The objective of the paper is to explore the best phenophase for estimating the nitrogen contents of apple leaves, to establish the best estimation model of the hyperspectral data at different phenophases. It is to improve the apple trees precise fertilization and production management. The experiments were done in 20 orchards in the field, measured hyperspectral data and nitrogen contents of apple leaves at three phenophases in two years, which were shoot growth phenophase, spring shoots pause growth phenophase, autumn shoots pause growth phenophase. The study analyzed the nitrogen contents of apple leaves with its original spectral and first derivative, screened sensitive wavelengths of each phenophase. The hyperspectral parameters were built with the sensitive wavelengths. Multiple stepwise regressions, partial least squares and BP neural network model were adopted in the study. The results showed that 551 nm, 716 nm, 530 nm, 703 nm; 543 nm, 705 nm, 699 nm, 756 nm and 545 nm, 702 nm, 695 nm, 746 nm were sensitive wavelengths of three phenophases. R551+R716, R551*R716, FDR530+FDR703, FDR530*FDR703; R543+R705, R543*R705, FDR699+FDR756, FDR699*FDR756and R545+R702, R545*R702, FDR695+FDR746, FDR695*FDR746 were the best hyperspectral parameters of each phenophase. Of all the estimation models, the estimated effect of shoot growth phenophase was better than other two phenophases, so shoot growth phenophase was the best phenophase to estimate the nitrogen contents of apple leaves based on hyperspectral models. In the three models, the 4-3-1 BP neural network model of shoot growth phenophase was the best estimation model. The R2 of estimated value and measured value was 0.6307, RE% was 23.37, RMSE was 0.6274.

Apple leaf ADP-glucose pyrophosphorylase was purified 1436-fold to apparent homogeneity with a specific activity of 58.9 units mg–1. The enzyme was activated by 3-phosphoglycerate (PGA) and inhibited by inorganic phosphate (Pi) in the ADPG synthesis direction. In the pyrophosphorolytic direction, however, high concentrations of PGA (> 2.5 mm) inhibited the enzyme activity. The enzyme was resistant to thermal inactivation with a T0.5 (temperature at which 50% of the enzyme activity is lost after 5 min incubation) of 52°C. Incubation with 2 mm PGA or 2 mm Pi increased T0.5 to 68°C. Incubation with 2 mm dithiothreitol (DTT) decreased T0.5 to 42°C, whereas inclusion of 2 mm PGA in the DTT incubation maintained T0.5 at 52°C. DTT-induced decrease in thermal stability was accompanied by monomerisation of the small subunits. Presence of PGA in the DTT incubation did not alter the monomerisation of the small subunits of the enzyme induced by DTT. These findings indicate that binding of PGA renders apple leaf AGPase with a conformation that is not only more efficient in catalysis but also more stable to heat treatment. The physiological significance of the protective effect of PGA on thermal inactivation of AGPase is discussed.

Field trials were conducted in Hawkes Bay and Lincoln into methods of treating overwintering apple leaves to reduce ascospore production by the apple black spot pathogen Venturia inaequalis The leaf treatments comprised three levels of nutrient amendments (including a water control) and five levels of saprophytic fungal isolates (including a nofungus control) in a factorial design Leaves were left to overwinter on the orchard floor and in spring the V inaequalis ascospores released were trapped on glass slides and counted Ascospore numbers were reduced (Plt;005) by the leaf amendment urea which alone caused 73 reduction but not by the BioStarttrade; product The effect of fungal isolates was not significant (P012) although when combined with the water treatment the isolates Chaetomium Phoma and Epicoccum spp and Trametes versicolor reduced numbers of ascospores by 33 27 15 and 28 respectively compared to the nofungus control When combined with urea the Chaetomium isolate reduced ascospore numbers by 92 and 82 compared to the nil fungus/water control treatments in Hawkes Bay and Lincoln respectively indicating that this treatment has potential for reducing primary inoculum of apple black spot

The Generalized Logarithmic Series Distribution (GLSD) adds an extra parameter to the usual logarithmic series distribution and was introduced by Jain and Gupta (1973). This distribution has found applications in various fields. The estimation of parameters of generalized logarithmic series distribution was studied by the methods of maximum likelihood, moments, minimum chi square and weighted discrepancies. The GLSD was fitted to counts of red mites on apple leaves and it was observed that all the estimation techniques perform well in estimating the parameters of generalized logarithmic series distribution but with varying degree of non-significance.

The development of leaf spot, a non-pathogenic disorder specific to the cv. Golden Delicious, was accelerated in detached leaves (whether previously unmarked or showing leaf spot) when these were cultured in solutions of GA3 or GA4+7 at 10, 50 or 100 mg/l for 72 h. About 15% of the foliage on the trees exhibited leaf spot at the time selected leaves were detached for culture. In a second experiment with cultured leaves the interaction of PBA and GA4+7 applied simultaneously was highly synergistic in increasing the severity of the disorder when compared with the effect of either growth substance alone.
