The development of ideas on the role of glucose in regulating milk secretion

The author has reviewed the development of theories, over the last century, on the role of glucose in mammary metabolism, focusing on three overlapping issues, via. the extent of glucose utilization by mammary tissue, the synthesis of lactose, and the way in which glucose supply controls secretion of the aqueous phase of milk. In the first half of the century, deficiencies in technique produced several misleading results, and later progress resulted largely from methodological advances. Methodology has also been important in shaping research programmes, as is evident in the current emphasis on techniques of molecular biology. However, the review indicates the importance of employing a combination of experimental approaches, including whole-animal studies. Through the adoption of such approaches, it is now appreciated that glucose plays a key role in regulating lactation. It is suggested that developments over the last 30 years, which have led to formulation of an apparently resilient theory of milk secretion, conform to the notion of a 'progressive research programme' built on a 'positive heuristic', as proposed by the philosopher of science, I. Lakatos.

Traditionally, anthropologists study evolutionary change throughmorphological analysis of fossils and comparative primate data. For the analysis of the genotypephenotype continuum, the current emphasis on genes is misplaced because genes don’t make structure. Developmental processes make structure through the activity of cells that use instructions specified by genes. A critical mechanism underlying any phenotypic trait is the genetically guided change in developmental events that produce the trait. But even when a developmental mechanism is identified, the links between genetically guided instructions and phenotypic outcome are lengthy, complicated, flexible, and sensitive to physical forces of functioning organs. We use the study of craniofacial phenotypes of craniosynostosis (premature closure of sutures) to demonstrate how patterns produced by the covariation of cranial traits cannot always reveal mechanism. Next we turn to encephalization, a critical feature of human evolution that covaries with cranial phenotypes, and show how experimental approaches can be used to analyze mechanism underlying this well-documented pattern in human evolution. With the realization that no single line of evidence can explain the dramatic changes in cranial morphology that characterize human evolution come fundamental changes in the way we conduct anthropological inquiry - collaborative efforts from scientists with diverse expertise will continue to push the field forward.

Hypoxia is a common and severe stress to an organism’s homeostatic mechanisms, and hypoxia during gestation is associated with significantly increased incidence of maternal complications of preeclampsia, adversely impacting on the fetal development and subsequent risk for cardiovascular and metabolic disease. Human and animal studies have revealed a causative role of increased uterine vascular resistance and placental hypoxia in preeclampsia and fetal/intrauterine growth restriction (FGR/IUGR) associated with gestational hypoxia. Gestational hypoxia has a major effect on mitochondria of uteroplacental cells to overproduce reactive oxygen species (ROS), leading to oxidative stress. Excess mitochondrial ROS in turn cause uteroplacental dysfunction by damaging cellular macromolecules, which underlies the pathogenesis of preeclampsia and FGR. In this article, we review the current understanding of hypoxia-induced mitochondrial ROS and their role in placental dysfunction and the pathogenesis of pregnancy complications. In addition, therapeutic approaches selectively targeting mitochondrial ROS in the placental cells are discussed.

In this paper we argue that adopting critical animal studies perspectives in critical public relations can not only be very fruitful, but that it is also a necessity if the aims of the latter are to be achieved. To this end, this text introduces the challenges and opportunities that the field of critical animal studies brings to critical public relations studies. First, a short explanation of what critical animal studies is and why it can contribute to critical public relations studies is provided. Then the main fields of research where this contribution can be most relevant are discussed, including ethics, discourse studies and political economy. The final aim of this theoretical paper is to expand research within the field of critical public relations by including a critical animal studies approach. Eventually, the authors suggest that embracing the animal standpoint in critical public relations is an essential step to furthering the study of power, hegemony, ideology, propaganda or social change and to accomplishing the emancipatory role of research.

Toxoplasma gondii is an apicomplexan parasite causing toxoplasmosis, a common disease, which is most typically asymptomatic. However, toxoplasmosis can be severe and even fatal in immunocompromised patients and fetuses. Available treatment options are limited, so there is a strong impetus to develop novel therapeutics. This review focuses on the role of oxidative stress in the pathophysiology and treatment of T. gondii infection. Chemical compounds that modify redox status can reduce the parasite viability and thus be potential anti-Toxoplasma drugs. On the other hand, oxidative stress caused by the activation of the inflammatory response may have some deleterious consequences in host cells. In this respect, the potential use of natural antioxidants is worth considering, including melatonin and some vitamins, as possible novel anti-Toxoplasma therapeutics. Results of in vitro and animal studies are promising. However, supplementation with some antioxidants was found to promote the increase in parasitemia, and the disease was then characterized by a milder course. Undoubtedly, research in this area may have a significant impact on the future prospects of toxoplasmosis therapy.

SUMMARY The role of the peripheral innervation of mammary tissue in the maintenance of lactation has been investigated by the procedure of selective thelectomy combined with denervation of the posterior thoracic nipples. When suckling is restricted to a single pair of nipples bilateral transection of the three adjacent nerves supplying a nipple arrests lactation completely; partial denervation is associated with a reduced level of lactational performance which is directly related to the concentration of the residual innervation. Increase in litter size is associated with an overall increase in milk-yield up to a limit beyond which the addition of further young to the litter is without effect. It is inferred that there is a quantitative relationship between the neural stimulus of suckling and the endocrine response of the hypothalamopituitary system.

Abstract Persistent Organic Pollutants (POPs) are exogenous, artificially made chemicals that can disrupt the biological system of individuals and animals. POPs encompass a variety of chemicals including, dioxins, organochlorines (OCs), polychlorinated biphenyl (PCBs), and perfluoroalkyl substances (PFASs) that contain a long half-life and highly resistant to biodegradation. These environmental pollutants accumulate over time in adipose tissues of living organisms and alter various insulin function-related genes. Childhood Metabolic Syndrome (MetS) consists of multiple cardiovascular risk factors, insulin function being one of them. Over the years, the incidence of the syndrome has increased dramatically. It is imperative to explore the role of persistent organic pollutants in the development of Childhood Metabolic Syndrome. Some epidemiological studies have reported an association between prenatal exposure to POPs and offspring MetS development throughout childhood. These findings have been replicated in animal studies in which these pollutants exercise negative health outcomes such as obesity and increased waist circumference. This review discusses the role of prenatal exposure to POPs among offspring who develop MetS in childhood, the latest research on the MetS concept, epidemiological and experimental findings on MetS, and the POPs modes of action. This literature review identified consistent research results on this topic. Even though the studies in this review had many strengths, one major weakness was the usage of different combinations of MetS criteria to measure the outcomes. These findings elucidate the urgent need to solidify the pediatric MetS definition. An accurate definition will permit scientists to measure the MetS as a health outcome properly and allow clinicians to diagnose pediatric MetS and provide individualized treatment appropriately.

Abstract Ten percent of deaths worldwide are due to trauma, and it is the third most common cause of death in the United States. Despite a profound upregulation in procoagulant mechanisms, one-quarter of trauma patients present with laboratory-based evidence of trauma-induced coagulopathy (TIC), which is associated with poorer outcomes including increased mortality. The most common causes of death after trauma are hemorrhage and traumatic brain injury (TBI). The management of TIC has significant implications in both because many hemorrhagic deaths could be preventable, and TIC is associated with progression of intracranial injury after TBI. This review covers the most recent evidence and advances in our understanding of TIC, including the role of platelet dysfunction, endothelial activation, and fibrinolysis. Trauma induces a plethora of biochemical and physiologic changes, and despite numerous studies reporting differences in coagulation parameters between trauma patients and uninjured controls, it is unclear whether some of these differences may be “normal” after trauma. Comparisons between trauma patients with differing outcomes and use of animal studies have shed some light on this issue, but much of the data continue to be correlative with causative links lacking. In particular, there are little data linking the laboratory-based abnormalities with true clinically evident coagulopathic bleeding. For these reasons, TIC continues to be a significant diagnostic and therapeutic challenge.

The goal of this study is to analyse sources of variation, residing within the individual or within the relationship, in the ability to balance co-operative and competitive behaviours in a dyadic context. The ability to balance these two tendencies can be considered fundamental to successful adaptation within a social unit because co-operation may be essential in raising offspring, competing with other groups or in generating resources, whereas egoistic behaviour may protect the individual from exploitation or otherwise enhance reproductive success. Research is reviewed on the influence of social structures and relationships on co-operation in peer groups, and the origin and developmental significance of individual differences in co-operative abilities. Finally, a research programme investigating the conjunction of kin and peer relations is described, emphasising the role of affective synchrony, behavioural contingency, and reciprocity in shaping and sustaining co-operative behaviour as a conditional strategy.

Antimicrobial activity is not an ‘all or none’ effect. An increase in the rate and extent of antimicrobial action is usually observed over a wide range of antimicrobial concentrations. Subinhibitory antimicrobial concentrations are well known to produce significant antibacterial effects, and various antimicrobials at subinhibitory concentrations have been reported to inhibit the rate of bacterial growth. Bacterial virulence may be increased or decreased by subinhibitory antimicrobial concentrations by changes in the ability of bacteria to adhere to epithelial cells or by alterations in bacterial susceptibility to host immune defences. Animal studies performed in rats, hamsters and rabbits demonstrate decreased bacterial adherence, reduced infectivity and increased survival of animals treated with subinhibitory antimicrobial concentrations compared to untreated controls. The major future role of investigation of subinhibitory antimicrobial concentrations will be to define more fully, at a molecular level, how antimicrobials exert their antibacterial effects.
