Neuromuscular properties of the quintuply innervated flexor muscle in lobster limbs

The flexor muscle of the lobster's walking leg was shown by enzyme histochemistry and electrophysiology to display a regional segregation of fibre types: medial fibres have a higher ATPase activity, lower oxidative capacity, and shorter membrane time constant than peripheral fibres lying near the cuticle. The muscle was confirmed to receive one inhibitory and four excitatory motor axons. As judged by the properties of their output excitatory junctional potentials (ejp's), the four excitors lie along the fast-to-slow gradient defined by the two specialized excitors of dually excited muscles. The Fα axon produces initially large ejp's which facilitate weakly or antifacilitate; they are similar to those of fast axons in other muscles. The Fρ axon at the other end of the spectrum produces strongly facilitating ejp's which are initially small, resembling those of known slow axons. The Fβ and Fγ axons show intermediate properties. The inhibitor, which is the common inhibitor of all leg muscles, innervates preferentially the more tonic muscle fibres, as does Fρ. Muscle fibres were observed to receive anywhere from one to five efferents, most receiving two to four. Serial electron microscopic observations in several regions revealed a rich supply of synaptic terminals, usually comprising a single inhibitory terminal and two or three excitatory ones. The inhibitory terminal typically has a few large synapses, each with more than one active site. Excitatory terminals, on the other hand, have many more smaller synapses, each with at least one active site. Although excitatory and inhibitory terminals were often closely juxtaposed, no synaptic interactions were observed between them.

It was shown in an earlier paper (7) that if maximal stimulation of either of two different afferent nerves can reflexly excite fractions of a given flexor muscle, there are generally, within the aggregate of neurones which innervate that muscle, motoneurones which can be caused to discharge by either afferent (i. e., motoneurones common to both fractions). The relationship which two such afferents bear to a common motoneurone was shown, by the isometric method of recording contraction, to be such that the activation of one afferent, at a speed sufficient to cause a maximal motor tetanus when trans­mitted to the muscle fibres, caused exclusion of any added mechanical effect when the other afferent was excited concurrently. This default in mechanical effect was called “occlusion.” Occlusion may conceivably be due to total exclusion of the effect of one afferent pathway on the common motoneurone by the activity of the other; but facilitation of the effect of one path by the activation of the other when the stimuli were minimal suggests that, in some circumstances at least, the effect of each could augment and summate with th at of the other at the place of convergence of two afferent pathways. Further investigation, using the action currents of the muscle as indication of the nerve impulses discharged by the motoneurone units, has now given some information regarding the effect of impulses arriving at the locus of convergence by one afferent path when the unit common to both is already discharging in response to impulses arriving by the other afferent path. Our method has been to excite both afferent nerves in overlapping sequence by series of break shocks at a rapid rate and to examine the action currents of the resulting reflex for evidence of the appearance of the rhythm of the second series in the discharge caused by the first when the two series are both reaching the motoneurone.

s-ACE (the somatic form of angiotensin-converting enzyme) consists of two homologous domains (N- and C-domains), each bearing a catalytic site. Negative co-operativity between the two domains has been demonstrated for cow and pig ACEs. However, for the human enzyme there are conflicting reports in the literature: some suggest possible negative co-operativity between the domains, whereas others indicate independent functions of the domains within s-ACE. We demonstrate here that a 1:1 stoichiometry for the binding of the common ACE inhibitors, captopril and lisinopril, to human s-ACE is enough to abolish enzymatic activity towards FA {N-[3-(2-furyl)acryloyl]}-Phe-GlyGly, Cbz (benzyloxycarbonyl)-Phe-His-Leu or Hip (N-benzoylglycyl)-His-Leu. The kinetic parameters for the hydrolysis of seven tripeptide substrates by human s-ACE appeared to represent average values for parameters obtained for the individual N- and C-domains. Kinetic analysis of the simultaneous hydrolysis of two substrates, Hip-His-Leu (S1) and Cbz-Phe-His-Leu (S2), with a common product (His-Leu) by s-ACE at different values for the ratio of the initial concentrations of these substrates (i.e. σ=[S2]0/[S1]0) demonstrated competition of these substrates for binding to the s-ACE molecule, i.e. binding of a substrate at one active site makes the other site unavailable for either the same or a different substrate. Thus the two domains within human s-ACE exhibit strong negative co-operativity upon binding of common inhibitors and in the hydrolysis reactions of tripeptide substrates.

An electron-microscopic study was made of the mitochondria in normal and denervated rat diaphragm muscles. Two types of structurally different muscle fibres were found: one type had a well-defined M-line, while the other did not. The mitochondria in normally innervated muscle are regularly arranged at both sides of the Z-line. The mitochondria are very long and branched, and surround the myofibrils at the level of the Z-line. In transverse sections of the muscle fibres the worm-like mitochondria give a very characteristic picture. After denervation the mitochondria are smaller and less regularly arranged. In transverse sections of the muscle fibres the mitochondria have small circular profiles. This contrasts sharply with the normal appearance and makes it possible to distinguish normal from denervated fibres. The mitochondrial changes can be detected less than 24 h after denervation.

Electron microscopic observations on ultrathin sections of the red thoracic flightmuscles and white leg muscles of Hydrophilus and Dytiscus are reported. In red muscle-fibres with high values in frequency of contraction, oxygen consumption, and dehydrogenase activity, the single fibrils are completely surrounded by huge mitochondria. Tracheoles penetrate the sarcolemma and supply the mitochondria with oxygen by intracellular branches. In the less active white muscle fibres, mitochondria are found irregularly scattered between the fibrils or along the I band. The intracellular tracheolization is sparse but an endoplasmic reticulum is widely spread between the synfibrillar contractile material. The same muscles of the two insects differ considerably in detail.

This chapter examines how the law deals with contracts that a party entered into by mistake. After a brief discussion of the historical background of the rules in the civil law and the common law, the question is raised whether there is a relevant mistake if a party’s ‘motive’ for entering into the contract turns out to be incorrect, if the party’s mistake refers to the value of what it promised or was to receive under the contract, or if the party’s mistake is due to its carelessness. Should the relevance of a mistake not depend on whether it was caused or shared by the other party? Finally, the chapter outlines some common threads in the development of a European law on mistake.

<p>Enzyme activities and fibre properties in four muscles from Svalbard reindeer, collected during the summer, have been compared with corresponding muscles during the winter. In two muscles, gluteobiceps and semimembranosus, oxidative capacity is higher in winter than in summer; in the other two muscles, semitendinosus and longissimus dorsi, there is no difference with time of the year. The capacity to oxidize fatty acids is low in winter compared with summer, especially in semitendinosus and longissimus. These changes are similar in both sexes. Histochemical studies of the three main fibre types, I (BetaR), HA (&deg;cR) and IIB (aW), from the four muscles show that in male reindeers the muscle fibres are narrower at the end of the winter season than during the summer. The decrease of muscle tissue amounts to about one third of the total volume (33%), of which I accounts for 5%, IIA for 2% and IIB for 26%. The results indicate that the Svalbard reindeer use lean tissue in general, and IIB fibres in particular, in order to survive the hostile arctic winter period at Svalbard.</p><p>Histokemiska och enzymatiska skillnader i skelettmuskel fr&aring;n Svalbardren mellan sommar och vinter.</p><p>Abstract in Swedish / Sammandrag: Enzymaktiviteter och fiberegenskaper i fyra av Svalbardrenens muskler, insamlade under sommaren, har j&aring;mforts med motsvarande muskler insamlade under vintern. The nests of a particular species of swallow which is principally met with in the island of Java, have from time immemorial formed an article of trade between that island and China, where they are purchased at a high price by that voluptuous people, it being believed, that the materials of which the nests are composed, are possessed of an aphrodisiac virtue in an eminent degree. They have been occasionally brought into this country, and are preserved in collections of natural history, as curiosities. In what manner the bird procures the materials out of which the nest is made, has till now remained unknown; a thousand conjectures have, however, been make upon this subject. It has been supposed by some, that it is a gluten collected from the mollusca picked upon the surface of the sea. By others, a substance extracted from certain fuci found on the sea shore. By others again, a portion of the food in a half digested state regurgitated to be employed for this particular purpose. Sir Stamford Raffles, who has just returned from Java, where he resided five years, as lieutenant governor, has brought over a number of these nests, and has been kind enough to offer me some of them, for the purpose of investigating the nature of the materials of which they are composed, and gives it decidedly as his own opinion, that, whatever it is, it is brought up from the stomach, and requires at times so great an effort, as to bring up blood, the stain of which is seen on the nest. This account of Sir Stamford Raffles, in the correctness of whose observation I have the greatest confidence, led me to investigate this subject, and to ascertain by examination whether this particular swallow has any glands that are peculiar to its œsophagus, or stomach, enabling it to secrete a mucus similar in its nature to the substance of which the nest is composed. I at the same time requested my friend, Professor Brande, to analyze one of the nests, and to inform me of its composition. In examining the gastric glands of the Java swallow, even with the assistance of a common magnifying glass, I saw an obvious difference between the appearance of the orifices by which the secretion is poured into the gizzard, and of those of other birds, but, as I had never examined those glands in the common swallow which migrates to this country, it became necessary, before I proceeded farther in the enquiry, to ascertain whether in all the swallow tribe there were similar structures. In the present season this opportunity has been afforded me, and I find that in the common swallow, both male and female, the orifices of the gastric glands differ in nothing from those of birds in general, but that the peculiar structure which I am about to describe is confined to the Java swallow. This bird, Sir Stamford Raffles informs me, does not migrate, but remains all the year an inhabitant of the caverns in that island. Some of the most extensive caves in which they reside, are forty miles from either sea. Those swallows that build their nests near the sea, are observed to fly inland towards extensive swamps where gnats and other insects are in great abundance. Those that build in inland caves, are observed to quit the caves in the morning, and generally return in swarms darkening the air, towards the close of the day; they are, however, going in and out the whole of the day. This bird is double the size of our common swallow. There are two separate nests, one for the male to lie and rest in, which is oblong and narrow, adapted to his form, the other wide and deeper, to receive the female and the eggs.

A number of biochemically distinct systems have been characterized for the microbial reduction of the oxyanions, selenate (SeO42−) and nitrate (NO3−). Two classes of molybdenum-dependent nitrate reductase catalyse the respiratory-linked reduction of nitrate (NO3−) to nitrite (NO2−). The main respiratory nitrate reductase (NAR) is membrane-anchored, with its active site facing the cytoplasmic compartment. The other enzyme (NAP) is water-soluble and located in the periplasm. In recent years, our understanding of each of these enzyme systems has increased significantly. The crystal structures of both NAR and NAP have now been solved and they provide new insight into the structure, function and evolution of these respiratory complexes. In contrast, our understanding of microbial selenate (SeO42−) reduction and respiration is at an early stage; however, similarities to the nitrate reductase systems are emerging. This review will consider some of the common themes and variations between the different classes of nitrate and selenate reductases.

In the article the global crisis of 2008-2009 is considered as superposition of a few regional crises that occurred simultaneously but for different reasons. However, they have something in common: developed countries tend to maintain a strong level of social security without increasing the real production output. On the one hand, this policy has resulted in trade deficit and partial destruction of market mechanisms. On the other hand, it has clashed with the desire of several oil and gas exporting countries to receive an exclusive price for their energy resources.

Neste artigo analisamos a relação do Ensino Religioso com a sua evolução ao longo do contexto recente do Brasil para compreender a posição do Supremo Tribunal Federal ao considerar a possibilidade do Ensino Religioso confessional. Inicialmente apresentaremos a perspectiva legislativa criada com a constituição de 1988 e seus desdobramentos nas indicações curriculares. Neste contexto é frisado a intenção de incluir o Ensino Religioso na Base Nacional Curricular Comum, o que acabou não acontecendo. A tendência manifesta nas duas primeiras versões da BNCC era de um Ensino Religioso não-confessional. Uma tendência que demarcava a função do Ensino Religioso em debater a religião, mas que não permitia o direcionamento por uma vertente religioso qualquer. Esta posição se mostrava uma evolução da primeira perspectiva histórica mais associada à catequese confessional. Assim como também ultrapassava a interpretação posterior de um ecumenismo interconfessional, que mantinha a superioridade do cristianismo ante as demais religiões. Sendo assim, neste artigo, adotaremos o argumento de que a decisão do STF, de seis votos contra cinco, acaba retrocedendo ante o que nos parecia um caminho muito mais frutífero.Palavras-chave: Ensino Religioso. Não-confessional.Abstract: On this article, we analyze the relation between Religious education and its evolution along the currently Brazilian context in order to understand the position of the Supreme Court in considering the possibility of a confessional Religious education. Firstly, we are going to present the legislative perspective created with the 1988 Federal Constitution and its impacts in the curricular lines. On this context it was highlighted the intention to include the Religious Education on the Common Core National Curriculum (CCNC), which did not really happened. The tendency manifested in the first two versions of the CCNC was of a non-confessional Religious Education. A tendency that delineated the function of the Religious Education as debating religion, but not giving direction on any religious side. This position was an evolution of the first historical perspective more associated to the confessional catechesis. It also went beyond the former interpretation of an inter-confessional ecumenism, which kept the superiority of the Christianity over the other religions. As such, in this paper we adopt the argument that the decision of the Supreme Court, of six votes against five, is a reversal of what seemed to be a much more productive path on the Religious Education.Keywords: Religious Education.
