For the first time, a mollicute-like organism (MLO) was cultured from moribund mud crabs (Scylla serrata) during an outbreak of clearwater disease in Zhejiang Province, China. The MLO displayed a fried-egg colony morphology in culture, did not possess a cell wall, and was not retained by 0.45 μm and 0.2 μm filters. It was able to ferment glucose, sucrose, lactose, and maltose, but it did not utilize arginine and urea. The MLO grew in the absence of bovine serum and was not susceptible to digitonin. Sequence analysis of the 16S rRNA gene revealed that this MLO had 99% identity withAcholeplasma laidlawiiPG-8A, which indicates that the organism isolated from mud crabs is a member of the genusAcholeplasma.

The development of novel bioremediation strategies has focused on the isolation and identification of microorganisms that can thrive in polluted environments to evaluate their potential as biotechnological tools in bioremediation techniques. In this work, a bacterium isolated from hydrocarbon-contaminated soils from the Burgos basin was identified and its hydrocarbon degradation potential was evaluated. Identification based on sequencing the 16S rRNA gene identified one of the isolates (R3) as Microbacterium petrolearium. This strain was mainly antibiotic-sensitive with elevated carbohydrate assimilation differing from previously reported strains. Moderate surfactant production (I24 = 22.97 %) was observed, which was absent in a cell-free extract. M. petrolearium R3 showed increased growth that correlated with pollutant concentration. For light crude oil, at a higher contaminant percentage, the R3 strain showed increased growth; however, in the case of diesel, no growth was detected. The aforementioned data indicate that M. petrolearium strain R3, isolated from local sources, has potential use as a tool for hydrocarbon-contaminated soil bioremediation.

A Gram-stain-positive, facultatively anaerobic and non-motile strain, designated SYSUP0004T, was isolated from the tubers of Gastrodia elata Blume collected from Yunnan Province, PR China. The 16S rRNA gene sequence result showed that the strain SYSUP0004T shared low similarity (97.7 %) with the type strain of Cellulomonas marina . SYSUP0004T grew at pH 6.0–9.0 (optimum, pH 8.0), temperature 4–30 °C (optimum, 28 °C) and could tolerate NaCl up to 4 % w/v (optimum in the absence of NaCl). The cell-wall peptidoglycan type was A4β with an interpeptide bridge l-ornithine–d-glutamic acid. Cell-wall sugars were mannose, ribose, glucose, galactose and fucose. The menaquinone was MK-9(H4). The major fatty acids were anteiso-C15:0, anteiso-C15 : 1 A, C16 : 0 and anteiso-C17 : 0. The polar lipids of SYSUP0004T were diphosphatidylglycerol, unidentified phosphoglycolipid, phosphatidylinositol mannosides and unidentified glycolipid. The genomic DNA G+C content was 76.5 %. The average nucleotide identity values between SYSUP0004T and members of the genus Cellulomonas were below the cut-off level (95–96 %) recommended as the ANI criterion for interspecies identity. Thus, based on the above results strain SYSUP0004T represents a novel species of the genus Cellulomonas , for which the name Cellulomonas endophytica sp. nov. is proposed. The type strain, SYSUP0004T (=KCTC 49025T=CGMCC 1.16405T).

BacterialN-acetylmuramoyl-L-alanine amidases are cell-wall hydrolases that hydrolyze the bond betweenN-acetylmuramic acid and L-alanine in cell-wall glycopeptides. Rv3717 ofMycobacterium tuberculosishas been identified as a unique autolysin that lacks a cell-wall-binding domain (CBD) and its structure has been determined to 1.7 Å resolution by the Pt-SAD phasing method. Rv3717 possesses an α/β-fold and is a zinc-dependent hydrolase. The structure reveals a short flexible hairpin turn that partially occludes the active site and may be involved in autoregulation. This type of autoregulation of activity of PG hydrolases has been observed inBartonella henselaeamidase (AmiB) and may be a general mechanism used by some of the redundant amidases to regulate cell-wall hydrolase activity in bacteria. Rv3717 utilizes its net positive charge for substrate binding and exhibits activity towards a broad spectrum of substrate cell walls. The enzymatic activity of Rv3717 was confirmed by isolation and identification of its enzymatic products by LC/MS. These studies indicate that Rv3717, anN-acetylmuramoyl-L-alanine amidase fromM. tuberculosis, represents a new family of lytic amidases that do not have a separate CBD and are regulated conformationally.

Infectious of ectoparasites and vibrios were  found as a problem in fattening mud crab. The aims of this study were to determine the clinical sign of infected mud crabs and to identify the ectoparasites and generic vibrio in the fattening mud crabs from Pemalang Coast, Indonesia. Ectoparasites were collected by smear method. Whereas, 20 isolates were collected from hepatopancreas, gills, hemolymph and injured carapace of the mud crabs. Thiosulphate Citrate Bile Salt Sucrose (TCBS) medium was used to isolate suspected vibrios. The clinical signs of mud crabs infected by ectoparasites were shown through the damaged gills and the attachment of other organisms on the gills, whereas the clinical signs of mud crabs infected by vibriosis were shown through red spot on carapace and wound in carapace, claw and abdomen, followed by weak condition. It also indicated that seven ectoparasites were Ichthyobodo sp., Epistylis sp., Carchesium sp., Vorticela sp., Octolasmis sp., Lepeophtherius sp, and Copepodit sp., whilst five Vibrios were V. harveyi, V. Cholerae, V. parahaemolyticus, V. alginolyticus and V. fischeri.

An obligately aerobic, Gram-negative, non-spore-forming, non-motile, spherical bacterium, designated strain 04OKA010-24T, was isolated from seawater surrounding the hard coral Galaxea fascicularis L., collected at Majanohama, Akajima, Japan, and was subjected to a polyphasic taxonomic study. Phylogenetic analyses based on the 16S rRNA gene sequence indicated that the new strain represented a member of the phylum ‘Verrucomicrobia’ and shared 84–95 % sequence similarity with cultivated strains of ‘Verrucomicrobia’ subdivision 4. Amino acid analysis of the cell-wall hydrolysate indicated the absence of muramic acid and diaminopimelic acid, which suggested that the strain did not contain peptidoglycan in the cell wall. The G+C content of the DNA was 53.9 mol%. MK-7 was the major menaquinone and C14 : 0, C18 : 1 ω9c and C18 : 0 were the major fatty acids. On the basis of these data, it was concluded that strain 04OKA010-24T represents a novel species in a new genus in subdivision 4 of the phylum ‘Verrucomicrobia’, for which the name Coraliomargarita akajimensis gen. nov., sp. nov. is proposed. The type strain of Coraliomargarita akajimensis is 04OKA010-24T (=MBIC06463T=IAM 15411T=KCTC 12865T).

To identify the pathogen of Mulberry wilt disease, a tested strain WJ-1 was isolated from a naturally infected mulberry in Guangzhou, Guangdong province, P. R. China. Hypersensitive reaction, along with Koch’s postulates, morphological, biochemical and physiological characterization, light microscope observation and molecular identification were performed on WJ-1. The results indicated that WJ-1 was a Gram-negative, short-rod shaped bacterium. Based on morphology and the 16S rRNA gene sequence analysis, WJ-1 strain was identified as members of the genusEnterobacter. The phylogenetic tree revealed that WJ-1 shared the highest homology withEnterobacter cloacaestrain M5. It was another evidence on plant disease caused byEnterobacterspecies. We found thatEnterobacterspecies could cause mulberry wilt disease in Guangzhou.

Abstract The aim of this study to find out the optimal level of salinity for the growth and survival of the mud crab (Scylla serrata). The weight of crab seeds were about 47.65-51.32 grams and the carapace length ranged from 64.35-71.30 mm, then put into 12 tanks (1 m x 1 m x 0.4 m) with a density of 4 crab seed per tank. During the maintenance of this test, mangrove crab was fed with fresh chicken intestines as much as 10% of its body weight per day. Water changes are carried out more than 150% per week. At the beginning of the experiment, each tank was conditioned to the same water salinity in all experimental units, namely 33 ppt. To see the effect of differences in salinity levels on the growth and survival of mud crabs, 4 treatments were made consisting of (A) 33 ppt salt content, (B) 31 ppt salt content, (C). The salt content is 29 ppt, and (D) the salt content is 27 ppt. From the results of this experiment, it is known that a decrease in the salt content of 4 ppt is the best treatment for the growth of mud crabs, but has no effect on the survival value.

ABSTRACT The recent description of the polychromatic sponge Plakina kanaky revealed original microsymbionts, with some morphotypes recorded for the first time in Homoscleromorpha and others never before observed in other sponge groups. Illumina 16S amplicon sequencing was used to characterize this microbial community by comparing contents of seven specimens of this Plakinidae with five other sponge species: one Homoscleromopha of the Oscarellidae family and four Demospongiae. A total of 256 458 sequences of the hypervariable V5-V6 region of the 16S rRNA gene were clustered into 2,829 OTUs at 97% similarity, with Proteobacteria, Poribacteria and Chloroflexi being the most abundant phyla. The Plakina kanaky specific community appeared to be mainly composed by five OTUs representing about 10% of the total microbiome. Among these, the filamentous bacterium Candidatus Entotheonella, which was among the dominant morphotypes previously observed in the mesohyl and the larvae of P. kanaky, was detected in all studied specimens. However, other original and dominant morphotypes could not be assigned to a known prokaryotic taxon. This cave dwelling sponge species harbors a distinctive microbiome composition of potential taxonomic and metabolic novelties that may be linked to its ecological success in such extreme environments.

The general aim of this study, which was conducted for the first time in Iran, was to evaluate the seroprevalence and geographical distribution of Ehrlichia canis in a dog population in Iran, followed by molecular confirmation using PCR and sequencing. Blood samples were collected from 240 dogs in different areas of Alborz and Tehran Provinces and initially analyzed using the immunofluorescent antibody (IFA) test to detect anti-Ehrlichia canis IgG antibodies. Subsequently, nested PCR was performed based on a fragment of the 16S rRNA gene of E. canis on serologically positive samples. The results showed that 40/240 dogs (16.6%) presented anti-Ehrlichia canis IgG antibodies and that nine of the blood samples from the 40 seropositive dogs (22.5%) contained E. canis DNA, which was confirmed by sequencing. The seroprevalence of E. canis tended to be higher in purebred, one to three-year-old male dogs living in the Plain zone, in rural areas; however, this difference was not statistically significant.
