Abstract Background This study presents the results of the minimum inhibitory concentration (MIC) assay of a series of nosodes: namely Escherichia coli, Klebsiella pneumoniae, Salmonella typhi, Neisseria gonorrhoeae, and Candida albicans. Each was tested against its corresponding infection as well as cross infections. Methods In-vitro efficacy of polyvalent nosodes was tested using the MIC assay technique. The nosodes, namely C. albicans polyvalent nosode (35c, 100c), N. gonorrhoeae (35c), K. pneumoniae (35c, 100c), E. coli polyvalent nosode (35c, 100c) and Salmonella typhi polyvalent nosode (30c, 100c), were tested along with positive and negative controls. Nosodes were studied in different potencies and at 1:1 dilution. Results C. albicans polyvalent nosode 35c, 100c, N. gonorrhoeae 35c, and positive control amphotericin B showed inhibition of the growth of C. albicans species. K. pneumoniae 35c, E. coli polyvalent nosode 100c, and meropenem (positive control) showed inhibition of the growth of K. pneumoniae; this effect was not seen with ceftriaxone, ofloxacin and amoxicillin antibiotics. E. coli polyvalent nosode 30c in 10% alcohol (direct and dilution 1:1) and the positive controls ciprofloxacin, ofloxacin, and amoxicillin showed inhibition of the growth of E. coli. The S. typhi polyvalent nosode 30c in 10% alcohol showed inhibition of growth of S. typhi. Conclusion This study reveals that the tested nosodes exhibited antibacterial potential against the corresponding micro-organisms and against other selected organisms studied using this assay.

Klebsiella pneumoniae, Shigella dysenteriae, Salmonella typhi, Escherichia coli, dan Candida albicans merupakan mikroorganisme yang paling sering menginfeksi di Indonesia. Penyakit infeksi dapat disembuhkan serta diminimalkan penularannya menggunakan antibiotik. Upaya pencarian senyawa antimikroba dari alam diharapkan mampu mengurangi pengaruh negatif antibiotik. Tanaman yang diduga memiliki kandungan antimikroba salah satunya pohon sengon. Penelitian ini bertujuan untuk mengetahui kemampuan ekstrak kayu ranting sengon (Falcataria moluccana) yang terinfeksi jamur U. tepperianum dengan pelarut etil asetat dan metanol sebagai antimikroba. Metode pengujian yang digunakan Kirby bauer dengan perlakuan 9%, 9.5%, 10%, 10.5%, dan 11%. Hasil penelitian ekstrak etil asetat kayu ranting sengon sakit menunjukkan adanya aktivitas antimikroba terhadap S. dysenteriae pada semua konsentrasi dan K. pneumoniae pada konsentrasi 11%, tidak ada aktivitas antimikroba terhadap E. coli. Ekstrak metanol kayu ranting sengon sakit menunjukkan adanya aktivitas antimikroba terhadap K. pneumoniae pada konsentrasi 10%, 10.5%, 11%, dan C. albicans pada semua konsentrasi, namun tidak menunjukkan aktivitas antimikroba terhadap E. coli, S. typhi, dan S. dysenteriae. Dapat disimpulkan bahwa ekstrak etil asetat kayu ranting sengon sakit dapat menghambat pertumbuhan S. dysenteriae dan K. pneumoniae. Ekstrak metanol kayu ranting sengon sakit dapat menghambat pertumbuhan K. pneumoniae dan C. albicans.

Several 6- and 7-alkoxy-2-oxo-2H-chromene-4-carbaldehydes were prepared from corresponding alkyl ethers of 6- and 7-hydroxy-4-methyl-2-oxo-2H-chromen-2-ones by oxidation using selenium dioxide. 6- and 7-Alkoxy-4-methyl-2H-chromenes were obtained with yields of 57-85%. Corresponding 4-carbaldehyde derivatives were prepared with yields of 41-67%. Thiosemicarbazones of these aldehydes with D-galactose moiety were synthesized by reaction of these aldehydes with N-(2,3,4,6-tetra-O-acetyl-β-Dgalactopyranosyl) thiosemicarbazide with yields of 62-74%. These thiosemicarbazones were screened for their antibacterial and antifungal activities in vitro against bacteria, such as Staphylococcus aureus, Escherichia coli, and fungi, such as Aspergillus niger, Candida albicans. Several compounds exhibited strong inhibitory activity with MIC values of 0.78- 1.56 μM, including 8a (against S. aureus, E. coli, and C. albicans), 8d (against E. coli and A. niger), 9a (against S. aureus), and 9c (against S. aureus and C. albicans).

Objectif : L’objectif de ce travail était d’évaluer in vitro l’activité antimicrobienne des extraits de feuilles et tige de Jatropha multifida sur la croissance de Candida albicans, Escherichia coli et Staphylococcus aureus, puis d’évaluer in vivo la toxicité de cette plante. Méthodologie et résultats : Les méthodes de diffusion en milieu gélosé et de microdilution en milieu liquide ont été utilisées pour évaluer l’effet antimicrobien. Une étude en subaigüe était réalisée afin d’explorer les effets toxiques de l’extrait aqueux des feuilles. Les résultats des tests antimicrobiens montrent une activité des extraits de feuilles et tige de J. multifida sur la croissance des souches utilisées avec des diamètres de zones d’inhibition allant de 8 à 25 mm et des concentrations minimales inhibitrices (CMI) variant de 0,039 mg/mL à 1,25 mg/mL à l’exception des souches de E. coli qui sont résistantes aux extraits de la tige. L’administration en subaigüe de l’extrait aqueux des feuilles de J. multifida à la dose de 600 mg/kg entraîne une perte significative de poids chez les souris. Conclusion et applications des résultats : Les extraits aqueux, éthanolique et hydroéthanolique des feuilles et tige de J. multifida possèdent d’activité antimicrobienne et pourraient être utilisés dans le traitement des Candidoses à C. albicans et des infections à S. aureus. Mais l’essai de toxicité subaigüe montre que l’extrait aqueux de la plante serait toxique. Des études toxicologiques approfondies restent donc nécessaires sur ces extraits afin de mieux élucider leur inocuité. But the subacute toxicity test shows that the aqueous extract of the plant would be toxic. Extensive toxicological studies therefore remain necessary on these extracts in order to better elucidate their safety. O conhecimento da microbiota que compõe as diferentes áreas do organismo tem importância reconhecida para a compreensão de doenças infecciosas que podem acometer os avestruzes, embora se disponha de dados limitados acerca deste assunto na literatura. Foi objetivo deste estudo determinar as espécies de microrganismos (bactérias aeróbias e fungos) que compõem a microbiota normal de avestruzes. Para tanto, foram coletadas amostras de cloaca (N=50) e orofaringe (N=50) de avestruzes hígidos de um criadouro. Das amostras de cloaca, foram isolados Escherichia coli (76% das amostras positivas), Bacillus spp. (60%), Streptococcus spp. (18%), Staphylococcus coagulase-negativo (16%), Pseudomonas aeruginosa (8%), Rhodotorula spp. (8%), dentre outros microrganismos isolados em cultura pura ou em associação com outras bactérias e/ou fungos. Das amostras de orofaringe, foram isolados E. coli (74% das amostras positivas), Candida albicans (44%), Bacillus spp. (38%), Staphylococcus coagulase-negativo (32%), Klebsiella pneumoniae (32%), Rhodotorula spp. (8%), Criptococcus spp. (4%), dentre outros microrganismos isolados em cultura pura ou em associação com outras bactérias e/ou fungos. Verificou-se predominância de bactérias Gram negativas em relação às Gram positivas, nas microbiotas da cloaca e orofaringe. Verificou-se freqüência de ocorrência semelhante entre bactérias Gram negativas nas microbiotas da cloaca e orofaringe, bem como de bactérias Gram positivas nestes mesmos sítios. Observou-se maior ocorrência de leveduras em amostras de orofaringe quando comparadas com as de cloaca.

This study investigates GT-1 (also known as LCB10-0200), a novel-siderophore cephalosporin, inhibited multidrug-resistant (MDR) Gram-negative pathogen, via a Trojan horse strategy exploiting iron-uptake systems. We investigated GT-1 activity and the role of siderophore uptake systems, and the combination of GT-1 and a non-β-lactam β-lactamase inhibitor (BLI) of diazabicyclooctane, GT-055, (also referred to as LCB18-055) against molecularly characterised resistant Escherichia coli, Klebsiella pneumoniae and Acinetobacter spp. isolates. GT-1 and GT-1/GT-055 were tested in vitro against comparators among three different characterised panel strain sets. Bacterial resistome and siderophore uptake systems were characterised to elucidate the genetic basis for GT-1 minimum inhibitory concentrations (MICs). GT-1 exhibited in vitro activity (≤2 μg/mL MICs) against many MDR isolates, including extended-spectrum β-lactamase (ESBL)- and carbapenemase-producing E. coli and K. pneumoniae and oxacillinase (OXA)-producing Acinetobacter spp. GT-1 also inhibited strains with mutated siderophore transporters and porins. Although BLI GT-055 exhibited intrinsic activity (MIC 2–8 μg/mL) against most E. coli and K. pneumoniae isolates, GT-055 enhanced the activity of GT-1 against many GT-1–resistant strains. Compared with CAZ-AVI, GT-1/GT-055 exhibited lower MICs against E. coli and K. pneumoniae isolates. GT-1 demonstrated potent in vitro activity against clinical panel strains of E. coli, K. pneumoniae and Acinetobacter spp. GT-055 enhanced the in vitro activity of GT-1 against many GT-1–resistant strains.

Se evaluó la actividad antibacteriana in vitro de aceites esenciales de diferentes especiesdel género Citrus frente a cepas ATCC de Staphylococcus aureus, Staphylococcus epidermidis,Klebsiella pneumoniae, Pseudomonas aeruginosa y Escherichia coli, determinandola concentración mínima inhibitoria (CMI) y la concentración mínima bactericida(CMB). Las bacterias se replicaron en medios de agar y caldos específicos. Se determinóel momento de máxima densidad óptica (DO620) para emplearlo como tiempode incubación; luego se hicieron pruebas de evaluación de sensibilidad con la exposiciónde las cepas a concentraciones a 1000 g/mL del extracto en caldo. Para solubilizarse empleó dimetilsulfóxido (DMSO) al 1%. Posteriormente, se le determinó laconcentración mínima inhibitoria mediante metodologías de microdilución en caldoy la concentración mínima bactericida. Encontrándose una actividad de los aceitesesenciales del género Citrus, con valores de CMI ≥ 600 mg/mL frente a S. aureus,S. epidermidis, K. pneumoniae, P. aeruginosa y E. coli. En función a los resultados obtenidos,se concluye que las diferentes especies del género Citrus son consideradas comopromisorias para el control del componente bacteriano.

Мета роботи. Визначення антимікробної активності бедринцю ломикаменевого підземних органів екстракту густого. Матеріали і методи.  Oб’єктoм для дoслідження обрано бедринцю ломикаменевого підземних органів екстракт густий (БЕГ-1, одержаний екстрагуванням 75 % етанолом і БЕГ-2, екстрагент 85 % етанол). В якості тест-культур використовували 5 музейних штамів: грампозитивні мікроорганізми Staphylococcus aureus АТСС 6538, спорову культуру Bacillus subtilis АТСС 6633, грамнегативну культуру Escherichia coli АТСС 25922 та Pseudomonas aeruginosa АТСС 9027. Антифунгальну дію з’ясовували відносно Candida albicans АТСС 885-653. Бактеріостатичні властивoсті дoсліджуваних oб’єктів встанoвлювали за результатами росту еталонних штамів мікроорганізмів у нативному водному розчині бедринцю та в розведені 1:2 та 1:4 в м'ясо-пептонному бульйоні; бактерицидні – за відсутністю росту вмісту пробірок із розведенням екстракту на щільних пoживних середoвищах (м’ясо-пептонний агар – МПА) для грампозитивних мікроорганізмів S. aureus, B. subtilis, для грамнегативної культури E. coli, P. аeruginosa. Агар Сабуро використовували для дріжджеподібних грибів роду Candida (C. аlbicans). Результати й обговорення. Бедринцю ломикаменевого підземних органів екстракт густий має широкий спектр антибактеріальної активності. Нативний екстракт БЕГ-1 по відношенню до тест-штамів S. aureus, B. subtilis, C. аlbicans проявляв бактерицидну дію. Бактеріостатичну дію він проявляв до P. aeruginosa, а по відношенню до E. coli антимікробного ефекту не виявлено. Екстракти в розведені 1:2 та 1:4 проявили бактерицидну дію відносно B. subtilis, бактеріостатичну – до S. aureus, C. albicans і не виявлено протимікробної дії до E. coli та P. aeruginosa. Нативний зразок БЕГ-2 по відношенню до усіх досліджуваних 5 тест–штамів проявив бактерицидну активність; у розведені 1:2 та 1:4 проявляв бактерицидну та бактеріостатичну дію відносно S. aureus, B. subtilis, C. albicans. Антимікробна дія по відношенню до E. coli та P. aeruginosa не виявлена. Антимікрoбну активність нативного бедринцю ломикаменевого екстракту густого також вивчали у дoслідах in vitro методом дифузії в агар –  метод “колодязів”. Штами S. aureus, C. albicans, B. subtilis, P. aeruginosa, E. coli проявили чутливість до нативного бедринцю ломикаменевого підземних органів екстракту густого. Грампозитивні бактеріальні штами S. aureus,а також гриби C. albicans, B. subtilis є найбільш чутливими до  БЕГ-2, а БЕГ-1 проявляв порівняно меншу чутливість. Грамнегативні культури P. aeruginosa та E. coli також проявили помірну чутливість до  БЕГ-1, але значно більшу чутливість до БЕГ-2. Висновки. 1. Експериментально встановлено, що обидва досліджувані зразки бедринцю ломикаменевого підземних органів екстракту густого проявляють антибактеріальну активність. 2. Бедринцю ломикаменевого підземних органів екстракт густий проявляв більш виражену антимікробну активність по відношенню до грампозитивної мікрофлори, тому є перспективним для створення лікарського засобу з антимікробними властивостями.

Colistin is being considered as “the last ditch” treatment in many infections caused by Gram-negative stains. However, colistin is becoming increasingly invalid in treating patients who are infected with colistin-resistant Escherichia coli (E. coli) and Klebsiella Pneumoniae (K. pneumoniae). To cope with the continuous emergence of colistin resistance, the development of new drugs and therapies is highly imminent. Herein, in this work, we surprisingly found that the combination of quercetin with colistin could efficiently and synergistically eradicate the colistin-resistant E. coli and K. pneumoniae, as confirmed by the synergy checkboard and time-kill assay. Mechanismly, the treatment of quercetin combined with colistin could significantly downregulate the expression of mcr-1 and mgrB that are responsible for colistin-resistance, synergistically enhancing the bacterial cell membrane damage efficacy of colistin. The colistin/quercetin combination was notably efficient in eradicating the colistin-resistant E. coli and K. pneumoniae both in vitro and in vivo. Therefore, our results may provide an efficient alternative pathway against colistin-resistant E. coli and K. pneumoniae infections.

Dialium angolense is used in Bagira for its various medicinal properties particularly in the management of infectious diseases. In this study, the methanol and aqueous extracts of leaves and fruits were evaluated for their in vitro antioxidant and antimicrobial properties and their in vivo toxicity on Cavia porcellus. The major phytochemical classes of extracts were screened using standard in-tube reactions. The antimicrobial study was tested on Candida albicans, Escherichia coli, Salmonella typhi, Staphylococcus aureus and Streptococcus pneumoniae using agar well diffusion and dilution methods, while the antioxidant activity was evaluated by a DPPH assay. For the acute toxicity study, animals (6/group) were orally given in a single dose 5000, 1000 or 15000 mg of extract/kg body weight (BW) then observed for 14 days. In sub-acute toxicity assays, 150 or 300 mg/kg BW/day were orally given, and animals observed for 28 days. Total phenolics and total flavonoids contents ranged 1.19 to 1.61 mg GAE.g-1 and 0.45 to 1.01 mg QEg-1, respectively. The extracts presented antioxidant activity with IC50 ranging 4.9 to 6.9 µg/mL. The minimal inhibitory concentration (MIC) on tested strains ranged from 1.9 to 500 µg/mL with the aqueous extract of fruits as a most active extract: MIC=1.9 µg/mL on E. coli and C. albicans. No signs of toxicity were noted during the acute and sub-acute toxicity assessments, suggesting a maximal tolerate doses (MDT) and LD50 > 15000 mg/kg BW. This study highlights the antioxidant and antimicrobial activities of Dialium angolense and suggests that further studies be directed towards the isolation of active compounds.

The present study was conducted to assess the in vitro activities of folk medicinal plants in combination with levofloxacin against TG1 and mutant KAM3-1(∆acrB-∆tolC) Escherichia coli strains. Plants were chosen based on their traditional use in combination with antibiotics among laymen. Standard protocols were followed to examine the antimicrobial activity of plant extracts and levofloxacin against E. coli in term of their minimum inhibitory concentrations (MICs) and to evaluate the plant extracts-levofloxacin interaction using checkerboard method. Among the twelve plants investigated, Thymus vulgaris, Zingiber officinale, Teucrium polium, Matricaria chamomilla and Curcuma longa had the best antimicrobial activities against E. coli strains with MIC values at 250 μg/ml. It is noteworthy to mention that other folk plants extracts reveled no effects against E coli strains. Furthermore, additive interactions were observed between levofloxacin and T. polium or T. vulgaris against E. coli wild-type TG1 strain. There was no antagonism being observed in this study. The detection of additive interaction between the extracts and levofloxacin demonstrates the prospective of these folk medicinal plants as a source of compounds to modulate antibiotic resistance.
