Reverse-transcription quantitative PCR (RT-qPCR) is currently the most sensitive method to detect severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), which causes coronavirus disease 2019 (COVID-19). We analysed 1927 samples collected in a local public hospital during the autumn 2020 peak of the pandemic in the Czech Republic. The tests were performed using the Seegene Allplex 2019-nCov assay, which simultaneously detects three SARS-CoV-2 genes. In all samples analysed, 44.5 % were negative for all three genes, and 37.6 % were undoubtedly positive, with all three viral genes being amplified. A high degree of correlation between C t values among the genes confirmed the internal consistency of testing. Most of the positive samples were detected between the 15th and 35th cycles. We also registered a small number of samples with only one (13.2 %) or two (4.7 %) amplified genes, which may have originated from either freshly infected or already recovering patients. In addition, we did not detect any potentially false-positive samples from low-prevalence settings. Our results document that PCR testing represents a reliable and robust method for routine diagnostic detection of SARS-CoV-2.

Peach latent mosaic viroid (PLMVd) is widespread in peach all over the world. It has never been reported from the Czech Republic. That is why we adapted specific and sensitive method for its detection, PCR, to be able to prove its possible occurrence and for certification purposes. Primers PLMVdR, PLMVdF1 and PLMVdF2 were designed on the basis of published RNA sequences. Products of amplification are 208 and 114 bp long for PLMVdF1 and PLMVdF2, respectively. Four PLMVd isolates from Dr Di Serio (CNR Bari) were used as standards. Potato spindle tuber viroid and Hop latent viroid infected plant material and also healthy material were used to check detection specifity. Both RNA extraction from plant material and PCR were optimalized so that this method of PLMVd detection can also be used for certification purposes.

The presented article is based on research evaluating the impact of cluster organisations on the financial performance of member entities. The author’s doctoral thesis examines whether there is a difference in the financial performance of cluster organisations created through the bottom-up and the top-down approaches, under the conditions existing in the Czech Republic. Both types of clusters that meet the condition of maturity (established before or in 2012) and of a high degree of activity were selected for the research. The financial performance of member business entities was assessed using the following indicators: ROA, ROE, ROS, EVA, EVA/employee and EVA/sales. The aim of the research was to demonstrate whether public support for clusters would be reflected in member entities’ better financial performance. The final part of the paper then summarises and discusses the findings.

Early neurological Lyme disease (neuroborreliosis) typically presents with well-recognised neurological syndromes. Spinal myoclonus is however a rare manifestation of neuroborreliosis. We present the case of a man who developed spinal myoclonus 3 weeks after returning from the Czech Republic where he developed erythema migrans on his arm following multiple tick exposures. Spinal fluid analysis showed a pleocytosis and MRI showed enhancement at C5-C6. His serology was positive for IgM antibodies to Borrelia afzelli. He was successfully treated with ceftriaxone and doxycycline with improvement of his spinal myoclonus and radiculitis. We conclude that early Lyme neuroborreliosis may present with uncommon neurological manifestations and so a high degree of suspicion is needed.

Air pollution is changing the ecosystems, especially in Europe. Tropospheric ozone may adversely affect tree growth, with critical levels for ozone being exceeded in many parts of Europe, especially in the Czech Republic. This research reports the information on exceedances of ozone threshold values for the period 1994 to 2000 and gives an evaluation of the observed exceedances of the thresholds during the vegetation period (April-September) and from May to July at the Bílý Køíž station. The threshold for warning of the public (240 ìg m-3 as hourly average concentration) was not exceeded at the Bílý Køíž station for the studied period. The threshold value for information to the population (180 ìg m-3 as hourly average concentration) was exceeded in 1994 (July and August), 1995 (May and July), 1998 (August) and 2000 (June). The threshold value set for the protection of human health population (120 ìg m-3 as eight-hourly average concentration) was exceeded for all period at the Bílý Køíž station.

Even in the case of an optimistic scenario of joining the EU at an early stage in the next century, CE countries are highly unlikely to participate in the take-off stage of EMU. Instead, they are expected to address some more elementary issues related to their transition processes. But the fact of being a small and open economy implies a high degree of macroeconomic discipline, in many respects not so much unlike that embedded in the Maastricht convergence criteria. Whatever disputes many surround the Maastricht convergence criteria, one can scarcely deny that they do suggest directions in which CE economies need to further upgrade their fundamentals. They put aside, however, so-called real convergence measured by the level of GDP per capita. The currency crisis in the Czech Republic delivered the lesson that the growth rates, if not underpinned by a healthy macroeconomic background, are short-lived and bring only lags Into the catching-up process.

The relationship between detrended district yields of spring barley (1961&minus;2000) and meteorological drought was assessed by Palmer Z-index. We found that the seasonal water balance (April&minus;June) significantly (<i>P</i> = 0.05) influences the spring barley production in 51 out of 62 evaluated districts. Coefficients of correlation varied in individual districts from 0.19 to 0.70, with the highest values being found in southern Moravia. Data analysis revealed the presence of six distinct groups of districts with a specific drought-yield relationship. The most drought-sensitive cluster included five districts in the South East of the country. On the other hand, the districts in Northern Moravia and Silesia belong among the least sensitive to agricultural drought. The study also defined threshold values of seasonal drought (sums of relative Palmer Z-index lower than &minus;8 and &minus;10, respectively), below which medium to severe spring barley yield reductions are very likely, regardless of the district of occurrence.

Czech-Armenians relations: a brief historical survey Armenian studies in Czech Republic became more widely known toCzech public in recent years. Following up the long tradition of Oriental studies ingeneral, a newly re-established research centre was opened at the Department ofEast-European Studies at Charles University, aiming to cooperate with other colleagues.After long and heated discussions, Czech Republic decided on (albeit nonofficial) recognition of Armenian Genocide and supports initiatives to prevent suchviolence in future. Several monographs dealing with this subject were published recently.And finally, despite the fact that the Czech Republic has no historical experience with the presence of Armenian diaspora before the year 1990, well establishedand coherent Armenian community exists here now and – especially in Prague –shows a high degree of integration into Czech majority. Already two generations ofCzech Armenians are following their identity strategies while preserving their owncultural identity.

The aim of this study was to screen selected Czech dairy cattle farms for presence of Mycoplasma bovis. Between 2012 and 2014, 160 individual samples of cow milk, 60 colostrum samples, and 105 bulk-tank milk samples collected on 21 dairy farms were analyzed in our laboratory. DNA was extracted from samples and tested by quantitative PCR assay for M. bovis. All analyzed samples were negative. Results support long-term trend of very low prevalence of M. bovis in the Czech Republic.

Among European countries the Czech Republic has a high degree of knowledge of the vegetation cover. Suffice it to recall that in 1960-1970 was published a multivolume series of books «Vegetace ČSSR», where even then with great detail characterized the vegetation of this country.

Abstract We propose using a simple Taylor rule to evaluate business cycle convergence of the Czech Republic, Hungary, and Poland with the Eurozone. Our findings indicate an ongoing convergence of those CEE countries to the Eurozone, but with instabilities and heterogeneity between the countries. Especially Poland has shown a high degree of convergence in recent years. But there are still relevant differences in Taylor rates of each country to the Eurozone of about two percentage points.
