Blood stasis (BS) is characterized as a disorder of blood circulation. In traditional Korean medicine (TKM), it is viewed as a cause factor of diseases such as multiple sclerosis and stroke. This study investigated differences in the plasma metabolites profiles of subjects displaying BS or non-BS patterns. Thirty-one patients with cerebral infarction diagnosed with BS and an equal number of sex- and age-matched non-BS patients were enrolled. Metabolic profiling was performed using UPLC-MS. The ratio of subjects with a rough pulse and purple coloration of the tongue was higher in patients presenting with BS pattern. Through metabolomics analysis, 82 metabolites that differed significantly between the BS and non-BS pattern were identified, and the two groups were significantly separated using an orthogonal partial least square-discriminant analysis model(P<0.001). Of these 82 metabolites, acetyl carnitine, leucine, kynurenine, phosphocholine, hexanoyl carnitine, and decanoyl carnitine were present in significantly higher levels in patients with a BS pattern than those with a non-BS pattern. Our results also demonstrated that seven plasma metabolites, including acyl-carnitines and kynurenine, were associated with a BS pattern, suggesting that variant plasma metabolic profiles may serve as a biomarker for diagnosis of BS in patients with cerebral infarction.

Abstract Background OPD and OPD' are the two main active components of Ophiopogon japonicas in Shenmai injection (SMI). Being isomers of each other, they are supposed to have similar pharmacological activities, but the actual situation is complicated. The difference of hemolytic behavior between OPD and OPD' in vivo and in vitro was discovered and reported by our group for the first time. In vitro, only OPD' showed hemolysis reaction, while in vivo, both OPD and OPD' caused hemolysis. In vitro, the primary cause of hemolysis has been confirmed to be related to the difference between physical and chemical properties of OPD and OPD'. In vivo, although there is a possible explanation for this phenomenon, the one is that OPD is bio-transformed into OPD' or its analogues in vivo, the other one is that both OPD and OPD' were metabolized into more activated forms for hemolysis. However, the mechanism of hemolysis in vivo is still unclear, especially the existing literature are still difficult to explain why OPD shows the inconsistent hemolysis behavior in vivo and in vitro. Therefore, the study of hemolysis of OPD and OPD' in vivo is of great practical significance in response to the increase of adverse events of SMI. Methods Aiming at the hemolysis in vivo, this manuscript adopted untargeted metabolomics and lipidomics technology to preliminarily explore the changes of plasma metabolites and lipids of OPD- and OPD'-treated rats. Metabolomics and lipidomics analyses were performed on ultra-high performance liquid chromatography (UPLC) system tandem with different mass spectrometers (MS) and different columns respectively. Multivariate statistical approaches such as principal component analysis (PCA) and orthogonal partial least square-discriminant analysis (OPLS-DA) were applied to screen the differential metabolites and lipids. Results Both OPD and OPD' groups experienced hemolysis, Changes in endogenous differential metabolites and differential lipids, enrichment of differential metabolic pathways, and correlation analysis of differential metabolites and lipids all indicated that the causes of hemolysis by OPD and OPD' were closely related to the interference of phospholipid metabolism. Conclusions This study provided a comprehensive description of metabolomics and lipidomics changes between OPD- and OPD'-treated rats, it would add to the knowledge base of the field, which also provided scientific guidance for the subsequent mechanism research. However, the underlying mechanism require further research.

Purpose This study aims to explain the factors that affect the performance of traditional fishing business at Kubu Raya Regency of West Borneo Province. Design/methodology/approach The method used in this research is quantitative method. The research location in Kubu, Padang Tikar and Teluk Pakedai sub-districts of Kubu Raya Regency of West Borneo Province, considering the location is the target of CCDP-IFAD in Kubu Raya district. The data analysis model in this research is done by using structural equation model (SEM) approach with assisted WarpPLS program (partial least square development). Findings Based on the results of the analysis of SEM, it is revealed that the direct influence of the six dependent variables the environment of the individual fisherman, regulatory and government policy, environmental economics, a social-cultural environment, managerial capacity and the sustainability of the business aspects of ecological management affect directly toward traditional capture fisheries business performance, only empowerment that is not directly influential on performance of traditional capture fisheries business. Originality/value The originality in this study is shown in the objectives and variables used in the research, i.e. individual environmental variables of fishermen, government policies and regulations, economic environment, social-cultural environment, empowerment, management capacity and business sustainability influence the performance variable of traditional fishing business. In addition, there is a direct influence, allegedly there is an indirect influence on the variables empowerment of the performance of fishery business.

End-stage renal disease patients undergoing maintenance hemodialysis (HD) are vulnerable to the protein energy wasting (PEW) syndrome. Identification and diagnosis of PEW relies on clinical processes of judgment dependent on fulfilling multiple criteria drawn from serum biochemistry, weight status, predictive muscle mass, dietary energy and protein intakes. Therefore, we sought to explore the biomarkers’ signature with plasma metabolites of PEW by using 1H-nuclear magnetic resonance for an untargeted metabolomics approach in the HD population, to understand metabolic alteration of PEW. In this case-controlled study, a total of 53 patients undergoing chronic HD were identified having PEW based on established diagnostic criteria and were age- and sex-matched with non-PEW (n = 53) HD patients. Fasting predialysis plasma samples were analyzed. Partial least square discriminant analysis demonstrated a significant separation between groups for specific metabolic pattern alterations. Further quantitative analysis showed that the level of 3-hydroxybutyrate, acetate, arabinose, maltose, ribose, sucrose and tartrate were significantly increased whilst creatinine was significantly decreased (all p < 0.05) in PEW subjects. Pathway analysis indicated that PEW-related metabolites reflected perturbations in fatty acid mechanism and induction of glyoxylate and dicarboxylate pathway attributed to gluconeogenesis. These results provide preliminary data in understanding metabolic alteration of PEW and corresponding abnormal metabolites that could potentially serve as biomarkers of PEW.

The widely used techniques for analyzing the quality of powdered food products focus on targeted detection with a low-throughput screening of samples. Owing to potentially significant health threats and large-scale adulterations, food regulatory agencies and industries require rapid and non-destructive analytical techniques for the detection of unexpected compounds present in products. Accordingly, shortwave-infrared hyperspectral imaging (SWIR-HSI) for high throughput authenticity analysis of almond powder was investigated in this study. Two different varieties of almond powder, adulterated with apricot and peanut powder at different concentrations, were imaged using the SWIR-HSI system. A one-class classifier technique, known as data-driven soft independent modeling of class analogy (DD-SIMCA), was used on collected data sets of pure and adulterated samples. A partial least square regression (PLSR) model was further developed to predict adulterant concentrations in almond powder. Classification results from DD-SIMCA yielded 100% sensitivity and 89–100% specificity for different validation sets of adulterated samples. The results obtained from the PLSR analysis yielded a high determination coefficient (R2) and low error values (<1%) for each variety of almond powder adulterated with apricot; however, a relatively higher error rates of 2.5% and 4.4% for the two varieties of almond powder adulterated with peanut powder, which indicates the performance of quantitative analysis model could vary with sample condition, such as variety, originality, etc. PLSR-based concentration mapped images visually characterized the adulterant (apricot) concentration in the almond powder. These results demonstrate that the SWIR-HSI technique combined with the one-class classifier DD-SIMCA can be used effectively for a high-throughput quality screening of almond powder regarding potential adulteration.

We clarified the hepatoprotective effect of Gentiana dahurica Fisch ethanol extract (GDEE) in our previous study, and we further revealed the mechanism with the help of metabolomics technology in this study. The livers from Control group, Alcohol group, and Alcohol + GDEE group were analyzed by metabolomics. The metabolites in the liver were separated by ultra-high-performance liquid chromatography (UHPLC) and were tentatively identified using mass spectrometry (MS)/MS analysis. Differential metabolites were defined with VIP > 1 and P < 0.05 . Principal component analysis (PCA) and orthogonal partial least square discriminant analysis (OPLS-DA) were applied to analyze differences among these groups. The results showed that the groups could be clearly distinguished by PCA and OPLS-DA analysis. Alcohol and GDEE could change the overall profile of liver metabolites. Alterations in liver tissues of ALD mice induced by alcohol were mainly involved in the dipeptides, purine and pyrimidine metabolism and glucose and lipid metabolism, which could be partly affected by GDEE. This study revealed that the mechanism of GDEE in alleviating ALD had the characteristics of multitarget and multipathway.

Endometrial cancer has a high prevalence among post-menopausal women in developed countries. We aimed to explore whether certain metabolic patterns could be related to the characteristics of aggressive disease and poorer survival among endometrial cancer patients in Western Norway. Patients with endometrial cancer with short survival (n = 20) were matched according to FIGO (International Federation of Gynecology and Obstetrics, 2009 criteria) stage, histology, and grade, with patients with long survival (n = 20). Plasma metabolites were measured on a multiplex system including 183 metabolites, which were subsequently determined using liquid chromatography-mass spectrometry. Partial least square discriminant analysis, together with hierarchical clustering, was used to identify patterns which distinguished short from long survival. A proposed signature of metabolites related to survival was suggested, and a multivariate receiver operating characteristic (ROC) analysis yielded an area under the curve (AUC) of 0.820–0.965 (p ≤ 0.001). Methionine sulfoxide seems to be particularly strongly associated with poor survival rates in these patients. In a subgroup with preoperative contrast-enhanced computed tomography data, selected metabolites correlated with the estimated abdominal fat distribution parameters. Metabolic signatures may predict prognosis and be promising supplements when evaluating phenotypes and exploring metabolic pathways related to the progression of endometrial cancer. In the future, this may serve as a useful tool in cancer management.

Lymphoma defines a group of different diseases. This study examined pre-treatment plasma samples from 66 adult patients (aged 20–74) newly diagnosed with any lymphoma subtype, and 96 frequency matched population controls. We used gas chromatography-mass spectrometry (GC-MS) to compare the metabolic profile by case/control status and across the major lymphoma subtypes. We conducted univariate and multivariate analyses, and partial least square discriminant analysis (PLS-DA). When compared to the controls, statistically validated models were obtained for diffuse large B-cell lymphoma (DLBCL), chronic lymphocytic leukemia (CLL), multiple myeloma (MM), and Hodgkin lymphoma (HL), but not follicular lymphoma (FL). The metabolomic analysis highlighted interesting differences between lymphoma patients and population controls, allowing the discrimination between pathologic and healthy subjects: Important metabolites, such as hypoxanthine and elaidic acid, were more abundant in all lymphoma subtypes. The small sample size of the individual lymphoma subtypes prevented obtaining PLS-DA validated models, although specific peculiar features of each subtype were observed; for instance, fatty acids were most represented in MM and HL patients, while 2-aminoadipic acid, 2-aminoheptanedioic acid, erythritol, and threitol characterized DLBCL and CLL. Metabolomic analysis was able to highlight interesting differences between lymphoma patients and population controls, allowing the discrimination between pathologic and healthy subjects. Further studies are warranted to understand whether the peculiar metabolic patterns observed might serve as early biomarkers of lymphoma.

Mice lacking the GABAB(1) subunit of gamma-aminobutyric acid (GABA) type B receptors exhibit spontaneous seizures, hyperalgesia, hyperlocomotor activity, and memory impairment. Although mice lacking the GABAB(1) subunit are viable, they are sterile, and to generate knockout (KO) mice, it is necessary to cross heterozygous (HZ) mice. The aim of our study was to detect the metabolic differences between the three genotypes of GABAB(1) KO mice in order to further characterize this experimental animal model. Plasma samples were collected from wild-type (WT), HZ, and KO mice. Samples were analyzed by means of a gas chromatography-mass spectrometry (GC-MS) platform. Univariate t-test, and partial least square discriminant analysis (PLS-DA) were performed to compare the metabolic pattern of different genotypes. The metabolomic analysis highlighted differences between the three genotypes and identified some metabolites less abundant in KO mice, namely elaidic acid and other fatty acids, and chiro-inositol.

Purpose The purpose of this paper is to analyze marketability constructed from market share and concentration and to test its effect on the profitability and the mediation effects of profit‒loss sharing under stewardship theory. Design/methodology/approach This research employs data of financial statements published by ten sharia commercial banks listed in the Indonesia Financial Services Authority during the period 2011–2016. The data are analyzed into path analysis model using multiple mediators. Findings The result reveals that sharia banks’ marketability in Indonesia tends to be low. Based on the test of significance through Partial Least Square, it is found that marketability has a positive effect on the level of profitability, indicating that market share and concentration of sharia banks positively lead the change on the level of Return on Asset and Return on Equity. This paper further identifies the mediation effects emerged through mudharabah and musharakah. The results point out that mudharabah has a partial effect and musharakah has a competitive effect on the relationship between market share and profitability. Practical implications This paper can be a decision-maker for Central Bank and Financial Services Authority for encouraging sharia banks to enhance the power market through the mode of finances with profit‒loss sharing. Originality/value The growth of sharia banks is currently becoming highlight of the literature of sharia banks. This paper provides insights into stewardship theory that sharia banking management provides the concept of the alignment of interest.

Online near infrared reflectance spectroscopy combined with characteristic wavelength variables was used to establish a fast and nondestructive analytical method for the classification of fish meal produced in China and Peru. In this study, 117 fish meal samples (47 from China and 70 from Peru) were scanned in the spectral range of 1000–2500 nm by the online near infrared spectroscopy instrument applied on the conveyor belt. The K–S (Kennard–Stone) method was used for the division of samples into calibration and validation sets. Principal component analysis and partial least square discriminant analysis were applied to classify fish meal samples. The results showed that the discrimination accuracies with calibration and validation set samples were 100% and 89.74%, respectively, for the partial least square discriminant analysis model using the full spectrum after the optimimal spectral pre-treatment. Then competitive adaptive reweighted sampling (CARS) was used to select the characteristic wavelength variables for partial least square discriminant analysis model analysis, and the discrimination accuracy for the validation set increased to 94.87%. All the results indicated that online near infrared spectroscopy combined with characteristic wavelength variables could be used for discriminating fish meal samples produced in different places, which offers feed purchasers an effective, reliable, and real-time analysis method for the identification and authentication of the commercial fish meal product.
