Over a billion people on earth are infected with helminth parasites and show remarkable variation in parasite burden and chronicity. These parasite distributions are captured well by classic statistics, such as the negative binomial distribution. But the within-host processes underlying this variation are not well understood. In this study, we explain variation in macroparasite infection outcomes on the basis of resource flows within hosts. Resource flows realize the interactions between parasites and host immunity and metabolism. When host metabolism is modulated by parasites, we find a positive feedback of parasites on their own resources. While this positive feedback results in parasites improving their resource availability at high burdens, giving rise to chronic infections, it also results in a threshold biomass required for parasites to establish in the host, giving rise to acute infections when biomass fails to clear the threshold. Our finding of chronic and acute outcomes in bistability contrasts with classic theory, yet is congruent with the variation in helminth burdens observed in human and wildlife populations.

Ommastrephid squid of the subspecies Illex illecebrosus illecebrosus (LeSueur) were obtained during the late summer and early autumn of 1966 and 1967 from numerous areas around the eastern coast of the island of Newfoundland. Helminths of five genera were found, namely, Phyllobothrium sp. and Dinobothrium plicitum (both of which had previously been recorded from this host), Pelichnibothrium speciosum, Scolex polymorphus, and Nybelinia sp. The last three represent new records from this ommastrephid. The incidence of these parasites was tested for correlation with standard length and with sex of the host animal. Annual and seasonal variations in degree of infection were also noted. The greatest number of procercoids encountered were Dinobothrium plicitum. Degree of infection with helminths in 1966 and 1967 was similar (39%:40%) and contrary to earlier published data, infection by cestodes is not always directly correlated with mantle length. Any possible relations between sex of the squid and parasite burden are unclear.

The number of joint replacements performed in resource-rich countries has increased significantly in the last few decades and has allowed a significant improvement in the quality of life for many patients. Despite preventative measures, a small fraction (around 1% for hip replacements) will become infected. These can present as acute or chronic infections. They can also occur at any time after the primary procedure. Late acute infections are usually via the haematogenous route and often present to the acute medical take or via infection services. A prompt, appropriate medical and surgical management strategy is important.

AbstractPlasmodium falciparum infection during pregnancy is a major cause of severe maternal illness and neonatal mortality. Mouse models are important for the study of gestational malaria pathogenesis. When infected with Plasmodium chabaudi chabaudi AS in early gestation, several inbred mouse strains abort at midgestation. We report here that outbred Swiss Webster mice infected with P. chabaudi chabaudi AS in early gestation carry their pregnancies to term despite high parasite burden and malarial hemozoin accumulation in the placenta at midgestation, with the latter associated with induction of heme oxygenase 1 expression. Infection yields reduced fetal weight and viability at term and a reduction in pup number at weaning, but does not influence postnatal growth prior to weaning. This novel model allows for the exploration of malaria infection throughout pregnancy, modeling chronic infections observed in pregnant women prior to the birth of underweight infants and enabling the production of progeny exposed to malaria in utero, which is critical for understanding the postnatal repercussions of gestational malaria. The use of outbred mice allows for the exploration of gestational malaria in a genetically diverse model system, better recapitulating the diversity of infection responses observed in human populations.

ABSTRACT Infections with Trypanosoma cruzi are usually lifelong despite generating a strong adaptive immune response. Identifying the sites of parasite persistence is therefore crucial to understanding how T. cruzi avoids immune-mediated destruction. However, this is a major technical challenge, because the parasite burden during chronic infections is extremely low. Here, we describe an integrated approach involving comprehensive tissue processing, ex vivo imaging, and confocal microscopy, which allowed us to visualize infected host cells in murine tissue with exquisite sensitivity. Using bioluminescence-guided tissue sampling, with a detection level of <20 parasites, we showed that in the colon, smooth muscle myocytes in the circular muscle layer are the most common infected host cell type. Typically, during chronic infections, the entire colon of a mouse contains only a few hundred parasites, often concentrated in a small number of cells each containing >200 parasites, which we term mega-nests. In contrast, during the acute stage, when the total parasite burden is considerably higher and many cells are infected, nests containing >50 parasites are rarely found. In C3H/HeN mice, but not BALB/c mice, we identified skeletal muscle as a major site of persistence during the chronic stage, with most parasites being found in large mega-nests within the muscle fibers. Finally, we report that parasites are also frequently found in the skin during chronic murine infections, often in multiple infection foci. In addition to being a site of parasite persistence, this anatomical reservoir could play an important role in insect-mediated transmission and have implications for drug development. IMPORTANCE Trypanosoma cruzi causes Chagas disease, the most important parasitic infection in Latin America. Major pathologies include severe damage to the heart and digestive tract, although symptoms do not usually appear until decades after infection. Research has been hampered by the complex nature of the disease and technical difficulties in locating the extremely low number of parasites. Here, using highly sensitive imaging technology, we reveal the sites of parasite persistence during chronic-stage infections of experimental mice at single-cell resolution. We show that parasites are frequently located in smooth muscle cells in the circular muscle layer of the colon and that skeletal muscle cells and the skin can also be important reservoirs. This information provides a framework for investigating how the parasite is able to survive as a lifelong infection, despite a vigorous immune response. It also informs drug development strategies by identifying tissue sites that must be accessed to achieve a curative outcome.

SummaryExperiments were undertaken to investigate the factors which influence the transmission dynamics of Gyrodactylus bullatarudis within populations of laboratory guppies. The parasites possess a number of biological attributes which are almost unique amongst helminth parasites of vertebrates. These include the ability to reproduce viviparously and directly on the surface of the host, the ability to survive death of the host (for a short period of time) and the ability to transfer between hosts (despite the absence of a specialized transmission stage in the parasite's life-cycle). Long-term laboratory experiments demonstrated the inability of the parasite to persist within populations of the host in the absence of the continual introduction of naive susceptible fish. With regular addition of susceptible fish, the parasite population exhibited recurrent epidemic behaviour. The magnitudes of the epidemics and the time interval between them, were dependent on the rate at which fish were added to the populations. The parasite was over-dispersed in its distribution within the experimental fish populations and was a significant cause of host mortality (in a manner related to parasite burden). The experimental results suggest that acquired resistance to infection is an important factor determining epidemic behaviour.

Chronic (long-lasting) infections are globally a major and rising cause of morbidity and mortality. Unlike typical acute infections, chronic infections are ecologically diverse, characterized by the presence of a polymicrobial mix of opportunistic pathogens and human-associated commensals. To address the challenge of chronic infection microbiomes, we focus on a particularly well-characterized disease, cystic fibrosis (CF), where polymicrobial lung infections persist for decades despite frequent exposure to antibiotics. Epidemiological analyses point to conflicting results on the benefits of antibiotic treatment, and are confounded by the dependency of antibiotic exposures on prior pathogen presence, limiting their ability to draw causal inferences on the relationships between antibiotic exposure and pathogen dynamics. To address this limitation, we develop a synthetic infection microbiome model, and benchmark on clinical data. We show that, in the absence of antibiotics, the microbiome structure in a synthetic sputum medium is highly repeatable and dominated by oral commensals. In contrast, challenge with physiologically relevant antibiotic doses leads to substantial community perturbation characterized by multiple alternate pathogen-dominant states and enrichment of drug-resistant species. These results provide evidence that antibiotics can drive the expansion (via competitive release) of previously rare opportunistic pathogens and offer a path towards microbiome-informed conditional treatment strategies.

Helminth parasites modulate immune responses in their host to prevent their elimination and to establish chronic infections. Our previous studies indicate that Taenia crassiceps-excreted/secreted antigens (TcES) downregulate inflammatory responses in rodent models of autoimmune diseases, by promoting the generation of alternatively activated-like macrophages (M2) in vivo. However, the molecular mechanisms triggered by TcES that modulate macrophage polarization and inflammatory response remain unclear. Here, we found that, while TcES reduced the production of inflammatory cytokines (IL-6, IL-12, and TNFα), they increased the release of IL-10 in LPS-induced bone marrow-derived macrophages (BMDM). However, TcES alone or in combination with LPS or IL-4 failed to increase the production of the canonical M1 or M2 markers in BMDM. To further define the anti-inflammatory effect of TcES in the response of LPS-stimulated macrophages, we performed transcriptomic array analyses of mRNA and microRNA to evaluate their levels. Although the addition of TcES to LPS-stimulated BMDM induced modest changes in the inflammatory mRNA profile, it induced the production of mRNAs associated with the activation of different receptors, phagocytosis, and M2-like phenotype. Moreover, we found that TcES induced upregulation of specific microRNAs, including miR-125a-5p, miR-762, and miR-484, which are predicted to target canonical inflammatory molecules and pathways in LPS-induced BMDM. These results suggest that TcES can modulate proinflammatory responses in macrophages by inducing regulatory posttranscriptional mechanisms and hence reduce detrimental outcomes in hosts running with inflammatory diseases.

The parasitofauna of ground-dwelling anurans from pesticide-treated cocoa plantations (CP) in Ojo Camp, Ugboke, Edo State of Nigeria were investigated and compared with those recovered from host specimens collected from the village settlement (VS). The anurans were caught by hand following visual or acoustic location. The anurans encountered in both the VS and the CP included Aubria subsigillata, Hylarana spp. (H. albolabris and H. galamensis), Sclerophrys spp. (S. maculata and S. regularis), Ptychadena spp. (P. aequiplicata, P. longirostris, P. mascareniensis, P. oxyrhynchus and P. pumilio) and Hoplobatrachus occipitalis. Hylarana galamensis, Ptychadena spp. and Sclerophrys spp. were encountered in the VS and the CP while Aubria subsigillata, H. albolabris and H. occipitalis occurred only in the CP. The helminth parasites recovered included four cestode species (adult of Cylindrotaenia jaegerskioeldi and three encysted  proteocephalid larvae), five Polystoma spp. 11 species of digeneans and 19 nematode species. More parasite species were recovered from toads collected from the VS; parasite prevalence was generally low in both habitats but the intensity of infection was higher in the specimens collected from the VS. Although cip A. subsigillata and H. ocitalis both occurred in the CP, A. subsigillata was the more susceptible host of the two, harbouring 16 helminth parasites as against four from H. occipitalis. Polystomes were recovered from H. albolabris and H. galamensis in addition to Diplodiscus fischthalicus and Mesocoelium spp. Infections occurred  mostly among the Ptychadeniidae collected from the CP, with prevalence ranging from 12.5% to 100% and infection intensity from 1.0 to 13.0. The generally low parasite burden in anurans from the CP can possibly be attributed to the pesticide contamination of this habitat which may have hindered the development of the free-living stages of parasites in this milieu. One thousand and one alcids of six species (Alca torda, Uria aalge, Uria lomvia, Plautus alle, Cepphus grylle, Fratercula arctica) were collected in the northwestern North Atlantic, during the period June, 1966 – September, 1969, and examined for parasites. Helminths belonging to 12 genera were recovered, the common murre (U. aalge) containing the greatest number of species. Statistically significant annual variations (P < 005) in the parasite burden of this species were noted. No differences in intensity of infection in male and female birds and in chicks and adults were detected. Twelve new host records are noted.

The distribution of helminth parasites within their host population is usually overdispersed and can be described by the negative binomial distribution. The causes of this overdispersion are poorly understood, but heterogeneity in the distribution of infective stages within the environment has been implicated as a possible factor. Here we describe the distribution of infective stages of the rat intestinal nematode parasite Strongyloides ratti among the faecal pellets of its host. The distribution of infective stages between faecal pellets is overdispersed and well described by the negative binomial distribution. This overdispersion increases during the course of infection and occurs over a range of infection intensities. Overdispersion of nematode infective stages among faecal pellets may result in increased spatial heterogeneity of the infective stages in the environment and thus may contribute to the generation of overdispersion of adult parasitic stages. In addition, these findings raise important issues regarding the accurate quantification of helminth egg counts from faecal samples.
