The free sterol, steryl ester, and steryl glycoside content of tomato cultivars resistant and susceptible to Phytophthora infestans

Leaf sterols were extracted from three late blight resistant ('Nova,' 'New Yorker,' and 'West Virginia 63') and three late blight susceptible ('Beefsteak,' 'Jubilee,' and 'San Marzano') cultivars of tomato (Lycopersicon esculentum Mill.). There were no significant differences (P < 0.05), either quantitatively or qualitatively, between the sterol contents of the resistant and the susceptible cultivars. Of the six cultivars only the steryl glycoside class of 'San Marzano' was significantly higher than that of the other cultivars. The total sterol content was 2.95–3.84 mg/g dry weight and was composed of 23–32% free sterol, 4–6% steryl ester, and 63–71% steryl glycoside. The relative susceptibility of tomato to late blight is not related to the free sterol or conjugated forms of sterols in the plant.

Abstract Total, free and conjugated forms (steryl esters, steryl glycosides and acyl steryl glycosides) of sterols from three microalgae that are extensively used in mariculture (Tetraselmis chuii, Nannochloropsis salina and Skeletonema costatum) were examined. The results revealed that cholesterol is the only common fraction detected in all investigated species and distributed in free and all conjugated forms. However, the total sterol content of T. chuii was about 325 μg/g dry wt, most of it was concentrated amongst 24-methylcholesta-5,24-diene-3β-ol and 24-methylcholest-5-en-3β-ol. On the other hand, the majority of the fractions were distributed in the free form. The total sterol content of N. salina was about 180 μg/g dry wt, cholesterol was the major fraction that was detected. Nevertheless, the dominant distribution forms were esterified. While in S. costatum, the total sterol content was 76 μg/g dry wt, approximately most fractions are quantitatively alike and dominated in the free form. Furthermore, our study shows clearly that most sterols are not distributed regularly within each form, a result that encouraged us to suggest a distribution of specific sterol fraction as a free or conjugated can be used as a serving tool in chemotaxonomic studies.

Function of coprophagous beetles (Coleoptera: Scarabaeidae, Geotrupidae, Hydrophilidae) in cattle pastures inferred from pitfall trapping dataAn analysis of data on the dry weight biomass of coprophagous beetles in standardized dung (4.5 l) was conducted in order to characterize the spatial and the seasonal distribution of the beetles' biomass in cattle pastures and to elucidate their function in dung decomposition. Nested Anova with factors of farm, site (nested in farm), seasonal period and year was used to evaluate the effect of these factors on the biomass of four functional species groups: the dung dwellers ofScarabaeidae(subfamilyAphodiinae), the dung dwellers ofHydrophilidae, the small tunnellers ofScarabaeidae(subfamilyCoprinae) and the large tunnellers ofGeotrupidae. The spatial variation of biomass (between the sites and the farms) was insignificant (P>0.05) in the two dung-dweller groups and in the large-tunnellers group. On the other hand, a significant (P<0.05) seasonal variation of biomass was found in all but the large tunneller group. In dung dwellers, the spring biomass was formed mainly by two species,Aphodius prodromusandA. sphacelatus. In summer, most of the biomass was accounted for bySphaeridium lunatum, S. scarabaeoidesandA. rufipes. In the two tunneller groups,Onthophagus fracticornis, Geotrupes stercorariusandG. spinigerformed a majority of the biomass in dung.

A previous study of lipids from pericarp tissue of tomato fruit ranging from mature-green to red-ripe showed a large increase in total sterols accompanied by dramatic changes in sterol composition and conjugation with ripening. This study was conducted to determine whether similar changes occur in microsomal membranes derived from tomato fruit pericarp. Acylated steryl glycoside (ASG), the predominant steryl lipid, declined during ripening, with increases in steryl glycoside (SG) and free sterol (FS). Only minor changes in fatty acid composition were associated with the drop in ASG. The stigmasterol:sitosterol ratio increased throughout ripening, but much more in Fs than in SG or ASG. The ratio of FS to phospholipid (PL) increased with ripening. However, FS was never greater than 10 percent of the total membrane sterol (TMS), and TMS:PL actually declined over the middle stages of ripening. It is not known why tomato tissues maintain such high levels of ASG and SG, but sterol conjugation is thought to regulate the physical properties of cell membranes.

The aim of this work was to evaluate the potential of ultrasound (US), alone or in combination with mild heating and/or EDTA towards reduction of As, Cd, I, and Hg content of Laminaria hyperborea. Concentrations of As, Cd, I, and Hg of 56.29, 0.596, 7340, and <0.01 mg kg−1 of dry weight, respectively, were found in L. hyperborea blades. Treatment with US at 50 °C increased approx. 2-fold the amount of As released, although did not affect significantly the content of Cd or I, as compared to control (no US) samples. Reducing the temperature to 8 °C significantly decreased the effect of US, but heating at 80 °C did not cause a significant effect as compared to treatments at 50 °C. On the other hand, treatment with 0.1 N EDTA at 50 °C enhanced the percentage of Cd released by approximately 7-fold, regardless of sonication. In the present work, the combination of US and EDTA at 50 °C for 5 min led to a significant reduction of the As (32%), Cd (52%) and I (31%) content in L. hyperborea, thus improving the product’s safety for consumers.

Twenty-eight commercial soybean [Glycine max(L.) Merr.] cultivars of maturity classes V through VIII were evaluated for differences in response to metribuzin [4-amino-6-tert-butyl-3-(methylthio)-as-triazin-5(4H)-one] 0.125 ppm w/w in hydroponic culture. Top dry weight (TDW) of treated ‘FFR 666’ soybeans equaled that of the cultivar check and five other cultivars were not significantly different (P = 5%). ‘Semmes' was most sensitive to the herbicide with TDW 40% of cultivar check. ‘Tracy’ and ‘Coker 156’ were not significantly different (P = 5%) from Semmes. The other cultivars tested were intermediate in response to metribuzin.

SummaryEight amounts of nitrogen ranging from 0 to 210 kg N/ha were applied to two tall and one semi-dwarf variety of winter wheat in the spring of 1975 and 1976. The tall varieties were Cappelle-Desprez and Maris Huntsman; the semi-dwarf variety was Maris Fundin in 1975 and Hobbit in 1976. Interactions between varieties and nitrogen were few and small compared with the main effects. All varieties produced their maximum grain yields with 180 kg N/ha. The yield of the semi-dwarf varieties, but not the others, decreased slightly with more nitrogen.Cappelle-Desprez yielded less grain than the other varieties in both years. In 1975 the yields of Maris Fundin and Maris Huntsman were similar and in 1976 Hobbit yielded more than Maris Huntsman. The varieties had similar numbers of ears at maturity and similar patterns of tillering. The semi-dwarf varieties had most grains per spikelet, and hence grains per ear, and Cappelle-Desprez had least. The semi-dwarf varieties had the smallest grains. The semi-dwarf varieties had less straw than the other varieties and hence the largest ratios of grain to total above-ground dry weight. The decrease in dry weight of stem and leaves between anthesis and maturity was similar for all varieties. In 1975 the efficiency of the top two leaves plus top internode in producing grain was the same for all varieties, but in 1976 Hobbit was more efficient than the other two. There were some small differences between varieties in nutrient uptake that were not related to differences in growth. Maris Fundin tended to have a greater phosphorus and potassium content than the tall varieties. Hobbit contained slightly less nitrogen than the tall varieties at maturity, and had a smaller concentration of nitrogen in the grain.Applying 210 kg N/ha doubled grain yield in 1975. Applying nitrogen resulted in a largeincrease in number of ears and a small increase in number of grains per ear due to the development of more fertile spikelets per ear. Nitrogen decreased dry weight per grain, especially of the semi-dwarf varieties. With extra nitrogen, straw dry weight at maturity, shoot dry weight atanthesis and leaf area were all increased relatively more than grain yield, and stems lost moredry weight between anthesis and maturity than without nitrogen. The year 1976 was exceptionallydry and nitrogen had only small effects in that it affected neither straw dry weight nor numberof ears but slightly increased grain yield by increasing the number of spikelets and number of grains per spikelet. It also increased leaf area proportionately to grain yield. In 1975 nitrogen increased evaporation of water from the crop before anthesis but decreased it after anthesis, even though it continued to increase the extraction of water from below 90 cm.

The comparative effects of ketoconazole, itraconazole, and fluconazole on the lipids of five Sporothrix schenckii strains were investigated. Quantitative analysis of lipids and sterols was completed, as well as qualitative analysis of sterols, by thin-layer chromatography and ultraviolet spectrophotometry. Growth of the S. schenckii isolates in the presence of azole derivative concentrations below the minimum inhibitory concentration (MIC) resulted in significant alterations in the lipid and sterol contents as compared with the control values. Furthermore, lanosterol was detected in these azole-treated cells. These results were in complete agreement with the proposed mechanism of action of azoles, which act by inhibiting ergosterol biosynthesis with a consequent accumulation of lanosterol. Concerning the MIC values, fluconazole was found to be the least effective drug. On the other hand, as determined from a comparison of the effects of the three azoles on the sterol content of the strains studied, no significant differences in efficacy were found among the tested drugs.Key words: Sporothrix schenckii, azole derivatives, lipids, sterols.

ABSTRACTResistance Test of Potato Clones Derived from Crossing of Atlantic x Repita to Late Blight (Phytophthora infestans)Late blight, incited by Phytophthora infestans is the most destructive disease of potato. The management that is effective and environmentally-friendly is the use of resistant variety. The objective of this study was to test the resistance of the six potato clones (AR 04, AR 05, AR 06, AR 07, AR 08 and AR 09) derived from crossing var. Atlantic x var. Repita to late blight caused by P. infestans. Var. Atlantic, Repita and Granola were used as susceptible, resistant and susceptible but the most-grown variety, respectively. Field test was located in Ciwidey, one of the potato growing center where late blight is endemic since potatoes are continuously grown. The treatments were arranged in a randomized block design with 3 replicates. The result showed that clones AR 07 and AR 08 were more resistant than the other potato clones, but it still below the resistance level var. Repita. However, based on statistical test on the diseases development (AUDPC), clone AR 08 could be categorized as resistant, equal with of the resistance level of var. Penelitian ini bertujuan untuk menguji ketahanan 6 klon kentang (AR 4, AR5, AR6, AR7, AR 8, dan AR9) yang merupakan hasil persilangan antara var. Atlantic (produksi tinggi tetapi rentan) dengan var. Repita, sebagai tetua tahan terhadap penyakit hawar daun yang disebabkan oleh P. infestans. Varietas Atlantic, Repita dan Granola digunakan sebagai pembanding. Pengujian ketahanan dilakukan di Ciwidey, yang merupakan salah satu sentra produksi kentang di Jawa Barat dan endemik penyakit hawar daun. Perlakuan ditata menggunakan Rancangan Acak Kelompok dengan 3 ulangan. Hasil penelitian menunjukkan bahwa klon AR 07 dan Klon AR 08 memiliki ketahanan yang lebih baik dibanding klon-klon kentang lainnya, namun tingkat ketahanannya masih di bawah cv Repita. Namun, berdasarkan uji statistik terhadap nilai perkembangan penyakit (AUDPC) klon AR 08 dapat dikategorikan tahan, sama dengan derajat tahan var. Moringa is used for human and animal consumption due to its high content of proteins, vitamins and minerals. The present study analyzed and evaluated the effectiveness of Azospirillum spp. over the nutritional content of moringa (Moringa oleifera Lam.) under nursery conditions. The experimental design consisted of four treatments with six repetitions in complete blocks at random, which consisted of the inoculation at 0.5, 1 and 1.5 mL of suspension with 9.8 × 105 CFU mL-1 of Azospirillum spp. The study variables: biomass, protein, macro (P, K. Ca and Mg) and micronutriments (Na, Fe, Cu and Mn) in leaves and stems were determined 70 days after sowing. The analysis of variance showed signif icant statistical differences between treatments only for the variables: potassium content in leaves and calcium content, iron and sodium in stem. According to the comparison of means (P ≤ 0.05) the best treatments were those that received 0.5 and 1 mL of suspension. The inoculation of Azospirillum, failed to increase the dry weight and protein contents; however, the highest values (30.2% crude protein) were associated with its application. The absence of significant effects in the other variables evaluated is associated with inoculum concentration, however, higher values are shown in the nutrient contents in relation to results reported in other research.

Two gibberellins, one GA1-like, the other GA3-like, were identified in the extracts of roots and tops of 8-,11- and 15-day-old barley seedlings by paper chromatography, paper electrophoresis, thin-layer chromatography, gas-liquid chromatography and bioassay procedures, followed by combined gas chromatography-mass spectrometry. The amounts of gibberellins in the seedlings ranged from 7 to 11 ng per plant. The concentrations of gibberellins in the seedlings were 32-320 ng/g dry weight and 5-28 ng/g fresh weight; concentrations in the roots were higher than those in the shoots.
