Enzymatic activity and protein expression of cellulase from rice straw produced by Trichoderma reesei in the presence of oxygen vectors

Abstract Background Agriculture by-products are considered a great potential value for utilization by ruminants as well as rabbits. They usually can be the maintenance and part of the production requirements. However, in developing countries, as well as in Egypt, animals suffer from shortage of feeds that are continuously increasing in costs. In general, biological treatments were shown to be the most effective and improved chemical composition of rice straw or corn stalks. Method This work aimed to investigate the possible ways of utilizing rice straws or corn stalks in rabbit feeding. The field work is designed to study the effect of biological treatment of Pleurotus ostreatus cultivated on rice straws and Trichoderma reesei cultivated on corn stalks and replacing clover hay by rice straws and corn stalks at levels of 0, 33, 66, and 100% either without or with microbes adding. Seventy-eight New Zealand White (NZW) rabbits aged 4–5 weeks (565 ± 13.57 g) were randomly divided into thirteen equal experimental groups. Results Untreated rice straws or biologically treated with Pleurotus ostreatus increased their contents of crude protein (CP) by 178.75 and 224.5% and nitrogen-free extract (NFE) by 6.30 and 24.53, respectively. Meanwhile, crude fiber (CF) content was reduced by 31.32 and 56.75%, and organic matter content was decreased by 2.81 and 5.51%, respectively, in comparison with the raw rice straws. Also, biological treatment of rice straws caused a decrease in values of neutral detergent fiber (NDF), acid detergent fiber (ADF), acid detergent lignin (ADL), and hemicellulose contents in comparison with either raw or treated rice straws. Furthermore, biological treatment with Trichoderma reesei realized a decrease in organic matter (OM), CF, NDF, and ADF and increased CP and ash contents in corn stalks. NFE content of corn stalks was decreased as a result of treatment without or with Trichoderma reesei experimental rations by 11.95% and 3.82% compared to raw corn stalks (CS). Biological treatments with fungi significantly (P < 0.05) improved average daily gain (ADG) and feed conversion. ADG and feed conversion were significantly (P < 0.05) improved when rabbits were fed diets containing rice straw (RS) compared to that fed CS. Rabbits fed diets replaced clover hay (CH) with 33% or 66% of RS or CS significantly increased ADG compared to control and that replaced 100% of both RS and CS containing rations. Levels of replacing had no significant effect on their dry matter intake (DMI) values. The highest improvement in feed conversion was recorded with rabbits that received diets replaced 33% of berseem hay (BH) by RS or CS, followed by that replaced 66% of BH by RS or CS. There were significantly interactions between biological treatments (T), roughage source (S), and replacement levels (L) (T × S × L) only on ADG. The best fed conversion was realized by rabbits fed diet replaced BH with 33% of RS that are treated by Pleurotus ostreatus (4.05 g DMI/g gain). Rabbits fed 33% biologically treated rice straw with Pleurotus ostreatus showed the highest economic efficiency (179%) followed by rabbits that received 33% of both rice straws treated without Pleurotus ostreatus and rabbits that received corn stalks biologically treated with Trichoderma reesei (161%). Conclusion Biological treatments of rice straws by Pleurotus ostreatus or corn stalks by Trichoderma reesei were safe, and it improves their chemical analysis and improved both daily gain and feed conversion, decreasing the costing of diet formulation which consequently decreased the price of 1-kg live body weight.

Transplanted mesenchymal stem/stromal cells (MSCs) are a promising and innovative approach in regenerative medicine. Their regenerative potential is partly based upon their immunomodulatory activities. One of the most investigated immunomediators in MSCs, such as in periodontal ligament-derived MSCs (hPDLSCs), is indoleamine-2,3-dioxygenase-1 (IDO-1) which is upregulated by inflammatory stimuli, like cytokines. However, there are no data concerning continuing IDO-1 expression in hPDLSCs after the removal of inflammatory stimuli, such as cytokines and toll-like receptor (TLR) agonist-2 and TLR-3. Hence, primary hPDLSCs were stimulated with interleukin (IL)-1β, tumor necrosis factor (TNF)-α, interferon (IFN)-γ, TLR-2 agonist Pam3CSK4 or TLR-3 agonist Poly I/C. IDO-1 gene and protein expression and its enzymatic activity were measured up to five days after removing any stimuli. IL-1β- and TNF-α-induced IDO-1 expression and enzymatic activity decreased in a time-dependent manner after cessation of stimulation. IFN-γ caused a long-lasting effect on IDO-1 up to five days after removing IFN-γ. Both, TLR-2 and TLR-3 agonists induced a significant increase in IDO-1 gene expression, but only TLR-3 agonist induced significantly higher IDO-1 protein expression and enzymatic activity in conditioned media (CM). IDO-1 activity of Poly I/C- and Pam3CSK4-treated hPDLSCs was higher at one day after removal of stimuli than immediately after stimulation and declined to basal levels after five days. Among all tested stimuli, only IFN-γ was able to induce long-lasting IDO-1 expression and activity in hPDLSCs. The high plasticity of IDO-1 expression and its enzymatic activity in hPDLSCs due to the variable cytokine and virulence factor milieu and the temporal-dependent responsiveness of hPDLSCs may cause a highly dynamic potential of hPDLSCs to modulate immune responses in periodontal tissues.

Nitric oxide (NO), produced by endothelial (e) NO synthase (NOS), is critically involved in the cardiopulmonary transition from fetal to neonatal life. We have previously shown that NO-dependent relaxation is attenuated in intrapulmonary arteries from fetal lambs with pulmonary hypertension (PHT) created by prenatal ligation of the ductus arteriosus. In the present study, we determined whether this is due to altered pulmonary eNOS expression. eNOS and neuronal NOS (nNOS) protein expression were assessed in lungs from near-term control lambs and PHT lambs that underwent ductal ligation 10 days earlier. eNOS protein expression was decreased 49% in PHT lung. In contrast, nNOS protein abundance was unchanged. NOS enzymatic activity was also diminished in PHT vs. control lung (60 +/- 3 vs. 110 +/- 7 fmol.mg protein-1.min-1, respectively). Paralleling the declines in eNOS protein and NOS enzymatic activity, eNOS mRNA abundance was decreased 64% in PHT lung. Thus pulmonary eNOS gene expression is attenuated in the lamb model of fetal PHT. Because NO modulates both vasodilation and vascular smooth muscle growth, diminished eNOS expression may contribute to both the abnormal vasoreactivity and the excessive muscularization of the pulmonary circulation in fetal PHT.

The aim of the study was to investigate the effect of icariin (ICA) on cardiac aging through its effects on the SIRT6 enzyme and on the NF-κB pathway. Investigating the effect of ICA on the enzymatic activity of histone deacetylase SIRT6 revealed a concentration of 10−8 mol/L ICA had a maximum activating effect on histone deacetylase SIRT6 enzymatic activity. Western analysis showed that ICA upregulated SIRT6 protein expression and downregulated NF-κB (p65) protein expression in animal tissues and cell models. ICA upregulated the expression of SIRT6 and had an inhibitory effect on NF-κB inflammatory signaling pathways as shown by decreasing mRNA levels of the NF-κB downstream target genes TNF-α, ICAM-1, IL-2, and IL-6. Those effects were mediated directly or indirectly by SIRT6. We provided evidence that inflammaging may involve a novel link between the effects of ICA on SIRT6 (a regulator of aging) and NF-κB (a regulator of inflammation).
