In Mediterranean regions, extreme weather conditions during the growing season may alter grapevine physiology and metabolism, thus modifying the quality of wines. The objective of this study was to investigate the effects of Ascophyllum nodosum treatments on plant physiology and berry metabolism in Vitis vinifera exposed to water stress. The experiment was performed on potted vines subjected to two irrigation regimes (well-watered, WW, and water stressed, WS) both associated with A. nodosum treatments (SWE), compared with control plants (CTRL). Gas exchanges, chlorophyll fluorescence, and water relations were monitored on SWE and CTRL leaves, both in WW and WS vines at three times. Moreover, the quantification of secondary metabolites and their partitioning were performed in berry skins. Plants treated with A. nodosum extract showed higher photosynthesis and stomatal conductance than CTRL in both irrigation regimes and maintained a better plant hydraulic conductivity at the end of the sampling period. In addition, secondary metabolites in berry skins and their partitioning were significantly affected by the treatments in both irrigation regimes. Our results suggest that foliar application of A. nodosum extract may help the acclimation of grapevines to post-veraison water stress, likely improving plant physiological and biochemical performances under environmental constraints.

AbstractThe objective of the present work was to adapt the Prussian Blue reaction for the determination of ascorbic acid. The procedure was successfully applied for the determination of ascorbic acid in red and white grapes (Vitis vinifera L.) just previous ingathering. In the present work was used the red and white grapes from Murfatlar vineyard: Mamaia, Cabernet Sauvignon, Merlot, Pinot Noir, Chardonnay, Sauvignon, Muscat Ottonel and Riesling Italian. The results were situated in the range of 0.67 - 1.79 mg vitamin C/100g product for red grapes and respectively 0.50 - 1.49 mg vitamin C/100g for white grapes.

Background: The prevalence of multiple sclerosis (MS) is not uniform, with a latitudinal gradient of prevalence present in most studies. Understanding the drivers of this gradient may allow a better understanding of the environmental factors involved in MS pathogenesis. Method: The New Zealand national MS prevalence study (NZMSPS) is a cross-sectional study of people with definite MS (DMS) (McDonald criteria 2005) resident in New Zealand on census night, 7 March 2006, utilizing multiple sources of notification. Capture—recapture analysis (CRA) was used to estimate missing cases. Results: Of 2917 people with DMS identified, the crude prevalence was 72.4 per 100,000 population, and 73.1 per 100,000 when age-standardized to the European population. CRA estimated that 96.7% of cases were identified. A latitudinal gradient was seen with MS prevalence increasing three-fold from the North (35°S) to the South (48°S). The gradient was non-uniform; females with relapsing—remitting/secondary-progressive (RRMS/SPMS) disease have a gradient 11 times greater than males with primary-progressive MS ( p < 1 × 10-7). DMS was significantly less common among those of Māori ethnicity. Conclusions: This study confirms the presence of a robust latitudinal gradient of MS prevalence in New Zealand. This gradient is largely driven by European females with the RRMS/SPMS phenotype. These results indicate that the environmental factors that underlie the latitudinal gradient act differentially by gender, ethnicity and MS phenotype. A better understanding of these factors may allow more targeted MS therapies aimed at modifiable environmental triggers at the population level.

Phaeomoniella chlamydospora (W. Gams, Crous. M.J. Wingfield & L. Mugnai) Crous & Gams (= Phaeoacremonium chlamydosporum) was isolated during the growing seasons of 2003-2004 from roots, trunks, and cordons of grapevines, including cvs. Cabernet Sauvignon, Merlot, Pinot noir, Thompson seedless, Ruby seedless and root stock 3309C, and Kober 5BB, from 10 locations in V, VI, VII, and metropolitan regions of Chile. P. chlamydospora was isolated from 82% of samples from vines 2 to 18 years old that showed decline symptoms in the field. Isolates were identified on the basis of a previous description (1) and internal transcribed spacer (ITS1-5.8S-ITS2) rDNA sequences identical to those of P. chlamydospora isolated from Vitis vinifera (culture CBS 22995, GenBank Accession No. AF 197973). P. chlamydospora is established as a member of the petri and esca disease complex and as a pathogen of grapevines (2,3). Pathogenicity tests were completed by injecting into the pith of 50 single-node, rooted cuttings of Pinot noir and 3309C, approximately 20 μl of a 106 conidia per ml suspension, obtained from four isolates from Chile and one from California. Ten control cuttings of Pinot noir and 3309C were injected with an equal volume of sterile distilled water. Twenty-four weeks after inoculations, all P. chlamydospora-inoculated cuttings exhibited dark streaking of the vascular tissue extending 40 to 45 mm from the point of inoculation. The vascular streaking observed in inoculated plants was identical to symptoms observed in declining vines in the vineyard. No symptoms were observed in the controls. P. chlamydospora was isolated from the region of vascular streaking in 85% of inoculated cuttings. P. chlamydospora was not isolated from the water-treated controls. The reisolated P. chlamydospora was verified with means of morphological characters and polymerase chain reaction amplification with the species-specific primers (3). P. chlamydospora is widespread and readily isolated from declining grapevines in Chile and other grape growing regions of the world. <p style="text-align: justify;"><span style="font-size: medium;"><strong>Aim: </strong>To determine which of the grapevine organ is the richest in stilbene, like the t-piceid, t-resveratrol and ε-viniferin and would it be useful as by-product of viticulture for processing stilbene concentrate.</span></p><p style="text-align: justify;"><span style="font-size: medium;"><strong>Methods and results: </strong>Nine organs, such as cane, buds, shoot tips, inflorescences, clusters at veraison, matured berry skins, seeds and cluster stems, of Merlot grafted on Teleki Kober 5BB rootstock were collected during a year and its ethanolic extractions were analyzed via HPLC-FLD. Stilbene content of the different organs occurred in a wide range, the lowest was 3.15 mg/kg dry weight (dw) measured in the seeds and the highest 2265 mg/kg dw in the buds.</span></p><p style="text-align: justify;"><span style="font-size: medium;"><strong>Conclusion:</strong> The present research demonstrated that different grapevine organs contain different amount of stilbene in the vineyard. The winter-buds, the roots and the matured cane internodes of Merlot are significantly richer in t-resveratrol and t-ε-viniferin than the green vegetative and generative parts we examined in one growing year. </span></p><p style="text-align: justify;"><span style="font-size: medium;"><strong>Significance and impact of the study: </strong>According to our knowledge there was no stilbenes measuring in several plant organs of a vine cultivar in field study, in vineyard. Our research pointed out the pruned cane could be primary source of stilbene of the health-industry. Beside that grape root is a by-product of grapevine nursery is also rich in stilbene. </span></p>
