The antioxidant potential ofActinidia macrospermaC. F. Liang (Actinidiaceae) was investigated in vitro for total phenolic content, along with total antioxidant activity (TAA), 1,1-diphenyl 2-picryl hydrazyl (DPPH), and lipid peroxidation (LP). The results indicated that different polarity extracts ofA. macrospermaexhibit different biological activities, which depends mainly on the presence of phenolic compounds. The antioxidant activity was in the following decreasing order: MeOH extract > EtOAc extract > aqueous extract > CHCl3extract > Hexane extract. Moreover, the cytotoxic activity of this plant by MTT dye assay using SMMC-7721 has been determined also. The hexane, EtOAc, and CHCl3extracts showed cytotoxicity in a dose-dependent manner. Methanol and aqueous extracts, however, showed weak activities in this test. And a very significant cytotoxic activity, not significantly different from the positive control of quercetin, was observed in CHCl3extract.

AbstractHaloferax sp strain NRS1 (MT967913) was isolated from a solar saltern on the southern coast of the Red Sea, Jeddah, Saudi Arabia. The present study was designed for estimate the potential capacity of the Haloferax sp strain NRS1 to synthesize (silver nanoparticles) AgNPs. Biological activities such as thrombolysis and cytotoxicity of biosynthesized AgNPs were evaluated. The characterization of silver nanoparticles biosynthesized by Haloferax sp (Hfx-AgNPs) was analyzed using UV–vis spectroscopy, transmission electron microscopy (TEM), X-ray diffraction (XRD), and Fourier-transform infrared spectroscopy (FTIR). The dark brown color of the Hfx-AgNPs colloidal showed maximum absorbance at 458 nm. TEM image analysis revealed that the shape of the Hfx-AgNPs was spherical and a size range was 5.77- 73.14 nm. The XRD spectra showed a crystallographic plane of silver nanoparticles, with a crystalline size of 29.28 nm. The prominent FTIR peaks obtained at 3281, 1644 and 1250 cm− 1 identified the Functional groups involved in the reduction of silver ion reduction to AgNPs. Zeta potential results revealed a negative surface charge and stability of Hfx-AgNPs. Colloidal solution of Hfx-AgNPs with concentrations ranging from 3.125 to 100 μg/mL was used to determine its hemolytic activity. Less than 12.5 μg/mL of tested agent showed no hemolysis with high significant decrease compared with positive control, which confirms that Hfx-AgNPs are considered non-hemolytic (non-toxic) agents according to the ISO/TR 7405-1984(f) protocol. Thrombolysis activity of Hfx-AgNPs was observed in a concentration-dependent manner. Further, Hfx-AgNPs may be considered a promising lead compound for the pharmacological industry.

The bioactive compounds, as well as the in vitro antioxidant activity of two Savoy cabbage cultivars, Dama and Leticia, grown in west of Spain under similar conditions were identified, quantified and compared. We found that cv. Dama presented in general betters results when compared with cv. Leticia. Cv. Dama presented higher concentrations of chlorophyll a (2.26 mg/100 g fresh weight), total phenolic content (102.71 mg of chlorogenic acid equivalent/100 g fresh weight) and total intact glucosinolates (195.22 µmol of sinigrin equivalent/100 g fresh weight). Thus, cv. Dama exhibited higher values of in vitro antioxidant activity.

Edible flowers have been used as ingredients because of their biological activities, taste, and overall appearance. This research was aimed to characterize the chemical composition and in vitro antioxidant activity of the marigold flower (Calendula officinalis L.) extracted with different proportions of water and ethyl alcohol, and the lyophilized extract with higher content of antioxidant compounds was incorporated into an organic yogurt. Results showed that the hydroalcoholic extract (50:50 v/v) presented the highest total phenolic content (TPC), flavonoids, and antioxidant activity (ferric reducing antioxidant power (FRAP), total reducing capacity (TRC), and Cu2+/Fe2+ chelating ability). Phenolic acids and flavonoids were quantified in the extract by LC-DAD, while 19 compounds were tentatively identified by ESI-MS/MS. The lyophilized marigold extract (LME) also inhibited 12% of Wistar rat’s brain lipid oxidation in vitro, inhibited α-amylase, and α-glucosidase activities, but showed no cytotoxicity towards cancerous cells (HCT8 and A549). However, marigold flower extract protected human erythrocytes against mechanical stress. When added into an organic yogurt model (0 to 1.5%), LME increased TPC and antioxidant activity (2,2-diphenyl-1-picrylhydrazyl (DPPH) and TRC), and the sensory analysis showed that the organic yogurt had an acceptance of 80.4%. Our results show that the use of LME may be a technological strategy to increase the content of bioactive compounds in yogurts.

There has been a growing interest in the beneficial effects of simple phenolic acids and their ability to exhibit various biological activities. The aim of this study was to assess in vitro biological activities of 2-, 3-, and 4-hydroxybenzoate lithium (HBLi) complexes on HT-1080 human fibrosarcoma cells by methods of using a metabolic activity assay, immunochemical and morphological techniques. Results showed that HBLi complexes exert their cytotoxic activities in a concentration- and chemical structure-dependent manner in the following order: 4-HBLi > 3-HBLi > 2-HBLi. Flow cytometry displayed evidence of apoptosis induced by 3-HBLi (21.8%) and 4-HBLi (33.2%). These results were verified by SEM, which revealed the formation of apoptotic bodies. In addition, these 3-HBLi and 4-HBLi caused an increase in HT-1080 cell cycle arrest in G0/G1 phase when compared to the controls (25% and 30.6%, resp.) when cells were treated with 6 mM for 24 hours. Immunochemical studies related to the molecular mechanism of apoptosis indicated that HBLi complexes downregulated the expression of Bcl-2 and upregulated Bax, p53, and caspases-3 in a concentration-dependent manner. HBLi complexes lowered Bcl-2/Bax ratios and induced the expression of p53 and caspase-3. These results suggest that HBLi complexes may exert their apoptotic effects through mitochondrial-mediated, caspase-dependent, apoptotic mechanisms.

The aim of this study was to determine the impending antioxidant properties of different extracts of crude methanolic extract (CME) of leaves ofLannea coromandelica(L. coromandelica) and its two ethyl acetate (EAF) and aqueous (AqF) subfractions by employing various establishedin vitrosystems and estimation of total phenolic and flavonoid content. The results showed that extract and fractions possessed strong antioxidant activityin vitroand among them, EAF had the strongest antioxidant activity. EAF was confirmed for its highest phenolic content, total flavonoid contents, and total antioxidant capacity. The EAF was found to show remarkable scavenging activity on 2,2-diphenylpicrylhydrazyl (DPPH) (EC5063.9 ± 0.64 µg/mL), superoxide radical (EC508.2 ± 0.12 mg/mL), and Fe2+chelating activity (EC506.2 ± 0.09 mg/mL). Based on ourin vitroresults, EAF was investigated forin vivoantioxidant assay. Intragastric administration of the EAF can significantly increase levels of superoxide dismutase (SOD), catalase (CAT), glutathione (GSH), and glutathione peroxidase (GSH-Px) levels, and decrease malondialdehyde (MDA) content in the liver and kidney of CCl4-intoxicated rats. These new evidences show thatL. coromandelicabared antioxidant activity.

The present study was designed to confirm the traditional use of the fruits of Ficus hispida Linn. (Moraceae) as an antioxidant agent. Fruits of the plant extracted with methanol and crude methanol extract (CME) were further fractionated with n-hexane, chloroform, and ethyl acetate. All the fractions, n-hexane (NHF), chloroform (CHF), ethyl acetate (EAF), aqueous (AQF) and CME were preliminary screened for in vitro antioxidant activity and total phenolic and total flavonoid content. In DPPH radical scavenging assay, CME exhibited highest scavenging activity (IC50 = 11.20 µg/mL) as compared to other fractions. In this assay, IC50 of reference standard BHT was 5.10 µg/mL. The reducing power of the samples was in the order as AQF > CME > CHF > EAF > NHF. The results for hydrogen peroxide scavenging activity indicated that CME, EAF and AQF had almost the same scavenging activity except NHF. Total antioxidant capacity of CME and other fractions were ranked as CHF > AQF > CME > EAF > NHF.  In the assay of antioxidant constituents (total phenol and total flavonoids content), the CME had highest phenolic and flavonoids content. The results indicate that Ficus hispida fruits could be considered as a potential source of natural antioxidant.

<p class="Abstract"><em>Xanthium indicum</em> is an important medicinal plant traditionally used in Bangladesh as a folkloric treatment. The current study was undertaken to evaluate thrombolytic, anthelmintic, anti-oxidant, cytotoxic properties with phytochemical screening of methanolic extract of <em>X. indicum</em> leaves. The analysis of phytochemical screening confirmed the existence of phytosetrols and diterpenes. In thrombolytic assay, a significant clot lysis was observed at four concentrations of plant extract compare to the positive control streptokinase (30,000 IU, 15,000 IU) and negative control normal saline. The extract revealed potent anthelmintic activity at different concentrations. In anti-oxidant activity evaluation by two potential experiments namely total phenolic content determination and free radical scavenging assay by 2, 2-diphenylpicrylhydrazyl (DPPH), the leaves extract possess good anti-oxidant property. In the brine shrimp lethality bioassay, the crude extract showed potent (LC<sub>50</sub> 1.3 μg/mL) cytotoxic activity compare to the vincristine sulfate as a positive control (LC<sub>50</sub> 0.8 μg/mL). </p><p> </p>

SummaryIntroductıon: Instead of synthetic antioxidants, using of natural products with antioxidant activity is demanded.Objectıve: The aim of this study is to evaluate the antioxidant activity and total phenolic and flavonoid contents of theTrachystemon orientalis(borage) obtained from Ordu.Methods: The water and ethanol extracts of the borage plant were investigated by using severalin vitroantioxidant activity tests such as total antioxidant capacity, O2•−,•OH, DPPH and ABTS radicals scavenging activities, hydrogen peroxide scavenging activity, FRAP, Fe2+chelating and reducing power assays in order to make effective comparisons.Results: Water extract of theT. orientalishad the more phenolic (90 mg GA/g extract) and flavonoid (56.88 mg CT/g extract) contents than ethanol extract. Furthermore, both of the extracts showed strong antioxidant and radical scavenging activities determined by differentin vitromodels. The highest total antioxidant capacity as ascorbic acid equivalents of 244.50 mg/g extract was also calculated for water extract. Generally, a relationship between total phenolic content and antioxidant activity was established. It is also an important finding for an edible food source that the phenolic and flavonoid content and antioxidative activities for water extract was higher than the ethanol extract.Conclusıon:T. orientaliscould be used as a food ingredient instead of synthetic antioxidants and all results will contribute to the recent increase in investigations on using natural products in many areas such as food, pharmacy, alternative medicine and natural therapy.

Abstract Objectives Inhibition of carbohydrate absorption from intestine is an efficient way to prevent type 2 diabetes. Some edible plants have shown the potential as anti-diabetic agents by inhibiting carbohydrate-digestive enzymes in intestine. In terms of edible plant's health effect, bioactive compounds’ stability during the process of digestive enzymes has been commonly questioned. Therefore, the objectives of this study were to 1) examine inhibitory effect of garlic scape (GS), the green flower head of garlic on carbohydrate-degrading enzymes and 2) investigate antioxidant activity and total phenolic content (TPC) of GS treated with intestinal digestive enzymes. Methods GS extract (GSE) was prepared with 80% methanol (v/v) by homogenization, sonication, and filtration, and evaporation. α-glucosidase, maltase, glucoamylase, sucrase, α-amylase inhibitory activities were determined by measuring glucose hydrolyzed from nitrophenyl-glucopyranoside, maltose, starch, sucrose, and starch, respectively. To prepare gastrointestinal enzyme-digested GSE, GSE was digested with pepsin and a pancreatin-bile solution. GSE and gastrointestinal enzyme-digested GSE were assessed for TPC using a Folin-Ciocalteu reagent and for total antioxidant and radical scavenging activities by the following methods, phosphomolybdeum, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and 2,2-diphenyl-1-picrylhydrazyl (DPPH) methods, respectively. Results GSE (20 mg/ml) showed 30%, 50%, and 42% inhibition on α-glucosidase, maltase, and glucoamylase enzyme activities, respectively. GSE (50 mg/ml) showed 55.1% DPPH radical and 5782 mg vitamin C equivalent (VCE)/g ABTS radical scavenging activities. Total antioxidant activity of the GSE (1.5 mg/ml) exhibited 2.76 mg VCE/g. Consistent with its antioxidant property, the GSE showed a dose-dependent increase in TPC. Gastrointestinal enzymatic treatment resulted in approximately 46% reduction in DPPH and ABTS radical scavenging activity, total antioxidant activity, and TPC of the GSE. Conclusions Garlic scape has the potential as a natural anti-diabetic agent. Garlic scape keeps beneficial effects after the process of intestinal digestive system. A methanolextract of the leaves of Parabaena sagitatta Miers and its petroleum ether, carbon tetrachloride, dichloromethane, ethylacetate and aqueous soluble partitionates were evaluated for antioxidant activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH), Folin-Ciocalteau reagent and phosphomolybdenum total antioxidant assays by using butylated hydroxytolune (BHT) and ascorbic acid as standards. The dichloromethane soluble fraction demonstrated the presence of significant amount of phenolic compounds 61.06 ± 0.54 mg GAE/g of extract and also has moderate antioxidant activity IC50 50.62 ± 0.25 ?g/ml. A positive correlation (R2), 0.969 was observed between total phenolic content and total antioxidant activity of P. sagitatta.
