Abstract Background Different parts of the Parkia biglobosa plant are employed in traditional medicine in different African communities. However, information ratifying its use and biochemical influence on health is still scanty in literature. Thus, the present study assessed the influence of the ethyl acetate fraction of Parkia biglobosa leaves (EAFPB) on some biochemical parameters of sub-chronic fructose-loaded rats. Methodology Twenty-five Wistar rats were randomized into five groups (n = 5). The normal control group was maintained on normal diet only while the high fructose solution (HFS) control (placebo), reference and treatment groups received high fructose solution (3 g/kg/d b.w of fructose) for 30 days before treatment. Based on pilot study, two doses (100 and 200 mg/kg/d b.w) of EAFPB were selected and were administered to two groups of test animals while the reference group received 300 mg/kg/d b.w. of metformin for 14 days. Thereafter, blood was collected from fasted animals for biochemical analyses for the examination of level of glycated hemoglobin (HbA1c), liver status (alanine and aspartate aminotransferases (ALT and AST) and alkaline phosphatase (ALP) activities, and bilirubin level), lipid profile (total cholesterol, triglyceride, and low- and high-density lipoproteins levels) and lipid peroxidation (malondialdehyde – MDA level). Results EAFPB was shown to have a good DPPH radical scavenging activity (EC50 = 0.395 mg/ml). Chromatographic analysis of EAFPB revealed 28 known flavonoids (mainly kaempferol (21.31 mg/100 g), quercetin (12.84 mg/100 g), and luteolin (6.75 mg/100 g)), four hydrocinnamic acids derivatives (mainly P-coumaric acid (6.73 mg/100 g)), and 11 phenolic acids derivatives (mainly chlorogenic acid (48.18 mg/100 g) and protocatechuic acid (21.58 mg/100 g)). Relative to normal control, it was observed that fructose overload significantly increased serum activities of ALP, ALT, and AST, and levels of MDA, total cholesterol, low density lipoprotein and triglyceride in placebo. However, EAFPB significantly tapered the elevated serum activities of ALP, ALT, and AST. In addition, relative to placebo, the increased levels of HbA1c, MDA, and lipid health markers were also rebated by EAFPB. Conclusions Ethyl acetate fraction of Parkia biglobosa leaves attenuates biochemical aberrations in fructose-loaded rats, an effect attributable to the rich store of polyphenolic compounds in the fraction. Objectives: To evaluate the effect of extract and its fraction (n-hexane, ethyl acetate, and water) of the mulberry leaf (Morus australis Poir.) in reducing triglyceride and total cholesterol levels on HFD and PTU induced rats. Methods: In this study, mulberry leaves were collected from East Nusa Tenggara-Indonesia, extracted using maceration method, and fractionated with various solvents. The thin layer chromatography with silica gel 60 F254 was utilized to identify the active compound on both extract and fraction. Furthermore, using a quasi-experimental with control group design, this study employed 40 healthy male Wistar rats and divided into 8 groups, namely: normal and negative control, positive controls (simvastatin and gemfibrozil), ethanolic extract, and fractions (n-hexane, ethyl acetate, and water). All groups were fed with a high cholesterol diet and 0.01% PTU, except for the normal group. During the study, the blood was drawn at day 0, 28, 35, and 42. The triglyceride level was observed using Glycerophosphate-Oxidase Phenol Aminoantipyrine (GPO-PAP) method, while the total cholesterol was measured by Cholesterol Oxidase Phenol Aminoantipyrine (CHOD-PAP). Results: On the 42nd day, ethyl fraction of mulberry leaf (Morus australis Poir.) showed the most significant result of reducing the level of triglyceride and total cholesterol compared to the high-fat diet and PTU induced rat on day 0: triglyceride of 104.89±2.70 mg/dl vs 64.76±1.97 mg/dl and total cholesterol of 96.70±2.45 vs 88.02±1.38 mg/dl. Ethyl acetate fraction was similar to simvastatin in lowering the level of total cholesterol. TLC identification also showed that mulberry leaf contains flavonoids and polyphenols that act as anti-hyperlipidemic. Conclusion: Ethyl acetate fraction of mulberry leaf (Morus australis Poir.) showed best activity on lowering both triglyceride and total cholesterol.

Objective: The objective of this research was to evaluate reduction of blood cholesterol level by fraction extracts of binahong (Anredera cordifolia) on male Wistar rats. Methods: The rats were given high fat food every day for 3 months and pure cholesterol on week 0, 3, 7 and 11.  After that each binahong fraction (n-hexane fraction, ethyl acetate fraction and water fraction) with doses of 50 mg/kg bw, 100 mg/kg bw, 200 mg/kg bw were given orally for 3 weeks and observed for lipid profile on day 21.   Results: The result showed that n-hexane, ethyl acetate and water fractions of binahong leaves decreased total cholesterol, triglyceride and LDL level, and did not influence the HDL level. Meanwhile statin decreased total cholesterol, triglyceride and LDL level, and increased HDL level. Conclusion: Ethyl acetate fraction of binahong leaves significantly reduced total cholesterol level, triglycerides, LDL level compared to negative control group (p< 0.05) and had no influence in HDL level. Keywords: Atherosclerosis, Anredera cordifolia, cholesterol

The genus Minthostachys belonging to the Lamiaceae family, and is an important South American mint genus used commonly in folk medicine as an aroma in cooking. The phytochemical-rich samples of the aerial parts of Minthostachys diffusa Epling. were tested for pharmacological and health-promoting bioactivities using in vitro chemical and enzymatic assays. A range of radical scavenging activities of the samples against biological radicals such as nitric oxide and superoxide anion and against synthetic 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radicals, the ferric reducing antioxidant power and the lipid peroxidation inhibition were determined and ranked using the ‘relative antioxidant capacity index’ (RACI). The ethyl acetate fraction showed the highest RACI of +1.12. Analysis of the various fractions’ inhibitory ability against enzymes involved in diabetes (α-amylase and α-glucosidase), and against enzymes associated with Parkinson’s or Alzheimer’s diseases (acetylcholinesterase and butyrylcholinesterase) also suggested that the ethyl acetate fraction was the most active. Liquid chromatography–tandem mass spectrometry analysis of the ethyl acetate fraction showed more than 30 polyphenolic compounds, including triterpenes. The inhibitory cholinesterase effects of the triterpenes identified from M. diffusa were further analysed by in silico docking of these compounds into 3D-structures of acetylcholinesterase and butyrylcholinesterase. This is the first study on pharmacological activities and phytochemical profiling of the aerial parts of M. diffusa, showing that this plant, normally used as food in South America, is also rich in health-promoting phytochemicals.

Using natural products as antioxidant agents has been beneficial to replace synthetic products. Efforts have been made to profile the antioxidant capacities of natural resources, such as medicinal plants. The polyphenol content of Himalayan rhubarb, Rheum emodi wall, was measured and the antioxidant activity was determined using DPPH and ABTS+ assay, and the oxidative stress was assessed using SOD enzymatic assay. Five different solvent fractions, n-hexane, n-butanol, ethyl acetate, dichloromethane, and water, were used for screening the antioxidant capacity in effort to determine the optimum extraction solvent. The total phenolic contents for R. emodi fractions ranged from 27.76 to 209.21 mg of gallic acid equivalents (GAE)/g of dry weight. DPPH and ABTS+ assay results are presented into IC50 values, ranged from 21.52 to 2448.79 μg/mL and 90.25 to 1718.05 μg/mL, respectively. The ethyl acetate fraction had the highest antioxidant activity among other fractions. Also, n-butanol and water fractions showed significantly lower IC50 values than the positive control in DPPH radical scavenging activity. The IC50 values of SOD assay of fractions ranged from 2.31 to 64.78 μg/mL. A similar result was observed with ethyl acetate fraction showing the highest SOD radical scavenging activity. The study suggests that the ethyl acetate fraction of R. emodi possess the strongest antioxidant activity, thus the most efficient in extracting antioxidant contents. Moreover, a highly significant correlation was shown between total polyphenol content and antioxidant activity screening assays. The compounds related to the antioxidant activity of R. emodi were identified to myricitrin, myricetin 3-galloyl rhamnoside, and myricetin, which have not been reported in studies about R. emodi before.

ABSTRACT Background: Recent studies showed that Interleukin-8 (IL-8), activated cytokine immune response which plays an important role in the development of acute and chronic gastritis. Harmless anti-inflammatory therapeutic alternatives have been proposed, for example, the consumption of Sechium Edule Jacq. Swartz (chayote). Antioxidant (flavonoid) and cell regeneration (alkaloid) agents were found in chayote. This study aimed to determine the effect of chayote Sechium Edule Jacq. Swartz extracts on the level of IL-8 in Wistar rats with aspirin- induced gastritis. Subjects and Method: This was a randomized controlled trial (RCT) conducted at the laboratory of Mathematics and Natural Science, Universitas Sumatra Utara from January to February 2020. A total of 35 male Wistar rats was selected for this study and randomly allocated into 7 groups: (1) Negative control; (2) Positive control; (3) 100 mg/ kg BW chayote ethanol extract ; (4) 200 mg/kg BW chayote ethanol extract; (5) 100 mg/ kg BW chayote ethyl acetate fraction; (6) 200 mg/kg BW chayote ethyl acetate fraction; and (7) 20 mg omeprazole. The rats in positive control and treatment groups were induced with aspirin (200mg/ kg BW). The negative control group received no intervention. The dependent variable was level of IL-8 measured by ELISA. The independent variables were treatment status. The data were analyzed by One Way Anova and post hoc test. Results: The mean differences of IL-8 level were not statistically significant between study groups (p= 0.327). Mean of IL-8 level was higher in positive control group (Mean= 160.80; SD= 6.90) than in negative control group (Mean= 141.20; SD= 10.98). The lowest IL-8 level was in 100mg/ kg BW chayote ethanol extract group (Mean= 149.94; SD= 40.4), followed by 200mg/ kg BW (Mean= 152.4; SD= 30.73) and 100mg/ kg BW (Mean= 164.60; SD= 25.04) chayote ethyl acetate fraction groups, 20 mg omeprazole group (Mean= 170.60; SD= 21.58), and 200 mg/ kg BW chayote ethanol extract group (Mean= 176.80; SD= 10.98). Conclusion: The low dose (100mg/ kg BW) chayote ethanol extract has the most potential antiinflammation effect on in vitro gastritis with the lowest IL-8 level of all doses of chayote ethanol extract, chayote ethyl acetate fraction, and omeprazole. Background: The prevalence of diabetes mellitus is increasing on a global trend. The aim of the present study is to identify the most effective antioxidants and hypoglycemic fraction of Azanza garckeana. Methods: The fractions (nhexane or ethyl-acetate or aqueous) of A. garckeana were administered to the alloxan-induced diabetic rats at doses of 100, 200, and 400 mg/kg for 15 days. Antioxidants activities were evaluated at concentrations of 62.5, 125, 250, and 500 µg/mL using the DPPH scavenging assay. Results: Results revealed that both the hexane, ethyl-acetate, and aqueous fractions exhibited hypoglycemic and antioxidants activities in a dose-dependent manner. The n-hexane fraction demonstrated highest percentage DPPH scavenging effect of 26.34±3.43, 38.44±4.35, 59.34±3.45, and 74.83±5.35 at 62.5, 125, 250, and 500 µg/mL respectively. The ethyl-acetate fraction demonstrated 19.33±2.98, 28.94±3.24, 47.34±2.90, and 57.82±4.54 respectively while the aqueous fraction exhibited the least activities of 12.45±23.45, 18.64±2.94, 27.94±3.89, and 39.43±3.89 at concentrations of 62.5, 125, 250, and 500 µg/mL respectively. In addition, the n-hexane fraction demonstrated the most significant hypoglycemic effect with the highest glucose reduction of 58.97 ±3.45 %, 63.86±5.35 %, and 66.51±4.35 %, ethyl acetate fraction demonstrated glucose reduction of 7.55±0.54%, 21.77±2.35 %, and 29.56±3.46 % while the aqueous fraction demonstrated the least hypoglycemic effect of 9.89±2.67 %, 18.09±3.45 %, and 18.87±3.24 at 100, 200 and 400 mg/kg bw respectively. Conclusion: The n-hexane fraction of Azanza garckeana extract could serve as a reservoir of bioactive agents that could be useful for the development of a new anti-diabetic agent

Background:  Rotator cuff tear are one of most common orthopedic musculoskeletal problem gaining importance due to large health scale expenditure. Causes of rotator cuff tears are multifactorial and unclear.  Previous studies have suggested relation between elevated serum lipid profile and rotator cuff tear in western population. We therefore undertook study in our Indian population to correlate association of hypercholesterolemia with rotator cuff tear.Objective: To find association of rotator cuff pathology with hypercholesterolemia in Indian patients.Materials and methods: After obtaining clearance from institutional ethics committee. We prospectively collected fasting lipid samples of population who came to our hospitals with complaints of shoulder pain during our study period. 50 patients had rupture of rotator cuff which was confirmed by ultrasound of involved shoulder. 50 were seen for non-cuff related complaints.  We followed strict inclusion and exclusion criteria.Results: Total cholesterol, Triglycerides and low density lipoprotein concentration of patients with rotator cuff tendon tear were on higher side than control group. High density lipoprotein trend showed being lower than control group.  21 of 50 (42%) had high cholesterol (total cholesterol greater than 240mg/dl) than compared to 18 of 50 (36%) in control group. however P value=0.539 and showed no statistical significance between groups.Conclusion: In Indian population there was no correlation between hypercholesterolemia and rotator cuff tear which is statistically significant.Key words Rotator cuff tear, Hypercholesterolemia, Indian population.

Background: Binjai (Mangifera caesia) is a herb derived from South Kalimantan possessing antioxidant properties which promote wound healing inhibiting oxidation radicals. The natural antioxidants present in binjai leaves can be extracted by fractionation. Purpose: This study aimed to analyze the antioxidant activity of ethyl acetate fraction in 96% ethanol extract of binjai leaf. Methods: The study constituted a pure experimental study incorporating a post-test design with only random sampling technique consisting of two groups, namely; an ethyl acetate fraction as the treatment group and ascorbic acid as the positive control group. The leaves were treated in accordance with the soxhlet method and subsequently fractionated to extract ethyl acetate fraction. This was used to measure antioxidant activity with DPPH radical damping method using a UV-Vis spectrophotometer. A linear regression calculation was performed with a standard curve to quantify the IC50 value, before the ethyl acetate fraction underwent a qualitative test of secondary metabolite. Results: An independent t-test indicated significant differences between groups, an average value of IC50 in ascorbic acid of 13.812 ppm with 0.996 linearity and a fraction of ethyl acetate 38.526 ppm with a linearity of 0.999. In contrast, at this linearity value ascorbic acid and ethyl fraction acetate demonstrate a very high linear connection between concentration and inhibition. A secondary metabolite test conducted on the ethyl acetate fraction produced positive results for flavonoid, tannins, and phenol. Conclusion: Based on the IC50 parameters, the fraction of ethyl acetate in 96% ethanol extract of binjai leaf produces very strong antioxidant activity in the content of the compounds in the fraction, namely: flavonoid, tannins and phenol.

ABSTRACT Allophylus edulis and Cupania vernalis (Sapindaceae) are Brazilian native trees used as medicinal plants for the treatment of respiratory, digestive, circulatory, and skin diseases. Ubiquitously distributed in the Brazilian territory, these species are indicated for mixed plantations aimed at the recovery of degraded ecosystems. In this study, the total phenolic content (TPC) and total flavonoid content (TFC), and the antioxidant activity of extracts and fractions obtained from A. edulis and C. vernalis leaves were assessed. The TPC and TFC was determined spectrophotometrically. Antioxidant activity was evaluated through radical scavenging activity of 2,2-diphenyl-1-picrylhydrazyl (DPPH). The extracts were obtained by two methods: maceration (method 1) and Soxhlet (method 2). Solvents of increasing polarity (hexane, dichloromethane, ethyl acetate, and n-butanol) were used to obtained the fractions. The results showed that the ethyl acetate fraction from A. edulis, obtained from the maceration method, had the highest TPC (442.0 ± 18.2 mg GAE g-1) and TFC (58.1 ± 0.4 mg RUE g-1), and antioxidant activity (EC50 = 43.6 ± 2.6 µg mL-1). By C. vernalis, superior results were obtained with the n-butanol fraction (TPC = 126.1 ± 5.8 mg GAE g-1, TFC = 37.7 ± 0.6 mg RUE g-1). The highest antioxidant potential was found in the crude hydroalcoholic extract (EC50 = 816.1 ± 50.9 µg mL-1) and butanol fraction (1,156.4 ± 3.8 µg mL-1). The results of this study show that the fractions obtained by maceration and liquid-liquid partition with more polar solvents (ethyl acetate and n-butanol) are the richest in TPC and TFC, and presented the greater antioxidant activity. Comparing the two plants, A. edulis showed the best results, with a high content of TPC, TFC, and antioxidant potential, and therefore may be used to treat diseases related to oxidative stress.
