The cultural, morphological, and physiological properties of Thermomonospora fusca (strain 190Th) are described. Its physiological properties show that this species is primarily a carbohydrate-degrading actinomycete which can use a wide range of plant sugars and polymeric carbohydrates as sources of carbon and energy. The culture does not use proteins or amino acids for carbon and energy, or as a nitrogen source. A few organic acids are utilized. Ammonia is the preferred nitrogen source. The culture has trace nutrient requirements which include biotin and an undetermined number of amino acids. These and other physiological characteristics are discussed in relation to the roles that T. fusca carries out as a saprophytic bacterium in nature. Its cultural and morphological properties are discussed in relation to the taxonomic status of this species in the literature.

Dietary proteins exert a wide range of nutritional and biological functions. Apart from their nutritional roles as the source of amino acids for protein synthesis, they take part mainly in the regulation of food intake, blood pressure, bone metabolism, glucose and lipid metabolism, and immune functions. The interaction of dietary proteins with the gastrointestinal (GI) tract plays a chief role in determining the physiological properties of proteins. The enzymes protease and peptidase hydrolyze dietary protein to generate dipeptides, tripeptides, and amino acids in the lumen of the gastrointestinal tract. These products digested from dietary proteins are utilized in the small intestine by microbes. Moreover, the microbes also convert the macro and micronutrients from the diet into an enormous number of compounds that may have either beneficial or adverse effects on human health. The present review discusses the various impacts caused by both dietary plant and animal protein sources on microbiota in the GI tract.Keywords: Animal protein; Plant protein; Dietary proteins; Gut microbiota; Human health.

Corynebacterium glutamicumis a nonpathogenic, aerobic, gram-positive soil bacterium used for the large-scale biotechnological production of several biomolecules. C. glutamicum is an industrial microbe traditionally used for the production of amino acids. It is a well known microbe as the workhorse in fermentation industry used since its first discovery in Japan in 1950s for production of monosodium glutamate (MSG)and L-lysine with a market size of 3.1 and 2.2 million. However, for the fermentative production of diverse products through genetic–metabolic engineering. Due to its industrial importance, several clones of C. glutamicum have been sequenced by both industry and academic groups. Furthermore it has also been exploited for the synthesis of a variety of other fuels and chemicals. Especially, the facultative anaerobic lifestyle of this Gram-positive bacterium formed the basis to engineer C. glutamicum for the production of reduced molecules such as organic acids (e.g., lactate, succinate) and alcohols (e.g., ethanol, isobutanol) under zero-growth anaerobic conditions. The present work is a review of various uses of C. glutamicum for which the researchers explored several resources including the PubMed database, Journals and online archives upto March 2019. The review in its analysis signifies the vital utility of C. glutamicum in various prospective. Most articles were on occurrence, metabolism, metabolic role, and industrial uses of bacterial polyhydroxyalkanoates. Through genetic alterations, this microbe showed its efficacy for the simultaneous utilization of hexose and pentose sugars of biomass hydrolysate for making value-added products, such as amino acids and polyamines, and thus making it a strain of choice to work in a bio-refinery concept. This soil microbe also showed its efficacy in bioremediation purposes, such as arsenic removal. Recently its extensive research has focused on engineering beyond the scope of amino acids for utilization of alternative carbon sources, (e.g. coming from wastes and unprocessed sources), and construction of C. glutamicum strains for production of new products such as diamines, organic acids and alcohols. Meanwhile, a variety of corynebacterial strains allows access to alternative carbon sources that allows production of a wide range of industrially relevant compounds. Some of these efforts set new standards in terms of titers and productivities achieved whereas others represent a proof-of-principle. These achievements manifest the position of C. glutamicum as an important industrial microorganism with capabilities far beyond the traditional amino acid production. According to the studies cited in different literatures C. glutamicum reported as the most effective and playing important roles as dominant bacteria in biotechnological applications as well as industrial processes and products. And, with recent advances in biotechnology, the economic contributions that these organisms can make in biotechnological applications and industrial processes can be exploited further for large scale benefit of mankind.

Benzofurans are everywhere in nature and they have been extensively studied by medicinal chemists over the years because of their chemotherapeutic and physiological properties. Herein, we describe a strategy that can be used to access elaborate benzo-2-carboxamide derivatives, which involves a synthetic sequence of 8-aminoquinoline directed C–H arylations followed by transamidations. For the directed C–H arylations, Pd catalysis was used to install a wide range of aryl and heteroaryl substituents at the C3 position of the benzofuran scaffold in high efficiency. Directing group cleavage and further diversification of the C3-arylated benzofuran products were then achieved in a single synthetic operation through the utilization of a two-step transamidation protocol. By bocylating the 8-aminoquinoline amide moiety of these products, it proved possible to activate them towards aminolysis with different amine nucleophiles. Interestingly, this aminolysis reaction was found to proceed efficiently without the need of any additional catalyst or additive. Given the high efficiency and modularity of this synthetic strategy, it constitute a very attractive approach for generating structurally-diverse collections of benzofuran derivatives for small molecule screening.

This study aimed to elucidate the variations in primary and secondary metabolites during Lycorisradiata flower development using high performance liquid chromatography (HPLC) and gas chromatography time-of-flight mass spectrometry (GC-TOFMS). The result showed that seven carotenoids, seven phenolic acids, three anthocyanins, and galantamine were identified in the L. radiata flowers. Most secondary metabolite levels gradually decreased according to the flower developmental stages. A total of 51 metabolites, including amines, sugars, sugar intermediates, sugar alcohols, amino acids, organic acids, phenolic acids, and tricarboxylic acid (TCA) cycle intermediates, were identified and quantified using GC-TOFMS. Among the hydrophilic compounds, most amino acids increased during flower development; in contrast, TCA cycle intermediates and sugars decreased. In particular, glutamine, asparagine, glutamic acid, and aspartic acid, which represent the main inter- and intracellular nitrogen carriers, were positively correlated with the other amino acids and were negatively correlated with the TCA cycle intermediates. Furthermore, quantitation data of the 51 hydrophilic compounds were subjected to partial least-squares discriminant analyses (PLS-DA) to assess significant differences in the metabolites of L. radiata flowers from stages 1 to 4. Therefore, this study will serve as the foundation for a biochemical approach to understand both primary and secondary metabolism in L. radiata flower development.

Autophagy is an essential protective mechanism that allows mammalian cells to cope with a variety of stressors and contributes to maintaining cellular and tissue homeostasis. Due to these crucial roles and also to the fact that autophagy malfunction has been described in a wide range of pathologies, an increasing number of in vivo studies involving animal models targeting autophagy genes have been developed. In mammals, total autophagy inactivation is lethal, and constitutive knockout models lacking effectors of this route are not viable, which has hindered so far the analysis of the consequences of a systemic autophagy decline. Here, we take advantage of atg4b−/− mice, an autophagy-deficient model with only partial disruption of the process, to assess the effects of systemic reduction of autophagy on the metabolome. We describe for the first time the metabolic footprint of systemic autophagy decline, showing that impaired autophagy results in highly tissue-dependent alterations that are more accentuated in the skeletal muscle and plasma. These changes, which include changes in the levels of amino-acids, lipids, or nucleosides, sometimes resemble those that are frequently described in conditions like aging, obesity, or cardiac damage. We also discuss different hypotheses on how impaired autophagy may affect the metabolism of several tissues in mammals.

Destruxin A (DA), a hexa-cyclodepsipeptidic mycotoxin produced by the entomopathogenic fungus Metarhizium anisopliae, exhibits insecticidal activities in a wide range of pests and is known as an innate immunity inhibitor. However, its mechanism of action requires further investigation. In this research, the interactions of DA with the six aminoacyl tRNA synthetases (ARSs) of Bombyx mori, BmAlaRS, BmCysRS, BmMetRS, BmValRS, BmIleRS, and BmGluProRS, were analyzed. The six ARSs were expressed and purified. The BLI (biolayer interferometry) results indicated that DA binds these ARSs with the affinity indices (KD) of 10−4 to 10−5 M. The molecular docking suggested a similar interaction mode of DA with ARSs, whereby DA settled into a pocket through hydrogen bonds with Asn, Arg, His, Lys, and Tyr of ARSs. Furthermore, DA treatments decreased the contents of soluble protein and free amino acids in Bm12 cells, which suggested that DA impedes protein synthesis. Lastly, the ARSs in Bm12 cells were all downregulated by DA stress. This study sheds light on exploring and answering the molecular target of DA against target insects.

Numerous varieties of doenjang are manufactured by many food companies using different ingredients and fermentation processes, and thus, the qualities such as taste and flavor are very different. Therefore, in this study, we compared many products, specifically, 19 traditional doenjang (TD) and 17 industrial doenjang (ID). Subsequently, we performed non-targeted metabolite profiling, and multivariate statistical analysis to discover distinct metabolites in two types of doenjang. Amino acids, organic acids, isoflavone aglycones, non-DDMP (2,3-dihydro-2,5-dihydroxy-6-methyl-4H-pyran-4- one) soyasaponins, hydroxyisoflavones, and biogenic amines were relatively abundant in TD. On the contrary, contents of dipeptides, lysophospholipids, isoflavone glucosides and DDMP-conjugated soyasaponin, precursors of the above-mentioned metabolites, were comparatively higher in ID. We also observed relatively higher antioxidant, protease, and β-glucosidase activities in TD. Our results may provide valuable information on doenjang to consumers and manufacturers, which can be used while selecting and developing new products.

Three methods of sampling xylem sap of maize roots were compared: sap bleeding from the stem cut just above the ground; sap bleeding from the cut tops of roots still undisturbed in the ground; and sap aspirated from excavated roots under reduced pressure. The bleeding saps were often unobtainable. When their composition was measured with time from cutting, the concentrations of the major solutes approximately doubled in 2 h. Aspirated sap was chosen as the most reliable sample of root xylem contents. Solute concentrations of the saps showed great variability between individual roots for all solutes, but on average the concentrations found (in �mol g-1 sap) were: total amino acids, 1.8; nitrate, 1.8; sugars (mainly sucrose), 5.4; total organic acids, 18.3. Individual amino acids also varied greatly between roots. Glutamine, aspartic acid and serine were generally most abundant. The principal organic acid found was malic, approximately 8 �mol g-1. From these analyses the ratios of carbon in the fractions (sugars : amino acids : organic acids) = (44 : 6 : 50). 14Carbon pulse fed to a leaf appeared in the root sap within 30 min, rose to a peak at 4-6 h, and declined slowly over a week. During all this time the neutral, cation and anion fractions were sensibly constant in the proportions 86 : 10 : 4. The 14C therefore did not move towards the equilibrium of 12C-compounds in the sap. It is argued that the results do not support a hypothesis of formation of amino carbon from recent assimilate and reduced nitrate in the roots and an export of this to the shoot in the transpiration stream.
