A (CsA) and 40 patients with tacrolimus (TcR) were recruited in the study. Periodontal examination (PI, GI, PBI, PPD, GO) was carried out only for maxillary and mandibular anterior teeth. Pharmacologic variables, such as daily dosage of immunosuppressant agents, TcR/CsA whole blood level were recorded. Results In the group medicated with CsA, 10 patients had clinically significant GO, while none of the patients medicated with Tac had clinically significant GO. Significant differences were observed for periodontal parameters (PI, GI, PBI, PPD), post-transplant period, percentage of GO. In CsA group, significant difference was detected for periodontal parameters, percentage of GO and daily glucocorticoid dosage between responders and nonresponders. Conclusion The results from our study show that GO does not occur in patients medicated with TcR, although TcR shares same mechanism with CsA. How to cite this article Bekit A, Bagis N, Arpak N. Introduction. The element composition of blood is the most relevant index in biomonitoring studies to assess public health risks. The authors determined the elemental composition of the whole blood in 100 the unexposed children residing in the West Ural region using the ICP-MS method and standard biomonitoring methodology of the World Health Organization (WHO). This study allows for improving the comparability and reliability of the analysis results, as well as to supplement information on human biomonitoring for various territories of the Russian Federation. A comparative assessment of the contents of vanadium, chromium, manganese, nickel, copper, zinc, arsenic, selenium, strontium, cadmium, thallium and lead with reference concentrations used in national programs for human biomonitoring (BMC) of European countries and the United States was made. Material and methods. The authors determined mass concentrations of V, Cr, Mn, Ni, Cu, Zn, As, Se, Sr, Cd, Tl, and Pb by ICP-MS in conformity with Methodical Guidelines 4.1. 3230-14 (FR.1.31.2014.17064) developed by the authors. The article contains setting parameters for quadrupole mass spectrometer with inductively coupled plasma (Agilent 7500cx), the optimal sample preparation scheme for eliminating the “matrix” effect. The operating mode of the mass spectrometer with a response/collision cell to mitigate interferences and the choice of internal standards is justified. The sample preparation of whole blood was made by acid dissolution followed by centrifugation. The detection limits (LOD) for each element are calculated. Results. The results are presented in the form of basic statistics: minimum and maximum values, arithmetic mean (AM), geometric mean (GM), 5, 50, 95 percentiles and interpreted in accordance with modern international requirements. The geometric mean (GM) of element content in blood amounted to 0.11 µg/l (V); 5.34 µg/l (Cr); 13.1 µg/l (Mn); 3.15 µg/l (Ni); 854.7 µg/l (Сu); 4655.0 µg/l (Zn); 0.13 µg/l (As); 84.97 µg/l (Se); 28.0 µg/l (Sr); 0.28 µg/l (Сd); 0.05 µg/l (Tl); 18.63 µg/l (Pb). The validity of the results was confirmed by means of SERONORM TM Whole Blood L2 standard samples (Norway). The results of the study relied upon the 95 percentile (P95) can be used as approximate reference data as a basis for assessing the risk associated with exposure to metals. Conclusion. Regional differences from levels found in other Europe countries (Italy, Norway) and the USA included higher concentrations of Mn, Cr and Ni and lower levels of Cu, Zn and Se in blood of children in the rural areas of the West Ural region. The optimal method for determining the content of essential and toxic elements in blood by mass spectrometry with inductively coupled plasma has been substantiated and recommended for practical use (MG 4.1. 3230-14, FR.1.31.2014.17064).

Background: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a poorly understood disease involving a high inflammatory status. Neutrophil extracellular traps (NETs) have been described as a new pathway to contain infectious diseases but can also participate in the imbalance of the inflammatory and the coagulation systems. NETs could be a therapeutic target in COVID-19 patients.Methods: Consecutive patients with SARS-CoV2 related pneumonia admitted to the intensive care unit were included in a prospective bicentric study. Neutrophil extracellular trap concentrations were quantified in whole blood samples at day-1 and day-3 by flow cytometry. The primary outcome was the association between the blood NET quantification at ICU admission and the number of days with refractory hypoxemia defined by a PaO2/FIO2 ratio ≤100 mmHg.Results: Among 181 patients admitted to the ICUs for acute respiratory failure related to SARS-CoV2 pneumonia, 58 were included in the analysis. Patients were 62 [54, 69] years old in median, mostly male (75.9%). The median number of days with severe hypoxemia was 4 [2, 6] days and day-28 mortality was 27.6% (n = 16). The blood level of NETs significantly decreased between day-1 and day-3 in patients who survived (59.5 [30.5, 116.6] to 47 [33.2, 62.4] p = 0.006; 8.6 [3.4, 18.0] to 4 [1.4, 10.7] p = 0.001 and 7.4 [4.0, 16.7] to 2.6 [1.0, 8.3] p = 0.001 for MPO+, Cit-H3+, and MPO+ Cit-H3+ NETs, respectively) while it remained stable in patients who died (38.4 [26.0, 54.8] to 44.5 [36.4, 77.7] p = 0.542; 4.9 [1.3, 13.0] to 5.5 [2.8, 6.9] p = 0.839 and 4 [1.3, 13.6] to 2.7 [1.4, 4.5] p = 0.421 for MPO+, Cit-H3+, and MPO+ Cit-H3+ NETs, respectively). In multivariable negative binomial regression, the blood level of MPO+ NETs was negatively associated with the number of days with severe hypoxemia within 7 days (0.84 [0.73, 0.97]), while neither Cit-H3+ NETs nor double-positive NETs were significantly associated with the primary outcome.Conclusion: The whole blood level of NETs at day-1 was negatively associated with the number of days with severe hypoxemia in patients admitted to the intensive care unit for SARS-CoV2 related pneumonia. The lack of decrease of the blood level of NETs between day-1 and day-3 discriminated patients who died within day-28.

Platelet adhesion initiates platelet aggregation and is an important component of the hemostatic process. Since the development of a new form of collagen as a topical hemostatic agent is of both basic and clinical interest, an ultrastructural and hematologic study of the interaction of platelets with the microcrystalline collagen preparation was undertaken.In this study, whole blood anticoagulated with EDTA was used in order to inhibit aggregation and permit study of platelet adhesion to collagen as an isolated event. The microcrystalline collagen was prepared from bovine dermal corium; milling was with sharp blades. The preparation consists of partial hydrochloric acid amine collagen salts and retains much of the fibrillar morphology of native collagen.

Thrombocytes (TC), the avian equivalent of blood platelets, support hemostasis by aggregating at sites of injury. Studies in our lab suggested that fibrinogen (fib) is a requisite cofactor for TC aggregation but operates by an undefined mechanism. To study the interaction of fib with TC and to identify fib receptors on cells, fib was purified from pigeon plasma, conjugated to colloidal gold and used both to facilitate aggregation and as a receptor probe. Described is the application of computer assisted reconstruction and stereo whole mount microscopy to visualize the 3-D organization of fib receptors at sites of cell contact in TC aggregates and on adherent cells.Pigeon TC were obtained from citrated whole blood by differential centrifugation, washed with Ca++ free Hank's balanced salts containing 0.3% EDTA (pH 6.5) and resuspended in Ca++ free Hank's. Pigeon fib was isolated by precipitation with PEG-1000 and the purity assessed by SDS-PAGE. Fib was conjugated to 25nm colloidal gold by vortexing and the conjugates used as the ligand to identify fib receptors.

Cell culture systems have become increasingly popular as a means of screening toxic agents and studying toxic mechanisms of drugs and other chemicals at the cellular and subcellular levels. These in vitro tests can be conducted rapidly in a broad range of relevant mammalian culture systems; a variety of biological and biochemical cytotoxicity endpoints can be examined. The following study utilized human keratinocytes to evaluate the relative cytotoxicities of nitrofurazone (NF) and silver sulfadiazine (SS), the active ingredients of FURACIN(R) Topical Cream and SILVADENE(R) Cream, respectively. These compounds are anti-infectives used in the treatment of burn patients. Cell ultrastructure and elemental composition were utilized as cytotoxicity endpoints.Normal Human Epidermal Keratinocytes (HK) were prepared from the EpiPackTM culture system (Clonetics Corporation, Boulder, CO). For scanning electron microscopy (SEM) and transmission electron microscopy (TEM), cells were seeded on sterile 35 mm Falcon plastic dishes; for elemental microanalysis, cells were plated on polished pyrolytic carbon discs (E. Fullam, Latham, NY) placed in the culture dishes.

The elemental composition of amino acids is similar to that of the major structural components of the epithelial cells of the small intestine and other tissues. Therefore, their subcellular localization and concentration measurements are not possible by x-ray microanalysis. Radioactive isotope labeling: I131-tyrosine, Se75-methionine and S35-methionine have been successfully employed in numerous absorption and transport studies. The latter two have been utilized both in vitro and vivo, with similar results in the hamster and human small intestine. Non-radioactive Selenomethionine, since its absorption/transport behavior is assumed to be the same as that of Se75- methionine and S75-methionine could serve as a compound tracer for this amino acid.
