Muntingia calaburaL. is a tropical plant species that belongs to the Elaeocarpaceae family. The present study is aimed at determining the hepatoprotective activity of methanol extract ofM. calaburaleaves (MEMC) using two models of liver injury in rats. Rats were divided into five groupsn=6and received 10% DMSO (negative control), 50 mg/kg N-acetylcysteine (NAC; positive control), or MEMC (50, 250, and 500 mg/kg) orally once daily for 7 days and on the 8th day were subjected to the hepatotoxic induction using paracetamol (PCM). The blood and liver tissues were collected and subjected to biochemical and microscopical analysis. The extract was also subjected to antioxidant study using the 2,2-diphenyl-1-picrylhydrazyl-(DPPH) and superoxide anion-radical scavenging assays. At the same time, oxygen radical antioxidant capacity (ORAC) and total phenolic content were also determined. From the histological observation, lymphocyte infiltration and marked necrosis were observed in PCM-treated groups (negative control), whereas maintenance of hepatic structure was observed in group pretreated with N-acetylcysteine and MEMC. Hepatotoxic rats pretreated with NAC or MEMC exhibited significant decreaseP<0.05in ALT and AST enzymes level. Moreover, the extract also exhibited good antioxidant activity. In conclusion, MEMC exerts potential hepatoprotective activity that could be partly attributed to its antioxidant activity and, thus warrants further investigations.

In an attempt to further establish the pharmacological properties ofBauhinia purpurea(Fabaceae), hepatoprotective potential of methanol extract ofB. purpurealeaves (MEBP) was investigated using the paracetamol- (PCM-) induced liver toxicity in rats. Five groups of rats (n=6) were used and administered orally once daily with 10% DMSO (negative control), 200 mg/kg silymarin (positive control), or MEBP (50, 250, and 500 mg/kg) for 7 days, followed by the hepatotoxicity induction using paracetamol (PCM). The blood samples and livers were collected and subjected to biochemical and microscopical analysis. The extract was also subjected to antioxidant study using the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay with the total phenolic content (TPC) also determined. In conclusion, MEBP exerts potential hepatoprotective activity that could be partly attributed to its antioxidant activity and high phenolic content and thus warrants further investigation.

Antioxidant and free radical scavenging activity of methanol, ethanol, ethyl acetate and chloroform extracts of aerial parts of Lamium purpureum L. was determined by DPPH, ABTS, FRAP and TRP assays. Contents of flavonoids and phenols were also investigated. The total phenolic content in the extracts, determined using Folin?Ciocalteu assay, ranged between 8.57 to 128.00 mg GAE/g d.e. while concentrations of flavonoids in the extracts varied from 24.20 to 39.80 mg QuE/g d.e. The highest phenolic content was found in methanol extract (128.00 mg GAE/g d.e.). The highest content of total flavonoids was identified in the methanol extract (39.80 mg QuE/g d.e.) and the lowest was in the chloroform (24.30 mg QuE/g d.e.). DPPH scavenging of the extracts was determined and obtained IC50 values ranged from 0.12 to 3.12 mg/mL of solution. The values of ABTS radical scavenging activity ranged from 0.35 to 1.80 mg AA/g. The highest ABTS antiradical activity was registered for methanol extract. The FRAP value was found within the range 0.08 to 1.04 ?mol Fe/mg. The best radical scavenger was methanol (1.04 ?mol Fe/mg). In reducing power assay different extracts of L. purpureum showed increasing of activity with increased concentration, and all extracts possessed substantial dose dependent antioxidant activity. The best reducing capacity was obtained with methanol extract of L. purpureum (0.0132 mg AA/mL). The results in this study confirmed that L. purpureum possesses moderate antioxidant properties.

Dryopteris marginalis L. is a local plant in Bangladesh which is widely used as folk medicine. In this study, the chemical composition of essential oil of D. marginalis L. as well as the antioxidant activity, antibacterial activity and total phenolic content of various organic extracts were analyzed. The gas chromatography–mass spectrometry analysis was used to identify the chemical compositions. The antibacterial activity of different extracts of Dryopteris marginalis was examined against three gram-negative bacteria and two gram-positive bacteria. The antibacterial activity of different extracts was investigated using disc diffusion method and minimum inhibitory concentration showing important antibacterial activity and was measured by using broth dilution method. The antioxidant activity was determined by using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and superoxide radical scavenging assay. The total phenolic portion of the extracts were ascertained by Folin-Ciocalteu in alkaline medium. Twenty-two volatile compounds were identified from the essential oil. The methanol extract exhibited significant inhibitory effect which was close to the value of standard antibiotic (Streptomycin). The zones of inhibition of different organic extracts against the tested bacteria were found in the range of (7-15) mm and the minimum inhibitory concentrations (MIC) were recorded (15.62-1000) µg/ml. The antioxidant activity of D. marginalis was (82.13+1.2) % and (59.21+0.45) % highest activity with DPPH and superoxide radical scavenging assay respectively. The methanol extract showed highest (147.43+3.1) mg/g GAE of total phenolic content. The extracts and essential oil of D. marginalis showing strong antibacterial activity can be considered as a potential source of natural medicine and the ingredient of modern therapeutic agents.

The flowers of Rhododendron arboreum Smith is a source of polyphenolic compounds. A flavonol, quercetin, was isolated from ethyl acetate fraction of the methanol extract of flower petals of R. arboreum by repeated Sephadex LH-20 column chromatography. Isolated quercetin was characterized by comparing melting point, Rf values, UV and IR spectra with authentic quercetin. The isolated quercetin was used as a standard for the estimation of total flavonoids.  Total phenolic and total flavonoid content in different parts of R. arboreum was carried out spectrophotometrically using Folin-Ciocalteu reagent and Aluminium chloride reagent respectively. Gallic acid and quercetin were used as standard for the construction of calibration curve of phenolic and flavonoid respectively. The results showed that the highest total phenolic content was detected in the 70% acetone extract of the flowers (600 mg GAE/g extract) and petals (600 mg GAE/g extract) and the lowest amount was detected in methanol extract of stem (188 mg GAE/g extract). Similarly, the highest total flavonoid was detected in the 70% acetone extract of the twigs (170 mg QE/g extract) and the lowest amount was detected in the methanol extract of stem (45 mg QE/g extract).  The antioxidant activity of the methanol extracts obtained from different parts of R. arboreum was determined by 2, 2- diphenyl-1-picryl hydrazyl (DPPH) assay and the radical scavenging activity (IC50) was calculated. The highest free radical scavenging effect was observed in leaves with IC50=8.34 ?g/ml and lowest was observed in stem with IC50=67.83 ?g/ml. The IC50 values, total phenolic and total flavonoid content (correlation coefficient R2= 0.923 for phenolic, R2= 0.965 for flavonoid) were correlated which showed strong correlation indicating that the major components responsible for antioxidant activity is phenolics. The highest the phenolic content, the lowest the IC50 value observed. The result indicated that R. arboreum is a rich source of high value polyphenols as natural antioxidant to use in preventive medicine as well as in food and pharmaceutical industry.Scientific World, Vol. 12, No. 12, September 2014, page 34-40

Objective: The present study was undertaken to evaluate the total phenolic and flavonoid content and the antioxidant property of two important seagrass species namely, Halophila ovalis and Halophila beccarii occurring in Chilika lagoon, Odisha, India. Methods: Total Phenolic Content (TPC) of the extracts of Halophila species was determined by Folin-Ciocalteu method with little modifications and the total flavonoid content (TFC) was measured by aluminum chloride colorimetric assay. The antioxidant activity of different extracts was investigated by DPPH and ABTS radical scavenging activity. IC50 values were calculated for the DPPH and ABTS methods. Result: The study revealed that the methanol extract of H. ovalis has greater antioxidant activity than H. beccarii. Methanol extract of both the species (H. ovalis and H. beccarii) was found to possess high phenolic content at value of 70.25 mg GAE/g of extract and 48.53 mg GAE/g of extract respectively. Similarly flavonoid contents was found highest in methanol extract for both H. ovalis (76.82 mg quercetin equivalent/ g of extract) and H. beccarii (64.28 mg quercetin equivalent/ g of extract). The antioxidant activity of different extracts of these two species were evaluated using DPPH and ABTS radical assay. The methanol extract of both H. ovalis and H. beccarii showed high radical scavenging activity with IC50 values of 37.77 μg/ml and 52.25 μg/ml for DPPH and 25.62 μg/ml and 45.45 μg/ml for ABTS respectively. Conclusion: The study revealed the potential of the Halophila species as natural sources of antioxidants having considerable commercial importance.

Gallic acid was isolated from ethylacetate soluble portion of the methanol extract of fruit pulp of Terminalia chebula and characterized by comparing the melting point, Rf values, UV and IR spectra with authentic gallic acid. Quantitative determination of phenolics from different extracts of medicinal herbs such as Adhatoda vasica Nees, Bergenia ciliata (Haw) Sternb, Phyllanthus emblica Linnaeus, Terminalia bellirica (Gaerth) Roxb, Terminalia chebula Retzius and Vitex negundo Linnaeus which are commonly used as home herbal remedies for the primary health care was carried out using Folin-Ciocalteu colorimetric method. Gallic acid was used as the standard for the estimation of phenolics. All the investigated plant extracts contained high amount of phenolics but the highest amount was detected in 70% acetone extract of B. ciliata (357.08 mg GAE/g sample) and the lowest amount was detected in 50% aqueous methanol extract of T. bellirica (108.69 mg GAE/g sample). The antioxidant activity of selected extracts was determined by 1,1-diphenyl-2-picryl-hydrazyl (DPPH) assay. The extract having the highest phenolic content showed the lowest IC50 demonstrating the positive correlation between radical scavenging activity and total phenolic content. Plant extracts are an important alternative to antibiotics, which are ever more restricted because of their developing microbial resistance and some adverse effects that have been observed following frequent application. The aim of the present study was to determine the antifungal and antioxidant activity of the methanolic extracts of Acorus calamus, Chlorella vulgaris, Lemna minuta and Scenedesmus dimorphus. The antifungal activity of the extracts against strains of Aspergillus flavus, Aspergillus parasiticus, Aspergillus ochraceus, Aspergillus niger, Aspergillus carbonarius, Fusarium graminearum, Fusarium oxysporum, Penicillium chrysogenum and Alternaria alternata was evaluated via the agar well diffusion method. The antioxidant activity of the extracts was measured through the determination of three parameters—total phenolic content, total flavonoid content and radical scavenging potential (determined through UV/Vis analysis). A. calamus extracts had the highest antimicrobial activity against eight fungal strains, followed by the C. vulgaris, L. minuta and S. dimorphus extracts, which were inhibitory against two to three strains. Among the extracts from the species studied, the extract from S. dimorphus showed the highest antioxidant potential, as determined via the DPPH (1,1’-diphenyl-2-picrylhydrazil-radical) method. This correlated to its high total phenolic and flavonoid content. From A. calamus and L. minuta, methanolic extracts were obtained that exhibited similar values of the aforementioned parameters, followed by C. vulgaris extracts, which showed the lowest antioxidant activity. Based on the Pearson correlation coefficients, the impacts of the total phenolic content and the total flavonoid content on radical scavenging capacity are similar, and flavonoids were a significant part of the total phenolic compounds extracted from the plant materials studied.

AbstractIn this study, the extraction conditions extracted maximize amounts of phenolic and bioactive compounds from the fruit extract of Ficus auriculata by using optimized response surface methodology. The antioxidant capacity was evaluated through the assay of radical scavenging ability on DPPH and ABTS as well as reducing power assays on total phenolic content (TPC). For the extraction purpose, the ultrasonic assisted extraction technique was employed. A second-order polynomial model satisfactorily fitted to the experimental findings concerning antioxidant activity (R2 = 0.968, P < 0.0001) and total phenolic content (R2 = 0.961, P < 0.0001), indicating a significant correlation between the experimental and expected value. The highest DPPH radical scavenging activity was achieved 85.20 ± 0.96% at the optimum extraction parameters of 52.5% ethanol (v/v), 40.0 °C temperature, and 22 min extraction time. Alternatively, the highest yield of total phenolic content was found 31.65 ± 0.94 mg GAE/g DF at the optimum extraction conditions. From the LC–ESI–MS profiling of the optimized extract, 18 bioactive compounds were tentatively identified, which may regulate the antioxidant activity of fruits of F. auriculata.

This work aims to determine antioxidant, total phenolic content and Fourier transform infrared spectroscopy (FTIR) analysis of breadfruit leaves which are essential in management of diabetes. The methanolic extracts of breadfruit leaves was used to analyze for total phenolic content and antioxidant. Total phenolic content of the extracts was measured using the Folin–Ciocalteu assay while the antioxidant activity of plant extracts was measured by the 1, 1-diphenyl-2-picryhyradrazyl (DPPH) radical scavenging assay. FTIR analysis was used to determine the chemical components in the leaves. Total phenolic content in fresh breadfruit leaves (144.16 mg/g ± 17.98) was comparable to those of green tea. The results showed the breadfruit leaves extracts exhibited potent antioxidant activity. The presence of OH group also suggests antioxidant capacity of breadfruit leaves to deactivate free radicals as glucose itself could react with hydrogen peroxide in the presence of iron and copper ions to form hydroxyl radical.

<p><strong>Objective: </strong>To evaluate the <em>in vitro </em>antioxidant activity of liquorice (<em>Glycyrrhiza glabra) </em>against H<sub>2</sub>O<sub>2</sub> induced oxidative stress in HepG2 cell line.</p><p><strong>Methods: </strong>Antioxidant activity of methanolic extracts of <em>Glycyrrhiza glabra</em> was investigated by measuring total phenolic content using folin-ciocalteu reagent (FCR), free radical scavenging activity by DPPH and ferric reducing antioxidant power (FRAP). The presence of phenolic compounds and flavonoids in the extract was confirmed by Liquid Chromatography-Mass Spectrometry (LC-MS) analysis. Furthermore, the protective effect of methanolic extract of <em>Glycyrrhiza glabra</em> against oxidative stress induced by H<sub>2</sub>O<sub>2 </sub>in HepG2 cells was investigated by MTT assay. HepG2 cells were exposed with five different treatments viz. liquorice, H<sub>2</sub>O<sub>2</sub>, ascorbic acid, H<sub>2</sub>O<sub>2</sub>+liquorice and H<sub>2</sub>O<sub>2</sub>+ascorbic acid, to explore the effect of the extract on malondialdehyde (MDA) production, catalase activity, and glutathione reductase levels.<strong></strong></p><p><strong>Results: </strong>The total phenolic content estimated in <em>Glycyrrhiza glabra </em>extract was found to be 241.47 µg per 1000 µg/ml of methanolic extract. It was found that as the concentration of the extract was increased both the free radical scavenging activity and ferric ion reducing power was also found to increase. LC-MS analysis confirmed the presence of eight different phenolic compounds in the methanolic extract which are possibly contributing to the antioxidant activity exhibited by the extract. It was also observed that liquorice treated HepG2 cells showed lower MDA and higher glutathione and catalase levels as compared to only H<sub>2</sub>O<sub>2 </sub>treated HepG2 cells where increased MDA production, decreased glutathione reductase and catalase production was observed.</p><p><strong>Conclusion: </strong>Our results thus conclude that, the methanolic extract of <em>Glycyrrhiza glabra </em>can be used as natural supplements in various disease conditions where oxidative stress has been reported. <strong></strong></p><p> </p>
