Given the rising demand for biological pigments, especially of microbial origin – the present study was conducted so as to report a potential source for the extraction of microbial pigment. The main objective was to isolate and identify a pigment–producing actinomycete because pigment production is prevelant in this group. A powdery, greenish–blue colony with a chalky azure aerial mass was isolated from one of the many rhizosphere soil samples. Upon preliminary investigation, viz. colony characterization and grams staining, the suspected colony was observed to have a filamentous margin with a slightly raised elevation and gram–positive filamentous hyphae.Biochemical analyses of the organism revealed positive results for carbohydrate fermentation and Triple Sugar Iron (TSI) test with no signs of gas production during the former but gas & H­2­S production during the latter. The identity of the isolate was established via 16S rDNA and phylogeny analysis, which strongly suggested it was Saccharomonospora azurea. Limited research pertaining to morphology, physiology, genomics and secondary metabolite production with no reports on the physicochemical properties of the pigment produced by S. azureaadequately suggests that it is relatively novel. Hence, further studies related to the same could be beneficial to the scientific community.

Milk itself is also considered to be one of the nature habitats of the LAB, It contains the many other nutrients and including protein and lactose. The aim of the research was a isolation and identification of naturally occurring lactic acid bacteria from cow raw milk. A total of ninety isolates of lactic acid bacteria were collected of which seven genera belonging to Lactobacillus (29.32%), Lactococcus (23.68%), Leuconostoc (21.09%), Streptococcus (12.09%), Aerococcus (9.09%), Pediococcus (17.09%) and Enterococcus (12.09%) were identified. Leuconostoc, Enterococcus, Pediococcus, Lactococcus, and Aerococcus were cocci and rod shape of Lactobacillus. In the current study, Lactobacillus (29.32%) was the main genera of lactic acid bacteria identified from cow raw milk samples. We can conclude from this research that raw cow milk is a good source of healthy lactic acid bacteria.

Background:Several reports have shown that severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection may trigger a vigorous immune response that could lead to the appearance of various autoantibodies such as antinuclear antibodies, antiphospholipid antibodies or anti-neutrophil cytoplasmic antibodies, among others. Moreover, the pulmonary involvement in SARS-CoV-2 may resemble that of patients with anti-MDA5 positive syndrome or acute form of antisynthetase syndrome.Objectives:Our aim was to analyse the presence of anti-MDA5 and other myositis-specific autoantibodies such as the antisynthetase antibodies in patients diagnosed with severe acute respiratory syndrome caused by SARS-CoV-2.Methods:Retrospective observational study performed in a tertiary care center. We included 28 patients admitted to the intensive care unit with severe acute respiratory syndrome, 14 at the onset of the disease (group A) and 14 after 30 days of being treated in an intensive care unit (group B). Chest CT was performed at the admission. We analyzed the presence of anti-MDA5 and antisynthetase antibodies by immunoblot (Euroimmune®) and in those who were positive we performed a confirmatory test by immunoprecipitation.Results:All chest CT showed bilateral ground glass pattern. Three out of 14 patients of group A (12 males, 86%, mean ± SD age 67.1 ± 12.2) were positive for antisynthetase antibodies (2 anti-PL7, 1 anti-Jo1), and 6 out of 14 patients of the group B (6 males, 48%, mean ± SD age 68.7 ± 8.1) were positive to antisynthetase antibodies (2 anti-PL7, 2 anti-PL-12, 1 anti-EJ, 1 anti-OJ+PL7). Immunoblots also show positivity for other myositis-specific or associated antibodies, such as anti-TIF1g, anti-PM75, anti-SAE and anti-SRP. All of these results found by immunoblotting were negative by immunoprecipitation. None of the 28 patients were positive for anti-MDA5 antibodies.Conclusion:Severe SARS-CoV-2 pneumonia is characterized by ground glass pattern in chest CT, as it is found in anti-MDA5 or antisynthetase syndrome. The positivity of several myositis related autoantibodies showed in immunoblot appears to be more related to the vigorous immune response producing polyclonal immunoglobulins than triggering a real myositis-associated interstitial lung disease. Beta-hemolytic group A streptococcus (Streptococcus pyogenes) is the most common bacterial agent associated with the upper respiratory tract infections in humans. The most frequently group A streptococcus-associated disease is pharyngitis. Males and females are equally affected by group A streptococcus. There is seasonal increase in the prevalence of group A streptococcus-associated pharyngitis. Streptococcal pharyngitis is most prevalent in winter and early spring with higher incidence of disease observed in crowded population such as school children. Early diagnosis and treatment of group A streptococcal pharyngitis has been shown to reduce the severity of symptoms and further complications such as rheumatic fever and glomerulonephritis. The conventional methods used for identification of group A streptococci depend on isolation and identification of the organism on blood agar plates. These methods usually require 18-24 hours of incubation at 37?C. Such delay in identifying the group A streptococcus has often made physicians to administer therapy without first disclosing the etiological agent. Development of immunologic tests, capable of detecting the group A streptococcal antigen directly from the throat swabs, produced rapid test results employed for better treatment of patients. STREP A test is a rapid immunochromatographic test for the detection of group A streptococci from throat swabs or culture. The accuracy of the test does not depend on the organism viability. Instead, group A strep antigen is extracted directly from the swab and identified using antibodies specific for the group A carbohydrates. We compared rapid test with conventional throat swab in 40 children, who met Centor criteria for streptococcal pharyngitis (absence of cough, high fever, purulent pharyngitis, enlarged and painful cervical lymph nodes). Overall congruence of rapid test and culture was 94%. Test is easy to perform and it is recommended as the first diagnostic test for management of children with streptococcal pharyngitis. In children with negative test, but with characteristics highly suggestive of streptococcal infection, throat culture should be performed.

Abstract Background: To explore the potential relationship of changes in classic serum tumor markers (CSTM) with differential diagnosis and disease assessment of upper tract urothelial carcinoma (UTUC).Methods: 60 UTUC (56 operated), 44 RCCC and 36 NTHN, were included into this retrospective analysis. The initial classic serum tumor markers (CSTMs), including CA242, CA199, CA125, CEA, AFP, SCC and CA724, were compared among the three groups. The preoperative, 1 month postoperative and 1 year postoperative/PD parameters (value and abnormal rate) of CSTMs were compared in UTUC group. A recommend test strategy was given and rechecked. The pathological manifestations of tumoral tissues and paracancerous tissues were analyzed.Results: The value of CA242, CA199 and CEA were higher in UTUC than in RCCC. The value of CEA was higher in UTUC than in NTHN. The value of CA199 and CEA were higher in UTUC than in RCCC+NTHN. The AR of CA199, CEA, SCC and CA724 were higher in UTUC than in RCCC. The AR of CEA and CA724 were higher in UTUC than in NTHN. The AR of CA199, CEA, SCC and CA724 were higher in UTUC than in RCCC+NTHN. The preoperative value of CA242 and CA199 were higher than 1 month postoperative ones in UTUC. The 1 year postop/PD value of CA242 and AFP were higher than 1 month postoperative ones in UTUC. For postop PD patients, the value during PD of CA242, CA199 and AFP were higher than 1 month postop ones. The preoperative AR of CA199 and CEA were higher than 1 month postoperative ones in UTUC. The recommended test strategy was given: CA242+CA199+CEA+AFP+SCC+CA724, and was rechecked. The P values were almost the lowest and the positive results covered all the comparisons. In 56 operated cases, the NAC in CSTMs by the recommended test strategy was statistically related with the tumor load. It appeared positive labelling by Ab of CA199, CA125, CEA, AFP and CA724 in UTUC tissues, while negative in paracancerous tissues.Conclusions: CSTMs may help to make differential diagnosis and disease assessment of UTUC. Group test (CA242+CA199+CEA+AFP+SCC+CA724) was recommended and more valuable.Trial registration: Not applicable

Adenoids and tonsils electively removed from 25 children with a history of recurrent group A β-hemolytic streptococci (GABHS) adenotonsillitis were cultured for aerobic and anaerobic bacteria. Two hundred twenty-four organisms (112 aerobes and facultatives, 110 anaerobes, and 2 Candida albicans) were isolated from the tonsils, and 229 (111 aerobes and facultatives and 118 anaerobes) were isolated from the adenoids. Mixed infection was present in all instances, with an average of 9.1 isolates per specimen. The predominant aerobes were Streptococcus sp, Haemophilus influenzae, and GABHS, and the prevalent anaerobes were Peptostreptococcus, Prevotella, and Fusobacterium spp. β-Lactamase-producing bacteria were detected in 72 isolates recovered from 22 tonsils (88%) and in 74 isolates recovered from 21 adenoids (84%). Discrepancies in the organisms recovered were found between the tonsils and adenoids. Of the aerobic isolates, 20% were isolated only in tonsils and 18% only in adenoids. Of the anaerobes, 20% were found only in tonsils and 26% only in adenoids. This study demonstrates a polymicrobial aerobic-anaerobic flora in both adenoids and tonsils, and discrepancies in recovery of pathogens such as GABHS. The adenoids may serve as a potential source of tonsillitis caused by GABHS.

The research work was performed for the isolation and identification of Pasteurella multocida from field cases, preparation of oil adjuvanted vaccine from isolated strain and determination of its efficacy. Samples were collected from suspected dead birds of three poultry farms of Bangladesh (Code name: M and R). The P. multocida isolates were Gram negative, non-motile, non- spore forming rod occurring singly or pairs and occasionally as chains or filaments. Biochemically P. multocida ferment basic sugar and consistently produced acid except from maltose and lactose. After isolation formalin killed oil adjuvanted Fowl cholera vaccine was prepared in Laboratory of the Department of Microbiology and Hygiene, BAU and this experimental vaccine (3.2x108 CFU/ml) was administered in nine weeks old White Leg Horn chickens at the different dose rate through intramuscular (IM) route in each selected group A (1ml alum precipitated vaccine), B (0.5ml alum precipitated vaccine), C (1ml oil adjuvanted vaccine) and D (0.5ml oil adjuvanted vaccine). Pre-vaccinated sera were collected from all groups of birds. The mean of Passive Hemagglutination (PHA) titers of post-vaccination were 51±17.8, 76.8±17, 89.6±17, and 115±17.81 in group A, B, C and D respectively which consist of 5 birds in each. The vaccine produced better immune response when boostering with the similar dose and route at 15 days after primary vaccination. The mean PHA titers were higher at group D than other groups after boostering. Challenge infection was conducted on all the vaccinated and control group (n=5) of birds after 15 days of vaccination which protect 93.75% of birds and the PHA titers from different groups analyzed to determine the protective capacity of vaccinated chickens against challenge exposure. It was demonstrated that experimental oil adjuvanted fowl cholera vaccine with 0.5ml dose produce higher immune response against challenge infection and found to be safe. Several studies have cited the Mediterranean diet as an example of healthy eating. In fact, the Mediterranean diet has become the reference diet for the prevention of cardiovascular disease. Red wine seems to be an essential component of the diet, since moderate consumption of wine is associated with lower risk and mortality from cardiovascular disease. Evidence is also accumulating that wine helps prevent the development of certain cancers. Of all the many components of wine, resveratrol, which is a natural component specifically present in wine, has been identified as being mainly responsible for these health-promoting properties. Many valuable properties such as cardioprotective and anticarcinogenic activity have been attributed to resveratrol; however, its bioavailability is quite low. The bioactivity of metabolites derived from resveratrol, and the accumulation of resveratrol in vital organs are still under study, but there are high expectations of positive results. Other stilbene compounds are also considered in this review, despite being present in undetectable or very small quantities in wine. The present paper reviews all aspects of the health properties of wine, bioactive compounds found in wine, and their concentrations, bioavailability and possible synergistic effects.

SUMMARYIn 1972 more than 20% of boys admitted to a closed community (Junior Detention Centre) developed acute tonsillitis and group-A streptococci were isolated from more than 40% of all boys at some time during their stay of two months. The most common serotype was M-type 5, which has frequently been associated with rheumatic fever in other epidemics; four cases of rheumatic fever occurred between 1972 and 1977. Various methods were tried to eliminate the infection, but only full penicillin prophylaxis (0·25 g oral penicillin 4 times a day for 10 days) to all boys on entry appeared to be effective.In a sample of cases of acute tonsillitis, group-A haemolytic streptococci were isolated from 31·7% by the use of dry swabs or unenriched transport medium, but with enrichment medium duplicate swabs, 77·0% yielded positive results. We question the current practice in some laboratories of reporting positive cultures only when more than ten colonies of β-haemolytic streptococci are present. In this survey viruses did not appear to be an important cause of acute tonsillitis.High titres of streptococcal antibodies (antistreptolysin O, anti-desoxyribonuclease B and anti-M associated protein) were found, not only in cases and carriers, but in boys on entry to the centre. This supports epidemiological evidence that adolescent boys on entry to the centre. This supports epidemiological evidence that adolescent boys are particularly prone to streptococcal throat infections.

The stormy fermentation reaction of Clostridium perfringens in iron milk medium was compared to that of several C. perfringens-like strains. These clostridia, C. barati, C. perenne, C. absonum, and C. paraperfringens are very similar to C. perfringens on the basis of certain biochemical reactions and, consequently, are often difficult to distinguish from C. perfringens. Furthermore, these related clostridia may also be present in foods. Results of this study demonstrate that after 18 h of incubation at 45°C, only C. perfringens gave a positive reaction in iron milk with inocula as low as 22 cells/g. Some of the other strains began to show only gas production at 18 h. After 24 to 42 h some strains gave positive results and after 72 h all were positive. Enumeration of C. perfringens from food samples in iron milk medium by a 3-tube most probable number (MPN) technique gave similar results to enumeration by plate count using Shahidi-Ferguson Perfringens (SFP) agar. Furthermore, a rapid positive response occurred after only 2 and 3 h incubation of iron milk inoculated with 108 and 107 cells/ml, respectively. The high selectivity, ease of identification and rapid growth of C. perfringens in iron milk make the iron milk MPN procedure a valuable assay for accurate enumeration and differentiation of C. perfringens from related Clostridia in food products.
