Effect of Storage of Loblolly and Slash Pine Cones on Seed Quality

Abstract Effects of cone storage on seed quality were evaluated in studies conducted in Mississippi and Alabama on drying rates and storage conditions (bag vs. crate). The drying study found that: (1) Cone drying was equally rapid in indoor and outdoor covered conditions and slower in unprotected outdoor conditions; (2) seed extraction efficiency was significantly affected by all treatments with the best efficiency from cones stored outdoors unprotected; and (3) germination rate was more sensitive to treatment conditions than total germination. The second study compared burlap bagsto 20-bushel wirebound crates as storage containers for loblolly (Pinus–taeda L.) or slash (P. elliottii Engelm.) pine cones stored for varying periods. Study results reveal that (1) either bags or crates provide satisfactory storage if handled properly and (2) cone storage (4 weeks for slash,5 to 7 weeks for loblolly) enhances seed quality through apparent improved maturation. Forage grass seed quality is influenced by plant genetics as well as seed density, storage conditions, age of seed, and endophyte infection status. We used eight seed lots of tall fescue (three cultivars, with natural endophytes, endophyte-free, or novel endophyte) to test the effect of seed density on germination and seedling vigour. Seed lots were separated into nine density fractions using a cylinder air column. Thousand seed weight values were determined. Three runs of four replications of plots consisting of 25 seeds of each entry were seeded in greenhouse experiments to determine seedling mass at 8 weeks after planting. Early germination counts as well as laboratory germination tests indicated significant differences among weight classes in speed of germination as well as seedling vigour. The lightest fraction of seed clearly contained inferior seed, but higher density seed lots tended to have better germination and seedling vigour than lower density ones. Abstract: The aim of this study was to evaluate the effects of storage conditions on the germination of A. cearensis seeds. The experimental design was completely randomized into split plots over time with four replicates. The storage conditions of the airtight containers in the refrigerator and laboratory, paper bags in the laboratory and liquid nitrogen were assessed for 27 months. In the laboratory, we evaluated the germination, the germination rate, uniformity of germination, and total soluble and reducing sugars in the radicle. In the greenhouse, we evaluated seedling emergence, emergence rate and height of 30-day-old seedlings. Seeds stored in the refrigerator maintained a high initial germination rate, which decreased from the 21st month. Seeds stored in paper bags in the laboratory showed low emergence and small seedlings. Total soluble sugars and reducing sugars were mobilized when the seeds were stored at low temperatures. Thus, it is not advisable to store A. cearensis seeds in a laboratory environment without airtight containers. A. cearensis seeds kept in a refrigerated environment maintained their viability for at least two years.

Abstract The effect of cone storage on seed viability during 5 years of storage at 35°F was tested with seeds from studies with loblolly (Pinus taeda L.), slash (P. elliottii Engelm.), longleaf (P. palustris Mill.), and eastern white (P. strobus L.) pines. Results showed that: (1) with the exception of slash pine, seeds from cones stored for the longer periods retained quality during storage better than seeds from cones that were stored only short periods; (2) all species exhibited slight, but statistically significant, decreases in seed quality after 3 years of storage, and with the exception of loblolly, more substantial decreases in germination rate at 5 years; (3) with the exception of eastern white pine, declining rates of germination indicated losses of seed quality at least one year before total percent germination decreased, and (4) slash pine seeds from cones in the center of 20-bushel crates performed better in storage than those from cones at the top of the crates or from cones stored in bags, but storage container made no difference for loblolly. ABSTRACT The drying process and storage may interfere in the quality of creeping signalgrass seeds [Urochloa humidicola (Rendle.) Morrone & Zuloago, syn. Brachiaria humidicola (Rendle.) Schweick.]. This study aimed to evaluate the physical, physiological and sanitary quality of creeping signalgrass cv. Llanero seeds submitted to drying surfaces and environments and stored for two periods (1 and 8 months). The experiment was arranged in a completely randomized design, in a 4 × 2 × 2 + 2 factorial arrangement, with four drying surfaces (soil, asphalt, concrete and ceramic floor), two drying environments (seeds exposed to the sun or shade), two seed storage periods (1 and 8 months) and two additional treatments with drying in paper bags under laboratory conditions (control) and stored for 1 or 8 months, with four replicates. The water content, viability by the tetrazolium test, electrical conductivity, germination rate, germination rate index and seed sanitary quality were measured. Drying at full sun impaired the physiological quality of the seeds due to the high temperatures (46.8-51.0 ºC) of the drying surface; however, reduced the incidence of pathogens. The drying of seeds in paper bags under shade results in a higher physiological quality, but the sanitary quality is compromised. However, the pathogens present in the seeds did not inhibit their viability and vigor. The storage for eight months reduces the seed quality.

AbstractThe goal of this work was to determine the best method of breaking the achene dormancy in Potamogeton praelongus Wulfen. The ways of breaking achene dormancy studied in this experiment included methods of achene storage, stratification, UVA radiation, anaerobic conditions, mechanical disruption of achenes?outer layers and their chemical disruption by NaClO. Nine different treatments of achenes were combined with two methods of achene storage. Particular achene treatments and storage conditions were proven to have a significant impact on breaking dormancy. Although the highest germination rate (83.3%) was achieved when the dormancy was broken chemically by long effect of 100% concentrations of Savo detergent (containing 5% NaClO), the growth of the sprouts was subsequently inhibited due to toxic effects of Savo. Thus the most successful treatment was based on changing temperature, e.g. 2.5 months of cold storage followed by 14 days at room temperature (germination rate 32.7%). This treatment was also most similar to the natural process. Germinated achenes were also found in Petri dishes exposed to UVA radiation, anaerobic conditions and chemical disruption of the outer layers. Results of these treatments were influenced by the storage method.

Abstract Although longleaf pine (Pinus palustris Mill.) seeds are considered the most susceptible of the southern pines to damage during collection, processing, and storage, results of these studies show that high seed quality can be assured for periods up to 20 yr through proper handling and storing techniques. Recommendations for long-term storage include drying seeds to moisture contents of 10% or less and storing at subfreezing temperatures, preferably near 0°F. Reevaluation of stratification treatments applied under operational conditions indicates that the soaking in water that is necessary for seed imbibition reduces total germination in an amount proportional to the length of the soak. Stratification is not recommended except under very controlled conditions. South. J. Appl. For. 17(4):174-179.

Fresh seeds of cocoa (Theobroma cacao L.) are covered by a large amount of mucilage, which plays important ecological roles. However, for stored seeds, mucilage can adversely affect germination. This study investigated the effect of mucilage removal (demucilaging) and seed storage of freshly extracted cocoa seeds on seedling emergence and growth. The study showed that demucilaging cocoa seeds confers several advantages over mucilage intact seeds provided the seeds have been stored for at least three days before sowing. Demucilaging increased germination rate (T50) and percentage germination and presence of mucilage increased days to seedling emergence and cessation of germination when seeds were stored for over two days. For up to 21 days of seed storage, there was generally no difference in many traits for demucilaged seeds. Demucilaged seeds had 80–100% germination after 21 days of storage, and rate of germination increased with increasing duration of seed storage. Intact mucilage seeds maintained 80–100% germination for only 2 days after seed extraction and rate of germination decreased with increasing seed storage duration. We conclude that (i) if seeds are sown on the day of extraction demucilaging before sowing would be superfluous and (ii) demucilaging and storage can provide advantages to smallholder farmers when environmental or socioeconomic conditions preclude immediate sowing.
