Various selective agents exert their influence on the processes governing the survival of new forms within an established plant group. As the balance of these agents changes so also will the genetic composition of the plant group change. Environmental conditions undoubtedly permit and direct evolution, but apparently the action of the environment is mainly selective in nature, and, although not directly concerned in the production of new growthforms, it aids the survival of suitable forms by establishing a certain amount of genotype isolation. Amongst the various selective agents whose action is confined to the direct selection of genotypes, or phenotypes and therefore indirectly of genotypes, there is the important question of pollination and fertilisation. As the very existence of individuals depends on compatible fertilisation, the investigation of the problem of pollination and fertilisation obtaining within a plant group is essential preparatory to any study of the survival of individuals within that plant group.

The problems of seed production of amenity grasses in New Zealand are considered from two aspects - (a) increasing seed potential by breeding; (b) achieving that potential by good management as practised in the Netherlands, (a) In terms of breeding, the strategies to increase seed yield depended to some degree on the species concerned. With browntop it was necessary to discard about two-thirds of the most promising genotypes because of poor heading, but the remaining genotypes flowered freely and in unison, and gave good seed yields. With fescue even the best original plants were sparsely heading, so direct selection for prolificacy has been needed. This was also carried out for perennial ryegrass, as well as selection of early-heading genotypes to avoid infection by stem rust. (b) It is suggested that areas for seed of amenity grasses in New Zealand should be used for seed production only rather than for both forage and seed as is the usual practice. Techniques used in the Netherlands could serve as guidelines to procedures. The implications of low seeding rates, good weed control, high fertilizer applications and minimum cutting or grazing in promoting strong individual plants capable of high seed yields are discussed. The results of investigations on revealing the features of the structure formation of seed productivity in new genotypes of peas with non-permeable beans are presented. Selection of Kaban, Fregat, Veles varieties and sample KT-6489 with a new trait was aimed at increasing the number of beans and seeds per plant. The increase in seed productivity was accompanied by a decrease in the mass of 1000 seeds and an increase in the performance of beans. Differences in the variability of the examined signs of seed productivity testify to the genotypic characteristics of the reaction to the effect of environmental conditions. The selection value of the “productivity of seed formation” and its hereditary character is grounded. The use of sources with a high seed-forming ability in breeding has contributed to an increase in the proportion of seeds from planted ovules on the plant. In the varieties with soft beans, the values of this feature, depending on environmental conditions, varied within the range 54.4-70.9%. In new breeding developments, these figures were significantly increased to 81.8-91.0%. Statistical analysis proved the reliability and significance of genotypic differentiation of the trait in changing environmental conditions. A high significant effect of the genotype with a share of 83.7% on the variability of seed production was established. The influence of the environment and the interaction of the factors “genotype x environment” on the trait was assessed by a weak impact (3.61 and 3.94%). The presence of stable genotypes in the study group results from the revealed advantage of variance of genotypes over the variance of the “genotype-environment” interaction. A sample of KT-6489 with a weak variability in seed production (1.6%) with variations by years 84.0-86.7% was isolated.


Confectionery sunflower - a special area of use of sunflower, which requires the creation of marketable seeds quality features. One of the possible ways to create large-fruited sunflower is to create production hybrids and lines. Objective: to evaluate the created new large-fruited sunflower lines by a complex of morphological characters and determine the best lines for use as large-seeds hybrids as parent components or source material. In 2016-2019 years on the basis of the Institute of Oilseed Crops NAAS a study was conducted to assess the economic characteristics of large-fruited sunflower lines. We studied a collection of 27 lines of large-seeds sources. The lines were created by direct selection or crossing and sampling: Reyny of Argentinean origin, Zaporizhzhya confectionery variety, confectionery hybrid with striped pericarp color of Israeli origin, white seed of Turkish origin, synthetic population - donor of complex resistance. To study from the collection, lines were drawn that went through at least 7 generations with selection for seed size. Experience has shown that the shortest growing season for lines 174d and KP11 was 99 days, and the longest for lines I2K670 was 109 days. In the studied collection, the greatest mass of 1000 seeds has the KP11-146.47g line, which is the mother component and does not have branching. The second by weight of 1000 seeds (109 g) stood out line 168v, which also had branches and pollen fertility restoration genes and will be used as the paternal form. The third largest is also one basket line ZKN51-100. The collection included lines originating from the same combination, but with a different morphotype for the presence and absence of branching. So, based on the combination of KP11 x Zaporizhzhya Confectionery, three lines were obtained. A mass of 1000 seeds was observed in 98-86 g, with the branching line having the largest mass of 1000 seeds. The lines created with one combination VK678 x ZKN32: with a branch 168a had a mass of 1000 seeds 95g, and a line 168b - without a branch 109 g. Of the two lines obtained from the descendants of the combination KP11 x the striped hybrid both had branches, but the seeds were much smaller (weight of 1000 seeds 59 and 79 g). The collection also studied samples created on the basis of varieties and populations 160c, 174, 175b, the mass of 1000 seeds of which turned out to be more acceptable for large-fruited use from 83 to 99 g. Summing up the results of studying the collection of newly created lines, we can highlight the lines 162d, 168v, 175b, KP11 that are potentially promising for use in hybrids. The selections showed that large-fruited lines can be obtained from large-fruited varieties, self-pollination of large-fruited hybrids and crossing lines with hybrids and varieties. Self-pollination and selection of large-fruited lines in several generations does not provide the necessary variability for positive changes in selections. The result of the selection by weight of 1000 seeds in the offspring from crosses and from populations creates opportunities for new large-seeds sunflower.

For 30 years, forage ryegrass breeding has known that the germplasm may contain a maternally inherited symbiotic Epichloë endophyte. These endophytes produce a suite of secondary alkaloid compounds, dependent upon strain. Many produce ergot and other alkaloids, which are associated with both insect deterrence and livestock health issues. The levels of alkaloids and other endophyte characteristics are influenced by strain, host germplasm, and environmental conditions. Some strains in the right host germplasm can confer an advantage over biotic and abiotic stressors, thus acting as a maternally inherited desirable ‘trait’. Through seed production, these mutualistic endophytes do not transmit into 100% of the crop seed and are less vigorous than the grass seed itself. This causes stability and longevity issues for seed production and storage should the ‘trait’ be desired in the germplasm. This makes understanding the precise nature of the relationship vitally important to the plant breeder. These Epichloë endophytes cannot be ‘bred’ in the conventional sense, as they are asexual. Instead, the breeder may modulate endophyte characteristics through selection of host germplasm, a sort of breeding by proxy. This article explores, from a forage seed company perspective, the issues that endophyte characteristics and breeding them by proxy have on ryegrass breeding, and outlines the methods used to assess the ‘trait’, and the application of these through the breeding, production, and deployment processes. Finally, this article investigates opportunities for enhancing the utilisation of alkaloid-producing endophytes within pastures, with a focus on balancing alkaloid levels to further enhance pest deterrence and improving livestock outcomes.

Abstract We have developed two novel schemes for the direct selection of peroxisome-biogenesis-defective (pex) mutants of the methylotrophic yeast Pichia pastoris. Both schemes take advantage of our observation that methanol-induced pex mutants contain little or no alcohol oxidase (AOX) activity. AOX is a peroxisomal matrix enzyme that catalyzes the first step in the methanol-utilization pathway. One scheme utilizes allyl alcohol, a compound that is not toxic to cells but is oxidized by AOX to acrolein, a compound that is toxic. Exposure of mutagenized populations of AOX-induced cells to allyl alcohol selectively kills AOX-containing cells. However, pex mutants without AOX are able to grow. The second scheme utilizes a P. pastoris strain that is defective in formaldehyde dehydrogenase (FLD), a methanol pathway enzyme required to metabolize formaldehyde, the product of AOX. AOX-induced cells of fld1 strains are sensitive to methanol because of the accumulation of formaldehyde. However, fld1 pex mutants, with little active AOX, do not efficiently oxidize methanol to formaldehyde and therefore are not sensitive to methanol. Using these selections, new pex mutant alleles in previously identified PEX genes have been isolated along with mutants in three previously unidentified PEX groups.

SUMMARYThe reproductive development and seed production characters of a Lolium multiflorum × Festuca pratensis tetraploid hybrid and a Lolium perenne × Festuca pratensis tetraploid hybrid were compared with those of their tetraploid parental cultivars. Experiments were carried out under glasshouse and natural conditions; single plants were studied as only small quantities of seed were available from the breeder. The lowtemperature/short-day requirements for inflorescence induction of the two hybrids were in between those of the corresponding parental species. The variance of the date at which vegetative primordia changed to the reproductive condition was greater in the hybrids than in the parental cultivars, but the variance of the dates of inflorescence emergence were similar for the hybrids and parental cultivars. Inflorescences ranged from the Lolium type with only sessile spikelets to the Festuca type with primary branches. Inner glumes, which are absent in the Lolium parents but present in Festuca pratensis, were found in the hybrids, though not in all spikelets. In the Lolium perenne × Festuca pratensis hybrid pollen release, potential pollen viability and floret fertility were low, but the Lolium multiflorum × Festuca pratensis hybrid produced large amounts of pollen with a higher potential viability and had floret fertilities similar to those of its two parental cultivars. There was sufficient variation in reproductive characters for further selection, particularly in the Lolium perenne × Festuca pratensis hybrid.

As the oil offloading operations of floating production storage and offloading (FPSO) units become more routine, the desire grows to increase the availability for offloading and thus decrease production downtime. Experience with these operations is the main tool available to increase the efficiency of this aspect of deepwater production. However, it is clear that a formal optimization approach can help to fine tune design parameters so that not only is availability increased but the significance of each design parameter can be better understood. The key issue is to define the environmental conditions under which the vessels involved in offloading are able to maintain position. By this, we reduce the notion of availability to a set of operating criteria, which can or cannot be met for a particular set of environmental conditions. The actual operating criteria such as relative vessel heading depend on selection of design parameters, such as the direction and magnitude of external force applied by thrusters or tugs. In the earliest offloading operations, engineering judgment was used to determine the feasibility of offloading at a particular time. For example, if wind and current were not expected to exceed a 1year return period, offloading may be considered safe. This approach can be both conservative and unconservative, depending on the nuances of the particular environmental conditions. This study will propose a formal approach to choosing the design parameters that optimize the availability of a FPSO for offloading. A simple analysis model will be employed so that optimization can be performed quickly using a robust second order method. The proposed analysis model will be compared to model test data to demonstrate its agreement with the more complex system.
