Da-Cheng-Qi decoction (DCQD) is a purgative compound prescription used in China and East Asia. In this paper, pharmacokinetic differences of six major active components (rhein, emodin, aloe-emodin, magnolol, naringenin and hesperetin) between DCQD and its three constitutional herbal medicines i.e. Radix et Rhizoma Rhei, Cortex Magnoliae officinalis and Fructus Aurantii Immaturus were investigated in rats after oral administration. Plasma samples were analyzed for the quantification of the six active components using validated LC-MS/MS methods. Unpaired Student's t-test was used for statistical comparison. Significant differences ( p<0.05) in the main pharmacokinetic parameters for rhein, emodin, aloe-emodin, magnolol, naringenin and hesperetin were found between DCQD and the decoction of its constitutional single herbal medicines, which demonstrated the presence of drug-drug interactions between these constitutional raw materials of DCQD occurring either in the procedure of decoction or during ADME process.

Introduction:: Asari Radix et Rhizoma (ARR) and dried ginger (Zingiber officinalis) (DG) are often used together in drug preparations in traditional Chinese medicine (TCM) to treat respiratory diseases including cold, bronchitis and pneumonia. Previous studies suggested that ARR and/or DG may influence the pharmacokinetics of other herbal components. In the current study, we examined pharmacokinetic interactions between ARR and DG in rats after oral administration. Methods:: We developed a method based on ultra-high-performance liquid chromatography-tandem mass spectrometry to simultaneously measure serum concentrations of two active components each in ARR (L-asarinin and sesamin) and DG (6-gingerol and 6-shogaol). Adult Sprague-Dawley rats were starved overnight, then given ARR extract, DO extract, or a co-decoction of ARR and DG by gastric gavage (6 g raw material per kg body weight; n = 6 per group). Blood samples were collected prior to drug administration and at the following times (h) afterward: 0.5, 1.0, 1.5, 2.0, 3.0, 4.0, 6.0, 8.0, 12.0 and 24.0. Pharmacokinetic parameters were compared using Student’s t test for independent samples. Results:: A simple, rapid, sensitive analytical method has been developed to detect four bioactive components simultaneously in the ARR-DG herbal pair. Pharmacokinetic parameters including Cmax, Tmax, T1/2 and AUC(0~t) were calculated using the non-compartmental model with the DAS 2.0 pharmacokinetic software. For L-asarinin, Tmax was 2.00 ± 0.00 h in ARR animals and 1.67±0.26 h in ARR-DG animals (P<0.05), T1/2 was 8.58 ± 1.75 h in ARR and 11.93 ± 2.13 h in ARR-DG (P<0.05). For 6-gingerol, Cmax was 350.48 ± 23.85 ng/mL in DG animals and 300.21 ± 20.02 ng/mL in ARR-DG (P<0.01), Tmax was 2.83 ± 0.41 h in DG and 2.17 ± 0.41 h in ARR-DG (P<0.05) and AUC(0~t) was 1.93 ± 0.15 mg/mL•h in ARR and 1.70 ± 0.15 mg/mL•h in ARR-DG (P<0.05). For 6-shogaol, Cmax was 390.28 ± 26.02 ng/mL in DG animals and 455.63 ± 31.01 ng/mL in ARR-DG (P<0.01), Tmax was 2.93 ± 0.10 h in DG and 1.92 ± 0.10 h in ARR-DG (P<0.01), T1/2 was 3.74 ± 0.29 h in DG and 3.28 ± 0.22 h in ARR-DG (P<0.01), and AUC(0~t) was 2.15 ± 0.18 mg/mL•h in DG and 2.73 ± 0.15 mg/mL•h in ARR-DG (P<0.01). Conclusions:: Pharmacokinetic interations between ARR and DG decrease Tmax, increase T1/2 but do not affect overall bioavailability of L-asarinin in ARR. The interactions in ARR-DG decrease Cmax and Tmax but increase T1/2 and AUC(0~t) of 6-gingerol in DG. The interactions increase Cmax and AUC(0~t) but decrease Tmax and T1/2 of 6- shogaol in DG. Interactions in ARR-DG do not affect the pharmacokinetics of sesamin.

Drs. Giangiacomo and Gleason are quite correct in their criticism that the number of patients with vitamin D deficiency rickets is too small for statistical comparison of the concentration of serum 25-hydroxyvitamin D (25-OH-D) in mild, moderate, and severe rickets. For this reason, we did not attempt this type of statistical comparison (Student's t test). The statistical analysis which supports our conclusion is the calculation of the regression between senim 25-OH-D, ng/ml and the stage of the disease, designated by arbitrary numbers 1, 2, and 3 for mild, moderate, and severe.

Aim of the study: Citrulline is a recognized marker of small bowel mass and function. The aim of this study was to identify plasma citrulline concentration that permitted adult patients to be partially or completely weaned off total parenteral nutrtion (TPN).<br>Materials and Methods: We measured plasma citrulline in 29 adult TPN patients in Warsaw hospital in 2014 and 2015. We evaluated how many of those patients were weaned from TPN or whether there was a decrease in the frequency of parenteral nutrition (PN) over the following year. Citrulline was assayed in deproteinised plasma samples using HPLC with fluorescence detection after derivatization in a timed reaction with o-phtalaldehyde and mercaptoethanol as described by Kilpatric. The group of weaned patients were compared to the TPN-dependent using Student’s t-test.<br>Results: Citrulline levels among the 29 patients ranged from 6.1 to 44.9 μmol/L (average 19.9 μmol/L). Six patients were completely weaned off PN over one year. Their citrulline levels were between 19,5 and 44.9 (average 28,7) μmol/L and were the highest measured in the patient population. Four patients with citrulline levels between 19.0 and 30.6 (average 23.8) μmol/L) had PN 5 days each week. The 19 patients who were totally dependent on PN had citrulline levels between 6.1 and 18.8 (average 15.3) μmol/L. There was a statistically significant difference between weaned and dependent on parenteral nutrition patients (p=0.0001).<br>Conclusions: Patients receiving TPN who have a high blood citrulline level may be able to be weaned off PN. This may reflect better intestinal mass and function. Citrulline levels may be a good predictor for weaning patients off PN or diminishing the frequency of PN.

The effects of cytokines on the pharmacokinetics of nucleoside analogs were evaluated in two separate studies using zidovudine in combination with interleukin-2 and didanosine in combination with alpha interferon. In each study, drug interactions were evaluated by using both a standard method (Student's t test) and bioequivalence testing. Serial blood samples were collected from human immunodeficiency virus-infected patients prior to and during cytokine therapy for determination of nucleoside analog concentrations. Concentrations were fit separately to a two-compartment model by using the iterative two-stage approach to population analysis. No alterations in area under the curve or oral clearance were observed for either drug during combination therapy. In general, there was good agreement between statistical methods for determining if antiviral pharmacokinetic parameters were altered by concomitant cytokine therapy. However, large individual changes in the maximum concentration of zidovudine in serum were detected by bioequivalence testing but no difference was found by Student's t test. For didanosine, significant but clinically irrelevant decreases determined by standard hypothesis testing were seen for both the volume of the central compartment (1.91 to 1.86 liters) and the absorption rate constant (0.79 to 0.73 h-1) in the presence of alpha interferon. No interaction was noted for these parameters by using bioequivalence guidelines. Bioequivalence testing may provide an alternative approach to assessment of drug interactions. Interleukin-2 and alpha interferon do not alter the pharmacokinetics of zidovudine and didanosine, respectively.

OBJECTIVE: The aim of this study was to assess the position of the cranial base, maxilla, and mandible of Brazilian achondroplastic adult subjects through cephalometric measurements of the cranio-dento-facial complex, and to compare the results to normal patterns established in literature. METHODS: Fourteen achondroplastic adult subjects were evaluated based on their radiographic cephalometric measurements, which were obtained using the tracings proposed by Downs, Steinner, Bjork, Ricketts and McNamara. Statistical comparison of the means was performed with Student's t test. RESULTS: When compared to normal patterns, the cranial base presented a smaller size in both its anterior and posterior portions, the cranial base angle was acute and there was an anterior projection of the porion; the maxilla was found to be smaller in size in both the anteroposterior and transversal directions, it was inclined anteriorly with anterior vertical excess, and retropositioned in relation to the cranial base and to the mandible; the mandible presented a normal-sized ramus, a decreased body and transverse dimension, a tendency towards vertical growth and clockwise rotation, and it was slightly protruded in relation to the cranial base and maxilla. CONCLUSION: Although we observed wide individual variation in some parameters, it was possible to identify significant differences responsible for the phenotypical characteristics of achondroplastic patients.

Biodiversitas 22: 1535-1542. Ensuring consistency of quality, safety, and efficacy of herbal medicines from raw materials to finished products is important because of the variability in medicinal plants' active components. One of the plants that have been used as an antidiabetic herbal medicine is A. paniculata. This study aims to determine α-glucosidase inhibitory activity, the content of several metals, FTIR spectrum profile, and putative identification of A. paniculata metabolites using UHPLC-Q-Orbitrap HRMS from different geographical origin. We found that ethanol extract of A. paniculata gave higher inhibition of α-glucosidase activity compared to water extract. Eight metals were determined using FAAS and FAES, and calcium showed the highest content in all A. paniculata samples. FTIR spectra of A. paniculata showed a similar profile and only differed in the absorbance. We detected the presence of OH, C=O, C=C aromatic, and C-O functional groups in A. paniculata extract. About 32 metabolites were putatively identified in A. paniculata, mainly from phenolic and diterpene lactones class compounds based on UHPLC-Q-Orbitrap HRMS. Using a combination of FTIR spectra and peak area from 32 identified peaks, we can clustered A. paniculata based on its geographical origin. Based on the result obtained, the geographical origin of A. paniculata affected the metals and metabolites composition and level, resulting in different levels of α-glucosidase inhibitory activity.

Two quantitative analytical methods for the determination of nitrofurantoin in pharmaceutical preparations in the visible-ultraviolet range of the spectrum are reported. One of them uses the diffuse reflectance technique and the other transmittance measurements in aqueous solution. Both procedures are very simple to be performed and also present green characteristics as only polyethylene glycol and water are used as solvents. Commercial pharmaceutical preparations of the drug were analyzed using the proposed methods. The results were compared with those obtained when the HPLC procedure recommended by the USP was applied. Statistical comparison using the paired Student’s t test and the Snedecor’s F test showed good agreement between all the methods.

Agroecology is the science that establishes the basis for construction of sustainable farming styles. The consortium usually to stand out as alternative interesting small for producers with structural and financial limitations, because it is an effective cropping system practiced long ago and found in all parts of the world, with greater diversity in the tropics. The experiment was conducted between August to November 2012 in the municipality of Garanhuns-PE/Brazil It had as objective to observe how the lettuce would behave in consortium with the carrot in an agroecological system of production. The parameters evaluated in the experiment were plant height, total weight of the plant, weight of leaves and number of leaves. The data obtained were submitted to a statistical comparison test between averages of the consortium and non-consortium beds, using Student's t-test at a significance level of 5%. It was observed that the consortium between lettuce and carrot presented a significant difference for most of the studied variables, negatively influencing its growth.

Background: Quality control and the standardization of herbal medicines involve steps like standard source and quality of raw materials, good manufacturing practices and adequate analytical screening. These practices play a vital role in guaranting the quality and stability of herbal preparations. Chandrashakaladi Vataka is an Ayurvedic herbal formulation mentioned to be beneficial in Kushtha. Till date no published data is available on its analytical profile. Aim: To develop the Pharmacognostical and Phyto-chemical profile of Chandrashakaladi Vataka. Material and Methods: Chandrashakaladi Vataka was prepared as per classical method and analytical findings were recorded. Samples were subjected to organoleptic analysis, physico-chemical analysis and HPTLC examination by optimizing the solvent systems. Results and Conclusions: Pharmacognostical profile of Chandrashakaladi Vataka was established. Loss on drying, Ash value, Acid insoluble extract, Methanol soluble extract, Chandrashakaladi Vataka was found within prescribed limits. HPTLC profile of Chandrashakaladi Vataka revealed 12 spots at 254 nm and 13 spots at 366 nm.
