Background: Staphylococcus aureus is a gram positive bacterium that causes pyogenic infectious disease, such as boils, pimples, endocarditis and sepsis. Resistance of S. aureus is continued growth that made it becomes a very serious problem that need to be solved by looking for another effective alternative for this infection. Several of marine life can be used as a source of antibacterial medication, antiviral, and antifungal. One alternative of antibacterial that comes from water resources is brown seaweed (Sargassum sp.). Purposes: This study aimed to find out the inhibitory fraction of n-hexane and ethyl acetate of Sargassum sp. against the growth of S. aureus Methods: This study applied quasi experimental method which used posttest-only control design. Samples used in the form of brown seaweed treatment of fraction n-hexane and Ethyl acetate derived from Desa Bungin Permai Kecamatan Tinanggea Kabupaten Konawe Selatan. The fraction inhibition test was conducted by diffusion agar method used variant concentration (20%, 40%, 60%, 80%, 100%) with three repetitions. Erythromycin as control (+) and DMSO 10% as control (-).Result: The study result showed that the n-hexane and Ethyl acetate fraction of Sargassum sp. extract was able in inhibiting the growth of S. aureus that seen with the clear zone around the paper disc. Based on this result, the minimum inhibitory fraction of n-hexane and ethyl acetate obtained in this study was at the concentration of 20%. The diameter average of inhibition zone in both fractions in the concentration 20%, 40%, 60%, 80%, 100% were 9.3 mm, 12.3 mm, 25.6 mm, 27 mm, 27.7 mm for n-hexane fraction, mean while the diameter average of inhibition zone in the ethyl acetate fraction was 4.6 mm, 16 mm, 19.3mm, 27.6mm, 29.6 mm. ethyl acetate fraction at a concentration of 40%, 80%,100% inhibitory zone diameter higher than the fraction of n-hexane while at a concentration of 20% and 60% inhibitory zone diameter higner than the fraction of ethyl acetate. Conclusion: Based on the result of the study, it was concluded that the fraction of n-hexane and ethyl acetate of Sargassum sp. has an inhibitory effect against the growth of S. aureus. The Minimum Inhibitory Concentration (MIC) of n-hexane and ethyl acetate fraction was at the concentration of 20%. Fraction of n-heksan and ethyl acetate equally have a strong inhibition of the growth of S. aureus bacteria. Kejadian resistensi antibakteri terhadap S. aureus terus meningkat sehingga menjadi masalah yang sangat serius sehingga diperlukan alternatif lain untuk mengatasi penyakit infeksi yang lebih efektif. Beberapa hasil biota laut dapat dimanfaatkan sebagai antibakteri, antivirus dan antijamur. Salah satu bahan alternatif sebagai antibakteri yang berasal dari sumber perairan yakni rumput laut cokelat (Sargassum sp.). Tujuan: Penelitian ini bertujuan untuk mengetahui daya hambat fraksi N-Heksana dan Etil Asetat Sargassum sp. terhadap pertumbuhan bakteri S. aureus. Metode: Penelitian ini menggunakan metode quasi eksperimental dengan desain post test-only control. Sampel yang digunakan berupa perlakuan fraksi N-Heksan dan Etil Asetat Sargassum sp. yang berasal dari Desa Bungin Permai Kecamatan Tinanggea Kabupaten Konawe Selatan. Pengujian daya hambat dilakukan dengan metode difusi agar dalam berbagai konsentrasi 20%, 40%, 60%, 80%, 100% dengan tiga kali pengulangan. Eritromisin yang digunakan sebagai kontrol positif dan DMSO 10% (Dimetil sulfoksida) sebagai kontrol negatif. Hasil: Hasil penelitian menunjukkan bahwa fraksi Sargassum sp. mampu menghambat pertumbuhan bakteri S. aureus yang dilihat dengan adanya zona bening disekitar kertas cakram. Konsentrasi hambat minimum dari kedua fraksi pada konsentrasi 20%. Perbedaan rerata diameter zona hambat pada kedua fraksi dengan konsentrasi 20%, 40%, 60%, 80%, 100% yaitu sebesar 9,3 mm,12,3 mm, 25,6 mm, 27 mm, 27,7 mm pada fraksi n- heksan dan 4,6 mm, 16 mm, 19,3 mm, 27,6 mm, 29,6 mm pada fraksi etil asetat. Fraksi etil asetat pada konsentrasi 40%, 80%, 100% diameter zona hambatnya lebih tinggi dari pada fraksi n-heksan sedangkan pada fraksi n-heksan konsentrasi 20% dan 60% diameter zona hambatnya lebih tinggi dibandingkan fraksi etil asetat. Simpulan: Simpulan dari penelitian ini bahwa fraksi n-heksan dan fraksi etil asetat Sargassum sp. mempunyai daya hambat terhadap pertumbuhan bakteri S. aureus ekstrak rumput laut cokelat mengandung senyawa flavonoid, steroid, saponin dan tannin. Konsentrasi hambat minimum (KHM ) dari kedua fraksi yaitu pada konsentrasi 20 %. Fraksi n-heksan dan etil asetat mempunyai daya hambat yang kuat terhadap pertumbuhan bakteri S. aureusKata Kunci : fraksi etil asetat, fraksi n-heksan, Kadar Hambat Minimun, Sargassum sp,  Staphylococcus aureus

This study aims to determine the antibacterial activity and TLC-bioautography profile of the active fractions of Muntingia calabura L. leaves against Staphylococcus aureus. Muntingia calabura L. leaves were macerated with ethanol 96% then fractionated with n-hexane, ethyl acetate, and methanol solvent, respectively. The antibacterial activity was tested by the Kirby-Bauer method to determine the most active fraction and the lowest concentration that inhibited the growth of Staphylococcus aureus. TLC-bioautography was tested using chloroform: ethyl acetate (2:8) as the mobile phase and silica gel F254 as the stationary phase. Antibacterial activity test of n-hexane and ethyl acetate fractions at a concentration of 10% w/v showed activities with inhibition zone diameter of 0.33±0.288 and 9.66±5.77 mm, respectively. At the same time, the methanol fraction showed no activity. The lowest concentration of ethyl acetate fraction which still showed the inhibition zone was 0.312% w/v. The TLC-bioautography profile showed active spots with an Rf value of 0.82 and had an inhibitory zone diameter of 4.013±0.864 mm. It can be concluded that the ethyl acetate fraction was the most active fraction that inhibited the growth of Staphylococcus aureus and had one active spot on the bioautography test.

The radish (Raphanus sativus L.) is a vegetable of the Brassicaceae family cultivated worldwide and has several medicinal properties. Its biological activities are related to various secondary metabolites present in the species, especially phenolics. Thus, the objectives of this study were the chemical analysis and evaluation of the antioxidant and antimicrobial activities of the dry extract and fractions of the fodder turnip leaves (R. sativus var. oleiferus Metzg.). Samples were analyzed by mass spectrometry and the antioxidant activity was evaluated using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical method and the reducing power method. Antimicrobial activity was determined by the agar diffusion and microdilution methods. The total phenols were concentrated in the butanol fraction (121.27 mg GAE/g) and the flavonoids were concentrated in the ethyl acetate fraction (98.02 mg EQ/g). The ethyl acetate fraction showed the best antioxidants results, with 83.45% of free radical scavenging and 11.34% of ferric ions reduction. The analysis of antimicrobial activity showed that the dry extract had the highest average zone of inhibition against Bacillus subtilis (18.67 mm). Smaller values of the minimum inhibitory concentration for Micrococcus luteus were, and the ethyl acetate fraction showed a lower minimum inhibitory concentration (0.1 mg/ml) for that microorganism. There was a strong correlation between the antioxidant activity and the content of phenols and flavonoids. The results showed the potential antioxidant and antimicrobial activities of this extract with the ethyl acetate fraction being most promising for further studies.

Benchalokawichian (BLW) remedy is a Thai traditional medicine that has been notified in the List of Medicine Products of the National List of Essential Drugs A.D. 2006 and has long been used as an antipyretic. The phytochemicals of the 70% and 95% ethanolic extracts of were studied by color reaction test and HPTLC analysis. The BLW remedy showed a highest amounts of 70% ethanolic extractives (ca. 3.99 ± 0.60% of dry material weight). The several ethanolic extracts showed similar qualitative phytochemicals. The major of phytochemicals identified in these extracts were alkaloids, flavonoids, tannins, and polyphenols. The antioxidant activity was assessed using the 2,2-diphenyl-1-picrylhydrazyl assay. The analysed 70% ethanolic extract showed a highest antioxidant activity at IC50= 526.09 µg/ml and both extracts not showed tyrosinase inhibitory effect. The 95% ethanolic extract showed the greatest anti-propionibacterium acnes activity with inhibition zone of 10.10 ± 0.45 mm and minimum inhibitory concentration (MIC) was 5.05 mg/ml

 Objective: This research focuses on the antibacterial activities of fractions of ethyl acetate Garcinia latissima Miq. extracts against Bacillus subtilis and Pseudomonas aeruginosa.Methods: For each bacteria, the values of the inhibitory zone diameter were determined using paper discs method, while the minimum inhibitory concentration (MIC) was determined by microdilution and antibacterial assay was determined by bioautography. Fractionation was carried out using gradient elution with silica gel column as stationary phase successively increasing the separation according to the polarity (using combination of eluents; n-hexane, ethyl acetate, and methanol).Results: To determine the diameter of the inhibitory zone for each fraction, the fractions were diluted with dimethyl sulfoxide until the concentration of each fraction was 20,000 ppm. The results showed that fraction B had no inhibitory zone against B. subtilis. The inhibitory zone diameters of fractions A, C, D, E, F, and G against B. subtilis were 7.600±0.000, 6.767±0.202, 7.950±0.477, 7.883±0.901, and 9.233±0.231 mm, respectively. Only Fraction G had an inhibition zone diameter of 7.200±0.173 mm against P. aeruginosa. The active fractions were tested by contact bioautography using silica gel (60 GF254) plate and hexane:chloroform (2:3) as mobile phase for Fractions A, C, D, E, and F and chloroform:ethyl acetate (4:1) as mobile phase for Fraction G. The thin layer chromatography plates were observed under the ultraviolet light at wavelengths of 254 and 366 nm, while the others plate (using in contact bioautography) placing on agar medium that had been inoculated with the bacteria, so the compounds could be diffuse to the medium. The active compound will appeared inhibitory zones in agar medium. The determination of the MIC values using microdilution with methylthiazol tetrazolium salt showed that fractions of the ethyl acetate extracts of G. latissima Miq. stem bark inhibited the growth of B. subtilis more actively than P. aeruginosa. The highest activity of the fractions against B. subtilis was shown by fraction G, which had a MIC value of 78.125 ppm, followed by fractions F (156.25 ppm), E (312.5 ppm), D (625 ppm), C (1250 ppm), B (2500 ppm), and A (5000 ppm). The highest activity of the fractions against P. aeruginosa was Fraction G (5000 ppm), while the MIC values of other fractions were >5000 ppm.Conclusion: Further investigations should be conducted to obtain the new antibacterial compounds in fractions of ethyl acetate extract of G. latissima Miq. stem bark that effective to inhibit the growth of B. subtilis and P. aeruginosa.

Background: gram-negative bacteria-salmonella typhi and gram-positive bacteria-staphylococcus aureus are types of bacteria that cause infectious disease among several areas in Indonesia. Staphylococcus aureus often found in skin lesions injury. Red ginger extract (Zingiber officinale var rubrum) has been known for its medicinal value especially as an antibacterial agent. Purpose: This study aims to test the ability of Zingiber officinale var rubrum on inhibiting the growth of Staphylococcus aureus. Methods: the activity of the antibacterial agent was calculated based on Minimum Inhibitory Concentration (MIC) using dilution method. Minimum Inhibitory Concentration of extract was determined by incubating bacteria with the extract overnight at 37 0C. Changes in turbidity and inhibition on bacterial growth were then observed. Result: The results show that minimum concentration for inhibitory effect was 1000 μg/mL.Conclusion: extract of Zingiber officinale var rubrum have an inhibitory effect on the growth of Staphylococcus aureus but no activity of killing bacterial was observed on the concentration given.

Antibacterial Extract of Mangosteen (Garcinia mangostana L.)Fruit Shell           The skin of the mangosteen fruit is extracted with n-hexane and ethyl acetate to determine the Minimum Inhibitory Concentration (MIC) against Staphylococcus aureus ATCC 25923 and Pseudomonas aeruginosa ATCC 27 853. Results showed that n-hexane extract gave inhibition zone larger than the ethyl acetate extract on all concentrations . Extract n-hexane has a value of MIC against S. aureus ATCC bacterial test 25923 62.5 mg / ml while the ethyl acetate extract of 125 mg / ml . N- hexane extracts had MIC values of the test bacteria P.aeuroginosa ATCC 27 853 was 125 mg / ml , and while the ethyl acetate extract had a MIC value of 500 mg / ml . Treatment of solvent, concentration and interaction between the solvent and concentration significantly affected the test bacteria ATCC 25923 S. aureus at the level of 5 %, the highest interaction N-Hexane solvent with a concentration of 15,625 mg / ml and was not significantly different interactions with the concentration of 31.25 mg/ml and 125 mg/ml. Treatment solvent and concentration significantly while the interaction between the solvent and the concentration has no effect on the test bacteria P.aeuroginosa ATCC 27 853 at 5% level .Keywords: Garcinia mangostana L., Staphylococcus aureus and Pseudomonas aeruginosa ABSTRAK           Kulit buah manggis diekstrak dengan n-heksan dan etil asetat   untuk mengetahui Konsentrasi Hambat Minimum (KHTM) terhadap bakteri Staphylococcus aureus ATCC 25923 dan Pseudomonas aeruginosa ATCC 27853. Hasil penelitian menunjukkan bahwa ekstrak n-heksana  memberikan zona hambatan lebih besar dibandingkan dengan ekstrak etil asetat pada semua konsentrasi. Ekstrak n-heksana  memiliki nilai kadar hambat minimal (KHM) terhadap bakteri uji S. aureus ATCC 25923  62,5 mg/ml sedangkan ekstrak etil asetat 125 mg/ml.   Ekstrak n-heksana memiliki nilai KHM   terhadap bakteri uji P.aeuroginosa  ATCC 27853 adalah 125 mg/ml dan  sedangkan ekstrak etil asetat memiliki nilai KHM  500 mg/ml. Perlakuan pelarut, konsentrasi dan interaksi antara pelarut dan konsentrasi  berpengaruh nyata terhadap bakteri uji S. aureus ATCC 25923 pada taraf 5%, interaksi tertinggi yaitu pelarut N-Heksan dengan konsentrasi 15,625mg/ml dan interaksi ini tidak berbeda nyata dengan konsentrasi 31,25 mg/ml dan 125 mg/ml. Perlakuan pelarut dan konsentrasi  berpengaruh nyata sedangkan interaksi antara pelarut dan konsentrasi  tidak berpengaruh terhadap bakteri uji P.aeuroginosa  ATCC 27853 pada taraf 5%. Kata kunci:  Garcinia mangostana L., Staphylococcus aureus dan Pseudomonas aeruginosa

Objective: The main objective of this research were screened in vitro and in silico of 1,5-bis (3'-ethoxy-4'-hydroxyphenyl)-1,4-pentadiene-3-one as potential antibacterial agents.Methods: The in vitro antibacterial study was carried against Staphylococcus aureus, Staphylococcus epidermidis (gram positive) and Escherichia coli, Salmonella thypi (gram negative) using broth dilution method to determine Minimum Inhibitory Concentration (MIC), disc diffusion method to determine the diameter of inhibition zone. In silico antibacterial study was carried using computational software Protein-Ligand ANT System (PLANTS), computational docking was carried using receptor with Protein Data Bank (PDB) file 3MZD. The structures were optimized prior docking using YASARA, and MarvinSketch. The results of antibacterial testing were compared to two positive control drugs i. e amoxicillin and cefadroxil.Results: In vitro evaluation showed that 1,5-bis (3'-ethoxy-4'-hydroxyphenyl)-1,4-pentadiene-3-one has a better antibacterial activity than amoxicillin and cefadroxil with a Minimum Inhibitory Concentration (MIC) of 0.15 ppm and diameter of inhibition zone of 11.27±0.31, 11.35±0.39, 11.25±0.33, and 11.05±0.45 mm in Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, and Salmonella thypi, respectively. These results in line with in silico evaluation that showed 1,5-bis (3'-ethoxy-4'-hydroxyphenyl)-1,4-pentadiene-3-one has more negative docking score than amoxicillin, cefadroxil, and cloxacillin acyl as a native ligand on the 3MZD receptor.Conclusion: This results obtained in this research work were 1,5-bis (3'-ethoxy-4'-hydroxyphenyl)-1,4-pentadiene-3-one compound potential as an antibacterial agent.

    The effects of aqueous and alcoholic of extracts Mentha pulegium leaves were studied on the growth of strain Staphylococcus aureus that isolated from burns. The Phytochemical analysis of The aqueous and alcoholic extracts of Mentha pulegium leaves show the presence of bioactive constituents like comarins, steroids, tannins, glycosides, saponins, alkaloids, flavonoids, phenols  and essential oil. the ethanolic extract show an inhibition zone larger than aqueous extract.Results show that the ethanolic extract inhibits bacterial growth with inhibition zone(30.33mm) but the aqueous extract give an inhibition zone(8mm). In this study, minimum inhibitory concentration (MIC) was detected and the result varies according to the type of extract.

One of the medicinal plants used by the people of Katingan Regency, Central Kalimantan Province is the Saluang Belum stem, which empirically used as an aphrodisiac to increase stamina, sexual and fertility of male, and as an antioxidant. The purpose of this study was to find out the inhibition and to know which concentration of ethanol extract of Saluang Belum stem which had been able to inhibit the growth of Staphylococcus aureus bacteria. This research uses an experimental method with a laboratory approach. The results of the inhibitory zone of ethanol extract of Saluang Belum stem at a concentration of 0.5% was 26.7 � 2.76 mm, at a concentration of 1% was 21.6 � 2.20 mm, at a concentration of 5% was 20.5 � 0, 90 mm, at 10% concentration was 21.2 � 1.11 mm, at 15% concentration was 23.2 � 0.23 mm, and at 20% concentration was 25.5 � 0.36 mm. Conclusions from this study were that the MIC (Minimum Inhibitory Concentration) obtained at 0.5% concentration of ethanol extract Batang Saluang Belum was able to inhibit the Staphylococcus aureus bacteria.

Penelitian terhadap ekstrak metanol, fraksi etil asetat, dan fraksi butanol dari rumput laut Eucheuma spinosum menggunakan metode Disc Diffusion Test dilakukan untuk mengetahui peranannya sebagai antibakteri Escherichia coli ATCC 25922, dan Staphylococcus aureus ATCC 6538. Sebagai kontrol digunakan ekstrak (negatif) pelarut metanol, dan Amoxicilin. Konsentrasi sampel uji ditetapkan 10.000 ppm. Hasil pengujian menunjukkan adanya  aktivitas antibakteri yang ditandai dengan terbentuknya zona hambat terhadap bakteri Escherichia coli ATCC 25922.  Besarnya zona hambat secara berurutan untuk ekstrak metanol, fraksi  butanol, dan fraksi etil asetat, ialah : 6,98 mm, 7,85 mm, dan 7,88 mm.  Nilai zona hambat tersebut, relatif sama untuk setiap sampel uji. Sampel uji juga menunjukkan terdapat aktivitas antibakteri, ditandai dengan terbentuknya zona hambat terhadap bakteri Staphylococcus aureus ATCC 6538.  Besarnya zona hambat secara berurutan untuk ekstrak metanol, fraksi  butanol, fraksi etil asetat,  dan kontrol negatif pelarut metanol, ialah : 7,42 mm, 14.40 mm, 7,90 mm, dan 6,75 mm.  Zona hambat fraksi butanol lebih tinggi dibanding yang lain, ini menunjukkan bahwa aktivitas antibakteri Staphylococcus aureus pada fraksi butanol lebih tinggi dibanding fraksi lainnya.Kata kunci:  Antibakteri;  Eucheuma spinosum;  Escherichia coli; Staphylococcus aureus Antibacteria Activities of Extract Sea Algae  Eucheuma spinosum Fresh from Nain Island Waters North Sulawesi  ABSTRACTA research on antibacterial activity of methanol extract, ethyl acetate, and butanol fraction from Eucheuma spinosum seaweed using the disc diffusion method to know the role as  antibacterial activity was tested against Escherichia coli ATCC 25922 and Staphylococcus aureus ATCC 6538. Methanol was used as negative control while amoxicillin was used as positive control. The concentration of the test sample was set at 10,000 ppm. The antibacterial activity was indicated by the formation of an inhibition zone against the tested microorganisms. The inhibition zones against E.coli for methanol extract, butanol fraction, and ethyl acetate fraction were 6.98 mm, 7.85 mm, and 7.88 mm, respectively.  The value of the zone of inhibition is relatively the same for each sample.  In addition, the inhibition zones of methanol extract, butanol and ethyl acetate fraction against Staphylococcus aureus were 7.42 mm, 14.40 mm, 7.90 mm, and 6.75 mm, respectively. The inhibition zone of the butanol fraction is higher than the others, this indicated that the antibacterial activity for butanol fraction is higher than other fractions. Keywords: Antibacterial activity; Eucheuma spinosum; Escherichia coli; Staphylococcus aureus
