What geckos are – an ecological-biogeographic perspective

Geckos are a hyper-diverse, ancient, and globally distributed group. They have diverged early from other squamates and thus can be expected to differ from them along multiple ecological, life history, and biogeographic axes. I review a wide range of gecko traits, comparing them to those of other lizard taxa, to identify the unique, and unifying, attributes of geckos among lizards, based on comprehensive databases of lizard distributions and biological attributes. Few traits completely separate geckos from other lizard taxa, yet they differ to a large degree along many axes: they are more restricted to low latitudes and altitudes, are especially diverse on islands, but relatively scarce in America. They are small lizards, that lay small, fixed clutch sizes, for which they compensate only partially by laying frequently. Because they mature at relatively similar ages and have similar lifespans to other lizards, geckos produce fewer offspring over a year, and over their lifetimes, perhaps implying that they enjoy higher survival rates. While being the only large lizard clade of predominantly nocturnal lizards a large proportion of species is active by day. Gecko body temperatures and preferred temperatures are lower than those of other lizards –even when they are compared to lizards with similar activity times. Worryingly, most geckos have small ranges that often reside completely outside of protected areas – much more frequently than in other reptile and vertebrate taxa.

Background:1,4-Diazepines are two nitrogen containing seven membered heterocyclic compounds and associated with a wide range of biological activities. Due to its medicinal importance, scientists are actively involved in the synthesis, reactions and biological evaluation of 1,4-diazepines since number of decades.Objective:The primary purpose of this review is to discuss the synthetic schemes and reactivity of 1,4- diazepines. This article also describes biological aspects of 1,4-diazepine derivatives, that can be usefully exploited for the pharmaceutical sector.Conclusion:This review summarizes the abundant literature on synthetic routes, chemical reactions and biological attributes of 1,4-diazepine derivatives. We concluded that 1,4-diazepines have significant importance due to their biological activities like antipsychotic, anxiolytic, anthelmintic, anticonvulsant, antibacterial, antifungal and anticancer. 1,4-diazepine derivatives with significant biological activities could be explored for potential use in the pharmaceutical industries.

PurposeThe treatment outcome of brentuximab vedotin (BV) has not been related with CD30 expression in previous studies enrolling patients with a wide range of CD30 expression level. Thus, this study explored the efficacy of BV in high-CD30–expressing non-Hodgkin lymphoma (NHL) patients most likely to benefit.Materials and MethodsThis phase II study (Clinicaltrials.gov: NCT02280785) enrolled relapsed or refractory high-CD30–expressing NHL, with BV administered intravenously at 1.8 mg/kg every 3 weeks. The primary endpoint was > 40% disease control rate, consisting of complete response (CR), partial response (PR), or stable disease. We defined high CD30 expression as ≥ 30% tumor cells positive for CD30 by immunohistochemistry.ResultsHigh-CD30-expressing NHL patients (n=33) were enrolled except anaplastic large cell lymphoma. The disease control rate was 48.5% (16/33) including six CR and six PR; six patients (4CR, 2PR) maintained their response over 16 completed cycles. Response to BV and survival were not associated with CD30 expression levels. Over a median of 29.2 months of follow-up, the median progression-free and overall survival rates were 1.9 months and 6.1 months, respectively. The most common adverse events were fever (39%), neutropenia (30%), fatigue (24%), and peripheral sensory neuropathy (27%). In a <i>post-hoc</i> analysis for the association of multiple myeloma oncogene 1 (MUM1) on treatment outcome, MUM1- negative patients showed a higher response (55.6%, 5/9) than MUM1-positive patients (13.3%, 2/15).ConclusionBV performance as a single agent was acceptable in terms of disease control rates and toxicity profiles, especially MUM1-negative patients.

ABSTRACTThe leaves ofTamarix aphylla, a globally distributed, salt-secreting desert tree, are dotted with alkaline droplets of high salinity. To successfully inhabit these organic carbon-rich droplets, bacteria need to be adapted to multiple stress factors, including high salinity, high alkalinity, high UV radiation, and periodic desiccation. To identify genes that are important for survival in this harsh habitat, microbial community DNA was extracted from the leaf surfaces of 10Tamarix aphyllatrees along a 350-km longitudinal gradient. Shotgun metagenomic sequencing, contig assembly, and binning yielded 17 genome bins, six of which were >80% complete. These genomic bins, representing three phyla (Proteobacteria,Bacteroidetes, andFirmicutes), were closely related to halophilic and alkaliphilic taxa isolated from aquatic and soil environments. Comparison of these genomic bins to the genomes of their closest relatives revealed functional traits characteristic of bacterial populations inhabiting theTamarixphyllosphere, independent of their taxonomic affiliation. These functions, most notably light-sensing genes, are postulated to represent important adaptations toward colonization of this habitat.IMPORTANCEPlant leaves are an extensive and diverse microbial habitat, forming the main interface between solar energy and the terrestrial biosphere. There are hundreds of thousands of plant species in the world, exhibiting a wide range of morphologies, leaf surface chemistries, and ecological ranges. In order to understand the core adaptations of microorganisms to this habitat, it is important to diversify the type of leaves that are studied. This study provides an analysis of the genomic content of the most abundant bacterial inhabitants of the globally distributed, salt-secreting desert treeTamarix aphylla. Draft genomes of these bacteria were assembled, using the culture-independent technique of assembly and binning of metagenomic data. Analysis of the genomes reveals traits that are important for survival in this habitat, most notably, light-sensing and light utilization genes.

1. Blood pressure was measured in the dorsal aorta of restrained, unanaesthetized tiger snakes (Notechis scutatus) at different body temperatures during graded, passive tilt. Aortic blood pressure in horizontal snakes showed no significant change over a range of body temperatures between 18 and 33 degrees C (mean of measurements on 16 snakes = 42.2 +/− I.98 mmHg), while heart rate increased logarithmically (Q10 approximately 2.5). Blood pressure was stable during heating and cooling between body temperatures of 15 and 30 degrees C, but the pressure was 10--50% higher during heating than during cooling. 2. Head-up tilt usually caused a brief fall in pressure at heart level followed by partial or complete recovery and tachycardia. At the cessation of tilt, there was a characteristic overshoot of the blood pressure followed by readjustment to control (pretilt) levels. Head-down tilt typically increased pressure which then either stabilized or returned toward pretilt levels. Heart rate changes during head-down tilt were not consistent in direction or magnitude. Stabilized pressures at mid-body usually increased following head-up tilt and decreased following head-down tilt, indicating physiological adjustment to posture change. Blood pressure control was evident at body temperatures ranging from 10 to 38 degrees C, but was most effective at the higher and behaviourally preferred temperatures. 3. Propranolol lowered heart rate but did not influence pressure in horizontal snakes. During head-up tilt propranolol eliminated or reduced tachycardia and sometimes reduced the efficacy of pressure compensation for tilt. Phentolamine increased heart rate, lowered blood pressure, and eliminated pressure regulation during tilt. The results suggest that sympathetically mediated reflexes assist central blood pressure regulation in the tiger snake, with vasomotor adjustments having greater importance than changes in heart rate.

Abstract Lepidium draba is found in a wide range of habitats ranging from roadsides (highly disturbed sites) to rangelands, meadows and pastures, cultivated fields, home gardens, national parks and wasteland. It exists at high and low latitudes, above and below sea level. It is found under irrigation and in soils of high moisture content and also exists in arid regions. It invades all soil types, although it prefers alkaline conditions and can germinate in saline soils. All of these characteristics and adaptations provide exceptional advantages for this noxious weed and reflect its high potential to invade different agricultural systems. The absence of L. draba plants from plantations or sites where it has not been reported may not be due to the species' ability to invade, survive, grow or develop in these sites but rather to the absence of initial infestation. The weed can invade both agricultural and natural ecosystems (Larson et al., 2000). In summary, L. draba is a serious threat to agriculture and the environment. It is of a high ecological tolerance and is potentially harmful to human and animal health. Its ability to invade and reproduce by different means and to host plant pathogenic agents make this weed invasive with significant problems to man and natural resources.

Hybridization between closely related species has been documented across a wide range of taxa but has not been well studied in elasmobranchs. Hammerhead sharks have drawn global conservation concern because they experience some of the highest mortality rates among sharks when interacting with fisheries. Here we report on the detection of hybrids between the globally distributed scalloped hammerhead ( Sphyrna lewini ) and recently described Carolina hammerhead ( S. gilberti ) which are only known from the western Atlantic Ocean. Using a genomics approach, 10 first-generation hybrids and 15–17 backcrosses were detected from 554 individuals. The identification of backcrosses demonstrates hybrids are viable, and all backcrosses but one involved a scalloped hammerhead. All hybrids but one possessed Carolina hammerhead mtDNA, indicating sex-biased gene flow between species. Repeated hybridization and backcrossing with scalloped hammerheads could lead to the loss of endemic Carolina hammerheads.

Shiga toxin (Stx)–producing Escherichia coli O157 isolates (n = 123) were divided into groups according to origin, genotype (pulsed-field gel electrophoresis [PFGE] type, or ribotype), type of Stx produced, or phage type (PT). The survival rate ([number of CFU after 24 h of drying/number of CFU before drying] × 100) for each isolate was determined in triplicate after drying on concrete for 24.0 h. The overall mean survival rate among the 123 E. coli O157 isolates studied was 22.9%, but there was a wide range of responses to drying on concrete, with a minimum of 1.2% and a maximum of 61.9% of the initial inocula being recovered after drying. Among the groups, those isolates that originated from cases of human disease were, on average, significantly more sensitive (P &lt; 0.001) to drying (with a mean survival rate of 15.3%) than isolates from the other three sources (with mean survival rates of 27.7, 26.0, and 22.9% for meats, bovine or ovine feces, and bovine hides, respectively). When the isolates were grouped by genotype, three of the PFGE types were, on average, significantly more resistant to drying than two other PFGE types were, and similarly, significant differences in average resistance to drying between groups of E. coli O157 with different ribotypes were seen. There were no differences between the abilities of isolates producing different Stxs (Stx 1 or Stx 1 and Stx 2) to survive drying. E. coli O157 isolates of PT4, PT21/28, and PT32 survived drying on concrete better than groups of other PTs did. Since the E. coli O157 isolates had various abilities to survive drying on concrete, drying could contribute to a kind of E. coli O157 natural selection along the meat chain. This possibility may have significant meat safety implications if a range of E. coli O157 isolates are simultaneously exposed to drying at any point along the meat production chain. Those E. coli O157 isolates that are more able to survive drying could be more likely to pass farther along the meat chain and ultimately reach consumers.

Purified porcine gastric mucin (PGM) is an alternative biomaterial to native mucin which displays multifunctional properties for exploring a wide range of biomedical applications. The present study evaluated the in vitro (RAW 264.7 macrophage cells) and in vivo (zebrafish embryos and larvae) bioactivities of PGM. The median lethal concentration (LC50) of PGM was 197.9 µg/mL for embryos, while it was non-toxic to RAW 264.7 cells, even at 500 µg/mL. Following PGM exposure (100 µg/mL), a higher embryo hatching rate (59.9%) was observed at 48 h post fertilization, compared to the control (30.6%). Protective effects of PGM from pathogenic Aeromonas hydrophila were demonstrated by high larvae survival rates of 85.0% and 94.0% at 50 and 100 μg/mL of PGM exposure, respectively. Heat tolerance effect of PGM (50 and 100 µg/mL) on larvae (40 °C for 48 h) was confirmed by 75% and 100% of survival rates, respectively. Additionally, PGM reduced the A. hydrophila–induced reactive oxygen species (ROS) generation in larvae. The qRT-PCR results in PGM exposed larvae exhibited induction of immune-related genes (tlr5a and tlr5b, myd88, c-rel, il1β, tnf-α, il6, il10, cxcl18b, ccl34a.4, defbl1, hamp, ctsd, muc2.1, muc5.1, muc5.2, and muc5.3), stress response (hsp70, hsp90aa1.1, and hsp90ab1), and antioxidant genes (cat and sod1). Moreover, our results revealed that PGM involved in the regulation of transcriptional gene induction increases Hsp90 protein in the zebrafish larvae. Furthermore, upregulation of Il6, Il10, Tnfα, Ccl3, Defa-rs2, Defa21 and Camp and antioxidant genes (Sod2 and Cat) were observed in PGM-exposed RAW 264.7 cells. Overall findings confirmed the activation of immune responses, disease resistance against pathogenic bacteria, heat tolerance, and ROS-scavenging properties by PGM, which may provide insights into new applications for PGM as a multifunctional immunomodulator.

Local climatic conditions influence the way in which ectotherms regulate their body temperature and activity. We examined correlations between local climatic conditions, body temperature and activity in adult, basking lowland copperheads (Austrelaps superbus) from two localities (warm-temperate versus cool-temperate) in south-eastern Australia. We also collected data from highland copperheads (Austrelaps ramsayi) at a locality with cold-temperate climate. We found that across the active season, mean body temperatures were similar among localities (approximately 27˚C) irrespective of species. In contrast, activity times differed. Cool-temperate A. superbus emerged earlier in spring and in the morning and retreated earlier in the evening and in autumn than their conspecifics from the warm-temperate locality. Spring emergence was correlated with yearly fluctuations in thermal conditions, suggesting that activity times depend on environmental temperatures. Predator–prey interactions influenced body temperature and activity to some extent in spring when warm-temperate A. superbus with relatively low body temperatures (as low as 18.5˚C) were captured around ponds in which they had been foraging for frogs. Austrelaps ramsayi from the cold-temperate locality not only displayed a later emergence in spring and reduced daily activity times compared with warm and cool-temperate A. superbus but also compared with A. ramsayi, as reported from a warmer locality in eastern Australia. These data indicate that activity times vary on a geographic basis while snake body temperatures largely remain inflexible. The surprising exception was that cold-temperate A. ramsayi retreated later in autumn than cool-temperate A. superbus, and at that time they showed body temperatures as low as 12.5˚C, well below those we had recorded for A. superbus. We suggest that A. ramsayi retreat later in autumn because they need to extend their reproductive season and that this is mediated via adaptive changes in the critical minimum body temperature, as has been reported for other snakes.

A field study of the thermal relationships of the small agamid lizard A. fordi has been carried out in two areas of mallee in central western New South Wales, where this lizard occurs only in close association with the grass Triodia scariosa. The body temperatures characteristic of various phases in this lizard's die1 cycle have been determined. The behavioural techniques employed to regulate temperature are described; they are similar to those used by a wide range of diurnal heliothermic lizards in other regions. The total effect of these thermoregulatory responses is to maintain an internal thermal environ- ment approaching homoiothermy while the lizard is active. For A. fordi the eccritic body temperature determined from animals in the field is 36.9+-0.16C. Lowering of activity thermal levels occurs in winter, and can be induced at any time by even mild starvation.
