The efficiency of two different laparoscopic techniques for embryo collection was examined. Two groups (A and B) of 12 Chios breed ewes were used as donors and 20 crossbred Chios ewes as recipients. Oestrus was synchronised by intravaginal sponges, the donors were supervoulated by 8,8 mg of ovine FSH and laparoscopic intrauterine artificial insemination was conducted with extended fresh ram semen. Embryo collection was carried out the 6th day after AI. The uteri of the animals of group A were flushed after catheterisation of the ipsilateral oviduct. In the animals of group Β a flushing catheter was inserted close to the tip of the horn. Fifty nine and 46 embryos were collected from groups A and Β corresponding to recovery rate of 51.75% and 39% respectively (P>0.05). Laparoscopic transfer of 43 fresh embryos resulted to 11 (55%) pregnancies with (39.5%) surviving fetuses.

Abstract This study compared the semen quality traits between Etawah grade (EGB) and Senduro bucks (SB) raised in Singosari National Artificial Insemination Center, Indonesia. A total of 96 ejaculated semen were collected from three EGB and three SB with an average age of three years. The ejaculates were evaluated for semen volume (SV), spermatozoa concentration (SC), total spermatozoa (TS), spermatozoa motility (SM), post-thawing spermatozoa motility (PTSM), the recovery rate of spermatozoa motility (RRSM), and frozen semen production (FSP). Data of SV, SM, PTSM, and RRSM were compared using the Mann-Whitney U test. While the data of SC, TS, and FSP were analyzed using an independent t-test. The results showed that SV, SM, PTSM, and RRSM were not significantly differed (P>0.05) between breeds. Whereas, SC, TS, and FSP of EGB were significantly higher (P<0.05) than SB. Therefore, it can be concluded that Etawah grade bucks can produce higher total spermatozoa and as a result higher frozen semen production for artificial insemination as compared to Senduro bucks.

The aim of the present study was to evaluate the effects of uterine double flushing on embryo recovery rates (total structures and viable embryos), after a resting period with the uterus filled with PBS. There were 210 embryo recovery procedures conducted using the uterine double flushing method, and the results were compaired with 432 conventional single flushing procedures. All procedures were conducted with Limousin (n=403) and Guzerá (n=239) cows, following the same superovulation protocol. Cyclic donors received a progestagen implant (CIDR) and 1mL of oestradiol benzoate (Estrogin®) at Day 0. Between Day 5 and Day 9 animals received doses ranging from 200 to 300UI (zebu cows) and 300 to 500UI (taurine cows) of FSH (Pluset® – Serono) in decreasing doses. Between Day 6 and Day 8 PGF2a (Ciosin®) was administered, followed by withdrawal of the progestagen after 24h. Artificial insemination was performed between 14 and 26h after the beginning of treatment. For the double flushing procedure, after the first uterine flushing of both horns with 1L DPBS (Nutricell), a Foley catheter was positioned in the uterine body in order to fill the uterus with the same solution (80 to 150mL). After this procedure the catheter was closed with a disposable 5-mL syringe gasket, and the animals were allowed to rest in the surroundings of the work place for 30min. After this period, a second flush was performed in order to recover the remaning liquid used during manipulation of the uterus. Animals from the control group (group A) were subjected to a single uterine flushing procedure. From 210 double flushing procedures (group B – test), 1409 viable embryos were produced, classified as grades I, II, III and IV (IETS), (average of 6.7 embryos per procedure), whereas, in the 432 single flushing procedure (group A – control), 1993 embryos were produced (average of 4.6). Statistical analysis showed the increase of viable embryo recovery rate. When consecutive double flushing was performed, the average of recovered embryos increased from 8.3 to 12.7 (P&lt;0.05) in Limousin cows, and from 7.9 to 11.5 (P&lt;0.05) in Guzerá cows. Comparing recovery after single flushing with that after double flushing, the mean number of viable embryos increased from 4.7 to 6.9 (P&lt;0.05) in Limousin cows and from 4.5 to 6.4 (P&lt;0.05) in Guzerá cows. In order to assure the nonexistence of negligence effects or operator influence on results, the mean values of total embryo recovery rate after single flushing (control group) was compared to the mean values of the same rate after the first uterine flushing on test group. Results indicated no difference between recovery rates. The present work showed the viability of using the uterine double flushing procedure for improvement of embryo recovery rates in cattle.

Abstract The effects of supraphysiological estradiol (E2) on neonatal outcomes and the significance of specific E2 concentrations remain unclear. The purpose of this study was investigate whether supraphysiological E2 levels on the hCG trigger day are associated with small size for gestational age (SGA) in singletons born from fresh embryo transfer (ET) cycles. Patients with singleton pregnancies with delivered after transfer of fresh embryos, during the period of July 2012 to December 2017 at our center were included. We excluded cycles involving a vanishing twin, maternal age >35 years, basal FSH ≥10 mIU/ml, AMH ≤1 ng/ml or incomplete records. We then divided all cycles into 5 groups by E2 level on the day of hCG trigger: group A, <2000 pg/ml (reference group); group B, 2000 pg/ml≤E2<2999 pg/ml; group C, 3000 pg/ml≤E2<3999 pg/ml; group D, 4000 pg/ml≤E2<4999 pg/ml; and group E, ≥5000 pg/ml. The prevalence of SGA among singletons from fresh ET was the primary outcome. The SGA rate significantly increased when the E2 level was ≥4000 pg/ml, as observed by comparing groups D (odds ratio [OR]: 1.79, 95% confidence interval [CI]: 1.16–2.76, P=0.01) and E (OR: 1.68, 95% CI: 1.10–2.56, P=0.02) with the reference group. Multivariate logistic regression indicated that a serum E2 level of at least 4000 pg/ml on the hCG trigger day was associated with increased SGA and with significant differences for groups D (adjusted OR [AOR]: 1.65, 95% CI: 1.05–2.59, P=0.03) and E (AOR: 1.60, 95% CI: 1.03–2.53, P=0.04) relative to the reference group. In conclusion,for fresh ET cycles, supraphysiological E2 ≥4000 pg/ml on the hCG trigger day increases the risk of singleton SGA.

Objective This study was performed to investigate the effect of microscopically assisted decompression using a micro-hook scalpel on ossification of the posterior longitudinal ligament (OPLL). Methods Sixty-one patients with OPLL were divided into Group A (posterior surgery with laminectomy of the responsible segment and lateral mass screw fixation) and Group B (anterior cervical corpectomy with intervertebral titanium cage fusion). Neurological function was assessed by the Japanese Orthopaedic Association (JOA) score, visual analog scale (VAS) score, and recovery rate. The fixation status and the result of spinal canal decompression were radiographically assessed. Results In Groups A and B, the JOA score was significantly higher and the VAS score was significantly lower at 1 week postoperatively and at the final follow-up than during the preoperative period. The mean recovery rate in Group A and B was 59.92% ± 13.46% and 62.28% ± 14.00%, respectively. Postoperative radiographs showed good positioning and no damage to the internal fixation materials. The spinal canal was also fully decompressed. Conclusions Microscopically assisted decompression with a micro-hook scalpel in both anterior and posterior surgeries achieved good clinical effects in patients with OPLL.

Abstract Reproductive efficiency is critical for profitability, and age at weaning critically impacts weaning weight. The objective was to evaluate estrous synchronization and/or AI with conventional (CON) or gender-skewed (SEXED) semen on calving distribution. Beef females (n=1,620) were either 1) synchronized (7-CIDR:Syn) or not synchronized (Nonsyn) and mated to bulls, 2) synchronized (7-d CIDR) and mated to bulls (SynNS) or artificially inseminated (SynAI), or 3) synchronized (7-d CO-Synch plus CIDR) and AIed with CON or SEXED semen. Calving distribution and gender were determined at birth and were analyzed using the GLIMMIX procedure in SAS. Synchronization resulted in more calves born from d1 to 14 (P &lt; 0.01; 62% vs 47%) of the calving season. There was no differences (P = 0.31) between Syn and Nonsyn in the percent of calves born after d21, but between d22 and 42, more (P = 0.04) calves were born in the Nonsyn group. A greater proportion (P &lt; 0.02; 46% vs 38%) of calves were born in SynNS between d1 and 14 compared to SynAI. There was no difference between treatments (P &gt; 0.12) from d1 to 42, but more calves were born for SynAI after d43. With SEXED semen, there were no differences (P &gt; 0.14) between CON and SEXED for the proportion of calves born from d1 to 14 or d1 to 21; however, more of the desired gender were born in the SEXED group during d1 to 14 of the calving season (P &lt; 0.01; 84% vs 68%). More total calves were born from d22 to 42 in the SEXED group compared to the CON group (P &lt; 0.05; 49% vs 33%). In summary, synchronization increased the proportion of females that calved early in the calving season, and SEXED semen increased the proportion of the desired gender born early in the calving season.

SUMMARYEffect of timing of pregnant mares's serum gonadotrophin (PMSG) injection combined with a progestagen-impregnated vaginal sponge on fixed-time artificial insemination with fresh, undiluted semen during the nonbreeding season was investigated. PMSG injection 2 days before sponge removal (group A) caused significantly earlier oestrus than injection at the time of sponge removal (group B). Lambing percentages in a total of 320 ewes after fixed-time insemination at 36 h (64·1%) and 48 h (47·2%) in group A, and 48 h (59·5%) and 60 h (39·5%) in group B were not significantly different from those resulting from ‘at oestrus’ insemination (500% and 511% in groups A and B, respectively). The present results indicate that the optimum time for fixed insemination may depend upon the time of PMSG injection associated with progestagen treatment.

This trial evaluated the reproductive performance in an early routine oestrus induction programme using two different PGF2α preparations in dairy cattle. D-cloprostenol sodium (n = 192; Group A) or dinoprost (n = 187; Group B) was administered between days 35 and 42 post partum. Also, a group of non-treated cows (n = 135; Group C) was included as control. Pedometers were used to detect oestrus, and also secondary oestrous signs and vaginal mucus quality were assessed prior to artificial insemination (AI). When oestrus was not detected for 14 days after PGF2α administration, the treatment was repeated, up to a maximum of three times. There were no differences between the study groups in oestrus detection (A = 73.48%, B = 73.01%, C = 79.26%; P = 0.428), good mucus quality (A = 96.45%, B = 91.30%, C = 93.45%; P = 0.203) and the presence of mounting lesions (A = 98.58, B = 94.93%, C = 98.13; P = 0.414). First-service pregnancy rates were 19.78%, 15.64% and 32.03% in Groups A, B and C, respectively (P = 0.003). There were no inter-group differences for the interval from parturition to first AI. However, a significantly shorter interval from parturition to conception (92.17 days, 99.45 days, 118.93 days; P = 0.002) and significantly less services per conception (2.12, 2.18, 2.66; P = 0.003) were observed in Groups A and B in comparison with Group C. The use of PGF2α resulted in better fertility in a repetitive, routine postpartum programme, although no differences between Dcloprostenol and dinoprost were detected.

This study evaluated the variation in semen characteristics between two individual Murrah buffalo bulls. Two hundred and ninety semen samples were involved in this study. The results showed that most of the semen had a milky white color with moderate consistency. The Bull B1 tended to have higher proportion of creamy color and moderate consistency than the Bull B2. The ejaculatory volume is varied (P<0.01) between Bull B1 and Bull B2. However, semen pH and sperm concentration of Bull B1 and B2 were equal (P>0.05). The individual bull had a highly significant effect (P<0.01) on sperm motility of fresh semen. The individual variation (P<0.05) also found on sperm motility of before freezing. Whereas, sperm motility of post-thawing semen and recovery rate value were not significantly influenced (P>0.05). Moreover, it was found that the frozen semen production differed (P<0.01) between two individual bulls, with bull B1 had higher production than the bull B2. This study clearly demonstrates that the frozen semen production of Murrah buffalo is highly dependent on the individual bull. However, the variation in frozen semen production between two bulls studied here is still qualified to be used for artificial insemination.
