Change in chromosome number associated with a double deletion in the Neurospora crassa mitochondrial chromosome.

Abstract The mitochondrial genome of Neurospora is usually found in a single covalently closed circular 62-kbp DNA molecule. We report here that the mitochondrial genome of a phenotypic revertant of a stopper mutant (stp-ruv) is contained primarily in two separate, nonoverlapping, autonomously replicating circular chromosomes. The circles, one about 21 kbp and the other somewhat less than 36 kbp are derived from the most frequent classes of recombinant chromosomes (21 and 41 kbp) in the chromosomal population of mitochondria in the original stopper mutant. The new, more stable chromosomal configuration, is associated with the deletion of two sequences (1 kbp and 4 kbp) at the splice junctions of the two circles. The data suggest that both deletions are likely to have originated from a single recombinational event involved in generating the 36-kbp circle. Secondary, spontaneously arising derivatives of stp-ruv have been found to yield, at high copy number, short sections of the 21-kbp circle in covalently closed supercoiled circles varying from unit length to very high multimers. The amplified segments span a common segment likely to contain the replication origin of the 21-kbp chromosome.

A new measurement technique of electrical parameters of superconducting thin films at the Very High Frequency (VHF) range is described, based on resonators with microstrip (MS) structures. The design of an optimal resonator was achieved, based on a thorough theoretical analysis, which is required for derivation of the exact configuration of the MS. A theoretical model is presented, from which an expression for the attenuation of a MS line can be derived. Accordingly, simulations were performed, and an optimal resonator for the VHF range was designed and implemented. Production constraints of YBa2Cu3O7 (YBCO) limited the diameter of the sapphire substrate to 3″. Therefore, a meander configuration was formed to fit the long λ/4 MS line on the wafer. By measuring the complex input reflection coefficients of a λ/4 resonator, we extracted the quality factor, which is mainly affected by the dielectric and conductor attenuations. The experimental results are well fitted by the theoretical model. The dielectric attenuation was calculated using the quasi-static analysis of the MS line. An identical copper resonator was produced and measured to compare the properties of the YBCO resonator in reference to the copper one. A quality factor of ~6·105 was calculated for the YBCO resonator, three orders of magnitude larger than that of the copper resonator. The attenuation per unit length of the YBCO layer was smaller by more than five orders of magnitude than that of the copper.

DNA from the hypotrichous ciliatae Stylonychia lemnae was separated by PFGE. In addition to the separation of the macronuclear DNA molecules with a size up to ∼40 kb, we were able to separate the micronuclear DNA with a size between ∼90 kb and 2 Mb. One very prominent 90-kb DNA band appeared on the pulsed-field gels. We propose that this 90-kb DNA fragment represents a linear plasmid residing in the micronucleus in a very high copy number. About 10% of the micronuclear DNA consists of the 90-kb DNA molecule. It appears in the micronucleus as well as in the macronuclear anlagen during macronuclear development but not in the mature macronucleus. Thus, the multicopy DNA is eliminated during fragmentation of the macronuclear anlagen DNA in the course of macronuclear development. Therefore, this 90-kb DNA molecule might serve as an excellent tool to study the recognition and elimination of DNA during nuclear differentiation of hypotrichous ciliates.

Kinetics of saccharose fermentation by Kombucha is not yet well defined due to lack of knowledge of reaction mechanisms taking place during this process. In this research kinetics of saccharose fermentation by Kombucha was analysed using the suggested empirical model. The data were obtained on 1.5 g L-1 of black tea, with 66.47 g L-1 of saccharose and using 10% (v/v) or 15% (v/v) of Kombucha. Total number of viable cells was as follows: approximately 5x105 of yeast cells per mL of the inoculum and approximately 2x106 of bacteria cells per mL of the inoculum. The samples were analysed after 0, 3, 4, 5, 6, 7 and 10 days. Their pH values and contents of saccharose, glucose, fructose, total acids and ethanol were determined. A saccharose concentration model was defined as sigmoidal function at 22oC and 30oC, and with 10% (v/v) and 15% (v/v) of inoculum quantity. Determination coefficients of the functions were very high (R2>0.99). Reaction rates were calculated as first derivatives of Boltzmann?s functions. No simple correlation between rate of reaction and independent variables (temperature and inoculum concentration) was found. Analysis of empirical model indicated that saccharose fermentation by Kombucha occurred according to very complex kinetics.

The application of both thermoluminescence and infrared stimulated luminescence dating to the extensively studied “classical” Hungarian loess/paleosol sequences from Basaharc, Mende, and Paks provides a reliable chronological framework and climatostratigraphic reconstruction for the last interglacial/glacial cycle. Based on this combined luminescence dating study a new chronology is proposed for the “Young Loess” in Hungary. Luminescence dating suggests that the loess below the MF2 horizon formed during the penultimate glaciation. The MF1 horizon probably formed during an interstade within oxygen isotope stage 3. For the youngest loess, overlying MF1, a very high accumulation rate was determined. Large time gaps occur above MF2 and MF1, indicating that most of the record of the last glaciation is missing in the standard sections at Basaharc, Mende, and Paks. Either large discontinuities or a very low accumulation rate occurred in all three type sections during the soil-forming periods. High-resolution studies of climatic proxies using this combined luminescence dating approach provide a reliable chronological framework for loess and loess derivatives of the last glacial cycle in Hungary, although a precise and complete chronostratigraphic reconstruction cannot be achieved from the incomplete records found at these sites.

The phospholipid distribution in the mollusc Lymnaea stagnalis was investigated by fractionation of the lipid extract with silica-gel column chromatography followed by qualitative and quantitative thin-layer chromatography. The results obtained show that choline and ethanolamine phosphoglycerides are the major phospholipids in this species and that they amount to about 50% and 30% of total phospholipids, respectively. The amount of ceramide 2-aminoethylphosphonate in this species is 7.5% of the total phospholipids and this compound is the main phosphonolipid in this species. The glyceryl ether derivatives of both choline and ethanolamine phosphoglycerides are found to be very high. Nearly half (45%) of the choline phosphoglycerides is glyceryl ether in nature while 15% of the ethanolamine phosphoglycerides is also of this type.

Auxotrophs were isolated from four effective strains of Rhizobium leguminosarum and R. trifolii for a study of the relationship between metabolic defects and loss of ability for symbiosis. Most auxotrophs were isolated indirectly by prior isolation of mutants for resistance to metabolic inhibitors, especially D-alanine and D-histidine. The likelihood of some nutritional requirement was greatly increased among such resistant mutants, and was very high for derivatives of the R. trifolii strains. The complexity of growth factor requirement varied greatly between auxotrophs, and few had simple requirements. The most common requirement was for vitamins, notably thiamine. Partial or full restoration of effectiveness in symbiosis often accompanied reversion to prototrophy in some ineffective auxotrophs. The frequency of some degree of restoration among prototrophs varied from 0% to 100%, depending on the auxotrophic mutant involved. In one ineffective auxotroph of R. trifolii strain T1 the level of reversion (partial to complete) in vitro varied considerably and generally paralleled the degree of restoration of effectiveness. Biochemical deficiency appeared to be meaningfully related to impaired symbiosis in some auxotrophs but the relationship was probably incidental in most others. These auxotroph–prototroph studies are considered from the standpoint of relationship to several areas of research on Rhizobium.

T lymphocytes from guinea pigs immunized with 2,4-dinitrophenyl (DNP) derivatives of mycobacteria respond to a variety of DNP conjugates. Preincubation of such cells with a given DNP conjugate under conditions which lead to the inactivation of responding cells causes a loss of the response to that conjugate, but has little effect on the response to DNP coupled to unrelated carriers. Thus, the responses of such cells to a variety of DNP conjugates can best be explained by the presence of a mixture of highly specific cells each responding to a different antigenic dterminant rather than by the presence of T cells with specificity limited to the hapten itself. Furthermore, the activity of T cells from DNP-mycobacteria-primed donors could not be blocked by a variety of nonstimulatory DNP conjugates. This suggests that while such T cells clearly recognize DNP with great precision, the receptor does not contain a very high affinity site for the hapten. A possible model for such a T-cell receptor is discussed.

The annual height increment of the shoot can be divided into two morphogenetic components: the number of stem units initiated in the bud prior to bud break and their mean length. Observations were made on the lateral shoots of 43 Abiescephalonica open-pollinated families from three provenances located in an experimental site in southern France. Genetic parameters of the two components were evaluated with an analysis of variance. Correlations both between components and with annual height increment were examined at the phenotypic, genotypic, and individual levels. Differences between provenances were the result of number of stem units and annual height increment. Number of stem units presented a very high heritability and was strongly correlated to annual height increment at all variance levels. Mean stem unit length appeared to be more dependent on environmental variations and was less closely correlated to annual height increment, especially when genotypic data were considered. This particular growth strategy could be the consequence of A. cephalonica's adaptation to a Mediterranean-type climate. Of the two components studied, number of stem units was a better genetic predictor of annual height increment. A negative correlation between number of stem units and mean stem unit length was made apparent at the phenotypic and individual levels and should be considered when using number of stem units as a selection criterium.
