The novel use of the α-methylene-β-lactone (MeLac) moiety as a warhead of multiple electrophilic sites is reported. In this study, we demonstrate that a MeLac-alkyne is a competent covalent probe and reacts with diverse proteins in live cells. Proteomics analysis of affinity-enriched samples identifies probe-reacted proteins, resolves their modified peptides/residues, and thus characterizes probe-protein reactions. Unique methods are developed to evaluate confidence in the identification of the reacted proteins and modified peptides. Tandem mass spectra of the peptides reveal that MeLac reacts with nucleophilic cysteine, serine, lysine, threonine, and tyrosine residues, through either Michael addition or acyl addition. A peptide-centric proteomics platform, using MeLac-alkyne as the measurement probe, successfully analyzes the Orlistat selectivity in live HT-29 cells. MeLac is a versatile warhead demonstrating enormous potential to expedite the development of covalent probes and inhibitors in interrogating protein (re)activity. MeLac-empowered platforms in chemical proteomics are widely adaptable for measuring the live-cell action of reactive molecules.

Cavendish banana peel can be used as a substitute for the expensive xylan, while molasses than as a source of carbon as well as nitrogen, minerals and nutrients needed for the growth of microbes that can produce the enzyme. Xylanase produced from Bacillus stearothermopillus DSM 22, using media cavendish banana peels with the addition of molasses 1%, 2%, and 3%. Fermentation is done in a shaker incubator at 550C temperature conditions, initial pH 8, and 250 rpm agitation. The result showed the highest enzyme activity of 4,14 ± 0,16 U/mL min., on the addition 2% molasses after 24 hours. Further fermentation carried out in the fermenter working volume of 3.5 liters, with the condition of temperature 550C, pH 8, aeration 1 vvm, agitation 250 rpm, the highest spesific enzyme of activity of 51,62 ± 0,16 U/mg after 24 hours. Partial purification of xylanase enzyme fermentation is done with the results of microfiltration, ultrafiltration, ammonium sulfate (0-80%) and dialysis. There is an increase in the purity of the enzyme at each stage of purification, the highest purity on dialysis 3.23 times of crude enzymes.Kulit buah pisang kapendis dapat digunakan sebagai pengganti xilan yang harganya mahal, sementara molases selain sebagai sumber karbon serta nitrogen, mineral dan nutrisi dibutuhkan untuk pertumbuhan mikroba yang dapat menghasilkan enzim. Xilanase yang dihasilkan dari Bacillus stearothermopillus DSM 22, menggunakan media kulit pisang kapendis dengan penambahan molase 1%, 2%, dan 3%. Fermentasi dilakukan dalam shaker inkubator pada temperatur 550C, pH awal 8, dan agitasi 250 rpm. Hasilnya menunjukkan aktivitas enzim tertinggi 4,14 ± 0,16 U/mL min., pada penambahan 2% molases setelah 24 jam. Selanjutnya fermentasi dilakukan di dalam fermentor, volume kerja dari 3,5 liter, dengan kondisi temperatur 550C, pH 8, aeration 1 vvm, agitasi 250 rpm, aktivitas spesifik tertinggi 51,62 ± 0,16 U/mg setelah 24 jam. Pemurnian parsial fermentasi enzim xilanase dilakukan dengan hasil mikrofiltrasi, ultrafiltrasi, amonium sulfat (0-80%) dan dialisis. Ada peningkatan kemurnian enzim pada setiap tahap pemurnian, kemurnian tertinggi pada dialisis 3,23 kali dari enzim kasar.Keywords: Xylanase, B. stearothermophillus DSM 22, Cavendish banana peel, molasses, enzyme activity

The study aims to use an adsorbent natural based of Moroccan oil shale of Timahdit area (Y layer) in a physical-chemical adsorption process for treating industrial discharges colorful. The used adsorbent is the insoluble party of the sub-critical extraction of decarbonized oil shale of Timahdit. The tests performed on the methylene blue (MB), showed a strong elimination in the first 10 minutes. The influences of various experimental parameters were studied: mass ratio of adsorbent, time and temperature of thermal treatment, contact time, pH of MB and heating temperature of solution on the parameters of material were studied. The experimental results have shown that the adsorption of methylene blue dye by the adsorbent is more than 90% at initial pH a range 6-7 at room temperature for 30 minutes. The process is simple and the adsorbent produced is a new material with interesting adsorption capacities of moderate cost which does not require an activating agent and can be used as industrial adsorbent for the decontamination of effluents containing organic pollutants.

A series of three chitosan-based magnetic composites was prepared through a simple coprecipitation method. It was investigated the influence of mass ratio between chitosan and magnetite on the physical and chemical properties of the composites in order to establish the optimum conditions for obtaining a composite with good adsorption capacity for Pb(II) and Cu(II) from mono and bicomponent aqueous solutions. It was found that the microspheres prepared using mass ratio chitosan / magnetite 1.25/1, having a saturation magnetization of 15 emu g--1, are the best to be used as adsorbent for the metal ions. The influence of different parameters such as initial pH values, contact time, initial concentration of metal ions, on the adsorption of Pb(II) and Cu(II) onto the chitosan-based magnetic adsorbent was investigated in details. The adsorption process fits the pseudo-second-order kinetic model in both mono and bicomponent systems, and the maximum adsorption capacities calculated on the basis of the Langmuir model were 79.4 mg g--1 for Pb(II) and 48.5 mg g--1 for Cu(II) in monocomponent systems, while in bicomponent systems were 88.3 and 49.5 mg g--1, respectively. The results revealed that the as prepared chitosan-based magnetic adsorbent can be an effective and promising adsorbent for Pb(II) and Cu(II) from mono and bicomponent aqueous solutions.

Iron-coated sand (ICS) prepared by using FeCl3 and Joomoonjin sand widely used in Korea was used in this study. In batch adsorption kinetics, As(V) adsorption onto ICS was completed within 20 minutes, while adsorption of Pb(II), Cd(II), and Cu(II) onto ICS was slower than that of As(V) and strongly depended on initial pH. At pH 3.5, ICS showed a selective adsorption of Pb(II) compared to Cd( II) and Cu(II) . However, above pH 4.5, near complete removal of Pb(II), Cd(II), and Cu(II) was observed through adsorption or precipitation depending on pH. As(V) adsorption onto ICS occurred through an anionic-type and followed a Langmuir-type adsorption behaviour. In column experiments, pH was identified as an important parameter in the breakthrough of As(V). As(V) breakthrough at pH 4.5 was much slower than at pH 9 due to a strong chemical bonding between As(V) and ICS as similar with batch adsorption behaviour. With variation of ICS amounts, the optimum amount of ICS at pH 4.5 was identified as 5.0 grams in this research. At this condition, ICS could be used to treat 200 mg of As(V) with 1 kg of ICS until 50 ppb of As(V) appeared in the effluent. In this research, as a new treatment system, ICS can be potentially used to treat As(V) and cationic heavy metals.
