The objective of the paper is to explore the best phenophase for estimating the nitrogen contents of apple leaves, to establish the best estimation model of the hyperspectral data at different phenophases. It is to improve the apple trees precise fertilization and production management. The experiments were done in 20 orchards in the field, measured hyperspectral data and nitrogen contents of apple leaves at three phenophases in two years, which were shoot growth phenophase, spring shoots pause growth phenophase, autumn shoots pause growth phenophase. The study analyzed the nitrogen contents of apple leaves with its original spectral and first derivative, screened sensitive wavelengths of each phenophase. The hyperspectral parameters were built with the sensitive wavelengths. Multiple stepwise regressions, partial least squares and BP neural network model were adopted in the study. The results showed that 551 nm, 716 nm, 530 nm, 703 nm; 543 nm, 705 nm, 699 nm, 756 nm and 545 nm, 702 nm, 695 nm, 746 nm were sensitive wavelengths of three phenophases. R551+R716, R551*R716, FDR530+FDR703, FDR530*FDR703; R543+R705, R543*R705, FDR699+FDR756, FDR699*FDR756and R545+R702, R545*R702, FDR695+FDR746, FDR695*FDR746 were the best hyperspectral parameters of each phenophase. Of all the estimation models, the estimated effect of shoot growth phenophase was better than other two phenophases, so shoot growth phenophase was the best phenophase to estimate the nitrogen contents of apple leaves based on hyperspectral models. In the three models, the 4-3-1 BP neural network model of shoot growth phenophase was the best estimation model. The R2 of estimated value and measured value was 0.6307, RE% was 23.37, RMSE was 0.6274.

Pectin recovered from mango peel biomass can be used as a potential source for pectic oligosaccharide hydrolysate with excellent probiotic growth-enhancing performance and prebiotic potentials. Consequently, the objectives of the current study were to optimise the enzyme hydrolysis treatment of mango peel pectin (MPP) and to evaluate the pectic oligosaccharide effects of Lactobacillus reuteri DSM 17938 and Bifidobacterium animalis TISTR 2195. Mango of “chok anan” variety was chosen due to its excessive volume of biomass in processing and high pectin content. The optimal treatment for mango peel pectic oligosaccharide (MPOS) valorisation was 24 h of fermentation with 0.3% (v/v) pectinase. This condition provided small oligosaccharides with the molecular weight of 643 Da that demonstrated the highest score of prebiotic activity for both of B. animalis TISTR 2195 (7.76) and L. reuteri DSM 17938 (6.87). The major sugar compositions of the oligosaccharide were fructose (24.41% (w/w)) and glucose (19.52% (w/w)). For the simulation of prebiotic fermentation, B. animalis TISTR 2195 showed higher proliferation in 4% (w/v) of MPOS supplemented (8.92 log CFU/mL) than that of L. reuteri (8.53 CFU/mL) at 72 h of the fermentation time. The main short chain fatty acids (SCFAs) derived from MPOS were acetic acid and propionic acid. The highest value of total SCFA was achieved from the 4% (w/v) MPOS supplementation for both of B. animalis (68.57 mM) and L. reuteri (69.15 mM). The result of this study therefore conclusively advises that MPOS is a novel pectic oligosaccharide resource providing the opportunity for the sustainable development approach through utilising by-products from the fruit industry.

Apple leaf ADP-glucose pyrophosphorylase was purified 1436-fold to apparent homogeneity with a specific activity of 58.9 units mg–1. The enzyme was activated by 3-phosphoglycerate (PGA) and inhibited by inorganic phosphate (Pi) in the ADPG synthesis direction. In the pyrophosphorolytic direction, however, high concentrations of PGA (> 2.5 mm) inhibited the enzyme activity. The enzyme was resistant to thermal inactivation with a T0.5 (temperature at which 50% of the enzyme activity is lost after 5 min incubation) of 52°C. Incubation with 2 mm PGA or 2 mm Pi increased T0.5 to 68°C. Incubation with 2 mm dithiothreitol (DTT) decreased T0.5 to 42°C, whereas inclusion of 2 mm PGA in the DTT incubation maintained T0.5 at 52°C. DTT-induced decrease in thermal stability was accompanied by monomerisation of the small subunits. Presence of PGA in the DTT incubation did not alter the monomerisation of the small subunits of the enzyme induced by DTT. These findings indicate that binding of PGA renders apple leaf AGPase with a conformation that is not only more efficient in catalysis but also more stable to heat treatment. The physiological significance of the protective effect of PGA on thermal inactivation of AGPase is discussed.

Field trials were conducted in Hawkes Bay and Lincoln into methods of treating overwintering apple leaves to reduce ascospore production by the apple black spot pathogen Venturia inaequalis The leaf treatments comprised three levels of nutrient amendments (including a water control) and five levels of saprophytic fungal isolates (including a nofungus control) in a factorial design Leaves were left to overwinter on the orchard floor and in spring the V inaequalis ascospores released were trapped on glass slides and counted Ascospore numbers were reduced (Plt;005) by the leaf amendment urea which alone caused 73 reduction but not by the BioStarttrade; product The effect of fungal isolates was not significant (P012) although when combined with the water treatment the isolates Chaetomium Phoma and Epicoccum spp and Trametes versicolor reduced numbers of ascospores by 33 27 15 and 28 respectively compared to the nofungus control When combined with urea the Chaetomium isolate reduced ascospore numbers by 92 and 82 compared to the nil fungus/water control treatments in Hawkes Bay and Lincoln respectively indicating that this treatment has potential for reducing primary inoculum of apple black spot

Long term use of aspirin for prevention of cardiovascular events is limited by its gastrointestinal adverse effects, and, therefore, patients at high risk should receive gastroprotective agents. However, gastroprotective drugs also have a few adverse effects that require searching for safe and effective alternatives. The composites combining the swelling properties of clays and the biological activity of polysaccharides seem to be promising candidates for gastrointestinal protection. This study aimed to characterize pectin/montmorillonite composites and evaluate their protective effect on the gastrointestinal tract of rats taking aspirin over a long period. A series of composites based on montmorillonite (ММТ) and low esterified pectin (Pec) was synthesized using the adsorption-precipitation method. The polysaccharide to clay weight ratios were 1:19, 1:9, and 1:4. The composites were characterized using Thermogravimetric Analysis (TGA), Infrared Spectroscopy (IR), Scanning Electron Microscopy (SEM), and X-Ray Diffraction (XRD). Pectin was completely fixed on MMT, and the polysaccharide content in the composites was approximately 5, 10, and 20 wt%, respectively. The shifting of the absorption band of the C=O group of the pectin indicated the interaction of the polysaccharide with the clay, confirming effective immobilization of Pec on MMT. Modification with pectin changed the morphology and structure of the MMT due to the surface coating and intercalation into the interlayer space. The composites swelled in acidified water (pH = 2.0), and their swelling ability was higher to compare with unmodified MMT. The sorption capacity of Pec/MMT composites towards acetylsalicylic acid (ASA) was decreased from 6.8 to 1.0 mg g-1 with increasing of pectin content from 5 to 20 wt%. The hybrid composites promoted the protection of the gastrointestinal tract of rats, which were administered ASA with Pec/MMTs for 16 days. Protective properties of the Pec/MMT have been improved with increasing pectin content from 5 to 20 wt%.
