SummaryYoga is known to induce beneficial effects on physiological, biochemical and mental functions in man. Its effects on blood coagulation are not known. A study was conducted in seven previously untrained male adults who underwent a combination of yogic exercises, daily for one hour, over a period of four months. Parameters of blood coagulation were estimated before and after the end of yoga training. The following changes were observed: Fibrinolytic activity increased significantly with a concomitant fall in fibrinogen; activated partial thromboplastin time and platelet aggregation time were prolonged; blood and plasma platelets showed a rise; and both haemoglobin and heamatocrit were raised at the end of the training.These findings suggest that yoga induces a state of blood hypocoagulability. The impact of yoga on prevention of cardiovascular and thrombotic disorders is obvious.

Введение. Актуальность темы исследования обусловлена проблемой борьбы с тромбозами и тромбоэмболиями безопасными для организма методами. Во многих растениях обнаружены антикоагулянты разной природы (гепариноподобные, пептиды). Цель исследования - изучение возможности проявления синергических эффектов на антикоагулянтную и фибринолитическую активность крови и процессы полимеризации фибрина экстракта из корней пиона «Иван Горожанкин» в сравнительном аспекте с действием экстракта из корней пиона «молочноцветковый». Методика. Объектом исследования служили корни пионов «Иван Горожанкин» и «молочноцветковый», произрастающих в Ботаническом саду МГУ. Пион «Иван Горожанкин» был создан скрещиванием пиона «молочноцветкового» и «лекарственного» Разработаны методы получения экстрактов из корней различных пионов. При различных разведениях экстрактов (0.1, 1, 5%) определены антикоагулянтная активность по тестам, характеризующим внутренний, внешний и общий пути свертывания крови, а также степень полимеризации фибрина плазмы крови крыс. Для сравнения был использован стандартный препарат низкомолекулярного гепарина (LMWH) животного происхождения фирмы «Celsus» (США). Проведены выделение и очистка активного начала (гепариноидов) из сухих препаратов и измерены их активности. Pезультаты. Показано, что экстракты из обоих препаратов пионов обладали антикоагулянтной и суммарной фибринолитической активностью на нестабилизированном фибрине, но в разной степени. В экстрактах из корней пиона «Иван Горожанкин» отмечались преимущественные синергические эффекты, а именно превышение антикоагулянтной активности на 20-30%, суммарной фибринолитической - на 18% по сравнению с таковыми, отмечаемыми в экстрактах из корней пиона «молочноцветковый». Подобные результаты выявлены и при изучении степени полимеризации фибрина под влиянием очищенных препаратов из пионов. Рассмотрены возможные механизмы активирующего действия экстракта из пиона «Иван Горожанкин» на антикоагулянтные свойства плазмы, суммарную фибринолитическую активность и степень полимеризации фибрина. Это связано с блокадой активности тромбина и факторов внутреннего механизма свертывания крови. При этом антикоагулянтный эффект от применения экстракта из пиона «Иван Горожанкин» по тесту APTT (activated partial thromboplastin time) превышал на 20-30% ту же активность, выявленную у пиона «молочноцветковый», которая соответствовала антикоагулянтной активности препарата сравнения LMWH. В экстракте из пиона «Иван Горожанкин» впервые обнаружено наличие антикоагулянтного гепариноподобного вещества. Заключение. Впервые установлена способность экстракта из корней пиона «Иван Горожанкин» проявлять синергические антикоагулянтные и фибриндеполимеризационные эффекты, превышающие таковые у экстракта из пиона «молочноветковый». На основе полученных данных возникает необходимость исследования пиона «Иван Горожанкин» в качестве антитромботического, а возможно, и антиатеросклеротического агента. Introduction. The research topic is relevant due to the problem of safely combating thrombosis and thromboembolism. Anticoagulants of various kinds, e.g., heparin-like and peptides, have been found in many plants. Aim. To investigate the possibility of synergistic effects on the blood anticoagulant and fibrinolytic activity and on processes of fibrin polymerization by an extract from the roots of the «Ivan Gorozhankin» peony compared with the root extract from «Paeonia lactiflora». Methods. The focus of the study was the roots of the “Ivan Gorozhankin” peony and the Paeonia lactiflora growing in the Botanical Garden of the Moscow State University. The “Ivan Gorozhankin” peony was created by crossing P. lactiflora and the “medicinal” peony. Methods for obtaining extracts from the roots of various peonies have been developed. In 1%, 3%, and 5% dilutions of the extracts, the anticoagulant activity was determined according to tests characterizing the internal, external and general blood coagulation pathways, as well as by the degree of polymerization of rat blood plasma fibrin. For comparison, we used a standard preparation of low molecular weight heparin (LMWH) of animal origin (Celsus, USA). Isolation and purification of the active substances, heparinoids, were isolated from dry preparations and purified, and their activities were measured. Results. Extracts from both peony preparations had anticoagulant and total fibrinolytic activity on unstabilized fibrin, but to different extents. In the extracts from the roots of the “Ivan Gorozhankin” peony, preferential synergistic effects were noted, namely, the anticoagulant activity was higher by 20-30%, and the total fibrinolytic activity was higher by 18% compared to those of extracts from Paeonia lactiflora roots. Similar results were obtained when studying the degree of fibrin polymerization as influenced by purified peony preparations. Possible mechanisms of the activating action of the «Ivan Gorozhankin» peony extract on the anticoagulant properties of plasma, the total fibrinolytic activity, and the degree of fibrin polymerization are considered. This action is due to the inhibition of thrombin activity and factors of the internal mechanism of blood coagulation. According to the activated partial thromboplastin time (APTT) test, the anticoagulant effect of extracts from the «Ivan Gorozhankin» peony exceeded by 20-30% the activity of Paeonia lactiflora extract, which corresponded to the anticoagulant activity of the LMWH comparator drug. Using the described biochemical methods, the presence of an anticoagulant heparin-like substance in an extract from the peony «Ivan Gorozhankin» has been discovered. Conclusion. For the first time, the ability of an extract from the roots of the «Ivan Gorozhankin» peony to exhibit synergistic anticoagulant and fibrin-depolymerization effects was demonstrated. These effects exceeded those of the Paeonia lactiflora extract. Based on these data, it appears necessary to study the «Ivan Gorozhankin» peony as an antithrombotic, and possibly as an anti-atherosclerotic agent.

Цель исследования - изучить состояние системы гемостаза при хронической интоксикации хлоридом никеля, исследовать взаимосвязь показателей гемокоагуляции с процессами липопероксидации у крыс в эксперименте. Методика. Опыты проводили на крысах-самцах Вистар (n=50, 230-250 г). Раствор NiCl2 (5 мг/кг) вводили внутрижелудочно ежедневно в течение 2 нед, 1 и 2 мес. По завершении эксперимента исследовали состояние тромбоцитарного и коагуляционного звеньев гемостаза, антикоагулянтную и фибринолитическую активность крови, а также определяли активность процессов перекисного окисления липидов и антиоксидантных ферментов. Результаты. Установлено, что через 2 нед и 1 мес интоксикации у крыс отмечались гиперкоагуляционные изменения показателей свертывающей системы крови: повышение агрегационной активности тромбоцитов, увеличение концентрации фибриногена, снижение активированного частичного тромбопластинового времени (АЧТВ) и протромбинового времени. В этот период регистрировалось увеличение антитромбиновой и фибринолитической активности крови. Через 2 мес наблюдалось подавление активности клеточного звена гемостаза - тромбоцитопения, ослабление степени АДФ-индуцируемой агрегации тромбоцитов. Выявлялась тенденция к уменьшению концентрации фибриногена. На фоне снижения АЧТВ и тромбинового времени отмечалось увеличение протромбинового времени. В то же время регистрировалось угнетение противосвертывающего звена системы гемостаза (снижалась активность антитромбина III), наблюдалось истощение резервных возможностей фибринолитического звена (замедление фXIIа-зависимого эуглобулинового лизиса) и увеличение содержания растворимых фибрин мономерных комплексов, что свидетельствует о наличии тромбинемии. Через 2 нед, один и два месяца интоксикации у животных выявлялись корреляционные связи между основными показателями системы гемостаза и активностью процессов перекисного окисления липидов и антиоксидантных ферментов. Заключение. Полученные данные подтверждают наличие взаимосвязи активности процессов липопероксидации и системы гемостаза, в том числе при хронической никелевой интоксикации. Результаты исследования позволяют рекомендовать применение антиоксидантов для разработки способов коррекции гемостатических сдвигов при воздействии на организм тяжелых металлов. The aim. To study the state of the hemostasis system in chronic nickel intoxication and to investigate the relationship between hemocoagulation indices and lipoperoxidation processes in rats. Methods. Experiments were carried out on male Wistar rats (n=50, 230-250 g). A solution of nickel chloride (5 mg/kg) was administered daily intragastrically for two weeks, one and two months. At the end of the experiments, indices of platelet and coagulation hemostasis systems, anticoagulant and fibrinolytic activity of blood plasma, and activities of lipid peroxidation and antioxidant enzymes were studied. Results. Hypercoagulative changes in indices of the coagulation system were observed in rats after two weeks and one month of intoxication, including increased platelet aggregation and fibrinogen concentration and shortened activated partial thromboplastin time and prothrombin time. During the same period, increased antithrombin and fibrinolytic activities were observed. The depressed activity of the cellular component of hemostasis evident as thrombocytopenia and impaired ADP-induced platelet aggregation was detected after two months of intoxication. A tendency to decrease in fibrinogen concentration was observed. The shortened activated partial thromboplastin time and thrombin time were associated with prolonged prothrombin time. At the same time, inhibition of the anticoagulant component of hemostasis (decreased antithrombin III activity), exhaustion of the fibrinolysis system reserve (delayed fXIIa-dependent euglobulin lysis), and a significant increase in soluble fibrin monomeric complexes indicative of thrombinemia were observed. After two weeks, one and two months of nickel intoxication, a correlation was found between the major indices of the hemostasis system and the activities of lipid peroxidation and antioxidant enzymes. Conclusion. The study confirmed a relationship between the lipid peroxidation activity and the hemostasis system, specifically in chronic nickel intoxication. This result allows to recommend the use of antioxidants in developing methods for correction of hemostatic induced affected by heavy metals.

Bleeding time in rats was markedly prolonged after the adminstration of the water extract of Hsien-Ho-T'sao. This antihemostatic effect was more marked in the group of i.p. injection of the drug than in the group of p.o. administration for 2 to 7 consecutive days. Blood coagulation studies showed that plasma prothrombin time, activated partial thromboplastin time and stypven time were prolonged, while thrombin time adnd fibrinogen level were not changed. The thromboelastographic recording showed that reation time was prolonged and maximal elasticity of clot was decreased. In addition, ADP- and collagen- induced aggregations of platelet-rich plasma was suppressed. In conclusion, the prolongation of the bleeding time might be due to both anticoagulant and antiplatelet action of the drug.

Background: Dextran is a branched polysaccharide and one of the polymers, present in the biofilm matrix. The dextran plays a perilous role in dental plaque formation, which is involved in the development of some common oral diseases like dental caries. The dextran-hydrolyzing enzymes are under investigation to treat and manage the dental plaques. Aims and Objective: The present study reporting the preliminary observations on the effect of the use of dextranase-containing mouthwash (DMW) on dental plaque and oral health. Materials and Methods: DMW was prepared with food-grade dextranase, preservatives, gellingagents, and water as detailed. Four weeks of experimental design was employed in fourteen healthy volunteers. The selected volunteers were recommended to use DMW for at least twice a day. The plaque index (PI), probing depth (PD), gingival index (GI) and bleeding on probing (BOP) of the volunteer's teeth have been assessed before and after four weeks of DMW use. Results:The volunteers were insisted to use a DMW solution twice a day for four weeks. The PI, PD, GI, and BOP was measured before and after the treatment. The plaque index of the subject at baseline and after treatment was 2.22 ± 0.48, and 1.88 ± 0.50, respectively. PI was significantly reduced after the use of DMW solution for four weeks. The value of PD was 2.00 and 2.00 at baseline and after the use of DMW, respectively. The value of PD was not changed when compared to the baseline values. The sensory evaluation of DMW was performed using questionnaires. Conclusion: The preliminary study results suggested that the use of DMW solution for four weeks (twice a day) notably reduced the PI without any change in PD. However, GI and BOP values were not affected after the use of DMW. The participants, based on the sensory evaluation, accepted the prepared DMW solution. Additional detailed research on the impact of DMW on oral hygiene is needed to confirm the beneficial effects of DMW.

Gastric dilatation and volvulus syndrome is associated with changes in haemostatic profiles. The aims of this study were to compare selected haemostatic and fibrinolytic parameters between healthy dogs and dogs with gastric dilatation and volvulus syndrome, estimate the incidence of disseminated intravascular coagulation (DIC), and determine the most sensitive test for detection of DIC in these patients. Blood was collected from 22 dogs with gastric dilatation and volvulus syndrome, and nine healthy control dogs. Platelet counts, prothrombin time, activated partial thromboplastin time, fibrinogen concentrations and fibrin/fibrinogen degradation products were measured in all control dogs and patients with gastric dilatation and volvulus syndrome, before and after surgery. Significant differences between control dogs and patients were seen in activated partial thromboplastin time and fibrin/fibrinogen degradation products before surgery and all measured parameters after surgery. The incidence of DIC was 59%. The most sensitive tests for detection of DIC before surgery were those for activated partial thromboplastin time and fibrin/fibrinogen degradation products.

Acute decompression is associated with a shortening of the activated partial thromboplastin time (aPTT). This study was performed to examine whether this change in aPTT results from hypoxia or hypobaria. We exposed healthy adults on three separate occasions to 2 h of 1) hypoxic hypobaria (410 Torr, n = 5), 2) hypoxic normobaria (fractional inspired O2 tension = 0.11, n = 4), or 3) normoxic hypobaria (410 Torr breathing supplemental O2, n = 5). The aPTT shortened during hypoxic hypobaria and hypoxic normobaria (P less than 0.05) but was unchanged during normoxic hypobaria. The prothrombin and thrombin times, hematocrit, and concentrations of fibrinogen, total plasma protein, and fibrinogen-fibrin fragment E were unchanged. During hypoxic hypobaria biologic levels of prekallikrein, high-molecular-weight kininogen, and factors XII, XI, X, VII, V, and II were unchanged, but procoagulant VIII (VIII:C) increased 50% without an increase in VIII-related antigen levels (VIIIR:Ag). Fibrin monomer was not detected in any group. In one subject who became ill after 1.5 h of hypoxic normobaria aPTT shortened by 10 s; the platelet count decreased by 93,000/mm3; VIII:C increased fivefold, but VIIIR:Ag only increased three-fold. We conclude that it is the hypoxia which shortens aPTT during acute decompression to 410 Torr and speculate that it results from an increase in plasma VIII:C-like activity.

AbstractObjective:Abnormalities in neurotransmission via N-methyl-d-aspartic acid receptor (NMDAR) play a role in the pathophysiology of neuropsychiatric disorders. The impact of repetitive transcranial magnetic stimulation (rTMS) on NMDAR-related amino acids remains unknown. We aim to investigate the effects of rTMS on NMDAR-related amino acids in serum of post-stroke patients.Methods:Ninety-five consecutive post-stroke patients with upper limb hemiparesis were recruited. In 27 patients, the Beck Depression Inventory (BDI) score was 10 or higher. Twelve depressed patients underwent rehabilitation in combination with rTMS and 15 non-depressed patients underwent rehabilitation only without rTMS for 14 days. 1 Hz rTMS was applied to the primary motor area in the non-lesional hemisphere. BDI was conducted before and after treatment. Serum glutamine, glutamate, glycine, l-serine, and d-serine levels were measured before and after treatment.Results:There were no differences between depressed patients and non-depressed patients in clinical characteristics, levels of the five amino acids in serum, and the ratio of amino acids. However, in 27 depressed patients, there was a significant correlation between levels of glutamate in serum and BDI (ρ = 0.428, p = 0.026). BDI decreased significantly in depressed patients after treatment with or without rTMS. d-serine decreased in the rehabilitation with rTMS group, but increased in the rehabilitation without rTMS group. l-serine increased in the rehabilitation with rTMS group, but decreased in the rehabilitation without rTMS group.Conclusion:The results suggest that rTMS can modulate NMDAR-related amino acids in blood, producing beneficial effects.

Abstract Abstract 4222 Background: Activated partial thromboplastin time (aPTT) and prothrombin time (PT) are commonly used to screen for coagulation factor deficiencies. Shorter aPTT is also a risk marker for incident and recurrent venous thromboembolism (VTE). Genetic factors influencing aPTT and PT are not well understood. So far only one genome-wide association study (GWAS) has been reported for aPTT in 1,477 participants and none for PT. Methods: We conducted a GWAS for the aPTT in 9,240 European Americans (EAs) from the Atherosclerosis Risk in Communities (ARIC) study and for PT in 1,221 EAs from the Genetic Study of Three Population Microisolates in South Tyrol (MICROS). Replication was assessed by in silico analysis in MICROS (aPTT, n=1,215) and the Lothian Birth Cohorts (LBC) (LBC1936 (aPTT and PT, n=925-989), LBC1921 (aPTT, n=445)), and by de novo genotyping in the Caerphilly study (aPTT, n=882). Subjects on anticoagulants were excluded. Genotyping was conducted with the Affymetrix single nucleotide polymorphism (SNP) array 6.0 or Illumina HumanHap300/370 and imputed to ∼2.5 million HapMap SNPs. SNPs with imputation quality score < 0.3 or minor allele frequencies ≤1% were excluded from data analysis. The imputed SNP dosages were analyzed in linear regression adjusted for age, sex, and field center, where applicable. Results: Five loci were associated with aPTT at genome-wide significance of p<5×10-8 that have not been previously reported: F5 (1q23, top SNP rs9332701, missense, β (effect size associated with one copy increase in minor allele)=0.54, p=3.7×10-8), F11 (4q35, rs1593, intronic, β =0.54 and 0.36, p=1.25×10-17 and 2.0×10-8 before and after adjusting for known variants in F11), NSD1 (5q35.2-q35.3, rs11950938, intronic, β =1.00 and 1.21, p=1.11×10-15 and 1.5×10-22 before and after adjusting for known variants in F12 of the same region), C6orf10 (6p21.3, rs2050190, intronic, β =-0.25, p=1.3×10-8), and ABO (9q34.1-q34.2, rs8176704, intronic, tag for A2 group, β =0.19 and 0.89, p=0.02 and 4.3×10-24 before and after adjusting for O blood group). Three of the five loci replicated in at least one replication sample and the other two were directionally consistent in 3 replication samples. Furthermore, meta-analysis pooling the discovery and replication GWAS samples yielded two additional independent loci at chromosomes 1q23 (F5, best SNP rs6028, coding-synonymous, β =0.23, p=5.97×10-9) and 15q25.3 (AGBL1, rs2469184, intronic, β =0.16, p= 4.24×10-8), with consistent associations across studies. The signals at the F5 region were not due to FV Leiden (rs6025, p=0.46 in ARIC). We also confirmed previously reported loci in KNG1, HRG, F11, F12, and ABO (O group). For PT, novel associations from two gene regions reached genome-wide significance in MICROS: F7 (top SNP rs3093253, within an exon but not translated, β =-5.44, p=8.2×10-19) and PROCR (rs6060244, near PROCR, β =4.11, p=3.5×10-9). Both loci replicated in LBC1936. Conclusions: In this large GWAS, six of the nine novel loci associated with the aPTT and PT are coagulation-related and the other three (NSD1, C6orf10, and AGBL1) are new candidate genes not directly involved in coagulation. The C6orf10 gene interacts with TNF-a at the transcription level and was previously associated with inflammatory diseases. These findings may be relevant to the prevention and treatment of coagulation disorders including VTE. Disclosures: No relevant conflicts of interest to declare.

The effects of single and multiple moxibustions on platelet function, blood coagulation and fibrinolytic activity in ddY mice were studied. The increase in platelet aggregation and ATP-release after a single moxibustion was dependent on moxa weight and the kind of platelet stimulus. Blood coagulative activity tended to increase in the early phase after a single moxibustion. However, multiple moxibustions maintained the homeostasis on blood coagulation and fibrinolytic activiity. This investigation suggests that the effects of moxibustion on platelet functions and coagulative and fibrinolytic activities cause an enhancement of the phagocytic activity in the host defense mechanism.
