Abstract The Process of Coming and Going in this World is a four-channel, site-specific installation by artist Ruth Burke. The work incorporates its audience, including nonhuman collaborators. While dependent on time and place, it has been preserved in audio recordings and photographs. In this interview between the artist and art historian Jessica Landau, they discuss the installation’s use of sound, time, and place to evoke interspecies relationships based on collaboration and co-constituted domestication. While using the installation and subsequent sound recording of it as a starting point, the conversation looks at the ways our relationships with animals on farms involve notions of time and place, companionship, and coevolution.

A recording represents a paradoxical perceptual source: we can either attend to the sound of the medium, or to the virtual world conveyed by it, and the work of a record producer can be understood as either a process of capturing performances or one of creating virtual worlds. This paper demonstrates that the record producer John Culshaw had clear ideas about how recordings might approach the condition of a work of art, rather than being simply the trace of a moment in time. Culshaw's fundamental aesthetic and technical approach is described and illustrated with reference to a number of key recordings. Taking the relationship between sound recording and film as a starting point, and making use of the concept of subject-position, the tension between Culshaw's radical approach to the listener and traditional approach to the authority of the score is explored. Possible reasons are proposed for the abandonment of his ideas, and for the absence of a Culshaw legacy (apart from the recordings themselves). The paper ends with a brief discussion of the current paradigm for the recording of classical music, which seeks in various ways to reproduce “the live experience” in “the finest seat in the house”.

AbstractWagner’s vaunted model of artistic synthesis persists in scholarly assessments of his work. But at its centre, the composer argued that the media of voice and orchestra do not mix: they retain their identities as separate channels of sound that can neither duplicate nor substitute for one another. Taking as a starting point Wagner’s claims for the non-adaptability of media, this article addresses the adaptation of Wagner’s music to the modern digital technologies of HD cinema and video game. Drawing on a wide circle of writers, from Schiller and Žižek to Bakhtin, Augé, Baudrillard and second-generation media theorists, it interrogates the concept of ‘reality’ within live acoustic performance, both historically, as a discursive concept, and technologically, via the sensory realism of digital simulcasting and telepresence. The philosophical opposition of appearance and reality fails when reality is defined by the intimate simulation of a sensory event as it is registered on the body. And by contrasting the traditions of high fidelity in (classical) sound recording with that of rendering sound in cinema, I suggest ways in which unmixable media appear to have an afterlife in modern technologies. This raises questions – in a post-Benjamin, post-McLuhan context – about our definition of ‘liveness’, the concept of authenticity within mediatised and acoustic sounds, and our vulnerability to the technological effects of media.

Jazz is the first type of music art the earliest stage of development of which had been recorded. A single play recorded in 1917 by the quintet Original Dixieland “Jass” Band from New Orleans is known in history as the first jazz record. There’s a perception in the academic community that the musical material on this record can hardly be considered as a typical representative of jazz music of that period. The music was performed by the white musicians, though most first jazz bands were black, and the music was far from a real solo improvisation. However, it was not typical in the first place because it had been recorded. The research subject of the article is the influence of sound recording technology on jazz culture at the stage of its foundation. In those years, if jazz musicians wanted to make a recording they had to bear in mind numerous peculiarities of sound recording technology. The author gives special attention to the analysis of the consequences of reproducibility of a recording for jazz musicians, and for the audience’s perception. As a research methodology, the author uses the comprehensive approach which includes the study of historical sources and jazz musicians’ memoirs related to the sound recording industry. The research proves that audio recordings are not sufficient as a source for critical research of the first jazz gramophone record, and suggests alternative approaches to its interpretation.

Dehydroalanine exists natively in certain proteins and can also be chemically made from the protein cysteine. As a strong Michael acceptor, dehydroalanine in proteins has been explored to undergo reactions with different thiolate reagents for making close analogues of post-translational modifications (PTMs), including a variety of lysine PTMs. The chemical reagent 2-nitro-5-thiocyanatobenzoic acid (NTCB) selectively modifies cysteine to form S-cyano-cysteine, in which the S–Cβ bond is highly polarized. We explored the labile nature of this bond for triggering E2 elimination to generate dehydroalanine. Our results indicated that when cysteine is at the flexible C-terminal end of a protein, the dehydroalanine formation is highly effective. We produced ubiquitin and ubiquitin-like proteins with a C-terminal dehydroalanine residue with high yields. When cysteine is located at an internal region of a protein, the efficiency of the reaction varies with mainly hydrolysis products observed. Dehydroalanine in proteins such as ubiquitin and ubiquitin-like proteins can serve as probes for studying pathways involving ubiquitin and ubiquitin-like proteins and it is also a starting point to generate proteins with many PTM analogues; therefore, we believe that this NTCB-triggered dehydroalanine formation method will find broad applications in studying ubiquitin and ubiquitin-like protein pathways and the functional annotation of many PTMs in proteins such as histones.

Aiming to explore the diverse nature of sound and image, thereby establishing a bridge with the symbiotic creation of sensations and emotions, this paper aims to present the development and construction of a proposal for the creation of site-specific performances, based on sound and visual articulation and intermedia practices.Using as focal points several places, the authors try to look beyond time and develop site-specific intermedia performances, attempting to understand the past and sketching new configurations for the (re)presentation of these places identity, guiding the audience through a journey of perceptual experiences, using ambient electronic music, soundscapes and videos.This presentation also aims to present a practice-as-research project, which had as its starting point the apprehension of memories and the history of these places, emphasizing concepts such as local context, site-specific, sense of place and identity.For this, the authors present the development of some experimental approaches and critical forms of expression and communication that relate or incorporate sound and visual mediums. They also bring forward some concerns about the aesthetic experience and communicative functionality, and how the relationship between art and technology has an effect on artistic culture and new contemporary site-specific intermedia performances.

One hundred years after the founding of the École Coloniale Supérieure in Antwerp, the adjacent Middelheim Museum invites Sandrine Colard, researcher and curator, to conceive an exhibition that probes silenced histories of colonialism in a site-specific way. For Colard, the term Congoville encompasses the tangible and intangible urban traces of the colony, not on the African continent but in 21st-century Belgium: a school building, a park, imperial myths, and citizens of African descent. In the exhibition and this adjoining publication, the concept Congoville is the starting point for 15 contemporary artists to address colonial history and ponder its aftereffects as black flâneurs walking through a postcolonial city. Due to the multitude of perspectives and voices, this book is both a catalogue and a reference work comprised of artistic and academic contributions. Together, the participating artists and invited authors unfold the blueprint of Congoville, an imaginary city that still subconsciously affects us, but also encourages us to envision a decolonial utopia.

T cells engineered with chimeric antigen receptors (CARs) show great promise in the treatment of some cancers. Modifying T cells to express CARs generally relies on T-cell transduction using viral vectors carrying a transgene, resulting in semi-random DNA integration within the T-cell genome. While this approach has proven successful and is used in generating the Food and Drug Administration (FDA, USA) approved B-lymphocyte antigen CD19-specific CAR T cells, it is possible the transgene could integrate into a locus that would lead to malignant transformation of the engineered T cells. In addition, manufacturing viral vectors is time-consuming and expensive. One way to overcome these challenges is site-specific gene integration, which can be achieved through clustered regularly interspaced short palindromic repeat (CRISPR) mediated editing and non-viral DNA, which serves as a template for homology-directed repair (HDR). This non-viral gene editing approach provides a rapid, highly specific, and inexpensive way to engineer T cells. Here, we describe an optimized protocol for the site-specific knock-in of a large transgene in primary human T cells using non-viral double stranded DNA as a repair template. As proof-of-principle, we targeted the T-cell receptor alpha constant (TRAC) locus for insertion of a large transgene containing green fluorescence protein (GFP) and interleukin-15 (IL-15). To optimize the knock-in conditions we tested template DNA concentration, homology arm length, cell number, and knock-in efficiency over time. We then applied these established guidelines to target the TRAC or interleukin-13 (IL-13) locus for the knock-in of synthetic molecules, such as a CAR, bispecific T-cell engager (BiTE), and other transgenes. While integration efficiency depends on the targeted gene locus and selected transgene, this optimized protocol reliably generates the desired insertion at rates upwards of 20%. Thus, it should serve as a good starting point for investigators who are interested in knocking in transgenes into specific loci.

Abstract. The physical structure of polar firn plays a key role in the mechanisms by which glaciers and ice sheets preserve a natural archive of past atmospheric composition. This study presents the first measurements of gas diffusivity and permeability along with microstructural information measured from the near-surface firn through the firn column to pore close-off. Both fine- and coarse-grained firn from Summit, Greenland are included in this study to investigate the variability in firn caused by seasonal and storm-event layering. Our measurements reveal that the porosity of firn (derived from density) is insufficient to describe the full profiles of diffusivity and permeability, particularly at porosity values above 0.5. Thus, even a model that could perfectly predict the density profile would be insufficient for application to issues involving gas transport. The measured diffusivity profile presented here is compared to two diffusivity profiles modeled from firn air measurements from Summit. Because of differences in scale and in firn processes between the true field situation, firn modeling, and laboratory measurements, the results follow a similar overall pattern but do not align; our results constitute a lower bound on diffusive transport. In comparing our measurements of both diffusivity and permeability to previous parameterizations from numerical 3-D lattice-Boltzmann modeling, it is evident that the previous relationships to porosity are likely site-specific. We present parameterizations relating diffusivity and permeability to porosity as a possible tool, though use of direct measurements would be far more accurate when feasible. The relationships between gas transport properties and microstructural properties are characterized and compared to existing relationships for general porous media, specifically the Katz–Thompson (KT), Kozeny–Carman (KC), and Archie's law approximations. While those approximations can capture the general trend of gas transport relationships, they result in high errors for individual samples and fail to fully describe firn variability, particularly the differences between coarse- and fine-grained firn. We present a direct power law relationship between permeability and gas diffusivity based on our co-located measurements; further research will indicate if this type of relationship is site-specific. This set of measurements and relationships contributes a unique starting point for future investigations in developing more physically based models of firn gas transport.

Lysine acetyltransferases (KATs) are exquisitely fine-tuned to target specific lysine residues on many proteins, including histones, with aberrant acetylation at distinct lysines implicated in different pathologies. However, researchers face a lack of molecular tools to probe the importance of site-specific acetylation events in vivo. Because of this, there can be a disconnect between the predicted in silico or in vitro effects of a drug and the actual observable in vivo response. We have previously reported on how an in vitro biochemical analysis of the site-specific effects of the compound C646 in combination with the KAT p300 can accurately predict changes in histone acetylation induced by the same compound in cells. Here, we build on this effort by further analyzing a number of reported p300 modulators, while also extending the analysis to correlate the effects of these drugs to developmental and phenotypical changes, utilizing cellular and zebrafish model systems. While this study demonstrates the utility of biochemical models as a starting point for predicting in vivo activity of multi-site targeting KATs, it also highlights the need for the development of new enzyme inhibitors that are more specific to the regulation of KAT activity in vivo.

Metallic clusters are interesting from various points of view, e.g. as a mean of spreading expensive catalysts on a support, or following heterogeneous and homogeneous catalytic events. It is also possible to study nucleation and growth mechanisms for crystals with the cluster as known starting point.Gold-clusters containing 55 atoms were manufactured by reducing (C6H5)3PAuCl with B2H6 in benzene. The chemical composition was found to be Au9.2[P(C6H5)3]2Cl. Molecular-weight determination by means of an ultracentrifuge gave the formula Au55[P(C6H5)3]Cl6 A model was proposed from Mössbauer spectra by Schmid et al. with cubic close-packing of the 55 gold atoms in a cubeoctahedron as shown in Fig 1. The cluster is almost completely isolated from the surroundings by the twelve triphenylphosphane groups situated in each corner, and the chlorine atoms on the centre of the 3x3 square surfaces. This gives four groups of gold atoms, depending on the different types of surrounding.
