Circulating carotenoid levels are negatively associated with previous reproductive success in Florida Scrub-Jays (Aphelocoma coerulescens)

The relationship between individual fitness and antioxidants and oxidative stress has come under increasing scrutiny of late. In particular, associations between oxidative balance indicators and reproductive success in the wild have been inconsistent in the limited prior work on this topic. Studies spanning multiple seasons and antioxidant types are particularly lacking. Here, we examined associations between reproductive success over two breeding seasons and several metrics of circulating antioxidants (antioxidant capacity, uric acid, carotenoids, and vitamin E, measured in the intervening nonbreeding season) in Florida Scrub-Jays (Aphelocoma coerulescens (Bosc, 1795)). We found that carotenoid levels in the nonbreeding season were negatively associated with reproductive success in the preceding breeding season but unassociated with that in the subsequent breeding season. This correlation may be driven by the cost of reproduction (i.e., carotenoid depletion while breeding) or some other unmeasured and intercorrelated variable such as diet. Antioxidant capacity, uric acid, and vitamin E were not associated with reproductive success. These data are consistent with an emerging theme in physiological ecology: that antioxidants and oxidative stress are but one part of a suite of integrative physiological systems that interact and trade-off in complex ways, making full understanding of their ecological roles challenging.

Background: Cigarette smoke free radicals can cause cellular damage and different diseases. All the body fluids have antioxidants which protect against free radicals. Objective: The aim of this study was to evaluate salivary total antioxidant capacity and peroxidase, uric acid and malondialdehyde levels in smokers and a nonsmoking control group. Methods: Unstimulated saliva was collected from 510 males. A total of 259 subjects were current smokers and 251 were non-smokers. The levels of salivary total antioxidant capacity, uric acid, peroxidase and malondialdehyde were measured using standard procedures. Data were analyzed with t test and ANOVA. Results: The smokers were younger and dental hygiene index was higher than healthy nonsmoking controls. The mean total antioxidant capacity in smokers and nonsmokers was 0.13±0.07 and 0.21±011, respectively (P=0.001). Smokers had significantly lower peroxidase and uric acid levels than healthy controls. In addition, the mean malondialdehyde levels in the smokers and nonsmokers were 4.55 ±2.61 and 2.79 ±2.21, respectively (P=0.001). Conclusion: Cigarette smoke produces free radical and oxidative stress, causing many side effects. Salivary antioxidant levels decreased and malondialdehyde levels increased in smokers, indicating the high oxidative stress among smokers compared to nonsmokers. Cigarette smoke had deleterious effects on main salivary antioxidants levels.

A 2 × 2 factorial experiment (10 boars per treatment) was conducted for 16 weeks to evaluate the effects of the dietary n-6 : n-3 ratio (14 : 1 vs 6 : 1) and vitamin E (200 vs 400 mg kg–1) on boar sperm morphology and oxidative stress. Sperm mitochondrial membrane potential (MMP), reactive oxygen species (ROS), DNA damage (8-hydroxydeoxyguanosine; 8-OHdG), seminal lipoperoxidation (malondialdehyde; MDA) and antioxidant capacity in the serum, spermatozoa and seminal plasma were assessed as indicators of oxidative stress. Sperm production was similar among groups but increased (P < 0.05) throughout the 16 weeks of the study. Although sperm α-tocopherol content, ROS and seminal MDA did not differ between the two dietary n-6 : n-3 ratio treatments, enhanced antioxidant enzyme activity and MMP, but decreased 8-OHdG, were found in spermatozoa from boars consuming the 6 : 1 diet. The diet with the 6 : 1 ratio positively affected sperm morphology at Weeks 12 and 16 (P < 0.05). The α-tocopherol content and antioxidant capacity increased in boars with increasing levels of vitamin E supplementation. Compared with low-dose vitamin E, high-dose vitamin E supplementation improved sperm morphology. Overall, the results indicate that an n-6 : n-3 ratio of 6 : 1 and 400 mg/kg vitamin E have beneficial effects on sperm morphology by improving antioxidative stress.

Background: Over the past few years, the benefits of co-administration of omega-3 and antioxidants have been reported in the management of non-alcoholic fatty liver disease (NAFLD) complications.This study evaluated the effects of Camelina sativa oil (CSO) supplementation as one of the richest dietary sources of omega-3 fatty acids on glucose homeostasis,inflammation, metabolic endotoxemia, and oxidative stress in NAFLD patients. Methods: In all, 43 subjects with NAFLD were allocated to either an intervention (20g/d CSO) or placebo (20g/d sunflower oil) group receiving a calorie-restricted diet for 12 weeks. Fasting serum levels of glycemic indices, hs-CRP, endotoxin, antioxidant enzymes activity, total antioxidant capacity (TAC), malondialdehyde (MDA), 8-iso-prostaglandin F2α (8-iso-PGF2α), and uric acid were measured at baseline and post-intervention. Results: CSO intake led to a significant decrease in insulin concentration (-17.49%), homeostasis model assessment of insulin resistance (HOMA-IR) (-20%), high-sensitive C-reactive protein (hs-CRP), (-12.94%), lipopolysaccharide endotoxin (-32.55%), malondialdehyde (MDA) (-18.75%), 8-iso-prostaglandin F2α (8-iso-PGF2α) (-19.55%) and a significant increase in the levels of total antioxidant capacity (TAC) (31.82%) and superoxide dismutase activity (10.22%) in the CSO group compared with the placebo group. Also, there was no significant difference between the two groups in fasting plasma glucose, quantitative insulin sensitivity check index (QUICKI), catalase, glutathione peroxidase activity, and uric acid level. In addition, within-group analyses showed a significant reduction of insulin, QUICKI, hs-CRP, and endotoxin levels in the intervention group. Conclusion: These findings indicate that CSO may improve glycemic, inflammation, metabolic endotoxima, oxidative stress status, and mental health in patients with NAFLD.

CAD is recognized as a multifactorial disease that is influenced by environmental and genetic factors. This study aimed to evaluate the levels of lipid parameters, oxidative stress and antioxidant markers in subjects with CAD compared to their age & sex matched controls and to analyze the relationship between atherogenic Index and oxidative stress among them 62 clinically proved CAD patients and 62 healthy age and sex matched subjects without CAD were selected for this study. 5 ml of fasting venous blood was collected from all the subjects and investigations such as FPG, lipid profile, oxidative markers Malondialdehyde (MDA), F2 isoprostanes (F2iso) and antioxidants glutathione S-transferase (GST), superoxide dismutase (SOD), vitamin-C, vitamin-E were performed. This study showed that levels of lipid parameters total cholesterol (TC), triglycerides (TG), low density lipoprotein cholesterol (LDL-c) and AI were significantly higher whereas high density lipoprotein cholesterol (HDL-c) were significantly low in CAD patients compared to normal controls. Oxidative stress markers MDA and F2 Isoprostanes level were significantly high, whereas enzymatic antioxidants GST and SOD and non-enzymatic antioxidants Vitamin-C and Vitamin-E levels were significantly low in CAD patients. Oxidative stress markers were found to significantly influence the AI. Results of this study showed that oxidative stress markers F2iso and MDA and antioxidants GST, VIT-C and VIT-E are found to influence the atherogenic index significantly.

Hyperuricemia, which contributes to vascular endothelial damage, plays a key role in multiple cardiovascular diseases. This study was designed to investigate whether C1q/tumor necrosis factor (TNF)-related protein 3 (CTRP3) has a protective effect on endothelial damage induced by uric acid and its underlying mechanisms. Animal models of hyperuricemia were established in Sprague-Dawley (SD) rats through the consumption of 10% fructose water for 12 weeks. Then, the rats were given a single injection of Ad-CTRP3 or Ad-GFP. The animal experiments were ended two weeks later. In vitro, human umbilical vein endothelial cells (HUVECs) were first infected with Ad-CTRP3 or Ad-GFP. Then, the cells were stimulated with 10 mg/dL uric acid for 48 h after pretreatment with or without a Toll-like receptor 4 (TLR4)-specific inhibitor. Hyperuricemic rats showed disorganized intimal structures, increased endothelial apoptosis rates, increased inflammatory responses and oxidative stress, which were accompanied by reduced CTRP3 and elevated TLR4 protein levels in the thoracic aorta. In contrast, CTRP3 overexpression decreased TLR4 protein levels and ameliorated inflammatory responses and oxidative stress, thereby improving the morphology and apoptosis of the aortic endothelium in rats with hyperuricemia. Similarly, CTRP3 overexpression decreased TLR4-mediated inflammation, reduced oxidative stress, and rescued endothelial damage induced by uric acid in HUVECs. In conclusion, CTRP3 ameliorates uric acid-induced inflammation and oxidative stress, which in turn protects against endothelial injury, possibly by inhibiting TLR4-mediated inflammation and downregulating oxidative stress.
