Introduction: Root canal treatment is done to maintain the teeth to last as long as possible in the oral cavity. This can be done with three main stages, such as biomechanical preparation, sterilization and root canal filling. The hermetic seal of root canal filling has an important role in the success of root canal treatment. Objective: The aim of this in-vitro study is to compare the microbial leakage of root canals filled with RealSeal sealer/Resilon Points and AH Plus sealer/conventional gutta-percha points using single cone obturation technique and warm vertical compaction technique. Methods: Thirty-two extracted human mandibular premolars with single canals were decoronated to a standardized root length of 15 mm and prepared using crown-down technique to a master apical file size 30/.09. Teeth were divided into four experimental groups (n = 7 each group) and controls (n = 1 each group). Teeth in the first group were obturated using single master gutta percha cone and AH Plus sealer and second group were obturated with single Resilon gutta percha cone using RealSeal sealer. Third experimental group was filled with warm vertical condensed gutta-percha using AH Plus sealer and fourth group was filled with warm vertical condensed Resilon gutta-percha using RealSeal sealer. The coronal chamber of each sample were inoculated with Enterococcus faecalis. Results: There were no significant differences between the four experimental groups (P = 0.182). The differences occurred only in the survival time from each group. Group 4 (Warm Vertical Condensation - RealSeal) has the lowest leakage rate among other groups and the fastest leakage occurred in group 1 (Single Cone - AH Plus). Conclusion: The single-cone techniques does not insure durable apical seal against bacterial leakage. Warm vertical compaction technique using Resilon gutta-percha and RealSeal sealer appears to be more effective in minimizing bacterial leakage than gutta-percha and AH Plus sealer.

<p>Objective: To evaluate coronal bacterial leakage comparing five endodontic sealers (AH Plus, Apexit Plus, Copaifera sp oil, EndoREZ and Polifil), and comparing root canals filled with EndoREZ sealer/EndoREZ® Points and EndoREZ sealer/conventional gutta-percha points.</p><p>Material and Methods: 84 human teeth were prepared and filled with gutta-percha points using the single cone technique. Roots were randomly divided into 6 groups: Apexit Plus, AH Plus, Copaifera sp oil, Polifil, EndoREZ, and EndoREZ/EndoREZ Points. After setting time, the roots were incorporated in a leakage model, which upper chamber contained a suspension of <em>Streptococcus mutans</em>, and lower chamber a broth. Leakage was assessed for turbidity in lower chamber for 60 days. Statistic analysis was performed using the nonparametric Kaplan-Meier method (p&lt;0.05).</p><p>Results: All experimental groups presented leakage during the study’s period. The medium time of leakage was: Apexit Plus and AH Plus 6,3 days, Polifil 5,1 days, Copaifera 1,2 days, and both EndoREZ groups infiltrated in the first day.</p><p>Conclusions: There was no statistically significant difference between the sealers Apexit Plus, AH Plus and Polifil, but they prevented leakage better than Copaifera sp oil and both EndoREZ groups. However, none of the tested sealers was capable of resisting coronal bacterial leakage for more than 22 days.</p>

AIM: The aims of this study were to assess the penetration of two endodontic sealers (salicylate and epoxy resin-based sealers) into dentinal tubules using CLSM; and to evaluate the bacterial leakage of roots filled with the same sealers associated with gutta-percha. MATERIAL AND METHODS: For sealer penetrability assessment, thirty bovine roots were instrumented and divided into three groups: AHP: EDTA + filling with AH Plus and gutta-percha (n=10), MTAF: EDTA + filling with MTA Fillapex and gutta-percha (n=10), control group: canals were not irrigated with EDTA and were filled with gutta-percha and AH Plus (n=5) or MTA Fillapex (n=5). Rhodamine B was added to the sealers in order to provide adequate fluorescence. The roots were transversely sectioned 3mm from the apex to enable CLSM analysis. Leakage was evaluated for turbidity of the broth in a split chamber model system for 30 days, using Enterococcus faecalis as a microbial marker. Thirty roots were instrumented and divided in four grupos: AHP: filling with AH Plus and gutta-percha (n=10); MTAF: filling with MTA Fillapex and gutta-percha (n=10); positive control: filling with gutta-percha without sealer (n = 5); negative control: sealing with cyanoacrylate to test the seal of the system (n = 5). RESULTS: The medians for dentinal tubule penetration were 6.8% (AHP) and 6.6% (MTAF) (P = 0.82). The average time for bacterial leakage was 8 days in both experimental groups (P = 0.79). CONCLUSION: MTA Fillapex and AH Plus presented similar behavior regarding dentinal tubule penetration and bacterial leakage.

Background: This work aimed to evaluate the efficacy of sonic agitation of a binary mixture of solvents (methyl ethyl ketone/tetrachloroethylene) on filling remnants removal and compare the effects of solvent agitation with the enlargement to the next instrument size. Methods: Twenty-four mandibular incisors were prepared with ProTaper Next (X1, X2) and obturated with the single-cone technique and AH Plus sealer. The teeth were retreated with ProTaper Universal Retreatment and ProTaper Next and divided into two groups (n = 12) according to the final instrument (X3 or X4). All canals were submitted to a supplementary procedure consisting of a mixture of solvents―methyl ethyl ketone/tetrachloroethylene, agitated with EndoActivator. The volume of filling remnants was assessed through micro-computed tomography in the apical 5 mm. Statistical analysis was performed with a significance level of 5%. Results: The supplementary procedure of agitation of the solvent mixture was beneficial in both groups (p < 0.05). There were no statistically significant differences between canals re-prepared until X4 and canals re-prepared until X3 plus solvent (p > 0.05). Conclusions: An additional step with a two-solvent solution potentiated by EndoActivator showed to be very effective for the removal of gutta-percha and resinous sealer remnants from apical root canals of mandibular incisors, avoiding further enlargement.

ABSTRACT Aim The aim of this in vitro study was to evaluate the apical seal obtained with four root canal sealers AH 26, Sealapex, Endoflas FS and AH Plus, with lateral condensation. Materials and methods Sixty root canals were prepared using the step-back technique. The specimens were divided into four experimental groups of 12 teeth and two control groups of 12 teeth. The experimental groups were obturated by laterally condensed gutta-percha with one of the tested sealers and control groups were obturated without any sealer. Methylene blue dye penetration with centrifuging method was used to evaluate the apical sealing ability. The quantitative apical leakage of each specimen was measured after 2 weeks. Results The results showed no significant differences between all groups except between AH Plus and Endoflas FS (<0.05). AH Plus showed significantly less leakage than Endoflas FS. Conclusion AH Plus showed the least leakage compared to AH 26, Sealapex and Endoflas FS. How to cite this article Joseph R, Singh S. The objective of root canal obturation is to achieve a fluid-tight seal. Recently, GuttaFlow bioseal (GB), a root canal sealer composed of polydimethylsiloxane, gutta-percha particles, and bioactive glass ceramics, has been developed, to enhance the sealing ability of root canal filling material. The objective of this study was to assess the sealing ability of GB using a subnanoliter-scaled fluid-flow measuring device and to compare with that of AH Plus (AH). The fluid flow in root canal-filled teeth using either gutta-percha cone (GP) with AH (GAR; n = 10) or GP with GB (GBR; n = 10) and in GP inserted in AH blocks (GA; n = 10) or GP inserted in GB blocks (GB; n = 10) was measured. In addition, fluid flow in sealer blocks of AH (A; n = 10) and GB (B; n = 10), which served as negative controls, was measured. Root canal-filled teeth using GP without any sealer (GR) acted as positive controls (n = 10). The leakage was obtained by calculating the volume of moved water by time (s), after stabilization of the fluid flow was achieved. Statistical analysis was performed using the Kruskal–Wallis test and Mann–Whitney U-test with Bonferroni correction. A p value less than 0.00238 (0.05/21) was considered significantly different. The mean leakages (nL/s) in the groups are as follows: GAR, 0.0958 ± 0.0543; GBR, 0.0223 ± 0.0246; GA, 0.0644 ± 0.0803; GB, 0.0267 ± 0.0182; A, 0.0055 ± 0.0057; B, 0.0052 ± 0.005; and GR, 0.2892 ± 0.3018. The mean leakage in the GBR group was lower than that in the GAR group (p = 0.001), while the mean leakages in the GA and GB groups were not significantly different. GuttaFlow bioseal can be useful in single-cone obturation technique.

Objectives: The purpose of this in vitro study was to evaluate gaps or voids occurring in roots filled with three different sealers.Methods: Thirty extracted human single-rooted teeth were decoronated, instrumented using NiTi rotary instruments, divided into three groups (n=10 per group) and obturated with one of the following: Epiphany with Resilon, MetaSEAL with gutta-percha or AH Plus with gutta-percha using the match-taper single-cone technique. After storage for one week in 100% humidity at 37°C, the teeth were horizontally sectioned (n=10). Photographs were taken from the coronal, median and apical parts of the roots using a stereomicroscope at 10X magnification, and the images were then transferred to a computer. The mathematical method known as the ‘Affine Transformation’ was used for the transformation of pixel coordinates to ground coordinates in the Netcad Software program. The mean areas (μm2) of the gaps between the sealer and root dentin or gutta percha/resilon and the gaps between the sealer and/or voids inside the sealer mass were measured, scored on a 0-3 scale and statistically analyzed with the Kruskal-Wallis test.Results: The mean total area of gaps or voids for each sealer was 4631.80 μm2 for the Epiphany- Resilon, 3826.80 μm2 for the MetaSEAL-gutta-percha and 31334 μm2 for the AH Plus-gutta-percha. The MetaSEAL-gutta-percha group showed more gap or void-free interfaces. No significant differences were found among the sealers in the scores for the gap areas (P<.05), and the MetaSEAL showed similar interfaces with Epiphany.Conclusions: No significant differences in the mean areas of gaps or voids were found among the tested resin-based sealers. ABSTRACT Objective: Aim of the present study was to evaluate the retreatability and reestablishment of apical patency of two calcium silicate-based sealers, TotalFill BC Sealer (BCS) and mineral trioxide aggregate Fillapex (MTA F), versus AH Plus, when used in combination with Gutta-percha (GP). Materials and Methods: The canals of 54 single-rooted anterior teeth were instrumented and filled with GP/AH Plus (Group A), GP/MTA F (Group B), or GP/BCS (Group C) using continuous wave obturation technique. The groups were subdivided into subgroups with the master-GP cone placed to the working length (WL) or intentionally 2 mm short. The retreatment procedures were performed using ultrasonics, chloroform, rotary, and hand files. The ability to establish the patency and reach WL was determined as well as the time taken to reach WL was calculated in minutes. Furthermore, the samples were observed under a dental, optical microscope, after vertically splitting them. Results: The WL and patency were reestablished in 100% of specimens in all groups. The Mann–Whitney U-test indicated that there was a significant difference in the amount of time required to reach WL between the groups (P < 0.05) with group GP/BCS short of the WL showing the most amount of time to be retreated. Conclusion: The novel calcium silicate-based sealers are negotiable under simple root canal anatomy. However, the conventional retreatment techniques are not able to fully remove them.
