Diagnostic Value of Electron Microscopy in Soft Tissue Tumors

For years the separation of carcinoma and sarcoma and the subclassification of sarcomas has been based on the appearance of the tumor cells and their microscopic growth pattern and information derived from certain histochemical and special stains. Although this method of study has produced good agreement among pathologists in the separation of carcinoma from sarcoma, it has given less uniform results in the subclassification of sarcomas. There remain examples of neoplasms of different histogenesis, the classification of which is questionable because of similar cytologic and growth patterns at the light microscopic level; i.e. amelanotic melanoma versus carcinoma and occasionally sarcoma, sarcomas with an epithelial pattern of growth simulating carcinoma, histologically similar mesenchymal tumors of different histogenesis (histiocytoma versus rhabdomyosarcoma, lytic osteogenic sarcoma versus rhabdomyosarcoma), and myxomatous mesenchymal tumors of diverse histogenesis (myxoid rhabdo and liposarcomas, cardiac myxoma, myxoid neurofibroma, etc.)

The effectiveness of the mechanism of precipitation strengthening in metallic alloys depends on the shapes of the precipitates. Two different material systems are considered: tetragonal γ′′ precipitates in Ni-based alloys and tetragonal θ′ precipitates in Al-Cu-alloys. The shape formation and evolution of the tetragonally misfitting precipitates was investigated by means of experiments and phase-field simulations. We employed the method of invariant moments for the consistent shape quantification of precipitates obtained from the simulation as well as those obtained from the experiment. Two well-defined shape-quantities are proposed: (i) a generalized measure for the particles aspect ratio and (ii) the normalized λ2, as a measure for shape deviations from an ideal ellipse of the given aspect ratio. Considering the size dependence of the aspect ratio of γ′′ precipitates, we find good agreement between the simulation results and the experiment. Further, the precipitates’ in-plane shape is defined as the central 2D cut through the 3D particle in a plane normal to the tetragonal c-axes of the precipitate. The experimentally observed in-plane shapes of γ′′-precipitates can be quantitatively reproduced by the phase-field model.

AbstractMyoepithelial neoplasms of soft tissue are rare tumors with clinical, morphological, immunohistochemical, and genetic heterogeneity. The morphological spectrum of these tumors is broad, and the diagnosis often requires immunostaining to confirm myoepithelial differentiation. Rarely, tumors show a morphology that is typical for myoepithelial neoplasms, while the immunophenotype fails to confirm myoepithelial differentiation. For such lesions, the term “myoepithelioma-like” tumor was introduced. Recently, two cases of myoepithelioma-like tumors of the hands and one case of the foot were described with previously never reported OGT-FOXO gene fusions. Here, we report a 50-year-old woman, with a myoepithelial-like tumor localized in the soft tissue of the forearm and carrying a OGT-FOXO1 fusion gene. Our findings extend the spectrum of mesenchymal tumors involving members of the FOXO family of transcription factors and point to the existence of a family of soft tissue tumors that carry the gene fusion of the OGT-FOXO family.

Geospatial monitoring of leaves variability has been performed for Platanus orientalis L. (Platanaceae) in Hissar Valley of Tajikistan. 17 local populations were clustered in four territorial groups and investigated by 11 morphometric variables and four indices. Six levels of variability were taken in the classification of S.A. Mamaev. All tested parameters were ranked due to the total score of variability levels. It was established, that indicators which have the combination of high, increased, and medium levels of variability, can be recommended for the purpose of geospatial monitoring. In the studied set of variables are two such ones: (1) the ratio of the length of leaf blade to the lengh of petiole, and (2) the petiole lengh. These two indicators reliably respond to point sources of industrial environmental pollution (Aluminum and Cement factories). Six indicators with domination of the middle level of variability, are of diagnostic value to differ loci inside territorial groups of populations. The results of the experiment confirmed, that set of variability levels for 8 of 15 studied parameters has diagnostic matter for geospatial monitoring of environment conditions in Hissar Valley of Tajikistan.

Abstract Over the years, many studies have been conducted to measure and classify the lightning-generated electric field waveform for a better understanding of the lightning physics phenomenon. Through measurement and classification, the features of the negative lightning return strokes can be accessed and analysed. In most studies, the classification of negative lightning return strokes was performed using a conventional approach based on manual visual inspection. Nevertheless, this traditional method could compromise the accuracy of data analysis due to human error, which also required a longer processing time. Hence, this study developed an automated negative lightning return strokes classification system using MATLAB software. In this study, a total of 115 return strokes was recorded and classified automatically by using the developed system. The data comparison with the Tenaga Nasional Berhad Research (TNBR) lightning report showed a good agreement between the lightning signal detected from this study with those signals recorded from the report. Apart from that, the developed automated system was successfully classified the negative lightning return strokes which this parameter was also illustrated on Graphic User Interface (GUI). Thus, the proposed automatic system could offer a practical and reliable approach by reducing human error and the processing time while classifying the negative lightning return strokes.

In this paper, a guided wave temperature robust PCA-based stress monitoring methodology is proposed. It is based on the analysis of the longitudinal guided wave propagating along the path under stress. Slight changes in the wave are detected by means of PCA via statistical T2 and Q indices. Experimental and numerical simulations of the guided wave propagating in material under different temperatures have shown significant variations in the amplitude and the velocity of the wave. This condition can jeopardize the discrimination of the different stress scenarios detected by the PCA indices. Thus, it is proposed a methodology based on an extended knowledge base, composed by a PCA statistical model for different discrete temperatures to produce a robust classification of stress states under variable environmental conditions. Experimental results have shown a good agreement between the predicted scenarios and the real ones

We report a case of multiple myoepithelioma with synchronous bone and soft tissue tumors, associated with a new genomic alteration of the LPP locus. The lesions occurred in the foot by presenting one lump in the plantar soft tissue, and three lesions were detected in the calcaneus and in the navicular bone. All tumors showed the double immunophenotype of epithelial markers and S100 protein expression. No rearrangement of the EWSR1 and FUS loci was detected as reported in myoepitheliomas. However, molecular karyotyping detected an unbalanced rearrangement of the LPP locus, not involving the HMGA2 locus, which is the most frequent translocation partner observed in benign mesenchymal tumors such as lipomas (of soft tissue as well as parosteal) and pulmonary chondroid hamartoma.

Abstract Context.—Common fibrous histiocytoma (cFH) or dermatofibroma and dermatofibrosarcoma protuberans (DFSP) are 2 spindle cell mesenchymal tumors that are distinguished in part by their microscopic growth patterns and clinically by the greater propensity for DFSP to recur. Matrix metalloproteinases (MMPs) potentially play a role in modulating the growth patterns of cFH and DFSP by remodeling the extracellular matrix. Objective.—To evaluate the immunohistochemical (IHC) expression of MMP-1, MMP-2, MMP-9, and MMP-14 in DFSP and cFH, because (1) MMP-1, MMP-2, MMP-9, and MMP-14 are synthesized by dermal fibroblasts, the major constituent of DFSP and cFH; and (2) platelet-derived growth factor B, which is overexpressed in most examples of DFSP because of t(17;22), activates ets-1, a transcription factor that regulates molecules associated with tumor invasion and metastasis, including MMP-1, MMP-3, and MMP-9. Design.—Immunohistochemical studies were performed on archived, formalin-fixed, paraffin-embedded tissue of DFSP (n = 48) and cFH (n = 47). Results.—Significant IHC expression (>10% of tumor cells) in cFH included MMP-14 (27 [59%] of 46 tumors positive), MMP-2 (21 [47%] of 45 tumors positive), MMP-9 (9 [20%] of 45 tumors positive), and MMP-1 (6 [13%] of 46 tumors positive). No DFSPs showed significant IHC expression of any of the MMPs evaluated. However, anti– MMP-2 highlighted a rich microvascular element within deep tumor tissue present in 81% of DFSPs with a prominent subcutaneous component. Conclusion.—Our IHC results indicate that MMP-1 and MMP-9 are not up-regulated in DFSP. Convincing expression of MMP-14 in cFH suggests that this MMP may affect the growth pattern of the lesion, perhaps by activating MMP-2 expression in tumor cells. In DFSP, MMP-2 may play a role in tumor angiogenesis.
