Heredity is generally felt to play a minor role in the development of cancer. This review critically examines this assumption. Topics discussed include evidence for heritable predisposition in animals and humans; the potential importance of genetic-environmental interactions; approaches that are being used to successfully locate genes responsible for heritable predisposition; comparability of genetic findings among heritable and corresponding sporadic malignancies; and future research directions. Breast, colon, and lung cancer are used to exemplify clinical and research activity in familial cancer; clinical phenotypes, segregation and linkage analyses, models for environmental interactions with inherited traits, and molecular mechanisms of tumor development are discussed. We conclude that the contribution of heredity to the cancer burden is greater than generally accepted, and that study of heritable predisposition will continue to reveal carcinogenic mechanisms important to the development of all cancers.

This article presents an overview of the environmental history of the Brazilian Cerrado, its environmental characteristics and the processes related to the historical change in the landscapes of this endangered ecosystem. It highlights competing classifications of the Cerrado, the role of politics in establishing them, and the environmental consequences of such classifications. More than just describing an environment, classifying an ecosystem is a political process that involves complex socio-environmental interactions. The sources used points out the different attempts to get to know and "conquered" the Cerrado, bringing together interdisciplinary perspectives from a variety of actors and institutions. Historiographic challenges go beyond environmental descriptions in that the socio-environmental interactions that made up this unique ecosystem are equally complex. This paper’s conclusions reinforce the interdisciplinary role of environmental history in the study of ecosystems and the complex relationship between culture and nature.

There is significant animal model data demonstrating a benefit of testosterone on both inflammatory and neuroprotective mechanisms relevant to multiple sclerosis (MS). Several studies have demonstrated lowered testosterone levels in up to 40% of men with MS. Lower testosterone levels were correlated with worsened scores of physical and cognitive disability. There is increasing data suggesting a role of testosterone in MS risk. A pilot study has demonstrated significant benefits of testosterone replacement therapy on cognitive, radiological, and immunological outcome measures in men with MS. Larger studies in other conditions have demonstrated concerns in terms of cardiovascular risk, which indicate the need for careful monitoring upon administration to MS patients. Further studies are needed to develop safer testosterone preparations, which preserve its multiple beneficial effects, as well as multicenter clinical trials to evaluate safety, dosing, and efficacy in larger populations of men with MS. Additionally, studies are needed to further explore the role of androgens as a risk factor for MS, particularly at key life transitions.

BackgroundAsthma is a chronic airway disease driven by complex genetic–environmental interactions. The role of epigenetic modifications in bronchial epithelial cells (BECs) in asthma is poorly understood.MethodsWe piloted genome-wide profiling of the enhancer-associated histone modification H3K27ac in BECs from people with asthma (n = 4) and healthy controls (n = 3).ResultsWe identified n = 4,321 (FDR < 0.05) regions exhibiting differential H3K27ac enrichment between asthma and health, clustering at genes associated predominately with epithelial processes (EMT). We identified initial evidence of asthma-associated Super-Enhancers encompassing genes encoding transcription factors (TP63) and enzymes regulating lipid metabolism (PTGS1). We integrated published datasets to identify epithelium-specific transcription factors associated with H3K27ac in asthma (TP73) and identify initial relationships between asthma-associated changes in H3K27ac and transcriptional profiles. Finally, we investigated the potential of CRISPR-based approaches to functionally evaluate H3K27ac-asthma landscape in vitro by identifying guide-RNAs capable of targeting acetylation to asthma DERs and inducing gene expression (TLR3).ConclusionOur small pilot study validates genome-wide approaches for deciphering epigenetic mechanisms underlying asthma pathogenesis in the airways.

Plants have certain characteristics which allow them to respond to various environmental conditions, like changes in climate, water scarcity in the soil, lack of minerals; among others. In some of these traits, the responses to climatic phenomena such as drought can be evidenced through morphological adaptations (spines, succulent tissues, trichomes) or physiological adaptations (regulation of water potential at the cellular level, the concentration of nutrients, etc.). A systematic literature review was performed to study plant functional traits (PFTs) in tropical dry forests (TDFs). The chapter suggests the role of functional traits in community dynamics and processes. The authors will also highlight the limitations of PFTs in TDFs and how they can be improved.
