The nutrition of grazing ewes during pregnancy and lactation: a comparison of alkane-based and chromium/in vitro-based estimates of herbage intake

The n-alkane and chromium/in vitro procedures for estimating herbage intake were compared in grazing ewes during late pregnancy, early lactation, and mid-lactation. To ensure differences in herbage intake, the ewes were grazed in 4 plots of phalaris-dominant pasture at 2 levels of stocking: 17.1 ewes/ha and 30.8 ewes/ha. To investigate whether either procedure for estimating herbage intake was influenced by supplement consumption, half of the ewes at each stocking level received 500 g/day air-dry of a pelletted supplement (1 : 1 milled oat grain : sunflower meal). Supplement intakes were estimated using tritiated gypsum as a marker. During intake measurement periods, ewes were dosed twice daily with both alkane capsules and capsules containing chromium sesquioxide. For the last 6 days of the 12-day dosing period, rectal faecal samples were taken twice daily, immediately before the dosing. Over these same periods, wether sheep fitted with faecal collection harnesses were similarly dosed and sampled, and their total faecal output collected to establish the faecal recovery of chromium and the alkanes. Herbage intakes were estimated using the C27/C28, C29/C28, C31/C32, and C33/C32 alkane pairs. Estimates of intake based on the shorter alkane pairs were lower than those estimated with the C33/C32 alkane pair, by amounts which differed between the periods. Evidence is presented that estimates based on the last pair of alkanes (C33/C32) are the most accurate and are also more accurate than those based on the chromium/in vitro procedure. The relationship between these 2 methods for estimating intake was different in mid-pregnancy compared with either stage of lactation. The consumption of supplement did not interfere with any of the methods for estimating herbage intake. Estimates of faecal output based on the use of chromium, C28 alkane, or C32 as an external marker were statistically identical, indicating that the difference between the 2 methods for estimating herbage intake was not related to a failure to accommodate the incomplete recovery of any of the markers used or to the failure of rectal grab samples to be representative of total faeces. Our results indicate that herbage collected by oesophageally fistulated (OF) sheep was representative of that grazed by the ewes and could thus be used to provide the herbage alkane data needed to estimate herbage intake by the alkane method. However, the in vitro digestibility values obtained from the OF samples did not represent the digestibilities actually occurring in vivo. This was the main cause of the observed difference between the 2 methods for estimating intake. Possible reasons for the differences between the in vitro and in vivo estimates of digestibility are discussed.

A key contribution to study the cycling of nutrients in soil/plant/animal systems is the evaluation of the consumption of forage and their nutrients by cattle. The objective of this study was to test techniques to evaluate faecal production, in vitro digestibility, forage consumption and the proportion of legume in the acquired diet. Five Zebu steer calves were confined and fed five diets of different combinations of Brachiaria dictyoneura and Desmodium ovalifolium. All quantities of faeces were collected per animal and these values were found to compare favourably with those derived from using the chromium oxide technique. In vitro digestibility ranged from 7 to 10% higher than the actual in vivo digestibility. Faecal samples from steers fed with diets with 25% or more of grass in the mixture were found to be depleted in δ13C between 1.7 and 2.1‰, but no depletion was observed when the diet was 100% D. ovalifolium. There was a positive linear regression (r² = 0.97***) of the δ13C of the diet with the δ13C of faeces, but if the acquired diet contained a very high proportion of legume, the legume content could be underestimated by as much as 10%. None of the internal indicators, such as lignin or ash content of the diets, were useful to predict feed intake, but the chromium oxide external indicator performed satisfactorily. The 13C analysis of the faeces was an effective predictor of the proportion of the legume in the consumed diet.

Two studies were conducted to compare the precision of estimating kikuyu grass (Pennisetum clandestinum) intake by Friesian cows fed 0, 3 or 6 kg of cereal-based concentrate/cow.day, using a rising plate meter (RPM), standard energy requirements in reverse (RS) and plant wax alkanes as internal markers. Study 1 compared herbage intake estimates obtained using the RPM and RS techniques over a 45-day period. RS estimates were based on the metabolisable energy (ME) of ration components derived from in vitro organic matter digestibility (OMD) values. Pregrazing calibration equations for the RPM determined at 2-weekly intervals differed significantly (P<0.01) from postgrazing calibrations; consequently separate equations were used to determine pasture intake as the difference in pre- and post-grazing pasture mass. Estimates of total intake were lower using the RPM than the RS technique for the groups fed 0 kg (12.5 v. 14.8 kg dry matter (DM)/cow.day) and 3 kg (10.4 v. 12.9 kg DM/cow.day) of concentrate, and higher for those receiving 6 kg (10.5 v. 7.8 kg DM/cow.day). In study 2 (12 days duration), intakes derived using alkanes were compared with intakes estimated using the RS and RPM techniques. The C32/C33 alkane pair gave the closest estimate of herbage intake to that obtained using the RPM and RS techniques. Whole diet in vivo DM digestibility (DMD), determined by the alkane method, was not significantly different between the 3 groups (mean 70%), suggesting that digestibility of the kikuyu declined with increasing concentrate supplementation. The in vivo DMD of kikuyu alone (determined in the non-concentrate-supplemented cows) was considerably higher (69.5%) than the OMD determined in vitro (63.9%). By using in vivo rather than in vitro digestibilities for kikuyu in the RS calculations, the intake estimates were reduced by 17%, and for the 0 kg concentrate group, intake estimates aligned closely to predictions of the RPM and alkanes. Concentrates in the diet resulted in lower intake estimates using the RS technique compared with the RPM and alkane techniques. This was most evident at the 6 kg level of supplementation where RS predicted kikuyu intake to be 6.5 kg DM/cow.day using in vivo-derived DMD and this was substantially lower than either the RPM (12.4 kg DM/cow.day) or alkanes (9.2 kg DM/cow.day). The alkane technique provided a direct and precise method of measuring the intake of individual cows grazing tightly-managed kikuyu pasture. With the use of accurate animal production and feed quality parameters, the RS technique can provide sensible pasture intake estimates over an extended time period. The RPM technique is useful for obtaining herd estimates of pasture intake and for the determination of pasture parameters associated with intake.

We compared in vivo and in vitro dry matter (DM) and neutral detergent fiber (NDF) digestibility in donkeys using feces as microbial inoculum. Four donkeys were used in a 4 × 4 Latin square design with a 2 × 2 factorial arrangement of treatments. The animals were fed two types of hay, with or without flaked barley. For the in vivo procedure, total feces were collected for 6 days from each donkey; digestibility was calculated as the difference between ingested and excreted DM and NDF. For the in vitro procedure, donkey feces were buffered and used as microbial inoculum in an Ankom DaisyII Incubator; digestibility was estimated after 60 h of incubation. In vivo results showed that the addition of barley to hays did not change the digestibility values. In vivo estimates were higher than in vitro ones. The equations used to predict in vivo estimates from in vitro data were not reliable (R2 = 0.47 and 0.21; P = 0.003 and 0.078 for NDF and DM digestibility, respectively). Further studies need to evaluate different sample size and digestion times.

SummaryThe authors previously reported a transient decrease in adhesive platelet count and an enhancement of blood coagulability after administration of a small amount of adrenaline (0.1-1 µg per Kg, i. v.) in man and rabbit. In such circumstances, the sensitivity of platelets to aggregation induced by ADP was studied by an optical density method. Five minutes after i. v. injection of 1 µg per Kg of adrenaline in 10 rabbits, intensity of platelet aggregation increased to 115.1 ± 4.9% (mean ± S. E.) by 10∼5 molar, 121.8 ± 7.8% by 3 × 10-6 molar and 129.4 ± 12.8% of the value before the injection by 10”6 molar ADP. The difference was statistically significant (P<0.01-0.05). The above change was not observed in each group of rabbits injected with saline, 1 µg per Kg of 1-noradrenaline or 0.1 and 10 µg per Kg of adrenaline. Also, it was prevented by oral administration of 10 mg per Kg of phenoxybenzamine or propranolol or aspirin or pyridinolcarbamate 3 hours before the challenge. On the other hand, the enhancement of ADP-induced platelet aggregation was not observed in vitro, when 10-5 or 3 × 10-6 molar and 129.4 ± 12.8% of the value before 10∼6 molar ADP was added to citrated platelet rich plasma (CPRP) of rabbit after incubation at 37°C for 30 second with 0.01, 0.1, 1, 10 or 100 µg per ml of adrenaline or noradrenaline. These results suggest an important interaction between endothelial surface and platelets in connection with the enhancement of ADP-induced platelet aggregation by adrenaline in vivo.

SummaryDuring the collection of samples for plasma β-thromboglobulin (β-TG) determination, it is well established that artificially high values can be observed due to in-vitro release. To estimate the reliability of a single β-TG measurement, blood samples were collected simultaneously from both arms on two separate occasions in 56 diabetic patients selected for a clinical trial. From each arm, blood was taken into two tubes containing an anticoagulant mixture with (tube A) and without (tube B) PGE!. The overall mean value of B-TG in tube B was 1.14 times higher than in tube A (p <0.01). The markedly large between-arms variation accounted for the most part of within-subject variation in both tubes and was significantly greater in tube B than in tube A. Based on the difference between B-TG values from both arms, the number of subjects with artifically high B-TG values was significantly higher in tube B than in tube A on each occasion (overall rate: 28% and 14% respectively). Estimate of between-occasions variation showed that B-TG levels were relatively stable for each subject between two occasions in each tube. It is concluded that the use of PGEi decreases falsely high B-TG levels, but a single measurement of B-TG does not provide a reliable estimate of the true B-TG value in vivo.

Abstract Background The study highlighted the potential of three common and under-utilized tropical leguminous seeds (Tomentosa nilotica, Dioclea reflexa and Monodora myristica) to be used as supplementary feed to ruminant livestock. These seeds (their plants inclusive) are valuable sources of food and medicine for the prevention of illness and maintenance of human health. The medicinal properties of these seeds include antimicrobial, anti-inflammatory, anti-oxidant and immuno-stimulant. Trypsin inhibitors, which are common anti-nutritional factors in legumes and for monogastric animals, do not exert adverse effects in ruminants because they are degraded in the rumen. Hence, the crux of this study is to examine the effect of processing methods on the nutritional composition (proximate, fibre fractions, minerals, anti-nutrients) and in vitro digestibility of Tomentosa nilotica, Dioclea reflexa and Monodora myristica seeds and their suitability as feedstuff (protein sources) in small ruminant feed, particularly during off-season. Results From the results, raw Tomentosa nilotica and Monodora myristica have the highest crude protein (30.35% CP) and fat (22.40% EE), respectively. It is noteworthy that roasting best improve the mineral and significantly reduce the anti-nutrients observed in this study better compared to boiling and soaking methods. The highest organic matter digestibility, short-chain fatty acids, metabolizable energy and in vitro dry matter digestibility values were obtained in Dioclea reflexa compared to other test seeds. Roasting best improved the nutritive values, while Dioclea reflexa seed was rated highest for all the nutritional attributes and in vitro digestibility. Conclusions Dioclea reflexa could be incorporated in ruminants’ diet as protein source, particularly during the off-season, for improved ruminant production in Nigeria. However, in vivo study is therefore recommended to validate this report.

Abstract Background OPD and OPD' are the two main active components of Ophiopogon japonicas in Shenmai injection (SMI). Being isomers of each other, they are supposed to have similar pharmacological activities, but the actual situation is complicated. The difference of hemolytic behavior between OPD and OPD' in vivo and in vitro was discovered and reported by our group for the first time. In vitro, only OPD' showed hemolysis reaction, while in vivo, both OPD and OPD' caused hemolysis. In vitro, the primary cause of hemolysis has been confirmed to be related to the difference between physical and chemical properties of OPD and OPD'. In vivo, although there is a possible explanation for this phenomenon, the one is that OPD is bio-transformed into OPD' or its analogues in vivo, the other one is that both OPD and OPD' were metabolized into more activated forms for hemolysis. However, the mechanism of hemolysis in vivo is still unclear, especially the existing literature are still difficult to explain why OPD shows the inconsistent hemolysis behavior in vivo and in vitro. Therefore, the study of hemolysis of OPD and OPD' in vivo is of great practical significance in response to the increase of adverse events of SMI. Methods Aiming at the hemolysis in vivo, this manuscript adopted untargeted metabolomics and lipidomics technology to preliminarily explore the changes of plasma metabolites and lipids of OPD- and OPD'-treated rats. Metabolomics and lipidomics analyses were performed on ultra-high performance liquid chromatography (UPLC) system tandem with different mass spectrometers (MS) and different columns respectively. Multivariate statistical approaches such as principal component analysis (PCA) and orthogonal partial least square-discriminant analysis (OPLS-DA) were applied to screen the differential metabolites and lipids. Results Both OPD and OPD' groups experienced hemolysis, Changes in endogenous differential metabolites and differential lipids, enrichment of differential metabolic pathways, and correlation analysis of differential metabolites and lipids all indicated that the causes of hemolysis by OPD and OPD' were closely related to the interference of phospholipid metabolism. Conclusions This study provided a comprehensive description of metabolomics and lipidomics changes between OPD- and OPD'-treated rats, it would add to the knowledge base of the field, which also provided scientific guidance for the subsequent mechanism research. However, the underlying mechanism require further research.

Centrioles from spermatozoa of the starfish, Asterina pectinifera, were isolated and partially purified by solubilization of chromatin followed by sucrose density-gradient centrifugation. The ultrastructure of the isolated centriolar complex was investigated in whole mount preparations by electron microscopy. The complex unit was composed of a pair of centrioles and a pericentriolar structure, which associated with the distal end of the distal centriole by 9 spoke-like satellites extending radially to a marginal ring. Each satellite bifurcated at a dense node forming 2 fan-like shapes with a periodic striated pattern. The tubular structure of the centrioles easily disintegrated, leaving the pericentriolar structure or axonemal microtubules intact. The distal centriole in a spermatozoon served as an initiating site for flagellar microtubule assembly; that is, a number of “9 + 2′ axonemal tubules were observed adhering just beneath the distal end of the basal body. In experiments in vitro, polymerization of microtubule proteins purified from porcine brain was initiated by the structure at the ends of both proximal and distal centrioles, but not from the satellites or the marginal ring. Also, few if any microtubules were formed from the sides of each centriole, even in the presence of a high concentration of exogenous tubulin. On the other hand, centrioles of spermatozoa, when they were in mature ooplasm, could initiate the formation of sperm asters by microtubules. Therefore, centrioles in spermatozoa seem to be able to initiate microtubules in a 2 ways. A possible explanation of the difference between the 2 types of microtubule organization in vivo, i.e. in the sperm cell itself and in the ooplasm, it discussed.

Twenty experimental silages were made from seven pasture species at different stages of maturity. In vivo dry matter, organic matter, and energy ad libitum intakes and digestibilities of the silages were determined with standardized pairs of Merino wethers. The following chemical characteristics of the silages were measured: nitrogen, ammonia nitrogen, total titratable acids, acetic, propionic, butyric, and lactic acids, total volatiles lost during oven drying, lactic acid as a percentage of the total organic acids, pH, acid pepsin dry matter disappearance, dry matter content, and in vitro digestibility and rate of digestion. When all 20 silages were considered, energy intakes on a body weight basis were significantly related to silage pH (r = 0.55) and rate of in vitro digestion (r = 0.58). When the five legume silages were removed from the analysis and only the 15 grass-dominant silages were considered, dry matter intakes were significantly related to acetic (r = –0.57) and propionic acid (r = –0.55) concentrations. Multiple regression analyses did not significantly increase the accuracy of predicting intake. The results suggested that silage intake was negatively related to the degree of fermentation that occurred during the ensiling process.
