Maize (Zea maysL.) is the most important cereal crop in the world. However, soil salinity has become a major problem affecting plant productivity due to arable field degradation. Thus, transgenic maize transformed with a salinity tolerance gene has been developed to further evaluate its salt tolerance and effects on agronomic traits. It is necessary to analyze the potential environmental risk of transgenic maize before further commercialization. Enzyme activities, physicochemical properties, and microbial populations were evaluated in saline and nonsaline rhizosphere soils from a transgenic maize line (WL-73) overexpressingBcWRKY1and from wild-type (WT) maize LH1037. Measurements were taken at four growth stages (V3, V9, R1, and R6) and repeated in three consecutive years (2012–2014). There was no change in the rhizosphere soils of either WL-73 or WT plants in the four soil enzyme activities, seven soil physicochemical properties, and the populations of three soil organisms. The results of this study suggested that salinity tolerant transgenic maize had no adverse impact on soil properties in soil rhizosphere during three consecutive years at two different locations and provided a theoretical basis for environmental impact monitoring of salinity tolerant transgenic maize.

AbstractBiochar is an effective soil conditioner. However, we have limited understanding of biochar effects on the tobacco growth and bacterial communities in rhizosphere. The aim of this study was to investigate the effects of different straw biochar amendment (0, 2, 10, and 50 g/kg dry soil) on tobacco growth, soil properties, and bacterial communities in rhizosphere by pot trials. Most of tobacco agronomic traits increased when the application rate varied from 0 to 10 g/kg, but were inhibited by 50 g/kg of biochar application. Soil pH, SOC, available nutrients and soil urease, invertase, and acid phosphatase activities were all increased with the biochar application, whereas catalase activity decreased or remained unchanged. The OTUs and bacterial community diversity indices differed with the biochar application doses in rhizosphere and non-rhizosphere soils. And significant differences in bacterial communities were found between the rhizosphere and non-rhizosphere soils despite the biochar addition. Firmicutes, Proteobacteria, Acidobacteria, Bacteroidetes, and Actinobacteria were the dominant phyla in all soil samples, but they had different abundances in different treatment influenced by the rhizosphere and biochar effect. The high dose of biochar (50 g/kg) decreased the similarity of soil bacterial community structure in rhizosphere compared with those in non-rhizosphere soil. These results provide a better understanding of the microecological benefits of straw biochar in tobacco ecosystem.

Soil properties may be varied by spatially and temporally with different agricultural practices. An accurate and reliable soil properties assessment is challenging issue in soil analysis. The soil properties assessment is very important for understanding the soil properties, nutrient management, influence of fertilizers and relation between soil properties which are affecting the plant growth. Conventional laboratory methods used to analyses soil properties are generally impractical because they are time-consuming, expensive and sometimes imprecise. On other hand, Visible and infrared spectroscopy can effectively characterize soil. Spectroscopic measurements are rapid, precise and inexpensive. Soil spectroscopy has shown to be a fast, cost-effective, environmentally friendly, non-destructive, reproducible and repeatable analytical technique. In the present research, we use spectroscopy techniques for soil properties analysis. The spectra of agglomerated farming soils were acquired by the ASD Field spec 4 spectroradiometer. Different fertilizers treatment applied soil samples are collected in pre monsoon and post monsoon season for 2 year (4 season) for banana and cotton crops in the form of DS-I and DS-II respectively. The soil spectra of VNIR region were preprocessed to get pure spectra. Then process the acquired spectral data by statistical methods for quantitative analysis of soil properties. The detected soil properties were carbon, Nitrogen, soil organic matter, pH, phosphorus, potassium, moisture sand, silt and clay. Soil pH is most important chemical properties that describe the relative acidity or alkalinity of the soil. It directly effect on plant growth and other soil properties. The relationship between pH properties on soil physical and chemical parameters and their influence were analyses by using linear regression model and show the performance of regression model with R2 and RMSE. Abstract Background Pinellia ternata (Thunb.) Breit. is a commonly used herb in traditional Chinese medicine, and the main raw material of various Chinese patent medicines. Continuous cropping obstacle (CCO) is the main factor leading to the decline of crop yields and quality. Methods Metagenomics sequencing technology was used to analyze the microbial community and functional genes of continuous cropping (CC) and control (CK) soils of P. ternata. In addition, differences in physicochemical properties, enzyme activities, microbial community composition and the abundance of functional genes in CC and CK were evaluated, as well as the relationship between these factors and CCO. Results Results indicated that CC of P. ternata led to the decline of rhizosphere soil pH, nutrient imbalance and enzyme activity reduction. Metagenomic analysis indicted that CC also changed the composition of the microbial community, causing an increase in the relative abundance of pathogenic microorganisms such as Fusarium, Klebsiella oxytoca and Pectobacterium carotovorum in the P. ternata rhizosphere. The relative abundance of potentially beneficial Burkholderia and Bradyrhizobium was recorded to decrease. Results also showed that there were considerable differences in CC and CK about the abundances of functional genes related to soil enzymes and the degradation of P. ternata allelochemicals, as well as the microbial groups which they belong. These results clarified the effects of CC on the microbial community structure and functional genes of soil. In addition, Burkholderia and Bradyrhizobium might play important roles in enhancing soil fertility and reducing the toxicity of phenolic acids in rhizosphere soil. Conclusions CC of P. ternata changed the physicochemical properties, microbial community and functional genes of rhizosphere soil. Burkholderia and Bradyrhizobium for enhancing soil fertility and reducing the toxicity of phenolic acids might be potentially beneficial. These results provide theoretical guidance for bioremediation of CCO soil of P. ternata and other staple crops. Graphic abstract

Objective: The paper aimed to immobilize amylase producing bacterial strain on a suitable matrix and characterization of its physicochemical properties so that much amount of amylase could be produced to be applied in different industries. Methods: Bacterial colonies were sub-cultured from samples collected from soil in freshly prepared dishes containing starch agar by dot method using sterile inoculating needles from which five different bacteria belonged to genus Bacillus were isolated and assigned as A1, A2, A3, A4, and A5. Results: It was found that A1 displayed the highest enzyme activity of 17.89 IU/ml with enzyme assay of 0.83 mg/ml and the bacterium was identified to be Bacillus subtilis. A5 displayed 10.13 IU/ml with protein contents of 0.11 mg/ml indicated that A1 possess the highest enzyme activities which were categorized under Bacillus and protein contents and A5 showed less amount of enzyme activities and protein contents as compared to other. Conclusion: The bacteria which were produced much amount of enzyme activities identified as Bacillus subtilis and recommended and have been recommended to be cultured for the production of amylase enzyme.
