Dayak onion (Eleutherine polmifolia (L.) Merr) was a typical plant of Kalimantan. Empirically dayak onion (Eleutherine polmifolia (L.) Merr) used by the comunity to treat skin infections. The purpose of this study was to determine the antibacterial activity of ethanol extract of dayak onion power against Salmonella typhi and Escherichia coli bacteria. The highest concentration used was 100%, and the lowest concentration was 0,5%. As a control of cloramfenikol for Salmonella typhi and ciprofloxacin for Escherichia coli bacteria. This research method used agar diffusion. The results showed the ethanol extract of dayak onion (Eleutherine polmifolia (L.) Merr) contained flavonoids, alkaloids, tannins and phenols with the highest levels was by tannins at 7,45%. Dayak onion (Eleutherine polmifolia (L.) Merr) are inhibiting the growth of Salmonella typhi bacteria at a minimum inhibitory concentration (MIC)of 1% with a inhibition zone diameter of 6,24 mm and 0,9% with a inhibition zone diameter of 6,20 mm at Escherichia coli bacteria.Keywords : Eleutherine polmifolia (L.) Background: Staphylococcus aureus is a gram positive bacterium that causes pyogenic infectious disease, such as boils, pimples, endocarditis and sepsis. Resistance of S. aureus is continued growth that made it becomes a very serious problem that need to be solved by looking for another effective alternative for this infection. Several of marine life can be used as a source of antibacterial medication, antiviral, and antifungal. One alternative of antibacterial that comes from water resources is brown seaweed (Sargassum sp.). Purposes: This study aimed to find out the inhibitory fraction of n-hexane and ethyl acetate of Sargassum sp. against the growth of S. aureus Methods: This study applied quasi experimental method which used posttest-only control design. Samples used in the form of brown seaweed treatment of fraction n-hexane and Ethyl acetate derived from Desa Bungin Permai Kecamatan Tinanggea Kabupaten Konawe Selatan. The fraction inhibition test was conducted by diffusion agar method used variant concentration (20%, 40%, 60%, 80%, 100%) with three repetitions. Erythromycin as control (+) and DMSO 10% as control (-).Result: The study result showed that the n-hexane and Ethyl acetate fraction of Sargassum sp. extract was able in inhibiting the growth of S. aureus that seen with the clear zone around the paper disc. Based on this result, the minimum inhibitory fraction of n-hexane and ethyl acetate obtained in this study was at the concentration of 20%. The diameter average of inhibition zone in both fractions in the concentration 20%, 40%, 60%, 80%, 100% were 9.3 mm, 12.3 mm, 25.6 mm, 27 mm, 27.7 mm for n-hexane fraction, mean while the diameter average of inhibition zone in the ethyl acetate fraction was 4.6 mm, 16 mm, 19.3mm, 27.6mm, 29.6 mm. ethyl acetate fraction at a concentration of 40%, 80%,100% inhibitory zone diameter higher than the fraction of n-hexane while at a concentration of 20% and 60% inhibitory zone diameter higner than the fraction of ethyl acetate. Conclusion: Based on the result of the study, it was concluded that the fraction of n-hexane and ethyl acetate of Sargassum sp. has an inhibitory effect against the growth of S. aureus. The Minimum Inhibitory Concentration (MIC) of n-hexane and ethyl acetate fraction was at the concentration of 20%. Fraction of n-heksan and ethyl acetate equally have a strong inhibition of the growth of S. aureus bacteria. Kejadian resistensi antibakteri terhadap S. aureus terus meningkat sehingga menjadi masalah yang sangat serius sehingga diperlukan alternatif lain untuk mengatasi penyakit infeksi yang lebih efektif. Beberapa hasil biota laut dapat dimanfaatkan sebagai antibakteri, antivirus dan antijamur. Salah satu bahan alternatif sebagai antibakteri yang berasal dari sumber perairan yakni rumput laut cokelat (Sargassum sp.). Tujuan: Penelitian ini bertujuan untuk mengetahui daya hambat fraksi N-Heksana dan Etil Asetat Sargassum sp. terhadap pertumbuhan bakteri S. aureus. Metode: Penelitian ini menggunakan metode quasi eksperimental dengan desain post test-only control. Sampel yang digunakan berupa perlakuan fraksi N-Heksan dan Etil Asetat Sargassum sp. yang berasal dari Desa Bungin Permai Kecamatan Tinanggea Kabupaten Konawe Selatan. Pengujian daya hambat dilakukan dengan metode difusi agar dalam berbagai konsentrasi 20%, 40%, 60%, 80%, 100% dengan tiga kali pengulangan. Eritromisin yang digunakan sebagai kontrol positif dan DMSO 10% (Dimetil sulfoksida) sebagai kontrol negatif. Hasil: Hasil penelitian menunjukkan bahwa fraksi Sargassum sp. mampu menghambat pertumbuhan bakteri S. aureus yang dilihat dengan adanya zona bening disekitar kertas cakram. Konsentrasi hambat minimum dari kedua fraksi pada konsentrasi 20%. Perbedaan rerata diameter zona hambat pada kedua fraksi dengan konsentrasi 20%, 40%, 60%, 80%, 100% yaitu sebesar 9,3 mm,12,3 mm, 25,6 mm, 27 mm, 27,7 mm pada fraksi n- heksan dan 4,6 mm, 16 mm, 19,3 mm, 27,6 mm, 29,6 mm pada fraksi etil asetat. Fraksi etil asetat pada konsentrasi 40%, 80%, 100% diameter zona hambatnya lebih tinggi dari pada fraksi n-heksan sedangkan pada fraksi n-heksan konsentrasi 20% dan 60% diameter zona hambatnya lebih tinggi dibandingkan fraksi etil asetat. Simpulan: Simpulan dari penelitian ini bahwa fraksi n-heksan dan fraksi etil asetat Sargassum sp. mempunyai daya hambat terhadap pertumbuhan bakteri S. aureus ekstrak rumput laut cokelat mengandung senyawa flavonoid, steroid, saponin dan tannin. Konsentrasi hambat minimum (KHM ) dari kedua fraksi yaitu pada konsentrasi 20 %. Fraksi n-heksan dan etil asetat mempunyai daya hambat yang kuat terhadap pertumbuhan bakteri S. aureusKata Kunci : fraksi etil asetat, fraksi n-heksan, Kadar Hambat Minimun, Sargassum sp,  Staphylococcus aureus

Trichoderma harzianum is a fungus that can produce secondary metabolites which able to inhibit the growth of Colletotrichum capsici. a pathogenic fungus causing anthracnose in plants. the Aims of this research were to obtain and analyze the ability of secondary metabolites of Trichoderma harzianum AC1 (b) J2 isolated from Scleria poaeformis. to inhibit the growth of Colletotrichum capsici IPBCC13.1098. The secondary metabolite eluat of Trichoderma harzianum were tested for the antifungal activity toward Colletotrichum capsici IPBCC 13.1098. Fractionation was carried out by Column Chromatography and produced 31 eluat. The secondary metabolite eluat of Trichoderma harzianum with the highest inhibition zone diameter was eluat 1 with 8.4 mm in diameter. The secondary metabolite eluat with the highest value of inhibition zone was carried out by MIC test and thin layer chromatography (TLC). Based on the MIC results, the minimum inhibitory concentration of the secondary metabolite of Trichoderma harzianum to fungus Colletotrichum capsici was 250 ppm with inhibition zone diameter was 0.10 mm. The results of TLC showed orange spots on the TLC plate that indicated alkaloite compounds.

The in vitro activity of fluconazole against 143Candida spp. obtained from the bloodstreams of 143 hospitalized patients from 1995 to 1997 was studied. Susceptibility tests were carried out by two macrodilution methods, the M27-A and a modified M27-A method (0.165 M, pH 7/morpholinepropanesulfonic acid-buffered RPMI 1640 medium supplemented with 20 g ofd-dextrose per liter), and by the agar diffusion method (with 15-μg fluconazole [Neo-Sensitab] tablets). With 2 μg of fluconazole per ml, 96.92% of 65 C. albicans isolates, 86.2% of 58 C. parapsilosis isolates 7 of 8 C. tropicalis isolates, and 1 of 6 C. glabrata isolates were inhibited. Only one strain of C. albicans and one strain of C. tropicalis were resistant. The agreement between the two macrodilution methods was greater than 90% within ±2 log2 dilutions for all strains except C. glabrata (83.3%) and C. tropicalis(87.5%). Generally, MICs were 1 log2 dilution lower in glucose-supplemented RPMI 1640 medium. No correlation between zone sizes and MICs was found. All strains susceptible by the diffusion test were susceptible by the dilution method, but the converse was not necessarily true. Interestingly, inhibition zones were smaller forC. albicans, for which the geometric mean MIC was 0.29 μg/ml and the mean inhibition zone diameter was 25.7 mm, while for C. parapsilosis the geometric mean MIC was 0.96 μg/ml and the mean inhibition zone diameter was 31.52 mm. In conclusion, the two macrodilution methods give similar results. The modified M27-A method with 2% dextrose has the advantage of shortening the incubation time and simplifying the endpoint determination.

Objective: The main objective of this research were screened in vitro and in silico of 1,5-bis (3'-ethoxy-4'-hydroxyphenyl)-1,4-pentadiene-3-one as potential antibacterial agents.Methods: The in vitro antibacterial study was carried against Staphylococcus aureus, Staphylococcus epidermidis (gram positive) and Escherichia coli, Salmonella thypi (gram negative) using broth dilution method to determine Minimum Inhibitory Concentration (MIC), disc diffusion method to determine the diameter of inhibition zone. In silico antibacterial study was carried using computational software Protein-Ligand ANT System (PLANTS), computational docking was carried using receptor with Protein Data Bank (PDB) file 3MZD. The structures were optimized prior docking using YASARA, and MarvinSketch. The results of antibacterial testing were compared to two positive control drugs i. e amoxicillin and cefadroxil.Results: In vitro evaluation showed that 1,5-bis (3'-ethoxy-4'-hydroxyphenyl)-1,4-pentadiene-3-one has a better antibacterial activity than amoxicillin and cefadroxil with a Minimum Inhibitory Concentration (MIC) of 0.15 ppm and diameter of inhibition zone of 11.27±0.31, 11.35±0.39, 11.25±0.33, and 11.05±0.45 mm in Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, and Salmonella thypi, respectively. These results in line with in silico evaluation that showed 1,5-bis (3'-ethoxy-4'-hydroxyphenyl)-1,4-pentadiene-3-one has more negative docking score than amoxicillin, cefadroxil, and cloxacillin acyl as a native ligand on the 3MZD receptor.Conclusion: This results obtained in this research work were 1,5-bis (3'-ethoxy-4'-hydroxyphenyl)-1,4-pentadiene-3-one compound potential as an antibacterial agent.

Fish disease is a serious problem faced in the aquaculture business. One of the diseases that attack cultured fish is saprolegniasis caused by the fungus Saprolegnia sp. The use of chemical drugs tends to be environmentally unfriendly and has other negative effects on these fish and humans. For this reason, a safer alternative treatment is needed to control fish disease caused by the fungus Saprolegnia sp. One of the plants that have the potential to treat disease caused by the fungus Saprolegnia sp. is basil (Ocimum basilicum L). This research aims to determine the effectiveness of basil extract against the fungus Saprolegnia sp. in vitro. This research was conducted from May to June 2019. The research method used was a completely randomized design (CRD) with 5 treatment levels and 3 replications including 0 ppm (control), 20 ppm, 40 ppm, 60 ppm, and 80 ppm treatments. The test parameter observed was the inhibition zone diameter. In this study, 200 grams of basil extract were produced with 16.67% yield, the conclusion is that basil extract with a concentration of 60 ppm has the largest inhibition zone diameter of 22 mm in inhibiting the growth of the fungus Saprolegnia sp in vitro compared to other ethanol extract concentrations of basil.Keywords: Basil extract, antifungal, saprolegnia sp, in vitro

The determination of the antibacterial activity (zone of inhibition) and minimum inhibitory concentration of medicinal plants a crucial step in drug development. In this study, the antibacterial activity and minimum inhibitory concentration of the ethanol extract of Myrsine africana were determined for Escherichia coli, Bacillus cereus, Staphylococcus epidermidis and Streptococcus pneumoniae. The zones of inhibition (mm±S.E) of 500mg/ml of M. africana ethanol extract were 22.00± 0.00 for E. coli,20.33 ±0.33 for B. cereus,25.00± 0.00 for S. epidermidis and 18. 17±0.17 for S. pneumoniae. The minimum inhibitory concentration(MIC) is the minimum dose required to inhibit growth a microorganism. Upon further double dilution of the 500mg/ml of M. africana extract, MIC was obtained for each organism. The MIC for E. coli, B. cereus, S. epidermidis and S. pneumoniae were 7.81mg/ml, 7.81mg/ml, 15.63mg/ml and 15.63mg/ml respectively. Crude extracts are considered active when they inhibit microorganisms with zones of inhibition of 8mm and above. Therefore, this study has shown that the ethanol extract of M. africana can control the growth of the four organisms tested.

Objective: Black-pigmented bacteria represent one of the most prominent causes of chronic periodontitis. The objective of this study is to investigate the in vitro antibacterial effect of an infusion of binahong leaves (Anredera cordifolia (Ten.) Steenis) against black-pigmented bacteria.Methods: The black-pigmented bacteria were isolated from the periodontal pocket of a patient with chronic periodontitis. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined by means of a dilution test using 105 cfu/ml of bacteria cultured on Brucella broth containing blood and kanamycin, which was exposed to a 20%, 30%, 40%, 50%, 65%, 80%, and 95% infusion of binahong leaves and then incubated for 24, 48, and 72 h. In the blank disc diffusion test, a 50%, 65%, 80%, 95%, and 100% infusion was dripped onto a paper disc placed at the center of a Brucella agar bacterial culture and then incubated for 24 h.Results: The MIC and MBC of the binahong leaf infusion were >65% and 80%, respectively, while the antibacterial activity was not correlated with the duration of the exposure to the infusion. The diffusion test showed that an increased concentration of the infusion resulted in an increased diameter of the inhibition zone that ranged from 0.42 mm (50% infusion) to 1.42 mm (100% infusion).Conclusion: Binahong leaves infusion has potential efficacy against black-pigmented bacteria

Objective: This experiment aimed to analyze the effect of propolis extract and propolis containing candies on the growth of Aggregatibacter actinomycetemcomitans using spectrophotometric analysis and colony-forming units (CFU) counts.Methods: After A. actinomycetemcomitans were exposed to propolis extract and candies, the minimum inhibitory concentration (MIC) and the minimum bactericidal concentration (MBC) were determined with spectrophotometry and post-exposure colony counting.Results: The MIC of propolis extract against A. actinomycetemcomitans was determined to be 10%, and the MBC was 20%. A decrease in the total CFU count of A. actinomycetemcomitans was observed after propolis extract and candy exposure.Conclusions: Propolis extract and propolis candies were effective in inhibiting the growth of A. actinomycetemcomitans ATCC 43718 in vitro.
